Abstract:The molecular biomarkers in relation to environmental pollutants have become imperative research topic for environmental toxicology studies. In order to set up a series of simple, sensitive and exact monitoring systems for environmental pollution, several studies were carried out. The potential of oxygen free radicals and other reactive oxygen species (ROS) to damage tissues and cellular components in organisms was called oxidative stress. The balance between pro-oxidant endogenous and exogenous factors (i.e., environmental pollutants) and anti-oxidant defenses in biological systems has been used to assess the adverse effect of environmental pollutants, especially for oxidant damage of the different chemicals. The anti-oxidative defense systems and their sensitivity for oxidative stress are of great importance in environmental toxicology. Numerous studies on the effects of oxidative stress to organisms imposed by environmental pollutants were performed. The results indicated that the transition metals, polycyclic aromatic hydrocarbons, organophosphate and organochlorine pesticides, polychlorinated biphenyls, dioxins, and other xenobiotics could cause oxidative stress to organisms. These toxicants can stimulate a variety of toxicity effects, such as oxidative damage to membrane lipid, DNA and protein and changes to antioxidant enzymes. This review summarizes the current knowledge and advances in understanding such oxidative stresses. And the application of related molecular biomarkers in aquatic organisms was also discussed.

Abstract:An imbalance between oxidants and antioxidants in favour of the oxidants, potentially leading to damage, is termed 'oxidative stress'. Oxidants are formed as a normal product of aerobic metabolism but can be produced at elevated rates under pathophysiological conditions. Antioxidant defense involves several strategies, both enzymatic and non-enzymatic. In the lipid phase, tocopherols and carotenes as well as oxy-carotenoids are of interest, as are vitamin A and ubiquinols. In the aqueous phase, there are ascorbate, glutathione and other compounds. In addition to the cytosol, the nuclear and mitochondrial matrices and extracellular fluids are protected. Overall, these low molecular mass antioxidant molecules add significantly to the defense provided by the enzymes superoxide dismutase, catalase and glutathione peroxidases.

Abstract:Thirty-five years ago, Sies and colleagues insightfully described the universal phenomenon that the generation of reactive oxygen species could modify macromolecules in living organisms, resulting in a wide range of measurable damage. They used the term "oxidative stress" to define the loss of the balance between oxidants and antioxidants in favor of the former. After decades of research, it became increasingly clear that cells are not simply passive receivers of oxidative modification but can act dynamically to resist and adapt to oxidants. Furthermore, many redox-sensitive pathways have been identified wherein certain oxidants (mainly hydrogen peroxide and nitric oxide) are used as messenger molecules to transduce the signals required for these adaptations. Since the turn of the century, redox signaling has developed into a vibrant multidisciplinary field of biology. To reflect the evolution of the study in this field, the definition of oxidative stress is postulated to define a state in which the pro-oxidative processes overwhelm cellular antioxidant defense due to the disruption of redox signaling and adaptation.

Abstract:Reactive oxygen species (ROS) typically produce in algae and act as secondary messengers in numerous cellular processes. Under abiotic stresses, the balance between production and suppression of ROS disappears and causes increase of ROS. Increasing excessive ROS can cause damage to various cellular components comprising cell membranes, proteins and lipids. Algae have an antioxidant defense system to overcome on oxidative damage. Antioxidant defense mechanisms are of two types, namely enzymatic and non-enzymatic antioxidants. The enzymatic antioxidants include superoxide dismutase, catalase, ascorbate peroxidase and glutathione reductase. The non-enzymatic antioxidants include carotenoids, tocopherol, ascorbic acid, glutathione, flavonoids and phenolic compounds. In this review, we describe the various types of ROS and their production, and antioxidant defense mechanisms for ROS suppression.

