Polyamines: Association in aqueous medium and promising transport vesicles

Abstract:Polyamines, which contain several nitrogen atoms, are present in almost all living organisms, playing an important role in nucleic acid transformations and other key processes. Long-chain polyamines are essential for the function of silicifying organisms, particularly diatom algae. The ability to associate with various molecules, primarily polyacids or polyanions, is a distinctive feature of polyamines that determines their physiological activity. The self-association of polyamines is poorly investigated, and the aim of this work is to study the association in aqueous solutions of polyamines containing mostly trimethylene insertions between nitrogen atoms. Seven new polyamine compounds have been synthesized using synthetic schemes that allow the preparation of many new polyamines with a variety of substituents. The polyamines with four or more nitrogen atoms are capable of forming vesicle-like aggregates in aqueous medium. These aggregates are stabilized mainly by hydrogen bonds between protonated and non-protonated nitrogens. As temperature increases or pH decreases, the association of polyamines decreases. Aggregates of long-chain polyamine (16 nitrogen atoms) are able to bind to oligonucleotides and curcumin, giving rise to positively charged particles of 100–200 nm in size. Curcumin is a weak acid showing antioxidant, antimicrobial, anti-inflammatory, antimutagenic, antiviral, anticancer and other valuable properties. Biomedical application of curcumin is hampered by poor water solubility, while polyamines allow to increase this parameter by more than 50 times. Polyamine aggregates can be considered as a new type of carriers and stabilizers of physiological active compounds. In contrast to polymeric amines such as polyethylenimine or polyvinylamine, polyamine aggregates are labile structures capable of degradation upon changes in pH, temperature, or exposure to fluctuating electric field. Taking into account the intrinsic physiological activity of polyamines, their aggregates are promising as a basis for the design of medical preparations capable of stimulus-responsive behavior in the body.
