Abstract:AimsThis study aimed to investigate the ability of CpG7909 adjuvant to enhance immunogenicity and protective efficacy of a subunit vaccine composed of ESAT6-Ag85A fusion protein (Pe685a) of Mycobacterium tuberculosis.Methods and ResultsELISA was used to detect specific antibody and IFN- expression in sera; ELISPOT, to detect IFN- expression in splenocytes; MTT assay and FACS, to detect T-lymphocytes proliferation in spleens; and RT-PCR, to detect cytokines expression in lungs of mice after immunization. Bacterial load and histopathological lesions in lungs or spleens of mice challenged with Myco.tuberculosis H37Rv strain were analysed. Compared with incomplete Freund's adjuvant, induced more potent production of Pe685a-specific IgG2a/IgG1 antibody and higher expression of IFN- in sera, stimulated more generation of antigen-specific IFN--secreting splenocytes, enhanced frequencies of CD3(+)CD4(+) and CD3(+)CD8(+) T-lymphocytes in spleen and increased transcription of TNF-, IFN-, IL-6 and TLR9 in lung. However, lower bacterial load in lung and less severe lung pathology were not observed in CpG7909 group mice.ConclusionsCpG7909 is able to enhance immunological effects of Pe685a subunit vaccine, but does not confer significant protective efficacy against Myco.tuberculosis infection.Significance and Impact of the StudyCpG7909 as an adjuvant of subunit vaccine against Myco.tuberculosis is worthy of further investigation.

Abstract:Fifty-six mice were randomly divided into four groups with 14 mice in each. Two groups were subcutaneously injected twice with a foot-and-mouth disease vaccine with 2-week intervals; each of them had been orally administered 0.89% saline or Atractylodis macrocephalae Koidz. polysaccharides (RAMPS) 0.05 g for 4 days before immunization. The rest were not immunized but treated in the same way. One-week after the primary and two weeks after the booster immunization, half in each group were sacrificed to measure serum IgG and the parameters for the intestinal mucosal immunity. Results indicated that oral administration of RAMPS increased both serum specific IgG response and intestinal mucosal immunity as shown by elevated total sIgA, mRNA expression of TGF-β, IL-6, TNF-α, IgA(+) cells and intestinal intraepithelial lymphocytes in duodenum. It is suggested that increased serum IgG response may be associated with enhanced local mucosal immunity by oral administration of RAMPS.

Abstract:Present study was designed to investigate the polysaccharide (RAMPS) extracted from the rhizome of Atractylodis macrocephalae Koidz. (RAM) for its effect on the immune responses to foot-and-mouth (FMD) vaccine. Thirty-five ICR mice were randomly distributed into 5 groups with 7 mice in each. After oral administration of RAMPS for 4 days at a dose of 0, 0.025, 0.05, 0.1 g or 0.25g of RAM, respectively, the animals were immunized twice with FMD vaccine at 2-week intervals. Three weeks later, serum IgG, IFN-gamma/IL-5, splenocyte proliferation, and mRNA expression of cytokines by splenocytes were measured. Results indicated that RAMPS and RAM significantly enhanced IgG titers, IgG subclasses, IFN-gamma, IL-5 and the splenocyte proliferations stimulated by concanavalin A, lipopolysacharide, and FMDV (P<0.05). Therefore, RAMPS and RAM increased both cellular and humoral immune responses to vaccination, and may have a potential as an oral adjuvant to improve vaccination. (C) 2011 Elsevier Ltd. A Comparative Study on the Mechanisms of Innate Immune Responses in Mice Induced by Alum and Actinidia Eriantha Polysaccharide.

