Abstract:Interactions of amide linkages in the peptide backbone with solvent play key roles in the folding and stability of proteins. The strength of these effects can be quantitatively characterized in terms of solvent electric fields on the peptide, a useful framework that has been applied to understand various non-covalent solvent-solute interactions. We demonstrate how the solvent-induced electric field on carbonyl-containing molecules, including peptides, can be experimentally measured using the vibrational Stark effect (VSE). Particularly, we characterize how solvent interactions with the carbonyl differ depending on the solute structure. We observe that amide-containing molecules, as prototypical peptides, interact with polar solvents to stimulate unusually strong electric fields on the carbonyl compared to non-amides. These results are demonstrated computationally via molecular dynamics simulations and experimentally via frequency shifts in the Fourier-transform infrared (FTIR) spectra, with support from vibrational Stark spectroscopy (VSS). Observed strong fields on amides are attributed to resonance capabilities of the solute carbonyl with the nitrogen at the α-position, increasing the carbonyl dipole moment to drive solvent organization. Based on the solvent configurations obtained from MD simulations, we use density functional theory (DFT)-based electronic structure calculations to separate the total electrostatic contributions into two main categories, namely hydrogen-bonding (HB) and non-hydrogen–bonding (non-HB) interactions. We find that both HB and non-HB solvent molecules have large contributions to the electric field on carbonyls, which both increase proportionally as the resonance capabilities of the solute increase. In contrast to solvation mechanisms that require covalent electron donor-acceptor interactions, we demonstrate how a predominantly electrostatic mechanism captures the chief aspects of peptide solvation. Characterization of amides as sensitive VSE probes plays an important first step towards experimental measurement of electric fields involving the protein backbone using the amide I band.
