Abstract:With the development of deformation structured color materials and sensor signal analysis, the combination of photonic crystals with optical structure characteristics and highly sensitive hydrogels in the environment can stimulate response under conditions of mechanical deformation, temperature, humidity, pH, etc. Optical-based hydrogel sensors avoid these disadvantages of conventional sensors, such as non-destructive measurements, high-speed transmission, almost interference-free, telemetry remote control, and many other advantages. This paper introduces the optical properties of photonic crystals and the synthesis of hydrogel polymers, and describes the working principle of photonic crystal hydrogel sensors. There is no systematic generalization of the direction of flexible sensors, so this paper summarizes the progress in the direction of physical, chemical, and biological sensors, and reviews the related research applications, and discusses the potential future directions and development challenges.

Abstract:Hydrogels, water-filled networks that can adapt to external stimuli by altering their volume, are known for their high flexibility and biocompatibility. DNA, a critical biomolecule renowned for its exceptional characteristics including information transmission, molecular recognition, and editability, has found widespread applications in the biosensing field as well. The integration of these two biomaterials offers promising opportunities for the development of novel biosensors with enhanced sensitivity, specificity, and adaptability. Therefore, by virtue of the collective features, researchers have recently focused on the construction of responsive DNA hydrogel systems. This feature article describes recent developments in fabricating DNA hydrogels and their applications in the biosensing area. Initially, it focuses on the design strategies employed in preparing DNA hydrogels, encompassing both pure DNA hydrogels and hybridized DNA hydrogels. Subsequently, it summarizes the use of DNA hydrogels in biosensing applications, highlighting their applications in visual detection, electrochemical sensing, and optical biosensing analyses. Furthermore, the underlying responsive mechanisms within these biosensing systems are also described. Lastly, this article presents a comprehensive discussion on the existing challenges and prospects of responsive DNA hydrogels, offering insights into their potential to revolutionize the field of biosensing.

Abstract:Photonic crystals, which are materials with periodic dielectric constants on the submicroscale, have been the focus of research for an extended period. Photonic soft materials have been extensively developed for use as colorimetric indicators and mechanochromic sensors, but their limited mechanical properties and molding characteristics only suitable for films restrict their practical implementation. Herein we report an approach to synthesize highly stretchable photonic soft materials based on a hydrogel system that is cross-linked by a crystalline colloidal array. The intrinsic inhomogeneous submicroscale structure is exploited for effective reinforcement in the multiphase system of the photonic crystals. The photonic hydrogels exhibit a high deformation capacity, with a stretching deformation above 2800% and compression above 98%. The gel has a full-color tunable range and shows 460 nm photonic shifts that can be reversibly actuated by a small compressive stress (kPa level) and can be ink-written to form patterns and freestanding structures. Mechanochromic sensors are constructed based on the three-dimensional and two-dimensional Bragg diffraction of the gel. Owing to its mechanical strength, formability, and tunable colors, the gel can be used in wearable optical devices, colorimetric tactile sensors, and full-color displays.

Abstract:Hydrogel is a unique family of biocompatible materials with growing applications in chemical and biological sensors. During the past few decades, various hydrogel-based optical ion sensors have been developed aiming at point-of-care testing and environmental monitoring. In this Perspective, we provide an overview of the research field including topics such as photonic crystals, DNAzyme cross-linked hydrogels, ionophore-based ion sensing hydrogels, and fluoroionophore-based optodes. As the different sensing principles are summarized, each strategy offers its advantages and limitations. In a nutshell, developing optical ion sensing hydrogels is still in the early stage with many opportunities lying ahead, especially with challenges in selectivity, assay time, detection limit, and usability.This article has not yet been cited by other publications.

