Abstract:Developing physical methods to modulate biomolecular clusters and condensates on cell membranes is of great importance for understanding physiological and pathological processes as well as for stimulating novel therapeutic strategies. Here, we propose an effective means to control receptor condensation on the cell membrane via specific adhesion to a supported lipid bilayer (SLB) with nanoscale topography. The specific adhesion is mediated by receptors in the cell membrane that bind their ligands anchored in the SLB. Using Monte Carlo simulations and mean-field theory, we demonstrate that the nanoscale topography of the SLB can enhance condensation of the receptors associated with lipid nanodomains. Our results indicate that SLBs with nanoscale topography proves an effective physical stimulus for tuning condensation of membrane adhesion proteins and lipids in cell membranes, and can serve as a feasible option to control and direct cellular activities, e.g., stem cell differentiation for biomedical and therapeutic applications.

Abstract:Cell adhesion on a supported lipid bilayer (SLB) functionalized with ligand proteins is a widely-used cell-mimetic model for the study of the trans receptor-ligand interaction that mediates the adhesion, and has been shown to play an important role in unravelling the molecular players in cell adhesion. Experimental studies of such cell-SLB adhesion systems often assume that there is no cis interactions between the ligands. An important question remains whether the cis-interaction affects the trans receptor-ligand interaction. Using a statistical-mechanical model and Monte Carlo simulations with biologically relevant parameters, we find that the attractive cis-interaction of strength 1 k(B)T between adjacent ligands on the SLB can lead to an amplification of both the affinity and cooperativity of the receptor-ligand binding, thereby facilitating the phase separation within the adhering cell membrane. In contrast, the adhesion system is less sensitive to the repulsive cis-interaction between adjacent ligands on the SLB. Our results suggest that the ligand-ligand cis-interaction should be carefully considered in the cell-SLB adhesion experiments.

Abstract:Integrins are adhesion molecules that connect a cell to its environment and transduce signals bidirectionally across the membrane. Their different functional states correspond to distinct conformations. Using a biomembrane force probe, we observed real-time reversible switches between bent and extended conformations of single cell surface integrin αLβ2 by measuring its nanometer-scale headpiece displacements, (un)bending frequencies and molecular stiffness changes. We determined stabilities of these conformations, their dynamic equilibrium, speeds and rates of conformational changes, and impacts of divalent cations and tensile forces. We quantified how initial conformations and their subsequent changes of αLβ2 regulate the force-dependent kinetics of dissociation from intercellular adhesion molecule 1. Our findings provide new insights into how integrins function as nanomachines to precisely control cell adhesion and signaling.

Abstract:Detailed understanding and control of the intermolecular forces that govern molecular assembly are necessary to engineer structure and function at the nanoscale. Liquid crystal (LC) assembly is exceptionally sensitive to surface properties, capable of transducing nanoscale intermolecular interactions into a macroscopic optical readout. Self-assembled monolayers (SAMs) modify surface interactions and are known to influence LC alignment. Here, we exploit the different dipole magnitudes and orientations of carboranethiol and -dithiol positional isomers to deconvolve the influence of SAM-LC dipolar coupling from variations in molecular geometry, tilt, and order. Director orientations and anchoring energies are measured for LC cells employing various carboranethiol and -dithiol isomer alignment layers. The normal component of the molecular dipole in the SAM, toward or away from the underlying substrate, was found to determine the in-plane LC director orientation relative to the anisotropy axis of the surface. By using LC alignment as a probe of interaction strength, we elucidate the role of dipolar coupling of molecular monolayers to their environment in determining molecular orientations. We apply this understanding to advance the engineering of molecular interactions at the nanoscale.

