Abstract:The C-O bond cleavage for activation of alcohols is synthetically useful and practically challenging. This work describes carbazate as a new type of electrochemically activated alkylating agent derived from ubiquitous alcohols for direct functionalization of heteroarenes under mild electrolytic conditions. The simple undivided cell at low oxidative potentials with carbon/platinum electrode set-ups offers excellent substrate tolerance, affording a variety of primary, secondary and tertiary alkyl-decorated heterocycles in good chemical yields. Furthermore, the mechanism for this electrochemical deoxyalkylation reaction has been investigated.

Abstract:The construction of C(sp(3))-heteroatom bonds represents a fundamental research field in synthetic chemistry. We report herein an electrochemical dehydroxymethylative functionalization reaction of aliphatic alcohols under mild and catalyst-free conditions, representing a new paradigm for forging various C(sp(3))-heteroatom bonds between diverse alkanols and N- or O-centered nucleophiles. In addition, detailed mechanistic investigations have been carried out, revealing that these reactions proceed through a beta-scission/anodic oxidation/nucleophilic addition sequence.

Abstract:Electrochemical C(sp3) − H oxygenation of phenols derivatives were accomplished using H2O as the sole oxygen source. Thus, selectively oxidative 4-cresols to 4-acylphenols derivatives proved viable with the aid of indirect electrolysis, through metal- and external oxidant free environmental-friendly and safe electrocatalytic conditions. Advances of this strategy were proved by its ability to transform various sulfonamides with excellent site and regioselectivity. Detailed mechanistic studies allowed to delineate the exact profile of the generation of the oxygenation events.

Abstract:Electrooxidation has emerged as an increasingly viable platform in molecular syntheses that can avoid stoichiometric chemical redox agents. Despite major progress in electrochemical C-H activations, these arene functionalizations generally require directing groups to enable the C-H activation. The installation and removal of these directing groups call for additional synthesis steps, which jeopardizes the inherent efficacy of the electrochemical C-H activation approach, leading to undesired waste with reduced step and atom economy. In sharp contrast, herein we present palladium-electrochemical C-H olefinations of simple arenes devoid of exogenous directing groups. The robust electrocatalysis protocol proved amenable to a wide range of both electron-rich and electron-deficient arenes under exceedingly mild reaction conditions, avoiding chemical oxidants. This study points to an interesting approach of two electrochemical transformations for the success of outstanding levels of position-selectivities in direct olefinations of electron-rich anisoles. A physical organic parameter-based machine learning model was developed to predict position-selectivity in electrochemical C-H olefinations. Furthermore, late-stage functionalizations set the stage for the direct C-H olefinations of structurally complex pharmaceutically relevant compounds, thereby avoiding protection and directing group manipulations.

Abstract:We report a straightforward, catalyst-free method for the cathodic benzylic C–H hydroxyalkylation/arylation of N-(alkyloxy)phthalimides with ketones or cyanopyridines. The reaction is driven by an electrochemically induced 1,2-hydrogen atom transfer involving alkoxy radicals generated by O–N bond cleavage. This protocol provides an efficient approach for the synthesis of a wide range of valuable unsymmetrical 1,2-diols and diarylmethanols.

Abstract:A metal-free electrophotochemical C(sp3)-H arylation was developed under mild conditions. This method enables a switchable synthesis of diaryl alcohols and diaryl alkanes from inactive benzylic carbons. More importantly, a cheap and safe mediator N-chlorosuccinimide (NCS) was developed, which was employed for the hydrogen atom transfer (HAT) process of the benzylic C-H bond. In addition, this active radical was captured and identified by electron paramagnetic resonance (EPR).

Abstract:With alkyl halides (I, Br, Cl) as a coupling partner, an electrochemically driven strategy for para‐selective C(sp2)−H alkylation of electron‐deficient arenes (aryl esters, aldehydes, nitriles, and ketones) has been achieved to access diverse alkylated arenes in one step. The reaction enables the activation of alkyl halides in the absence of sacrificial anodes, achieving the formation of C(sp2)‐C(sp3) bonds under mild electrolytic conditions. The utility of this protocol is reflected in high site selectivity, broad substrate scope, and scalable.

Abstract:A practical, electrochemical method is developed for the direct dehydrogenative lactonization of C(sp(2)/sp(3))-H bonds under external oxidant- and metal-free conditions, delivering diverse lactones, including coumarin derivatives with excellent regioselectivity. The scalable nature of this newly developed electrochemical process was demonstrated on a 40 g scale following an operationally simple protocol. The remote lactonization of C(sp(3)) H bonds would constitute an important synthetic advance toward electrochemical C-O bond formation.

Abstract:Abstract Herein, a metal-free electrochemical dihydroxylation of unactivated alkenes is described. The transformation proceeds smoothly under mild conditions with a broad range of unactivated alkenes, providing valuable and versatile dihydroxylated products in moderate to good yields without the addition of costly transition metals and stoichiometric amounts of chemical oxidants. Moreover, this method can be applied to a range of natural products and pharmaceutical derivatives, further demonstrating its synthetic utility. Mechanistic studies have revealed that iodohydrin and epoxide intermediate are formed during the reaction process.

Abstract:A novel palladium catalyzed hydroxylation of unactivated aliphatic C(sp(3))-H bonds was successfully developed. Different from conventional methods, water serves as the hydroxyl group source in the reaction. This new reaction demonstrates good reactivity and broad functional group tolerance. The C-H hydroxylated products can be readily transformed into various highly valuable chemicals via known transformations. Based on experimental and theoretical studies, a mechanism involving the Pd(II)/(IV) pathway is proposed for this hydroxylation reaction.