Abstract:Photonic crystals with orderly periodic structure and manipulated structural colors have garnered widespread attention in sensors field. However, the unsatisfactory detection range, low sensitivity and inconvenient integration highly limit their further practical applications. In this paper, by combining TiO2 inverse opal photonic crystal with polyacrylamide hydrogel, a novel organic-inorganic composite solvents responsive photonic hydrogel (SRPH) was developed. By encapsulating TiO2 inverse opal photonic crystal with high refractive index in this hydrogel, the photonic hydrogels, therefore, possess bright structural colors. Due to the discrepancy in swelling rate of SRPH film in various solvents (ethanol, dimethyl formamide, dimethyl sulfoxide, etc.), it can identify these organic solvents through the rapid change of structural colors and detect the concentrations of the solvent in a wide range. Even for solvents with similar refractive index, it can be clearly distinguished, along with the excellent repeatability. Benefiting from the wide detection range and high sensitivity, the chip sensors were then developed by integrating multiple hydrogel films into the microfluidic system, which have high efficiency in solvent naked-eye multi-identification and concentration multi-detection. The new composite photonic hydrogels provide a reference for the design of functional photonic crystals and next-generation application of sensors.

Abstract:The development and synthesis of hydrogels for chemical and biosensing are of great value. Hydrogels can be tailored to its own physical structure, chemical properties, biocompatibility and sensitivity to external stimuli when being used in a specific environment. This review mainly covers hydrogels and their applications in chemical and biosensing. In particular, the review focuses on the manner in which hydrogels serve as sensing materials to a specific analyte. Different types of responsive hydrogels are hence introduced and summarized. Researchers can modify different chemical groups on the skeleton of the hydrogels, which make them as good chemical and biosensing materials. Abstract:Diabetes is a global chronic disease that seriously endangers human health and characterized by abnormally high blood glucose levels in the body. Diabetic wounds are common complications which associate with impaired healing process. Biomarkers monitoring of diabetic wounds is of great importance in the diabetes management. However, actual monitoring of biomarkers still largely relies on the complex process and additional sophisticated analytical instruments. In this work, we prepared hydrogels composed of different modules, which were designed to monitor different physiological indicators in diabetic wounds, including glucose levels, pH, and temperature. Glucose monitoring was achieved based on the combination of photonic crystal (PC) structure and glucose-responsive hydrogels. The obtained photonic crystal hydrogels (PCHs) allowed visual monitoring of glucose levels in physiological ranges by readout of intuitive structural color changes of PCHs during glucose-induced swelling and shrinkage. Interestingly, the glucose response of double network PCHs was completed in 15 min, which was twice as fast as single network PCHs, due to the higher volume fraction of glucose-responsive motifs. Moreover, pH sensing was achieved by incorporation of acid-base indicator dyes into hydrogels; and temperature monitoring was obtained by integration of thermochromic powders in hydrogels. These hydrogel modules effectively monitored the physiological levels and dynamic changes of three physiological biomarkers, both in vitro and in vivo during diabetic wound healing process. The multifunctional hydrogels with visual monitoring of biomarkers have great potential in wound-related monitoring and treatment.

Abstract:A bilayer asymmetric photonic crystal slab made of porous Si3N4/SiO2 is designed as a biosensor by considering the optical performance of this photonic crystal slab with a square lattice based on rigorous coupled-wave analysis theory and wavelength interrogation methods. The results show that this bilayer asymmetric photonic crystal can be used as a biosensor according to its excellent linearity relationship between the guided resonance peak shift and refractive index of aqueous solution with or without glycerol. The theoretical sensitivity value of the bilayer asymmetric photonic crystal biosensor is achieved as (S>286 nm/RIU) in the wavelength range from 1400 nm to 1600 nm. These results also indicate that the asymmetry bilayer structure has an important influence on its optical characteristic and sensitivity of the bilayer photonic crystal biosensor, and hence, it can be modified by changing the lattice constant and slab thickness. This research paper is very useful for understanding the application and design of biosensors based on porous structures.