Abstract:Cell adhesion involved in biological processes such as cell migration, immune responses, and cancer metastasis, is mediated by the specific binding of receptor and ligand proteins. Some of these proteins exhibit affinity for nanoscale lipid clusters in cell membranes. A key question is how these nanoscale lipid clusters influence and react to the receptor-ligand binding during cell adhesion. In this article, we review recent computational studies that shed new light on the interplay of the receptor-ligand binding and the formation of lipid domains in adhering membranes. These studies indicate that the receptor- ligand binding promotes coalescence of lipid clusters into mesoscale domains, which, in turn, enhances both the affinity and cooperativity of the receptor-ligand binding in cell-cell adhesion with mobile ligands. In contrast, in the case of cell-extracellular matrix adhesion with immobile ligands, the receptor- ligand binding and the lipid cluster coalescence can be correlated or anti-correlated, depending strongly on the ligand distribution. These findings deepen our understanding of correlations between cell adhesion and membrane heterogeneities.

Abstract:The nanoparticle (nano)-cell membrane interface is one of the most important interactions determining the fate of nanoparticles (NPs), which can stimulate a series of biological events, allowing theranostic and other biomedical applications. So far, there remains a lack of knowledge about the mechanisms governing the nanoparticle-cell membrane interface, especially the impact of ligand exchange, in which molecules on the nanosurface become replaced with components of the cell membrane, resulting in unique interfacial phenomena. Herein, we describe a family of gold nanoparticles (AuNPs) of the same core size (∼13 nm core), modified with 12 different kinds of surface ligands, and the effects of their exchangeable ligands on both nanoparticle-supported lipid bilayers (SLBs) and nanoparticle-natural cell membrane interfaces. The ligands are categorized according to their molecular weight, charge, and bonding modes (physisorption or chemisorption). Importantly, we found that, depending on the adsorption affinity and size of ligand molecules, physisorbed ligands on the surface of NPs can be exchanged with lipid molecules. At a ligand exchange-dominated interface, the AuNPs typically aggregated into an ordered monolayer in the lipid bilayers, subsequently affecting cell membrane integrity, NP uptake efficiency, and the NP endocytosis pathways. These findings advance our understanding of the underlying mechanisms of the biological effects of nanoparticles from a new point of view and will aid in the design of novel, safe, and effective nanomaterials for biomedicine.
Modeling the evolution of membrane during cell adhesion on the nanostructured substrate

Abstract:Using nanostructured substrates for enhancing cell adhesion is an important method. However, the physical mechanism of cell adhesion on nanostructured substrate is not well understood yet, particularly the evolution process of membrane during cell adhesion. Here, we propose an analytical model to study quantitatively the evolution of membrane during adhesion process on the nanostructured substrate. The effects of substrate surface topography, receptor-ligand binding, substrate property and membrane property were investigated. We found a small nanopillar radius, low nanopillar density, large binding rate, large receptor density, low non-specific energy, or large stretching modulus can result in a large adhesion rate and a large adhesion depth. Besides, the evolution of membrane on truncated nanocone arrays was also studied by the model. The theoretical results are in good agreement with the experimental observations, which implied that our studies can provide a useful guide to control cell adhesion process and cell motion on nanostructured substrates.

Abstract:Cell adhesion with extracellular matrix depends on the collective behaviors of a large number of receptor-ligand bonds at the compliant cell-matrix interface. While most biological tissues and structures, including cells and extracellular matrices, exhibit strongly anisotropic material properties, existing studies on molecular adhesion via receptor-ligand bonds have been largely limited to isotropic materials. Here the effects of transverse isotropy, a common form of material anisotropy in biological systems, in modulating the adhesion behavior of a cluster of receptor-ligand bonds are investigated. The results provide a theoretical basis to understand cell adhesion on anisotropic extracellular matrices and to explore the possibility of controlling cell adhesion via anisotropy design in material properties. The combined analysis and simulations show that the orientation of material anisotropy strongly affects the apparent softness felt by the adhesive bonds, thereby altering their ensemble lifetime by several orders of magnitude. An implication of this study is that distinct cellular behaviors can be achieved through remodeling of material anisotropy in either extracellular matrix or cytoskeleton. Comparison between different loading conditions, together with the effects of material anisotropy, yields a rich array of out-of-equilibrium behaviors in the molecular interaction between reactant-bearing soft surfaces, with important implications on the mechanosensitivity of cells.