Abstract:Unprecedented hydroxy-alkynylation and -alkenylation reactions of arylalkenes have been developed through electrochemically enabled addition of an organotrifluoroborate reagent and H2O across the double bond of the alkene. The use of electrochemistry to promote these oxidative alkene 1,2-difunctionalization reactions not only obviates the need for transition-metal catalysts and oxidizing reagents but also ensures high regio- and chemoselectivity to afford homopropargylic or homoallylic alcohols. The possibility of extending the electrochemical alkene difunctionalization strategy to other alkene carbo-heterofunctionalization reactions has been demonstrated.

Abstract:Direct C3-functionalized of quinoxalin-2(1H)-ones are a convenient way to develop new and sustainable strategies, which are important for natural product and drug modification. Here, we report an electrocatalytic C-H activation method, which synthesized 3-hydroxyalkylquinoxalin-2(1H)-ones via a radical pathway using unprotected quinoxalin-2(1H)-ones and aliphatic aldehydes as the substrates. This electrolysis strategy avoids the use of stoichiometric oxidants and heavy metal catalysts. Further scale-up studies in electrochemical continuous flow reactors have demonstrated the potential application value.

Abstract:Decarboxylative C-H functionalization reactions are highly attractive methods for forging carbon-carbon bonds considering their inherent step- and atom-economical features and the pervasiveness of carboxylic acids and C-H bonds. An ideal approach to achieve these dehydrogenative transformations is through hydrogen evolution without using any chemical oxidants. However, effective coupling of decarboxylative carbon-carbon bond formation with proton reduction remains an unsolved challenge. Herein, we report an electrophotocatalytic approach that merges organic electrochemistry with photocatalysis to achieve efficient direct decarboxylative C-H alkylation and carbamoylation of heteroaromatic compounds through hydrogen evolution. The electrophotocatalytic method, which combines the high efficiency and selectivity of photocatalysis in promoting decarboxylation and electrochemistry in effecting proton reduction, allows efficient coupling of a wide range of heteroaromatic bases with a variety of carboxylic acids and oxamic acids. Advantageously, this method is scalable to decagram scale and applicable to the late-stage functionalization of drug molecules.

Abstract:A synergetic catalytic system of organophotocatalyst 4CzIPN, thianthrene, and methanol was developed for visible‐lightinduced Giese reactions of various alkanes and alkenes, upon using alkoxy radicals as hydrogen atom transfer reagents. The reaction can be performed under mild conditions to provide the alkylated products in moderate to excellent yields, avoiding the use of any strong oxidants and transition metal reagents. Different from the known LMCT and PCET processes, the key to success of this approach is the in situ thianthrenation of methanol by thianthrene radical cation to trigger the generation of methoxy radical.

Abstract:Transition-metal-catalyzed allylic C(sp3)–H alkylation with carbon-center nucleophiles is a straightforward approach to C(sp3)–C(sp3) bond formation, which has found widespread application in organic synthesis. However, stoichiometric oxidants are typically required to realize the transformation. Herein, by the triplet synergistic merger of Brønsted base/cobalt/photoredox catalysis, a mild protocol for the hydrogen-evolution allylic C(sp3)–H alkylation with protic C(sp3)–H feedstocks was developed in an oxidant-free manner. This operationally simple method enables direct allylic C(sp3)–H alkylation of a wide range of branched α-olefins with diverse protic C(sp3)–H feedstocks. The synthetic robustness of this strategy was further demonstrated by the late-stage functionalization of complex molecules and the synthesis of the natural product dihydropallescensin D.

Abstract:We have developed a protocol for electrochemical decarboxylative C3 alkylation of a wide range of quinoxalin-2(1H)-ones under metal- and additive-free conditions. N-Hydroxyphthalimide esters derived from chain, cyclic, primary, secondary, and tertiary carboxylic acids with a broad scope proved to be suitable substrates. This operationally simple protocol performed in an undivided cell under constant-current conditions is suitable for late-stage functionalization of quinoxalin-2(1H)-ones. The reactions can even be carried out with a 3 V battery as a power source, which demonstrates that organic electrosynthesis can be accomplished without the need for specialized equipment.

Abstract:A hindered para-hydroxyl group-directed remote benzylic C(sp(3))-H oxyfunctionalization has been developed for the straightforward transformation of 2,6-disubstituted 4-cresols, 4-alkylphenols, 4-hydroxybenzyl alcohols and 4-hydroxybenzyl alkyl ethers into various aromatic carbonyl compounds. The ligandand additive-free Cu(OAc)(2)-catalyzed atmospheric oxidation mediated by ethylene glycol unlocks a facile, atom-economical, and environmentally benign C=O formation for the functionalization of primary and secondary benzyl groups. Due to the pharmaceutical importance of 4-hydroxybenzaldehydes and 4-hydroxyphenones, the methodology is expected to be of significant value for both fundamental research and practical applications.

Abstract:An electrochemically enabled C(sp3)-C(sp) cross-coupling reaction is herein described for the synthesis of internal alkynes. This method is suitable not only to a diverse of acetylenic sulfones, but also to a wide range of alkyl radical precursors, such as alkyl iodides, N-hydroxyphthalimide esters and Katritzky salts. This reaction can also be applied to vinylation, thioetherification, selenation and hydrogenation reactions. This reaction shows a lot of significant advantages such as mild conditions, broad functional group tolerance, simple operation and convenience of purification and enlargement.