Abstract:Hydrogels are of great interest in the field of biosensing for their good biocompatibility, plasticity, and capability of providing 3D scaffolds. Nevertheless, the application of hydrogels has not been linked with broad surface biosensing systems yet. To overcome the limitations, here for the first time, surface-immobilized pure DNA hydrogels were synthesized using a surficial primer-induced strategy and adopted for biosensing applications. The DNA hydrogel 3D scaffold is successfully constructed on a transparent ITO electrode, which facilitates both colourimetric and electrochemical measurements. Results show that the hydrogel is able to wrap enzymes solidly and exhibits favourable stability under different conditions. Owing to the free diffusion of the micromolecular targets throughout the hydrogel, while isolating the enzymes from the macromolecular interferences outside the hydrogel, the direct colourimetric and electrochemical detection of hydrogen peroxide and bilirubin in serum is achieved. The detection limit of hydrogen peroxide in serum is 22 nM by colourimetric analysis and 13 nM by electrochemical measurement. The detection limit of bilirubin is 32 nM, a favourable limit that could be used in jaundice diagnosis. In addition, the enzyme@hydrogel can be easily regenerated and the catalytic activity is retained for a few cycles, thus allowing the recycling of the hydrogel-based biosensing system. The successful integration of DNA hydrogels with surface biosensing systems will greatly expand the applications of hydrogels for diagnostic and environmental monitoring purposes.

Abstract:Cell-based biosensors (CBBs) have emerged as promising biotechnical tools whereby various cell types can be used as basic sensing units to detect external stimuli. Specifically, CBBs have been applied in environmental monitoring, drug screening, clinical diagnosis and biosecurity. For these applications, CBBs offer several advantages over conventional molecular-based biosensors or living animal-based approaches, such as the capability to better mimic physiological situations, to enhance detection specificity and sensitivity, and to detect unknown compounds and toxins. On the other hand, existing CBBs suffer from several limitations, such as weak cell-substrate attachment, two-dimensional (2D) cell microenvironment, and limited shelf life. An emerging method for scaffold-free three-dimensional (3D) cell culture uses hydrogels to encapsulate cells. Advances in novel biomaterials and nano/microscale technologies have enabled encapsulation of cells in hydrogels to fabricate 3D CBBs, which hold great potential for addressing the limitation in existing 2D CBBs. Here, we present an overview of the emerging hydrogel-based CBBs, their applications in pathogen/toxin detection, drug screening and screening of cell-biomaterials interaction, and the associated challenges and potential solutions.

Abstract:A reversible and degradable supramolecular photonic crystal hydrogel (PCH) is formulated with agarose and monodispersed silica nanoparticles. Micro‐PCHs with precise and sophisticated structures can be easily fabricated via direct laser writing using the supramolecular PCH as a sacrificial scaffold. With this strategy, the unexplored structural‐designed micro‐PCH mechanical sensors are created. Photonic crystal hydrogels (PCHs), with smart stimulus‐responsive abilities, have been widely exploited as colorimetric sensors for years. However, the current fabrication technologies are mostly applicable to produce PCHs with simple geometries at the sub‐millimeter scale, limiting the introduction of structural design into PCH sensors as well as the accompanied advanced applications. This paper reports the microfabrication of three‐dimensional (3D) PCHs with the help of supramolecular agarose PCH as a sacrificial scaffold by two‐photon lithography (TPL). The supramolecular PCHs, formulated with SiO2 colloidal nanoparticles and agarose aqueous solutions, show bright structural color and are degradable upon short‐time dimethyl sulfoxide treatment. Leveraging the supramolecular PCH as a sacrificial scaffold, PCHs with precise 3D geometries can be fabricated in an economical and efficient way. This work demonstrates the application of such a strategy in the creation of structural‐designed PCH mechanical microsensors that have not been explored before.