Abstract:Supported lipid bilayer (SLB) is one of the most widely used structures to mimic cell membranes. To study the cell cell, cell matrix and cell material interactions, supported lipid bilayers (SLBs) functionalized with RGD peptides (SLBs RGD) were prepared by vesicle fusion on a SiO2 quartz crystal, and subsequently bone mesenchymal stem cells (BMSCs) adhesion was analyzed. A quartz crystal microbalance with dissipation (QCM-D) was utilized to detect the dynamic adsorption behavior of lipid vesicles and BMSCs in real time. Observations obtained by QCM-D signals are confirmed by conducting fluorescence microscopy.QCM-D measurements showed the SLB formation starts at the critical concentration of the vesicles. More BMSCs adhered on SLBs RGD than on SLBs. With the presence of SLBs, the adhesion cells on SLBs surfaces had a rounded morphology, and cells on SLBs RGD will take long time to rearrange their cytoskeleton, which led to incomplete spreading compared with SiO2. Differences in adhesion density and adhesion properties of the cells on the different substrates could be traced at the dissipation versus frequency (Delta D/Delta f) plots. These results indicate that RGD in/on SLBs could provide anchorage sites for more cells adhesion. QCM-D is demonstrated to be a useful tool for evaluating the interactions between various biological and non-biological systems in situ and in real-time. (C) 2014 Published by Elsevier B.V.
Tuning Interfacial Patterns of Molecular Bonds Via Surface Morphology.

Abstract:Many studies have demonstrated that the mechanical properties of the extracellular matrix can significantly influence the morphology, strength and lifetime of focal adhesions. However, how the morphology of the contact surface affects the pattern formation of the molecular bonds still remains largely unknown. Here, by simplifying the cell and extracellular matrix to two opposing elastic bodies and considering the lateral diffusion as well as the bonding/debonding of molecular bonds, we study the clustering behavior of receptor-ligand bonds between curved surfaces and the phase diagrams of cluster patterns. We reveal the important role of surface morphology and bond kinetics in regulating the patterns of bond clusters. We further investigate the segregation dynamics of the interfacial bonds under various loading speeds, and we show that the average interfacial stress is rate-dependent while the rupture stress is rate-independent. Finally, we demonstrate that programmable patterning of bond clusters can be achieved through the designed surface morphology.
Probing Mechanical Principles of Focal Contacts in Cell–matrix Adhesion with a Coupled Stochastic–elastic Modelling Framework

Abstract:Cell-matrix adhesion depends on the collective behaviours of clusters of receptor-ligand bonds called focal contacts between cell and extracellular matrix. While the behaviour of a single molecular bond is governed by statistical mechanics at the molecular scale, continuum mechanics should be valid at a larger scale. This paper presents an overview of a series of recent theoretical studies aimed at probing the basic mechanical principles of focal contacts in cell-matrix adhesion via stochastic-elastic models in which stochastic descriptions of molecular bonds and elastic descriptions of interfacial traction-separation are unified in a single modelling framework. The intention here is to illustrate these principles using simple analytical and numerical models. The aim of the discussions is to provide possible clues to the following questions: why does the size of focal adhesions (FAs) fall into a narrow range around the micrometre scale? How can cells sense and respond to substrates of varied stiffness via FAs? How do the magnitude and orientation of mechanical forces affect the binding dynamics of FAs? The effects of cluster size, cell-matrix elastic modulus, loading direction and cytoskeletal pretension on the lifetime of FA clusters have been investigated by theoretical arguments as well as Monte Carlo numerical simulations, with results showing that intermediate adhesion size, stiff substrate, cytoskeleton stiffening, low-angle pulling and moderate cytoskeletal pretension are factors that contribute to stable FAs. From a mechanistic point of view, these results provide possible explanations for a wide range of experimental observations and suggest multiple mechanisms by which cells can actively control adhesion and de-adhesion via cytoskeletal contractile machinery in response to mechanical properties of their surroundings.
Computer Simulation of the Effects of Nanoparticles' Adsorption on the Properties of Supported Lipid Bilayer