Abstract:Oxidative stress is a normal phenomenon in the body. Under normal conditions, the physiologically important intracellular levels of reactive oxygen species (ROS) are maintained at low levels by various enzyme systems participating in the in vivo redox homeostasis. Therefore, oxidative stress can also be viewed as an imbalance between the prooxidants and antioxidants in the body. For the last two decades, oxidative stress has been one of the most burning topics among the biological researchers all over the world. Several reasons can be assigned to justify its importance: knowledge about reactive oxygen and nitrogen species production and metabolism; identification of biomarkers for oxidative damage; evidence relating manifestation of chronic and some acute health problems to oxidative stress; identification of various dietary antioxidants present in plant foods as bioactive molecules; and so on. This review discusses the importance of oxidative stress in the body growth and development as well as proteomic and genomic evidences of its relationship with disease development, incidence of malignancies and autoimmune disorders, increased susceptibility to bacterial, viral, and parasitic diseases, and an interplay with prooxidants and antioxidants for maintaining a sound health, which would be helpful in enhancing the knowledge of any biochemist, pathophysiologist, or medical personnel regarding this important issue.

Abstract:Exposure to a variety of environmental factors such as salinity, drought, metal toxicity, extreme temperature, air pollutants, ultraviolet-B (UV-B) radiation, pesticides, and pathogen infection leads to subject oxidative stress in plants, which in turn affects multiple biological processes via reactive oxygen species (ROS) generation. ROS include hydroxyl radicals, singlet oxygen, and hydrogen peroxide in the plant cells and activates signaling pathways leading to some changes of physiological, biochemical, and molecular mechanisms in cellular metabolism. Excessive ROS, however, cause oxidative stress, a state of imbalance between the production of ROS and the neutralization of free radicals by antioxidants, resulting in damage of cellular components including lipids, nucleic acids, metabolites, and proteins, which finally leads to the death of cells in plants. Thus, maintaining a physiological level of ROS is crucial for aerobic organisms, which relies on the combined operation of enzymatic and nonenzymatic antioxidants. In order to improve plants’ tolerance towards the harsh environment, it is vital to reinforce the comprehension of oxidative stress and antioxidant systems. In this review, recent findings on the metabolism of ROS as well as the antioxidative defense machinery are briefly updated. The latest findings on differential regulation of antioxidants at multiple levels under adverse environment are also discussed here.

Abstract:Cytochrome P450 enzyme system is a clan of heme oxidases that is substantial in oxidative metabolism and activation of drugs in the liver and extrahepatic sites of the body. Compounds that are metabolized through CYP450 can fabricate reactive intermediates that can deplete oxygen at molecular level and yield reactive oxygen species (ROS). Such detrimental events taking place in the body are defied and compensated by body cells via diverse defense mechanisms. Defensive mechanisms may include the induction of free radical removers, for instance, some vitamins (C and E), bilirubin, thiols, some lipophilic scavengers such as ubiquinol and enzymes, e.g., glutathione peroxidases, superoxide dismutase, or catalases to sophisticate and embellish DNA repair mechanisms. In the circumstances, when the defense is inadequate to combat these free radicals, ROS hold the charge and oxidative stress is an obvious incident to happen. The resulting oxidative stress then bows the seeds of idiosyncratic drug toxicity by damaging the cells, proteins, and DNA. Consequently, by the study of CYP-controlled modified drug metabolism and resulting in oxidative stress, plenty of malfunctions can be prevented. Ample studies have intensified the contribution of hepatic metabolism, directing toward the complex diseases. The motive of this review is to highlight the oxidative stress due to ROS that are produced during CYP-induced drug metabolism and detoxification with antioxidant therapy. The writing will also elaborate on the unexplored aspects of this field that are yet to be considered.