Abstract:The innate immune mechanisms by which adjuvants enhance the potency and protection of vaccine remain at cellular level, but the molecular mechanisms, especially in vivo, are ill-identified. Actinidia eriantha polysaccharide (AEPS) is a potent adjuvant with dual Th1 and Th2 potentiating activity, while Alum elicits a strict Th2 response. The current experiments were designed to compare the innate immune responses in the peritoneal cavity of mice induced by two adjuvants and explore their molecular mechanisms using gene expression microarray including long noncoding RNAs (lncRNAs). AEPS induced the recruitment of monocytes, neutrophils and dendritic cells. However, Alum recruited neutrophils and eosinophils. AEPS and Alum specifically induced the differential expression of 546 and 922 genes in peritoneal cells, respectively. AEPS induced higher mRNA expression of CCL2, CCL3, CCL4, CCL7, CXCL2, CXCL3, CXCL5, CXCL10, IL-12β, and IL-23α in immune effector process, while Alum tended to Th17 response mRNAs such as IL-7A, IL-17F and IL-17RA. Furthermore, a robust adjuvant-specific expression pattern of lncRNAs was found in above mentioned biological processes, suggesting the involvement of lncRNAs in immune responses induced by AEPS and Alum. This study led to a better understanding of different molecular mechanisms of adjuvants and benefited the rational design of effective vaccines.
Novel Polysaccharide Adjuvant from the Roots of Actinidia Eriantha with Dual Th1 and Th2 Potentiating Activity.

Abstract:The plant polysaccharides are recognized as an effective biological response modifier with low toxicity. In this study, the water-soluble polysaccharide from the roots of Actinidia eriantha (AEPS) was evaluated for its toxicity and adjuvant potential on the specific cellular and humoral immune responses to ovalbumin (OVA) in mice. AEP did not cause any mortality and side effects when mice were administered subcutaneously twice at the dose up to 5.0mg at intervals of 7 days. The mice were immunized subcutaneously with OVA 100μg alone or with OVA 100μg dissolved in saline containing Quil A (10μg) or AEPS (25, 50, or 100μg) on days 1 and 15. Two weeks later, splenocyte proliferation, natural killer (NK) cell activity, production and mRNA expression of cytokines from splenocytes, and serum OVA-specific antibody titers were measured. The Con A-, LPS-, and OVA-induced splenocyte proliferation and the serum OVA-specific IgG, IgG1, IgG2a, and IgG2b antibody titers in the immunized mice were significantly enhanced by AEPS (P<0.05, P<0.01 or P<0.001). AEPS also significantly promoted the production of Th1 (IL-2 and IFN-γ) and Th2 (IL-10) cytokines and up-regulated the mRNA expression of IL-2, IFN-γ, IL-4 and IL-10 cytokines and T-bet and GATA-3 transcription factors in splenocytes from the immunized mice (P<0.05, P<0.01 or P<0.001). Besides, AEPS remarkably increased the killing activities of NK cells from splenocytes in the immunized mice (P<0.01 or P<0.001). The results indicated that AEPS had strong potential to increase both cellular and humoral immune responses and elicit a balanced Th1/Th2 response, and that AEPS may be a safe and efficacious adjuvant candidate suitable for a wide spectrum of prophylactic and therapeutic vaccines.

Abstract:In experiment A,Rhizoma Atractylodis Macrocephalae polysaccharides(RAMPS)were extracted with boiling water and precipitated by 95% ethanol,after removal of protein by Sevage method,the polysaccharide were further purified by Macroporous Adsorption Resin column D101 and dialyzed against distilled water.Total sugar content was estimated by the phenol-sulfuric acid analysis using glucose as a standard.The polysaccharides were hydrolyzed with TFA and derivatized to acetylated aldononitriles and isothermally separated by gas chromatography(GC)to analyse monosaccharides component.In experiment B,thirty-five female ICR mice were randomly divided into five groups with 7 mice in each.Mice were immunized with OVA alone as control,or in combination with 150,300 or 600 μg RAMPS or Astragalus polysaccharides.Two weeks after the first immunization,mice were boosted with the same route.Blood samples were then collected two weeks after the boost for detection of serum OVA-specific antibody and subclasses.The results indicated that the yield of RAMPS in this experiment was 4.2% with polysaccharides content of 86.2%,which was composed of rhamnose,arabinose,xylose,mannose,glucose and galactose.RAMPS could significantly improve the level of serum specific IgG and subclass response of mice,suggesting the immunological adjuvant effect of RAMPS.

