Abstract:The resolution of conventional optical equipment is always restricted by the diffraction limit, and improving on this was previously considered improbable. Optical super-resolution imaging, which has recently experienced rapid growth and attracted increasing global interest, will result in applications in many domains, benefiting fields such as biology, medicine and material research. This review discusses the contributions of different researchers who identified the diffractive barrier and attempted to realize optical super-resolution. This is followed by a personal viewpoint of the development of optical nanoscopy in recent decades and the road towards the next generation of optical nanoscopy.

Abstract:Achieving nanometer-scale resolution remains challenging in expansion microscopy due to photon loss. To address this concern, here we develop a multi-color expansion stimulated emission depletion technique based on small-molecule probes to realize high labeling density and intensity. Our method substantially lowers the barrier to visualizing diverse intracellular proteins and their interactions in three dimensions. It enables us to achieve sub-10-nm resolution in structures such as microfilaments, lysosomes, and mitochondria, providing new insights into cell biology.

Abstract:Far-field super-resolution imaging techniques such as stimulated emission depletion (STED), stochastic optical reconstruction microscopy (STORM), and photoactivation localization microscopy (PALM) etc., have enabled fluorescence labeled nanoscale imaging and analysis. However, it is necessary or preferable to develop label-free high resolution imaging techniques to avoid the problems of phototoxicity with live cells and moreover, for many non-biological applications where fluorescent labelling is simply not feasible. Recently, by capturing the exponentially decaying evanescent waves and propagating it to far field, novel imaging technique like hyperlens and microsphere contacting technologies, micro-fiber and nanowire illumination technology have brought new opportunities to label-free super resolution imaging, but still suffer from weak signal compared to background noise. On the other hand, there is lack of criteria reported to quantify the imaging quality of label free far-field nanoscopy, that will slow down the developing of label-free far-field nanoscopy. We introduce CNR first to label-free far-field nanoscopy to quantify the imaging performance and investigate the key influencing elements systematically. In our study, we investigated and analyzed the key elements to achieve high CNR label-free wide-field far-field sub-diffraction imaging. By optimizing the key factors including polarization, materials, as well as fabrication conditions, sub-diffraction imaging with resolution of 122 nm in a large FOV has been achieved experimentally. This work has provided an efficient and convenient way to realize high contrast wide-field far-field label-free super-resolution based on evanescent wave illumination.

Abstract:The development of nanomaterials has put forward high requirements for characterization techniques. Optical microscopy (OM), with easy accessibility and open operating spaces as compared to scanning electron microscopy, is a good choice to quickly locate materials and to be integrated with other equipment. However, OM is limited by its low resolution. Herein, we present a facile and non-destructive approach for optical observation of nanomaterials under conventional OMs with the aid of volatile nano-particles (NPs), which can be deposited and removed in a controlled manner. The NPs deposited on the surface of nanomaterials render strong light scattering to enable the nanomaterials to become optically visible. For example, this approach enables the observation of individual carbon nanotubes (CNTs) with OMs at low magnification or even with the naked eye. Both supported CNTs on various substrates and suspended CNTs can be observed with this approach. Most importantly, the NPs can be completely removed through moderate heat treatment or laser irradiation, avoiding potential influence on the properties or subsequent applications of nanomaterials. Furthermore, we systematically investigate the deposition of various volatile NPs (up to 14 kinds) for the optical observation of nanomaterials. We also demonstrated the application of this approach on other nanomaterials, including nanowires and graphene. We showed that this approach is facile, controllable, non-destructive, and contamination-free, indicating wide potential applications.

Abstract:The progress of near-field optics research and its application in some Chinese universities and institutions is overviewed. The research activities on the instrumentation aspect of scanning near-field optical microscope, novel sample-tip regulation mechanism, fiber-tip preparations, high resolution imaging and near-field spectroscopy in confined mesoscopic systems, light emission new materials and devices, theoretical development of near-field light-matter interactions, new observations of physical phenomena in the near-field region, optical nanostructuring, are summarized. The current research projects and future prospective of the possible application of SNOM and nearfield fluorescence imaging to biology, such as the in vitro cell nuclear assembly and apoptosis as well as gene recognition and DNA sequencing, and opto-electro devices are discussed.

