Abstract:A novel and efficient Cu(I)/Ag(I)-mediated synergistic annulation reaction of [60]fullerene with diary-lethanones, benzoylacetonitriles, and beta-dicarbonyl compounds has been developed for the direct construction of diverse disubstituted dihydrofuran-fused [60]fullerene derivatives. This transformation exhibits a remarkably broad substrate scope and functional group tolerance and provides a useful method to a scarce class of fullerene derivatives.

Abstract:A highly regio-and stereoselective Pd-catalyzed cascade reaction of allenoate adducts has been developed. Various allenoate adducts of phosphine-catalyzed addition of allenoates are found to be efficient substrates for the preparation of 2,3-dihydrobenzofuran. It is the first example of allenoate adducts used as ideal and practical precursors of hard-to-get functionalized 1,3-butadienes for heterocycle synthesis.

Abstract:An unusual I2‐mediated triple C(sp3)−H functionalization reaction between aryl methyl ketones and dimethyl sulfoxide to form polysubstituted furans has been developed. In this transformation, dimethyl sulfoxide functions as a dual synthon via C(sp3)−H functionalization with formation of two C−C bonds, one C−O bond, and one C−S bond in a one‐step process. Iodine is crucial as promoter of the reaction, by which a number of ketones could be converted easily into 2,4,5‐trisubstituted furans. This provides an extremely simple and expeditious approach to high‐value‐added, biologically significant O‐heterocycles. Based on preliminary experiments, a plausible mechanism is proposed.

Abstract:A novel silver-mediated highly selective oxidative C-H/C-H functionalization of 1,3-dicarbonyl compounds with terminal alkynes for the creation of polysubstituted furans and pyrroles in one step has been demonstrated. Promoted by the crucial silver species, perfect selectivity and good to excellent yields could be achieved. This protocol represents an extremely simple and atom-economic way to construct polysubstituted furans and pyrroles from basic starting materials under mild conditions.

Abstract:α‐Arylated tetrahydrofurans, commonly found in biologically active molecules, exhibit diverse biologically activities and pharmacological effects. The functionalization of 2,3‐dihydrofuran offers a potential route for the synthesis of substituted tetrahydrofurans. Nevertheless, the development of earth‐abundant metal‐catalyzed regioselective hydrofunctionalization of 2,3‐dihydrofuran for the synthesis of substituted tetrahydrofurans remains to be pursued. Herein, we report a cobalt‐catalyzed hydroarylation of 2,3‐dihydrofuran, which serves as the first example of cobalt‐hydride‐catalyzed alkene hydroarylation. This reaction provides an efficient method for the synthesis of α‐arylated tetrahydrofurans with high reaction efficiency, exclusive α‐arylation selectivity, and remarkable functional group compatibility.

Abstract:A facile and efficient strategy for modular access to furo[3,2- c ]chromen-4-ones using 4-hydroxycoumarin and β-nitroalkenes via Lewis acid-catalyzed formal [3 + 2] annulation protocol is described. This reaction proceeds via cascade Michael addition/nucleophilic addition/elimination in the presence of Yb(OTf) 3 , which involves the formation of two new σ (C–C and C–O) bonds for the construction of a novel furan ring in a single operation. This protocol affords a variety of functional groups, thereby providing a practical and efficient method for the fabrication of a furo[3,2- c ]chromen-4-one framework.

Abstract:We report a metal-, acid-, and base-free 2(bromomethyl)naphthalene (2-BMN)-promoted organocatalytic Cloke-Wilson rearrangement of chain doubly activated cyclopropanes for the construction of 2,3-dihydrofurans via a carbocation-initiated tandem intramolecular ring-opening/recyclization process. The strategy is especially suitable for the construction of furan units in complex molecules, providing a solution to the problem of heavy-metal residues in dihydrofuran-containing drugs synthesized by traditional metal-based protocols. Thus, it is of potential interest in synthetic and medicinal chemistry.

Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:A distinct strategy via unprecedented semipinacol rearrangements for the synthesis of functionalized hydrodibenzofurans is reported. The versatile reactivity of benzofuran-3-one as a building block enabled the convergent coupling of simple starting materials and, thus, allowed for the facile variation of R group and the construction of hydrodibenzofurans with fused rings.

Abstract:The first Pd-catalyzed asymmetric three-component reaction of 2,3-allenol, aryl iodides, and 2-arylmethylenemolononitriles has been developed via an allenol carbopalladation and an allylic cycloaddition cascade. This process allows rapid access to substituted tetrahydrofurans bearing diverse functional groups in good yields with high diastereoselectivities and excellent enantioselectivities. The concise total synthesis of a lignan, (-)-2-episesaminone, has been achieved by the elaboration of a functionalized tetrahydrofuran obtained from this reaction.

Abstract:The expeditious construction of tetrasubstituted furans via Cu/base-promoted cascade reaction of terminal alkynes with 1,2-diketones is described. This reaction proceeds smoothly involving the formation of multiple chemical bonds with tolerating a wide range of functional groups. The mechanism underlying these transformations has been thoroughly investigated, suggesting that 1,3-diyne serves as a key intermediate generated in situ through oxidative coupling facilitated by the Cu catalyst.

Abstract:A copper‐catalyzed olefinic C(sp2)–H activation/carbene insertion/ester hydrolysis/cyclization sequence has been developed for the synthesis of substituted 2(5H)‐furanones employing readily available enaminones and diazo esters as substrates. This method features operationally simple and good functional group tolerance. The transformation could be scaled up to gram quantities and the obtained 2(5H)‐furanone derivatives could be easily converted to a wide range of useful synthetic building blocks, indicative of the applicability of the synthetic strategy. Isotope labeling experiments demonstrated the ester group of diazo esters was hydrolyzed during the reaction.

Abstract:We have developed a facile and effective synthesis of 3-iodofurans from 3-monosubstituted 1,2-allenyl perfluoroalkyl ketones or 2-hydroxy-4-iodo-2,5-dihydrofurans from 3,3-disubstituted 1,2-allenyl perfluoroalkyl ketones. The perfluoroalkyl substituent and the amount of water in the solvent are important for the success of this electrophilic cyclization.

Abstract:Cyclobutanone is a strained motif with broad applications, while direct assembly of the aromatic ring fused cyclobutanones beyond benzocyclobutenone (BCB) skeletons remains challenging. Herein, we report a Rh-catalyzed formal [3+2] annulation of diazo group tethered alkynes involving a 4-exo-dig carbocyclization process, providing a straightforward access to furan-fused cyclobutanones. DFT calculations disclose that, by comparison to the competitive 5-endo-dig process, 4-exo-dig carbocyclization is mainly due to lower angle strain of the key sp-hybridized vinyl cationic transition state in the cyclization step. Using less reactive catalysts Rh2(carboxylate)4 is critical for high selectivity, which is explained as catalyst-substrate hydrogen bonding interaction. This method is proved successful to direct access previously inaccessible and unknown furan-fused cyclobutanone scaffolds, which can participate in a variety of post-functionalization reactions as versatile synthetic blocks. In addition, preliminary antitumor activity study of these products indicates that some molecules exhibite significant anticancer potency against different human cancer cell lines.

Abstract:The exploration of novel chemical reactivities of readily available feedstocks has always been a hot topic in the synthetic community. We herein report a three-component one-pot reaction with TMSCF 2 Br, p -QMs and 1,3-dicarbonyl compounds and analogues, providing a modular protocol for the construction of densely functionalized tetrasubstituted furans and pyrroles. While TMSCF 2 Br has been widely used as a difluorocarbene or trimethylsilyldifluoromethyl radical precursor, it herein acts as a surrogate of "naked" carbon. Control experiments indicate that repeated substitution/elimination processes are involved. This reaction features high step-economy, mild conditions and excellent chemoselectivity. The phenolic hydroxyl groups of products offer a platform for the late-stage modification of furans and pyrroles, which could enrich the structural diversity of the products rapidly.