Abstract:Actinidia eriantha polysaccharide (AEPS) is a potent adjuvant with dual Th1 and Th2 potentiating activity. lincAAM has been previously proved to facilitate the expression of immune response genes (IRGs) in AEPS-activated RAW264.7 macrophages. However, its role in mediating adjuvant activity of AEPS remains to be elucidated. In this study, bone marrow-derived macrophages (BMDMs) from wide-type (WT) and linc-AAM knockout C57BL/6J mice treated with AEPS were subjected to transcriptome sequencing and bioinformatic analysis. linc-AAM deficiency inhibited M1 and M2 immune responses in BMDMs induced by AEPS. In mechanisms, AEPS facilitated the expression of IRGs and activated BMDMs through NF-& kappa;B-linc-AAM-JAK/STAT axis. Furthermore, lincAAM knockout inhibited cytokine and chemokine production, immune cell recruitment as well as immune cell migration to draining lymph nodes at peritoneal cavity in mice induced by AEPS. More importantly, linc-AAM deletion reduced the adjuvant activity of APES on antigen-specific cellular and humoral immune responses to ovalbumin in mice. This study has for the first time demonstrated the role of lncRNAs in regulating the adjuvant activity of polysaccharides and its mechanisms. These findings expanded current knowledge on the mechanism of action of adjuvant and provide a new target for the design and development of vaccine adjuvants.

Abstract:The total saponin from the stem bark of Albizia julibrissin (AJSt) has previously showed the adjuvant potentials in mice. In this study, AJSt was subjected to resin column chromatography to afford four fractions (AJS30, AJS50, AJS75 and AJS95), and these fractions were further compared for the hemolytic activities and adjuvant potentials on the immune response to ovalbumin (OVA) and recombinant fowl pox virus vector-based avian influenza vaccine (rFPV). AJSt, AJS50, AJS75 and AJS95 showed a slight hemolytic effect. AJSt, AJS50 and AJS75 significantly enhanced not only the concanavalin A (Con A)-, lipopolysaccharide (LPS)- and antigen-stimulated splenocyte proliferation, but also serum antigen-specific IgG, IgG1, IgG2a and IgG2b antibody titers in the mice immunized with OVA and rFPV. AJSt, AJS50 and AJS75 also significantly promoted the NK cell activity and delayed-type hypersensitivity (DTH) in the OVA-immunized mice. Furthermore, the mechanisms of adjuvant action were explored by determining the effects of AJS75 on cytokines and chemokines at the site of injection using antibody array. AJS75 induced or up-regulated the protein expression of 12 cytokines (IL-12p40, IL-12p40/p70, IFN-γ, IL-13, IL-1β, IL-6, IL-10, TNF-α, sTNFR I, sTNFR III, IL-3 and IL-9) and 10 chemokines (Eotaxin, I-TAC, MIG, MIP-1α, RANTES, TECK, Fracatlkine, FasL, M-CSF and GM-CSF) in the injected muscles. The results suggested that AJS75, the most adjuvant-active fraction of AJSt, could improve antigen-specific both cellular and humoral immune responses and simultaneously elicit a Th1/Th2 response by inducing cytokine and chemokine at the site of injection.

Abstract:Abstract Complete Freund’s adjuvant (CFA) has historically been one of the most useful tools of immunologists. Essentially comprised of dead mycobacteria and mineral oil, we asked ourselves what is special about the mycobacterial part of this adjuvant, and could it be recapitulated synthetically? Here, we demonstrate the essentiality of N -glycolylated peptidoglycan plus trehalose dimycolate (both unique in mycobacteria) for the complete adjuvant effect using knockouts and chemical complementation. A combination of synthetic N -glycolyl muramyl dipeptide and minimal trehalose dimycolate motif GlcC14C18 was able to upregulate dendritic cell effectors, plus induce experimental autoimmunity qualitatively similar but quantitatively milder compared to CFA. This research outlines how to substitute CFA with a consistent, molecularly-defined adjuvant which may inform the design of immunotherapeutic agents and vaccines benefitting from cell-mediated immunity. We also anticipate using synthetic microbe-associated molecular patterns (MAMPs) to study mycobacterial immunity and immunopathogenesis.