Abstract:Supported lipid bilayer (SLB) represents a kind of well-established model cell membrane and is also used for many biosensors or biodevices. Here, for the first time, we use molecular dynamics simulation to study the effects of nanoparticle (NP) adsorption on SLB. In our simulations, the surface charge properties and the heating effects of NPs are investigated. Results show that NPs adsorption behavior, SLB's diffusion ability, and local order parameter distribution are largely dominated by the property of the NPs' surface charge. Meanwhile the NPs' heating can increase the nearby lipids' and water's thermal motions, thus disrupting the surface charge's domination on the aforementioned properties. Besides, we find that the solid support may induce more intense thermal motions but poorer diffusion ability for the lipid leaflet closer to the support. This study provides useful insights on the NPs' disruption to the functioning of the biological membrane and the performance of SLB-based biosensors or biodevices.

Abstract:Focal adhesions are clusters of specific receptor-ligand bonds that link an animal cell to an extracellular matrix. A capability to control focal adhesions, for which a quantitative description of the collective behavior of multiple molecular bonds is a critical step, is essential for tissue and cellular engineering. While the behavior of single molecular bonds is governed by statistical mechanics at small scale, continuum mechanics should be valid at large scale. How can this transition be modeled and can this tell us something about the mechanics of cell adhesion? Here we develop a stochastic-elasticity model of a periodic array of adhesion clusters between two dissimilar elastic media subjected to an inclined tensile stress, in which stochastic descriptions of molecular bonds and elastic descriptions of interfacial traction are unified in a single modeling framework. A fundamental scaling law of interfacial traction distribution is established to govern the transition between uniform and cracklike singular distributions of the interfacial traction within molecular bonds. Guided by this scaling law, we perform Monte Carlo simulations to investigate the effects of cluster size, cell/matrix modulus and loading direction on lifetime and strength of the adhesion clusters. The results show that intermediate adhesion size, stiff substrate, cytoskeleton stiffening, and low-angle pulling are factors that contribute to the stability of focal adhesions. The predictions of our model provide feasible explanations for a wide range of experimental observations and suggest possible mechanisms by which cells can modulate adhesion and deadhesion via cytoskeletal contractile machinery.

Abstract:Cells are always living in a dynamic microenvironment and are able to respond to stimuli and modulate their functions. Cell adhesion is the fundamental of almost all cell behaviors, such as cell proliferation, migration, differentiation etc. Therefore, it is essential to provide a platform for studying and regulating cell adhesion in dynamic environment. Owing to the controllability of surface properties, materials with dynamic surface stand out as ideal candidates of this platform. This review summarized the recent advances of design strategies of dynamic surface, and their applications in regulating cell adhesion and related functions.

Abstract:Experimental findings indicate that cell function and behavior such as cell growth, division, migration and differentiation, are subtly regulated via integrin-dependent cell adhesion. Cell adhesion is influenced by nanoscale ligand spacing and rigidity of extracellular substrates, as cell adhesion drops greatly when the ligand spacing is larger than ~60nm, and cell adhesion is stronger on stiff than soft substrates. However, how nanoscale ligand spacing and substrate stiffness jointly affect integrin clustering and hence nascent cell adhesion remains to be elucidated. To quantitatively investigate the phenomena and the underlying mechanochemical mechanism of integrin clustering modulated by ligand spacing and substrate stiffness, we introduced Monte Carlo simulations varying the values of ligand spacing and substrate stiffness. Moreover, the effects of integrin number, integrin binding free energy, integrin association free energy, and local ligand spacing were investigated. The simulation results showed that integrin clustering decreased sharply, when ligand spacing was relatively large such as dL>60nm in the current simulations, regardless of substrate rigidities, though with close spacing, the clustering increased with the substrate stiffness. The investigation contributes to the goals of understanding and predicting experimental phenomena, directing and optimizing biomaterial design, and manipulating integrin-dependent cell-substrate adhesion in tissue engineering.