Abstract:There is current great international concern about the contribution of environmental pollution to the global burden of disease particularly in the developing, low- and medium-income countries. Industrial activities, urbanization, developmental projects as well as various increased anthropogenic activities involving the improper generation, management and disposal of pollutants have rendered today's environment highly polluted with various pollutants. These pollutants include toxic metals (lead, cadmium, mercury, arsenic), polycyclic aromatic hydrocarbons, polychlorinated biphenyls, pesticides and diesel exhaust particles most of which appear to be ubiquitous as well as have long-term environmental persistence with a wide range of toxicities such as oxidative stress among others. Oxidative stress, which may arise from increased production of damaging free radicals emanating from increased pollutant burden and depressed bioavailability of antioxidant defenses causes altered biochemical and physiological mechanisms and has been implicated in all known human pathologies most of which are chronic. Oxidative stress also affects both flora and fauna and plants are very important components of the terrestrial environment and significant contributors of nutrients for both man and animals. It is also remarkable that the aquatic environment in which sea animals and creatures are resident is also highly polluted, leading to aquatic stress that may affect the survival of the aquatic animals, sharing in the oxidative stress. These altered terrestrial and aquatic environments have an overarching effect on human health. Antioxidants neutralize the damaging free radicals thus, they play important protective roles in the onset, progression and severity of the unmitigated generation of pollutants that ultimately manifest as oxidative stress. Consequently, human health as well as that of aquatic and terrestrial organisms may be protected from environmental pollution by mitigating oxidative stress and employing the principles of nutritional medicine, essentially based on antioxidants derived mainly from plants, which serve as the panacea of the vicious state of environmental pollutants consequently, the health of the population. Understanding the total picture of oxidative stress and integrating the terrestrial and aquatic effects of environmental pollutants are central to sustainable health of the population and appear to require multi-sectoral collaborations from diverse disciplinary perspectives; basically the environmental, agricultural and health sectors.

Abstract:The paradox of aerobic life, or the 'Oxygen Paradox', is that higher eukaryotic aerobic organisms cannot exist without oxygen, yet oxygen is inherently dangerous to their existence. This 'dark side' of oxygen relates directly to the fact that each oxygen atom has one unpaired electron in its outer valence shell, and molecular oxygen has two unpaired electrons. Thus atomic oxygen is a free radical and molecular oxygen is a (free) bi-radical. Concerted tetravalent reduction of oxygen by the mitochondrial electron-transport chain, to produce water, is considered to be a relatively safe process; however, the univalent reduction of oxygen generates reactive intermediates. The reductive environment of the cellular milieu provides ample opportunities for oxygen to undergo unscheduled univalent reduction. Thus the superoxide anion radical, hydrogen peroxide and the extremely reactive hydroxyl radical are common products of life in an aerobic environment, and these agents appear to be responsible for oxygen toxicity. To survive in such an unfriendly oxygen environment, living organisms generate--or garner from their surroundings--a variety of water- and lipid-soluble antioxidant compounds. Additionally, a series of antioxidant enzymes, whose role is to intercept and inactivate reactive oxygen intermediates, is synthesized by all known aerobic organisms. Although extremely important, the antioxidant enzymes and compounds are not completely effective in preventing oxidative damage. To deal with the damage that does still occur, a series of damage removal/repair enzymes, for proteins, lipids and DNA, is synthesized. Finally, since oxidative stress levels may vary from time to time, organisms are able to adapt to such fluctuating stresses by inducing the synthesis of antioxidant enzymes and damage removal/repair enzymes. In a perfect world the story would end here; unfortunately, biology is seldom so precise. The reality appears to be that, despite the valiant antioxidant and repair mechanisms described above, oxidative damage remains an inescapable outcome of aerobic existence. In recent years oxidative stress has been implicated in a wide variety of degenerative processes, diseases and syndromes, including the following: mutagenesis, cell transformation and cancer; atherosclerosis, arteriosclerosis, heart attacks, strokes and ischaemia/reperfusion injury; chronic inflammatory diseases, such as rheumatoid arthritis, lupus erythematosus and psoriatic arthritis; acute inflammatory problems, such as wound healing; photo-oxidative stresses to the eye, such as cataract; central-nervous-system disorders, such as certain forms of familial amyotrophic lateral sclerosis, certain glutathione peroxidase-linked adolescent seizures, Parkinson's disease and Alzheimer's dementia; and a wide variety of age-related disorders, perhaps even including factors underlying the aging process itself. Some of these oxidation-linked diseases or disorders can be exacerbated, perhaps even initiated, by numerous environmental pro-oxidants and/or pro-oxidant drugs and foods. Alternatively, compounds found in certain foods may be able to significantly bolster biological resistance against oxidants. Currently, great interest centres on the possible protective value of a wide variety of plant-derived antioxidant compounds, particularly those from fruits and vegetables.