Population-based incidence of intussusception and a case-control study to examine the association of intussusception with natural rotavirus infection among indian children.

Abstract:BACKGROUND A rotavirus vaccine previously licensed in the United States was withdrawn because it caused intussusception. Data on background intussusception rates in developing countries are required to plan pre- and postlicensure safety studies for new rotavirus vaccines. Also, it is unclear whether natural rotavirus infection is associated with intussusception. METHODS Passive surveillance for intussusception in a large, well-defined, poor, urban population in Delhi, India, was conducted in 2 phases. Intussusception was confirmed by ultrasonography or surgery. Fecal samples obtained from patients with intussusception at study hospitals (irrespective of their residence in study areas) and healthy control subjects were tested for rotavirus with use of enzyme immunoassay. If available, resected intestinal tissue samples were tested for rotavirus with use of immunohistochemistical analysis and reverse-transcription polymerase chain reaction. RESULTS The incidence of intussusception requiring hospitalization was 17.7 cases per 100,000 infant-years of follow-up (95% confidence interval, 5.9-41.4 cases per 100,000 infant-years). Detection rates of rotavirus in stool samples did not differ significantly between case patients and control subjects (4 of 42 case patients vs 6 of 92 control subjects), and no evidence of rotavirus was detected in any of the 22 patients with intussusception for whom intestinal tissue samples were available. CONCLUSIONS The incidence of intussusception among Indian infants appears to be lower than that reported in other middle- and high-income countries. Natural rotavirus infection does not appear to be a major cause of intussusception in Indian infants.
Effects of a Fusobacterium nucleatum Extract on Immunoregulation in Mice

Abstract:The purpose of this study was to examine the effect of an extract of Fusobacterium nucleatum on humoral and cell-mediated immunity and phagocyte functions in mice. The extract was obtained from the heat-inactivated supernatant of sonicated F. nucleatum. Mouse splenocytes were cultured with sheep red blood cells (SRBC), and antibody formation was assayed by counting the plaque-forming cells (PFC). After C3H mouse skin was grafted onto the BALB/c mouse, cytotoxicity of BALB/c mouse splenocytes to L-cells was enumerated, T- and B-cell blastogenesis was determined by the uptake of 3H-thymidine. In vivo phagocyte functions were measured using carbon clearance test, bactericidal, and acid phosphatase activity assays. The number of SRBC-specific and non-specific PFC in splenocytes stimulated with the extract increased 2.5-8. 7-fold over that of unstimulated splenocytes. Cytotoxicity of splenocytes from the extract-injected mice decreased in a dose-dependent fashion at 12 days after grafting. T cell mitogenicity of the extract was observed. The carbon clearance rate was reduced, and bactericidal and acid phosphatase activities were elevated by the extract injection of the mice. In conclusion, F. nucleatum extract has an adjuvant effect on humoral immunity, and a suppressive effect on cell-mediated immunity and activated phagocyte functions.

Abstract:Double-mutant heat-labile toxin (dmLT, LTR192G/L211A) of enterotoxigenic Escherichia coli (ETEC) is an effective mucosal adjuvant. Recent studies have shown that dmLT also exhibits adjuvanticity for antigens administered parenterally. In this study, we subcutaneously (SC) immunized mice with the ETEC adhesin-based vaccine, CFA/I/II/IV MEFA (multiepitope fusion antigen), adjuvanted with dmLT and examined the impact of dmLT on antibody responses specific to the seven adhesins in the vaccine construction [CFA/I, CFA/II (CS1, CS2, CS3) and CFA/IV (CS4, CS5, CS6)]. Mice were immunized with a fixed dose of CFA/I/II/IV MEFA and ascending doses of dmLT adjuvant (0, 0.05, 0.1, 0.5 or 1.0 mu g) to assess the potential dmLT dose response relationship. Data showed that dmLT enhanced systemic antibody responses to all seven antigens (CFA/I, CS1-CS6) targeted by MEFA in a dose-dependent way. The adjuvant effect of dmLT on the MEFA construct plateaued at a dose of 0.1 mu g. Results also indicated that dmLT is an effective parenteral adjuvant when given by the SC route with the ETEC adhesin MEFA vaccine and that antibody enhancement was achieved with relatively low doses. These observations suggest the potential usefulness of dmLT for parenteral ETEC vaccine candidates and also perhaps for vaccines against other pathogens.