Abstract:Nanoscale organization of transmembrane receptors is becoming recognized as critical for cellular functions, enabled by the nanoscale engineering of bioligand presentation. Previously, a spatial threshold of ≤60 nm for integrin binding ligands in cell‐matrix adhesion was demonstrated using mono‐ligand gold nanoparticles. However, the ligand geometric arrangement was limited to hexagonal arrays of mono‐ligands, while plasmonic quenching by gold limited further investigation by fluorescence‐based high‐resolution imaging. Here, these limitations were overcome with dielectric TiO2 nanopatterns, which allows fine‐tuning ligand cluster size, eliminating fluorescence quenching, thus enabling super‐resolution fluorescence microscopy on nanopatterns. By dual‐color super‐resolution imaging, high precision and consistency among nanopatterns, bioligands, and integrin nanoclusters was observed validating the high quality and integrity of both nanopattern functionalization and passivation. By screening TiO2 nanodiscs with various diameters, an increase in fibroblast cell adhesion, spreading area, and YAP nuclear localization on 100nm‐diameter compared with smaller diameters was observed, and Focal Adhesion Kinase was identified as the regulatory signal. These findings explore the optimal ligand presentation when the minimal requirements are sufficiently fulfilled in the heterogenous ECM network of isolated binding regions with abundant ligands. Integration of high‐fidelity nano‐biopatterning with super‐resolution imaging will allow precise quantitative studies to address early signaling events in response to receptor clustering and their nanoscale organization. Two simple l/d-phenylalanine-based derivatives (L/DPFM) have been designed, and their solvent-dependent morphology evolutions are illustrated. It was found that, as the content of H2O in aqueous ethanol solutions was increased, LPFM self-assembles first into right-handed nanofibers, then flat fibrous structures, and finally inversed left-handed nanofibers. Assemblies in ethanol and H2O exhibit opposite conformations and circular dichroism (CD) signals even though they are constructed from the same molecules. Thus, the morphology-dependent cell adhesion and proliferation behaviors are further characterized. Left-handed nanofibers are found to be more favorable for cell adhesion than right-handed nanostructures. Quantitative AFM analysis showed that the L929 cell adhesion force on left-handed LPFM fibers is much higher than that on structures with inversed handedness. Moreover, the value of cell Young's modulus is lower for left-handed nanofibrous films, which indicates better flexibility. The difference in cell-substrate interactions might lead to different effects on cell behavior.
Protein Nanosheet Mechanics Controls Cell Adhesion and Expansion on Low-Viscosity Liquids

Abstract:Adherent cell culture typically requires cell spreading at the surface of solid substrates to sustain the formation of stable focal adhesions and assembly of a contractile cytoskeleton. However, a few reports have demonstrated that cell culture is possible on liquid substrates such as silicone and fluorinated oils, even displaying very low viscosities (0.77 cSt). Such behavior is surprising as low viscosity liquids are thought to relax much too fast (<ms) to enable the stabilization of focal adhesions (with lifetimes on the order of minutes to hours). Here we show that cell spreading and proliferation at the surface of low viscosity liquids are enabled by the self-assembly of mechanically strong protein nanosheets at these interfaces. We propose that this phenomenon results from the denaturation of globular proteins, such as albumin, in combination with the coupling of surfactant molecules to the resulting protein nanosheets. We use interfacial rheology and atomic force microscopy indentation to characterize the mechanical properties of protein nanosheets and associated liquid-liquid interfaces. We identify a direct relationship between interfacial mechanics and the association of surfactant molecules with proteins and polymers assembled at liquid-liquid interfaces. In addition, our data indicate that cells primarily sense in-plane mechanical properties of interfaces, rather than relying on surface tension to sustain spreading, as in the spreading of water striders. These findings demonstrate that bulk and nanoscale mechanical properties may be designed independently, to provide structure and regulate cell phenotype, therefore calling for a paradigm shift for the design of biomaterials in regenerative medicine.