Abstract:Facultative bacteria can grow under either oxic or anoxic conditions. While oxygen provides substantial advantages in energy yield by respiration, it can become life-threatening because of reactive oxygen species that derive from the molecule naturally. Thus, to survive and thrive in a given niche, these bacteria have to constantly regulate physiological processes to make maximum benefits from oxygen respiration while restraining oxidative stress. Molecular mechanisms and physiological consequences of oxidative stress have been under extensive investigation for decades, mostly on research model Escherichia coli, from which our understanding of bacterial oxidative stress response is largely derived. Nevertheless, given that bacteria live in enormously diverse environments, to cope with oxidative stress different strategies are conceivably developed.

Abstract:Oxidative stress is defined as “an imbalance between oxidants and antioxidants in favor of the oxidants, leading to a disruption of redox signaling and control and/or molecular damage”. This Commentary presents basic features of this global concept which has attracted interest in biology and medicine. The term “antioxidants” in cellular defense against oxidants predominantly includes antioxidant enzymes with their substrates and coenzymes. Exogenous low-molecular-mass compounds also have a role, but this is more limited. Multiple biomarkers of damage due to oxidative stress have been identified for different molecular classes (protein, lipid, carbohydrate, and DNA), and the current state of practical aspects in health and disease is delineated.

Abstract:Chemical technology-based oxidative stress evaluation methods are of particular significance in practical applications. Reactive oxygen species/reactive nitrogen species (ROS/RNS) are inevitable metabolic by-products of the metabolism of the vital processes in living organisms. It has been hypothesized that the redox reaction balance could be coordinated with drugs and other interventions to prevent or treat diseases. Several indicators have been tested as evaluation standards in this field but standardized indicators have been difficult to achieve due to the complex regulation of the redox reactions in different organisms. Therefore, it is inevitable to use multiple methods based on different principles in order to evaluate oxidative stress and antioxidant capacity. In this review, we summarized the oxidative stress and the antioxidant capacity evaluation methods used in various organisms and antioxidant-using methods. We also described the strengths and weaknesses of each method, as well as potential directions for the development of new technologies.

Abstract:'Oxidative stress' (OS) refers to the deviations in redox biology inside biological systems. This term has been used to illustrate the imbalance between oxidants and antioxidants, which has been a subject of extensive research. The obtained findings suggest that many diseases are linked to OS, including diabetes mellitusinflammation, cancers, autoimmune disorders and neurodegenerative diseases. More recently, significant advances in biochemistry and molecular biology have enabled researchers to find master switches of various OS mechanisms and associate OS development to many biological paradigms. In this review, a brief overview of OS research is provided to offer insight into its future directions. Most importantly, the need for a greater understanding of the OS molecular mechanisms and its vital role in the progression of many disorders is emphasized. Likewise, more work is needed on the utility of reactive oxygen species markers as a diagnostic tool.

Abstract:Oxidative stress is a phenomenon caused by an imbalance between production and accumulation of oxygen reactive species (ROS) in cells and tissues and the ability of a biological system to detoxify these reactive products. ROS can play, and in fact they do it, several physiological roles (i.e., cell signaling), and they are normally generated as by-products of oxygen metabolism; despite this, environmental stressors (i.e., UV, ionizing radiations, pollutants, and heavy metals) and xenobiotics (i.e., antiblastic drugs) contribute to greatly increase ROS production, therefore causing the imbalance that leads to cell and tissue damage (oxidative stress). Several antioxidants have been exploited in recent years for their actual or supposed beneficial effect against oxidative stress, such as vitamin E, flavonoids, and polyphenols. While we tend to describe oxidative stress just as harmful for human body, it is true as well that it is exploited as a therapeutic approach to treat clinical conditions such as cancer, with a certain degree of clinical success. In this review, we will describe the most recent findings in the oxidative stress field, highlighting both its bad and good sides for human health.