Abstract:Vaccination is an important approach to the control of foot-and-mouth disease (FMD). This study evaluated the effect of oral administration of ginseng stem-leaf saponins (GSLS) on the immune response to FMD vaccine and the gut mucosal immunity in mice. In experiment 1, mice were orally administered GSLS or not treated as a control. The animals were then immunized twice with FMD vaccine. Blood was sampled weekly within five weeks after the boost immunization for measurement of serum IgG and the isotypes. In experiment 2, mice were orally administrated GSLS or not treated as a control. After that, splenocytes were prepared from sacrificed mice for lymphocyte proliferation assay and intestinal tissues were sampled for immunohistochemistry and histological examination. The results showed that oral administration of GSLS significantly enhanced serum IgG and the isotype responses to FMD vaccine as well as the number of intestinal intraepithelial lymphocytes (IELs) and immunoglobulin A (IgA)+cells. Therefore, GSLS may be a potent oral adjuvant and deserve further study to improve vaccination in susceptible animals.
Determination of fbe mucosal immunoadjuvantivity of recombinant EschericMa cott LTKA63 mutant and LTB

Abstract:To construct the prokaryotic expression systems of detoxified mutant LTKA63 of Es-cherichia coli heat-labile enterotoxin subunit A (LTA) gene and E. coli heat-labile enterotoxin subunit B (LTB) gene, and to determine the mucosal immunoadjuvantivity of the rLTKA63 and rLTB. Methods The LTA and LTB genes from E. coli strain 44815 genome DNA were amplified by high fidelity PCR. Site mutation technique was applied to construct the mutant LTKA63. All the cloned genes were sequenced after T-A cloning. Then the prokaryotic expression systems of LTKA63 and LTB genes were constructed and their expressed products were identified using SDS-PAGE. The pathways activating T lymphocytes of rLTKA63 were determined using the three colors flow cytometry. And GM1-ELISA was used to demonstrate the ability binding bovine GM1 of rLTB. By using recombinant urease subunit B (rU-reB) and adhesin A (rHpaA) as the antigens, the effects of rLTKA63 and rLTB on increasing the immunoprotecrion of rUreB and rHpaA in Helicobacter pylori strain SS1 infected mice were determined respectively. Results The amplification fragments of LTA and LTB genes with expected size were obtained from E. coli strain 44815 genome DNA templates. The mutant LTKA63 with correct sequence was obtained through the site mutation of the cloned LTA gene. The expression outputs of rLTKA63 and rLTB were approximate 30% and 20% of the total bacterial proteins, respectively. After co-incubation with LTKA63, T_H1 and T_H2 lymphocytes were increased by 19.9% and 42.3% respectively. The result of GM1-ELISA indicated that rLTB could combine bovine GM1. 33.3% (4/12) of the gastric biopsy samples in the mice immunized with rUreB alone and 41.7% (5/12) of the gastric biopsy samples in the mice immunized with rHpaA alone were positive for H. pylori isolation, and the rUreB or rHpaA specific S-IgAs in all the animals were negative. When rUreB or rHpaA co-administrated with rLTKA63 or rLTB, the positive H. pylori isolation rates were as lower as 16.7%-25.0% ( P > 0.05) and the rUreB and rHpaA specific S-IgAs positive rates reached 41.6%-58.3% (P<0.01). Conclusion The prokaryotic expression systems of LTKA63 and LTB genes were successfully established in this study. Both the expressed rLTKA63 and rLTB have strong mucosal adjuvant activities.

