Abstract:Aging and metabolic disorders feedback and promote each other and are closely related to the occurrence and development of cardiovascular disease, type 2 diabetes, neurodegeneration and other degenerative diseases. Liupao tea is a geographical indication product of Chinese dark tea, with a "red, concentrated, aged and mellow" flavor quality. In this study, the aqueous extract of aged Liupao tea (ALPT) administered by continuous gavage significantly inhibited the increase of visceral fat and damage to the intestinal–liver–microbial axis in high-fat modeling of SAMP8 (P8+HFD) mice. Its potential mechanism is that ALPT significantly inhibited the inflammation and aggregation formation pathway caused by P8+HFD, increased the abundance of short-chain fatty acid producing bacteria Alistipes, Alloprevotella and Bacteroides, and had a calorie restriction effect. The results of the whole target metabolome network pharmacological analysis showed that there were 139 potential active components in the ALPT aqueous extract, and the core targets of their actions were SRC, TP53, AKT1, MAPK3, VEGFA, EP300, EGFR, HSP90AA1, CASP3, etc. These target genes were mainly enriched in cancer, neurodegenerative diseases, glucose and lipid metabolism and other pathways of degenerative changes. Molecular docking further verified the reliability of network pharmacology. The above results indicate that Liupao tea can effectively delay the body's degenerative changes through various mechanisms and multi-target effects. This study revealed that dark tea such as Liupao tea has significant drinking value in a modern and aging society.

What problem does this paper attempt to address?
### Problems the Paper Attempts to Solve This paper aims to explore the role and potential mechanisms of aqueous extracts of aged Liubao tea (ALPT) in delaying degenerative changes in senescence-accelerated mouse prone 8 (SAMP8) mice. #### Specific Issues: 1. **Effects of a high-fat diet on SAMP8 mice**: Investigate the impact of a high-fat diet on metabolic disorders, fat accumulation, and liver function damage in SAMP8 mice. 2. **Effects of ALPT**: Verify whether ALPT can significantly inhibit fat increase and damage to the gut-liver-microbiota axis caused by a high-fat diet in mice. 3. **Potential Mechanisms**: Reveal the active components and targets of ALPT through network pharmacology analysis and verify its anti-inflammatory and multi-target effects. 4. **Molecular Docking Validation**: Further validate the results of network pharmacology using molecular docking technology. #### Research Background and Significance: - **Aging and Metabolic Disorders**: Aging and metabolic disorders mutually promote each other and are closely related to the occurrence and development of chronic non-communicable diseases such as cardiovascular diseases, type 2 diabetes, and neurodegenerative diseases. - **Potential of Liubao Tea**: Liubao tea is a type of Chinese black tea with "red, thick, aged, and mellow" flavors. Studies have shown that it has various health benefits, including antioxidant properties, regulation of glucose and lipid metabolism, and liver protection. - **Experimental Design**: Using the SAMP8 mouse model and high-fat diet treatment to evaluate the inhibitory effects of ALPT on degenerative changes and its mechanisms.
