Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.

Abstract:Cells, basic units of living structures and functions, build up a complicated small world, and their order and complexity resemble a small universe. The detailed understanding and elucidation of the matter transport and energy conversion mechanisms within a single cell will expand our knowledge about the origin and evolution of life, the disease mechanisms and much more. In past decades, single-cell analysis has been rapidly and significantly improved and various methodologies have been developed to reveal the complexity of mass, energy, and information within single cells. In this review, we focused on the methods developed in recent years for single-cell analysis, including electrochemical method, optical method, and mass spectrometry method. We reviewed the recent advances and representative studies in this research field, and also discussed the strengths and limitations of each method. Finally, we presented the existing technical challenges and further directions for single-cell analysis.

Abstract:Cells are basic structural and functional units of living organisms. Understanding the composition, structure and function of cells, and exploring cellular activities, are quite important for the cognition of phenomena and rules of life. Microfluidics, combined with advanced molecular, imaging and bioinformatics techniques, constitute a robust 'toolbox' and revolutionize the way for cell biology researches. In microfluidic systems, small amounts of fluids are manipulated using precisely designed channels with dimensions at micrometer level. Various chemical and biological processes can be transferred and integrated in a small single device, achieving multiple chemical and biological functions. Microfluidic technology displays a number of unique merits over conventional approaches, and has been extensively applied to various fields of cell research. In this chapter, we will review the recent developments and outstanding achievements of microfluidic technology in cell researches. Based on the cell study procedure, the main content is divided into four parts: cell culture, cell manipulation, cell stimulation and cell analysis. This review will also discuss the challenges and directions of microfluidic-based cell analysis, providing important references and ideas for the development of biological and medical researches and applications.

Abstract:Journal of Innovative Optical Health Sciences, Ahead of Print. Blood cells are the most integral part of the body, which are made up of erythrocytes, platelets and white blood cells. The examination of subcellular structures and proteins within blood cells at the nanoscale can provide valuable insights into the health status of an individual, accurate diagnosis, and efficient treatment strategies for diseases. Super-resolution microscopy (SRM) has recently emerged as a cutting-edge tool for the study of blood cells, providing numerous advantages over traditional methods for examining subcellular structures and proteins. In this paper, we focus on outlining the fundamental principles of various SRM techniques and their applications in both normal and diseased states of blood cells. Furthermore, future prospects of SRM techniques in the analysis of blood cells are also discussed.

Abstract:Cells are the fundamental unit of life, and studies on cell contribute to reveal the mystery of life. However, since variability exists between individual cells even in the same kind of cells, increased emphasis has been put on the analysis of individual cells for getting better understanding on the organism functions. During the past two decades, various techniques have been developed for single-cell analysis. Capillary electrophoresis is an excellent technique for identifying and quantifying the contents of single cells. The microfluidic devices afford a versatile platform for single-cell analysis owing to their unique characteristics. This article provides a review on recent advances in single-cell analysis using capillary electrophoresis and microfluidic devices; focus areas to be covered include sampling techniques, detection methods and main applications in capillary electrophoresis, and cell culture, cell manipulation, chemical cytometry and cellular physiology on microfluidic devices.

Abstract:Cells are the basic structural and functional units of living organisms. However, conventional cell analysis only averages millions of cell populations, and some important information is lost. It is essential to quantitatively characterize the physiology and pathology of single-cell activities. Precise single-cell capture is an extremely challenging task during cell sample preparation. In this review, we summarize the category of technologies to capture single cells precisely with a focus on the latest development in the last five years. Each technology has its own set of benefits and specific challenges, which provide opportunities for researchers in different fields. Accordingly, we introduce the applications of captured single cells in cancer diagnosis, analysis of metabolism and secretion, and disease treatment. Finally, some perspectives are provided on the current development trends, future research directions, and challenges of single-cell capture.

Abstract:Cells are fundamental units of life, and the key point in the field of biomaterial. Biological cells are always with high density, small nucleus and much impurities. Based on the technology of image processing, we propose a new method to count cells on the image of microscopic cells with high level of recognition. To precisely count the number, our method includes edge detecting and marking, efficient usage of three channel information of enhanced nucleus, binaryzation of dynamic threshold in separated areas and finally denoising. The experiment shows that the method is precise and quickly-reacted, moreover it can effectively rule out the impact of impurities. With little adjustment, it can apply to some other fields, not only decrease the labor involved, but the budget as well.

Abstract:In recent years, much attention has been gained on the study of single living cell with the development of biology, physics, electrophysiology, and nanotechnology. Researchers from biological and medical sciences regard the cell as a basic unit of life and the work of quantifying, imaging, and modulating living cells has been studied for decades. The dynamic changes in single living cell can reflect the changes and abnormalities of the organism. As such, it is extremely important to analyze living cells on an individual basis so as to illustrate the roles they play in these systems as well as their changes. In addition, the development of highly sensitive measurements applied in the field of analyzing single living cells, may contribute to clinical diagnostics. We present a summary of nanotechnologies resulting from the advances of intracellular analysis based on optoelectronic nanodevices and nanomaterials.