Abstract:IgG antibody response in chickens immunized with F4 fimbriae extracted from local enterotoxigenic Escherichia coli (ETEC) strain was studied during a 98-day immunization period for comparing the efficacy of four adjuvants: Freund' adjuvant, Quil A (QA), propolis and extract from Cochinchina momordica seed (ECMS). For this purpose, chickens were immunized with F4 fimbriae alone or combined with one of the above adjuvants on days 1 and 21. IgG antibody levels in serum and egg yolk (by ELISA) were measured on days 0, 7, 14, 21, 28, 35, 42, 49, 56, 70, 84 and 98. The egg production of each group was also determined during days 1-7 and the following four weeks. The results showed that QA could enhance antibody titre, as good or almost as good as Freund's adjuvant, whereas the titres of ECMS and propolis groups were relatively lower, with the overall order: Freund's adjuvant>QA>ECMS>propolis both in serum and egg yolk. However, the significant decrease of egg production was merely observed in the Freund's adjuvant group. It is concluded that the four adjuvants tested can stimulate immune response to F4 fimbriae in chickens, with Freund's adjuvant giving the best results, followed by QA.

Abstract:Ethnopharmacological relevance: Herbal polysaccharides have exhibited great immune-enhancing potential. Adjuvants are a key tool for developing efficacious vaccines. In our previous study, a water-soluble polysaccharide extracted from wild Cistanche deserticola Y.C. Ma showed potent immunostimulatory activity. Aim of study: In this study, the immune profiles and efficacy of aqueous extracts of cultivated Cistanche deserticola Y.C. Ma (AECCD) on ICR mice against ovalbumin (OVA) were investigated. In vitro experiments, the possible DC activation mechanism by AECCD was evaluated. Materials and methods: AECCD were extracted using hot water after which the crude polysaccharides were precipitated by ethanol. Mice were firstly immunized subcutaneously with OVA (10 mu g per mouse) alone or OVA (10 mu g per mouse) respectively containing different dose of AECCD (200, 400 and 800 mu g per mouse) on Days 1 and 14 and the magnitude and kinetics of antibodies and cell-mediated responses were then assessed. Results: AECCD elicited vigorous and long-term IgG responses with mixed Th1/Th-2 responses and up-regulated levels of Th-associated cytokines (CD4(+)IL-4, CD4(+)IFN-gamma and CD8+IFN-gamma). Moreover, AECCD induced the strong cellular immune response characterized by increased splenocyte proliferation as well as the activated T cell response. Notably, AECCD significantly enhanced the maturation of dendritic cells (DCs) and inhibited Tregs. In vitro experiments, Preliminary tests indicated that AECCD induced DC activation by promoting phenotypic maturation, cytokine section and allostimulatory activity. Toll-like receptor 4 (TLR4) was an essential receptor for DCs to directly bind AECCD. The inhibitors of NF-kappa B decreased the expression levels of CD40, CD80, CD86 and MHC-II and the production of IFN-gamma, TNF-alpha and IL-6 through DCs. Conclusions: Finally, these findings suggested that AECCD could elicit potent and durable antigen specific immune responses through DC activation, which was involved in the regulation of maturation markers and cytokine expression via TLR4-related NF-kappa B pathway. The study indicates that AECCD is a potential immunomodulator.