Abstract:Fish are among the best-studied aquatic animals due to their economic and ecological values. Fish meat is the most affordable protein source for the economically weaker section of people. The environment of almost all aquatic ecosystems has a specific influential role on or by fishes. Therefore, studying their stress biology, especially oxidative stress, is vital because it can influence their growth, production, reproduction, etc. To review the above topic, peer-reviewed electronic databases, including Web of Science, science direct, PubMed, Google Scholar, Scopus, and AGRICOLA, were searched with specific keywords associated with fish, oxidative stress, diseases, etc. The influence of abiotic stress, such as the effects of water dissolved oxygen, temperature, salinity, water hardness, alkalinity, pH, pollutants, heavy metals, and anthropogenic activities, was reviewed in the current article to draw a conclusion on the updated relation that exists between fish physiology, disease, and abiotic stressors. Oxidative stress and redox regulatory levels under the above parameters were reviewed as the stress or anti-stress responses differ in various fish models. Undoubtedly, the reviewed abiotic factors modulate fish oxidative health status to a greater extent, and therefore, these factors must be considered on a priority basis to improve the general health and immunity status of fish. The statement above remains valid in both saline and freshwater habitats.

Abstract:Oxidative stress, resulting from the excessive production of reactive oxygen species, is a common and major cause of cellular damage in plants exposed to various abiotic stresses. To address this challenge, we introduce the concept of antioxidant agriculture as a comprehensive strategy to improve stress tolerance and thus crop productivity by minimizing oxidative stress levels in the field environment. This strategy encompasses a diverse range of approaches, including genetic engineering, the exogenous application of antioxidant agents, microbial inoculation, and agronomic practices, to reinforce the plant's intrinsic antioxidant defense system and mitigate oxidative stress. We present recent successful studies of antioxidant measures that have been validated in field conditions, along with our perspective on achieving antioxidant agriculture.

Abstract:Oxidative stress is a biological mechanism triggered by an imbalance between the biosynthesis and accumulation of reactive oxygen species (ROS) in a biological system. Normally, ROS are produced as byproducts of oxygen metabolism and fulfill various physiological roles such as cell signaling. However, environmental stressors like UV radiation, ionizing radiation, pollutants, heavy metals, and xenobiotics can also lead to increased ROS production. Common types of ROS include superoxide radicals (O2−), hydrogen peroxide (H2O2), hydroxyl radicals (•OH), and singlet oxygen (1O2), which are generated during various metabolic pathways. Key cellular processes such as protein phosphorylation, activation of transcriptional factors, apoptosis, immunity, and differentiation depend on maintaining appropriate levels of ROS within cells. Under conditions of excessive ROS production, which can occur during both normal physiological processes and pathological conditions like inflammation, ischemia, and aging, ROS can damage important cellular components such as proteins, lipids, and nucleic acids. This production is primarily mitochondrial but can also result from enzymatic activities involved in the respiratory chain, prostaglandin synthesis, phagocytosis, and the cytochrome P450 system. Both immune system activation and environmental factors contribute to endogenous and exogenous free radical production. The signs of oxidative stress include fatigue, memory loss, muscle or joint pain, the appearance of wrinkles and grey hair, decreased eyesight, headaches, increased sensitivity to noise, and a heightened susceptibility to infections. To reduce oxidative stress, it is advised to avoid sugar and processed foods while balancing blood sugar levels, prevent infections, incorporate daily stress reduction practices, avoid toxins, and promote the production of antioxidants through diet and herbal supplements that are rich in antioxidants.

Abstract:In this study, the antioxidant responses induced by menadione in zebrafish (Danio rerio) were analyzed. Changes in activities of superoxide dismutase (SOD), and catalase (CAT) in the liver, gill, muscle and kidney of fish were determined. The results showed that there were significant changes in SOD and CAT activities along with the different time and concentrations to different organs and tissues of zebrafish in the mass, and that were correspond with the position and function of organs and tissues. © 2011 IEEE.