Abstract:There is an increasing interest in sensing applications for a variety of analytes in aqueous environments, as conventional methods do not work reliably under humid conditions or they require complex equipment with experienced operators. Hydrogel sensors are easy to fabricate, are incredibly sensitive, and have broad dynamic ranges. Experiments on their robustness, reliability, and reusability have indicated the possible long-term applications of these systems in a variety of fields, including disease diagnosis, detection of pharmaceuticals, and in environmental testing. It is possible to produce hydrogels, which, upon sensing a specific analyte, can adsorb it onto their 3D-structure and can therefore be used to remove them from a given environment. High specificity can be obtained by using molecularly imprinted polymers. Typical detection principles involve optical methods including fluorescence and chemiluminescence, and volume changes in colloidal photonic crystals, as well as electrochemical methods. Here, we explore the current research utilizing hydrogel-based sensors in three main areas: (1) biomedical applications, (2) for detecting and quantifying pharmaceuticals of interest, and (3) detecting and quantifying environmental contaminants in aqueous environments.

Abstract:Photonic crystal (PC) materials have huge potentials as sensors for noninvasive and real-time monitoring glucose in tears. We developed a glucose-sensitive PC material based on monolayered colloidal crystals (MCCs). Polystyrene nanoparticles were first self-assembled into a highly ordered MCC, and this two-dimensional (2D) template was then coated by a 4-boronobenzaldehyde-functionalized poly(vinyl alcohol) hydrogel. Such a sensor efficiently diffracts visible light, whose structural color could change from red through yellow to green, as the glucose concentration altered from 0 to 20 mM, covering both tears' and bloods' physiological ranges. The sensor also represents a rapid response within 180 s at each titration of glucose, combining the characteristics of high accuracy and sensitivity in detecting the glucose concentration in tears, and this intelligent sensing material presents certain possibility for the frontier point-of-care glucose monitoring.

Abstract:Hydrogel materials offer many advantages for chemical and biological sensoring due to their response to a small change in their environment with a related change in volume. Several designs have been outlined in the literature in the specific field of hydrogel-based optical sensors, reporting a large number of steps for their fabrication. In this work we present a three-dimensional, hydrogel-based sensor the structure of which is fabricated in a single step using thermal nanoimprint lithography. The sensor is based on a waveguide with a grating readout section. A specific hydrogel formulation, based on a combination of PEGDMA (Poly(Ethylene Glycol DiMethAcrylate)), NIPAAm (N-IsoPropylAcrylAmide), and AA (Acrylic Acid), was developed. This stimulus-responsive hydrogel is sensitive to pH and to water. Moreover, the hydrogel has been modified to be suitable for fabrication by thermal nanoimprint lithography. Once stimulated, the hydrogel-based sensor changes its topography, which is characterised physically by AFM and SEM, and optically using a specific optical set-up.

Abstract:Responsive photonic crystal hydrogel sensors are renowned for their colorimetric sensing ability and can be utilized in many fields such as medical diagnosis, environmental detection, food safety, and industrial production. Previously, our group invented responsive photonic nanochains (RPNCs), which improve the response speed of photonic crystal hydrogel sensors by at least 2 to 3 orders of magnitude. However, RPNCs are dispersed in a liquid medium, which needs a magnetic field to orient them for the generation of structural colors. In addition, during repeated use, the process of cleaning and redispersing can cause entanglement, breakage, and a loss of RPNCs, resulting in poor stability. Moreover, when mixing with the samples in liquid, the RPNCs may lead to the contamination of the samples being tested. In this paper, we incorporate one-dimensional oriented RPNCs with agarose gel film to prepare heterogeneous hydrogel films. Thanks to the non-responsive and porous nature of the agarose gel, the protons diffuse freely in the gel, which facilitates the fast response of the RPNCs. Furthermore, the "frozen" RPNCs in agarose gel not only enable the display of structural colors without the need for a magnet but also improve the cycling stability and long-term durability of the sensor, and will not contaminate the samples.