Abstract:(1) Background: The adjuvant properties of flagellin from various bacterial species have been extensively studied; however, a systematic comparison of the immunoadjuvant effects of flagellins from different bacterial species is lacking. This study aims to analyze the amino acid sequences and structural features of flagellins from Escherichia coli (FliCE.C), Salmonella enterica serotype Typhimurium (FliCS.T), and Pseudomonas aeruginosa (FliCP.A), and to evaluate their adjuvant activities in terms of Toll-like receptor 5 (TLR5) activation, antibody production, and cytokine responses in a murine model. (2) Methods: Bioinformatics analysis was conducted to compare the amino acid sequences and structural domains (D0, D1, D2, and D3) of flagellins from the three bacterial species. PyMol atomic models were used to confirm structural differences. Toll-like receptor 5 (TLR5) activation assays were performed to measure IL-8 and TNF-α production in vitro. The IgG antibody titers against the model antigen FaeG and cytokine responses, including IL-4 and TNF-α secretion were evaluated in a murine model. (3) Results: Bioinformatics analysis revealed that the D0 and D1 domains are highly conserved, whereas the D2 and D3 domains exhibit significant variability across the three species. Structural analysis via PyMol confirmed these differences, particularly in the D2 and D3 domains. TLR5 activation assays showed that FliCS.T and FliCP.A induced higher levels of IL-8 and TNF-α production compared to FliCE.C, indicating species-specific variations in TLR5 activation. In the murine model, FliCS.T as an adjuvant produced higher antibody titers against FaeG and increased IL-4 secretion in splenocytes compared to FliCE.C and FliCP.A. FliCP.A induced higher TNF-α expression than FliCS.T and FliCE.C, suggesting FliCS.T and FliCP.A are more effective at inducing T-cell responses. (4) Conclusions: This study highlights the potential of FliCS.T and FliCP.A as potent vaccine adjuvants. The results provide insights into the structure–function relationships of these flagellins and support their application in enhancing immune responses against diverse pathogens.

Abstract:Ag retention within lymph nodes (LNs) upon vaccination is critical for the development of adaptive immune responses, because it facilitates the encounter of the Ag with cognate lymphocytes. During a secondary exposure of the immune system to an Ag, immune complexes (ICs) that contain the unprocessed Ag are captured by subcapsular sinus macrophages and are transferred onto follicular dendritic cells, where they persist for weeks, facilitating Ag presentation to cognate memory B cells. The impact of adjuvants on Ag retention within the draining LNs is unknown. In this article, we provide the first evidence, to our knowledge, that the oil-in-water emulsion adjuvant MF59 localizes in subcapsular sinus and medullary macrophage compartments of mouse draining LNs, where it persists for at least 2 wk. In addition, we demonstrate that MF59 promotes accumulation of the unprocessed Ag within these LN compartments and facilitates the consequent deposition of the IC-trapped Ag onto activated follicular dendritic cells. These findings correlate with the ability of MF59 to boost germinal center generation and Ag-specific Ab titers. Our data suggest that the adjuvant effect of MF59 is, at least in part, due to an enhancement of IC-bound Ag retention within the LN and offer insights to improve the efficacy of new vaccine adjuvants.

Abstract:The blood–brain barrier (BBB) restricts molecular and cellular trafficking between the blood and the CNS. Although astrocytes are known to control BBB permeability, the molecular determinants of this effect remain unknown. We show that angiotensinogen (AGT) produced and secreted by astrocytes is cleaved into angiotensin II (AngII) and acts on type 1 angiotensin receptors (AT1) expressed by BBB endothelial cells (ECs). Activation of AT1 restricts the passage of molecular tracers across human BBB-derived ECs through threonine-phosphorylation of the tight junction protein occludin and its mobilization to lipid raft membrane microdomains. We also show that AGT knock-out animals have disorganized occludin strands at the level of the BBB and a diffuse accumulation of the endogenous serum protein plasminogen in the CNS, compared with wild-type animals. Finally, we demonstrate a reduction in the number of AGT-immunopositive perivascular astrocytes in multiple sclerosis (MS) lesions, which correlates with a reduced expression of occludin similarly seen in the CNS of AGT knock-out animals. Such a reduction in astrocyte-expressed AGT and AngII is dependent, in vitro, on the proinflammatory cytokines tumor necrosis factor-α and interferon-γ. Our study defines a novel physiological role for AngII in the CNS and suggests that inflammation-induced downregulation of AngII production by astrocytes is involved in BBB dysfunction in MS lesions.