Abstract:We propose an approach that uses a conventional optical microscope to achieve optical super-resolution. This approach will provide a direct, non-invasive, and far-field access in the observation of both metallic and non-metallic objects with a spatial resolution of tens-of-nanometres, achieved in a single snap shot. This method, combines near-field illumination by the micro-fiber and a passive spatial frequency shift by the sample itself, offering a promising approach to surface tomography imaging without localized field enhancement by surface plasmon polaritons. Our results present the potential of breaking the diffractive barrier using a conventional wide-field optical microscope within the visible spectrum.
Optical Spectroscopy of Individual Nano-materials with Defined Atomic Structure

Abstract:When the characteristic length of a material shrink to 1 nm scale, many distinct physical phenomena, such as quantum confinement, enhanced many-body interactions, strong van der Waals inter-material couplings and ultrafast charge separation, will appear. To investigate the related fascinating low-dimensional physics, we need a tool to quantitatively link the atomic structures to the physical properties of these very small nano-materials. In this talk, I will introduce our recently developed in-situ TEM + high-sensitive ultrafast nanooptics technique, which combines capability of structural characterization in TEM and property characterization in nanooptics on the same individual nano-materials. Several examples of using this technique to study the 1D carbon nanotube system and 2D atomic layered systems will be demonstrated.

While electron microscopes often provide static images of their chemical composition, morphology and structure, imaging the chemical activity of single nanoparticles is highly desirable for exploring the structure-activity relationship via a bottom-up strategy, to understand their microscopic reaction mechanisms and kinetics, and to identify a minority subpopulation with extraordinary activity. Recently, various optical microscopes have been emerging as powerful techniques towards this goal, owing to their non-invasive nature, excellent sensitivity, diversified spectroscopic principles and sufficient spatial and temporal resolution. In this review, we first introduce the motivational concept and the strength of using optical microscopy to study the chemical activity of single nanoparticles. In the second section, five types of commonly used optical microscopy, fluorescence microscopy, dark-field microscopy, surface plasmon resonance microscopy, Raman microscopy and photothermal microscopy are described, with an emphasis on their applicable nanomaterials and mechanisms for application. Recent achievements of these techniques in nanosensing, nanoelectrochemistry and nanocatalysis are surveyed and summarized in the subsequent sections, respectively. We finally conclude with our perspective on the remaining challenges and the future trends in this field.

Abstract:Here we present a simple yet powerful approach for the imaging of nanostructures under an optical microscope with the help of vapor condensation on their surfaces. Supersaturated water vapor will first form a nanometer-sized water droplet on the condensation nuclei on the surface of nanostructures, and then the water droplet will grow bigger and scatter more light to make the outline of the nanostructure be visible under dark-field optical microscope. This vapor-condensation-assisted (VCA) optical microscopy is applicable to a variety of nanostructures from ultralong carbon nanotubes to functional groups, generating images with contrast coming from the difference in density of the condensation sites, and does not induce any impurities to the specimens. Moreover, this low-cost and efficient technique can be conveniently integrated with other facilities, such as Raman spectroscope and so forth, which will pave the way for widespread applications.

Abstract:In the last few decades, nanoscience and nanotechnology have been growing with breath taking speed, and how to break through the diffraction limit and tame the light on a nanoscale have become the major challenges in optics. In this field, several super-resolution optical nanoscopy techniques have been developed, leading to a series of breakthroughs in physics, chemistry, and life sciences. In the work, we give a retrospect of the newly developed techniques in diffraction theory of linear optical systems, including the solid immersion lens, structured light illumination microscopy, scanning near-field optical microscopy, metamaterial-based wide field near-field imaging technique and super-oscillatory lens. Brief discussion on their principles, advantages and applications is also provided.

Far-field Nanoscale Thermal and Structure Imaging

Abstract:The imaging resolution of a conventional optical microscope is limited by diffraction to about half of the illuminating wavelength. A micro/submicron particle induced near-field effect can focus an incident laser beam to a small region of tens of nanometres. Using self-assembled transparent microspheres as far-field superlenses (FSLs), here we report the first experimental demonstration of combined structural, thermal, and stress imaging at a 20 nm resolution (<λ/26, λ=532 nm), even though the laser beam is at μm size. FSLs are integrated into a confocal Raman spectrometer and microscope system to overcome the diffraction limit. Periodical surface structure, thermal and stress information is examined. Furthermore, electromagnetic simulation is conducted to well interpret the experimental results. FSLs provide new opportunities in nanoscale imaging, nanolithography and nanotexturing.
