Tailored amphiphilic polymers enable stabilized biologics in hyperthermal water and freeze-drying process

Abstract:Biologics are susceptible to ambient and physical deterioration due to their complex macromolecular structures. Obtaining stable biologics under normal and extreme conditions would provide significant health and economic benefits. Here, we developed a versatile approach to stabilize a wide range of biologics (therapeutic proteins, enzymes, vaccines, and antibodies) by amphiphilic polymers. Stabilization effects were screened by tailoring amphiphilic polymers modified with varied hydrophobic groups, different lengths of hydrophobic and hydrophilic modules. The structures and bioactivities of biologics protected by optimized amphiphilic polymers were maintained under treatment with 100 °C water incubation, freeze-drying, or long-term storage at ambient temperature. Our study establishes a simplified and efficient approach to stabilize biologics under extreme and normal conditions, offering a promising platform associated with hyperthermal enzyme engineering and biologics avoidance with full cold chain transportation and storage.