Abstract:Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome, proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial scRNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field, offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Abstract:Cells, as the fundamental structural elements and functional units of life, are necessary to be explored the variation of components and behaviors in the physiological and pathological processes. Benefited from the development of microfluidic culture and fluorescence labeling techniques, chips coupled with fluorescent tools have become a powerful platform for cell analysis. Thus, in this review, we mainly focus on the two aspects of overview: one is the microenvironment simulation on microfluidic systems including the fabrication of the chemical and physical microenvironment, the cell-cell co-culture/cell-extracellular matrix interaction model, and organ/tissue-on-a-chip system; the other is the fluorescence labeling strategies for cellular component detection and behavior observation, introducing from several aspects: nucleic acid-based modification, in situ chemical modification, hydrophobic insertion labeling and genetic-based labeling. At last, the existing achievements and future challenges in fluorescent analysis of cells based on microfluidic chip are discussed.

Abstract:No cell lives in a vacuum, and the molecular interactions between cells define most phenotypes. Transcriptomics provides rich information to infer cell–cell interactions and communication, thus accelerating the discovery of the roles of cells within their communities. Such research relies heavily on algorithms that infer which cells are interacting and the ligands and receptors involved. Specific pressures on different research niches are driving the evolution of next-generation computational tools, enabling new conceptual opportunities and technological advances. More sophisticated algorithms now account for the heterogeneity and spatial organization of cells, multiple ligand types and intracellular signalling events, and enable the use of larger and more complex datasets, including single-cell and spatial transcriptomics. Similarly, new high-throughput experimental methods are increasing the number and resolution of interactions that can be analysed simultaneously. Here, we explore recent progress in cell–cell interaction research and highlight the diversification of the next generation of tools, which have yielded a rich ecosystem of tools for different applications and are enabling invaluable discoveries.

Abstract:Understanding how cells interact and communicate with each other in multicellular organisms is essential to understanding diverse life and biological processes. However, the complicated nature of cells, their various in vivo microenvironments, and the multitude of changing environmental and biological signals that confront cells make it challenging to probe and control cell–cell interaction and communication using conventional macro-scale and population-averaged tools. With the rapid advances in bioengineering, we now have ever finer tools to peek increasingly closely at cellular processes, including the interaction and communication of cells with each other. In this chapter, we discuss established as well as emerging micro-, nano-, and molecular-scale genetic, chemical, and engineering tools designed to address remaining challenges in cell–cell interactions and communication. Microengineering tools, such as microfluidic chips, microwells, and microdroplets, make possible the manipulation of the spatial and temporal distribution of individual cells or defined homotypic and heterotypic cell clusters, thus simplifying the complicated in vivo microenvironments into well-controlled in vitro platforms. Molecular tools based on genetic or chemical approaches, frequently utilizing fluorescent or luminescent reporter signals, are being used at increased resolution to interrogate target cells. Moreover, with advances in complementary imaging systems, such as two-photon imaging, it is now possible to probe cell–cell interactions in their in vivo microenvironments. The knowledge derived from these studies is expected to not only increase the information content on various biological processes but also reveal novel paradigms of cellular and physiological processes, thereby also helping in the design of novel strategies to diagnose and treat diseases.

Abstract:Recent advances in cellular analysis revealed that the seemingly identical cells are heterogeneous in term of functionality, compositions, and genetic performance. These differences cause difficulty in the diagnostic for a specific model of disease. Detection of biomolecules such as DNA, RNA, and protein or analysis of cell(s), detection of cell surface molecules, and secreted protein, can help us to improve the understanding of a targeted disease and development of new diagnostic and therapeutic approaches. A single-cell includes the minute quantity of these target molecules. Microfluidic devices have the ability to capture a single-cell and its lysate into a pico or femtoliter volumes droplet, or micro-well thus preventing dilution and limiting diffusion. In this review we described the advancement and limitations in microfluidic techniques used toward single-cells analysis.

Single cell technology can reveal the heterogeneity between cells and further explore the essence and law of life activities. It is widely used in the research of tumor cells, tumor networks and tumor stem cells. At the same time, it can help people better understand their occurrence, development and metastasis. Single cell technology has developed rapidly and made many innovative progress in the field of biology. Fully understanding the current development trend of flow cytometry and single cell technology can promote the development of immunology, developmental biology, neurobiology, cancer and other fields.

Abstract:<p>Currently, single-cell analysis has become increasingly important. It can reveal cell-to-cell variances in a heterogeneous cell population and obtain a rare but important cytological information. Statistical analysis of cell populations based on single cell analysis results in a more realistic state of cell distribution, which is more conducive to biochemical and biomedical development than traditional average analysis based on a large population of cells. Due to extremely small volume of cells and low content/concentration of target analyte in individual cell, ultra-sensitive and anti-interference analysis protocols are urgently required. Herein, we present the state-of-the-art advances in single-cell analysis in the last three years, involving optical analysis, electrochemical analysis, and mass spectrometric analysis. In addition to the introduction of the latest analytical approaches, we highlight quantitative single-cell analysis in terms of calibration methods, sensitivity and/or detection limit. Finally, future perspectives in single-cell analysis are depicted.</p>
