Abstract:Tetrazine-derived fluorogenic labels are extensively studied for their potential in biological and medical imaging. Nonetheless, the fluorescence quenching mechanism in numerous precursors continues to be debated, particularly as the wavelengths extend into the red and near-infrared (NIR) regions. This challenge poses obstacles to systematically optimizing their fluorogenicity, i.e., achieving red-shifted wavelengths and improved fluorescence turn-on signals through click reactions. This paper highlights the significance of photoinduced charge centralization (PCC), a quenching mechanism observed in tetrazine-fused fluorogenic labels with integrated π-conjugations. PCC is primarily responsible for the quenching effects observed in such labels emitting in the red-to-NIR spectrum. Drawing from structure-property relationships, this study proposes two molecular design strategies that incorporate the PCC mechanism and constitutional isomerization to develop high-performance tetrazine-based labels. These strategies facilitate multiplex fluorescence imaging following click reactions, promising significant advancements in bio-orthogonal imaging techniques.

Abstract:Conventional thermally activated delayed fluorescence (TADF) molecules achieve small energy differences between the lowest singlet and triplet excited states (EEST) by enhancing the intramolecular charge transfer, which inevitably leads to a wide emission spectrum and low fluorescence rate. Here, we prepared a deep blue TADF molecule via a small EEST pyridine-phenol fluoroboron complex as the acceptor. The small EEST is maintained when carbazole donors are attached to the 4-position of the phenyl rings in the fluoroboron complex. Benefiting from the strong electron coupling between the donor (D) and acceptor (A) moieties, the compound Cz-4-BF exhibits a high fluorescence rate of 4.8 x 108 s- 1 and a small D-A dihedral angle change in the excited state. Consequently, a photoluminescence (PL) quantum yield of nearly 100% and a PL spectrum with full-width at half-maximum (FWHM) < 60 nm were obtained in solution and low-concentration doped films. A TADFsensitized fluorescence (TSF) device containing Cz-4-BF achieves an external quantum efficiency of 21%, which is higher than the devices employing classical fluorescent emitters and multiple resonance-type TADF emitters. The Cz-4-BF-based TSF device shows significantly improved color coordinates of (0.14, 0.10) versus a control device without Cz-4-BF.

Abstract:The incorporation of aggregation-induced emission (AIE) luminogens provides a molecule with enhanced fluorescent emission in the solid state. A representative AIEgen compound is tetraphenylethene (TPE). A series of new conjugates of quinolone derivatives with TPE units (1a - 1f) was synthesized and characterized. The influence on the photophysics of 1a - 1e with a variety of functional groups was investigated systematically by spectroscopic methods and simulated by density-functional theory calculations. A centrosymmetric compound donor-acceptor-donor-type compound 1f with two TPE moieties proved to be the most promising multi-functional material, it exhibited tri-colored (489-569 nm) fluorescence characteristics in tetrahydrofuran-H2O solution with a different water fraction and intense tri-colored solvatochromic (blue, green, and yellow) behaviors. These compounds could be applied as promising acid-base gas probe based on their evident color switch. Compounds 1a - 1f showed the advantage of being a potential piezochromic material.
Nature of the Lowest Singlet and Triplet Excited States of Organic Thermally Activated Delayed Fluorescence Emitters: A Self-Consistent Quantum Mechanics/Embedded Charge Study

Abstract:Thermally activated delayed fluorescence (TADF) is dictated by the properties of the lowest singlet (S-1) and triplet (T-1) excited states. Both small energy difference (Delta E-ST) and large spin-orbit coupling (SOC) between S-1 and T-1 are desired to increase the rate for reverse intersystem crossing (RISC). In this work, we investigated the ground- and excited-state electronic properties of three representative D-(pi)-A type TADF molecules in solid phase by means of a self-consistent quantum-mechanics/embedded-charge (QM/EC) approach, which consists of a series of iterative QM/EC single-point computations to account for the solid-state polarization effect. The results show that, unless the D and A units are perpendicular to each other, both the S-1 and T-1 states are characteristic of mixed charge transfer (CT) and local excitation (LE). Thereby, the LEST values are relatively large in gas phase. Importantly, the CT contribution is relatively larger in the S-1 state than in the T-1 state; thus, the S-1 energy is more stabilized by electronic polarization, leading to smaller REST in solid phase. At the same time, the SOC can be considerable due to the difference in the nature of the S-1 and T-1 states. These results shed light on the origin of fast RISC in efficient organic TADF systems.

Abstract:A novel thermally activated delayed fluorescent (TADF) compound, 9-(3-((4,6-diphenyl-1,3,5-triazin-2-yl)oxy)phenyl)-3,6-diphenyl-9H-carbazole (PhCz-o-Trz), with donor-σ-acceptor (D-σ-A) motif is developed. A flexible small space σ-junction is adopted to partly suppress the intramolecular charge transfer (intra-CT) while inversely enhancing the intermolecular CT (inter-CT) between D/A moieties, realizing the coexistence of both intra- and inter-CT in an amorphous aggregate. The coexistence of dual CTs increases the complexity of the singlet and triplet states mixing, enhancing the triplet-to-singlet spin-flip transition and thereby the TADF emission. Additionally, PhCz-o-Trz is evaluated not only as an emitter but also a sensitizing host for fluorescent and phosphorescent dopants, all realizing high efficiencies with alleviated efficiency roll-offs. These results shed light on the development of new TADF materials with dual CTs and may further deepen our understanding about TADF mechanisms.

Abstract:Carbon dots (CDs) possess unique optical properties such as tunable photoluminescence (PL) and excitation dependent multicolor emission. The quenching and recovery of the fluorescence of CDs can be utilized for detecting analytes. The PL mechanisms of CDs have been discussed in previous articles, but the quenching mechanisms of CDs have not been summarized so far. Quenching mechanisms include static quenching, dynamic quenching, Förster resonance energy transfer (FRET), photoinduced electron transfer (PET), surface energy transfer (SET), Dexter energy transfer (DET) and inner filter effect (IFE). Following an introduction, the review (with 88 refs.) first summarizes the various kinds of quenching mechanisms of CDs (including static quenching, dynamic quenching, FRET, PET and IFE), the principles of these quenching mechanisms, and the methods of distinguishing these quenching mechanisms. Abstract:Energy transfer and electron transfer are both fundamental mechanisms enabling numerous functional materials and applications. While most materials systems employ either energy transfer or electron transfer, the combined effect of energy and electron transfer processes in a single donor/acceptor system remains largely unexplored. Herein, we demonstrated the energy transfer followed by electron transfer (ETET) process in a molecular dyad TPE-NBD. Due to energy transfer, the fluorescence of TPE-NBD was greatly enhanced in non-polar solvents. In contrast, polar solvents activated subsequent electron transfer and markedly quenched the emission of TPE-NBD. Consequently, ETET endows TPE-NBD with significant polarity sensitivities. We expect that employing ETET could generate many functional materials with unprecedented properties, i.e., for single laser powered multicolor fluorescence imaging and sensing.

Abstract:Although there are some proposed explanations for fluorescence enhancement of metal ions, a phenomenon with wide applications in biosensors and chemosensors, current understanding of the quantum–mechanical origin of this photophysical behaviour is limited. To address this issue, we selected thymolphthalein-based probe (T) reported recently as model system, and investigated the photochemical and photophysical properties of T and T-Zn(II) complex. The geometry structures of T and its Zn(II) complex were optimized, and the experimental absorption spectrum were well reproduced through density functional theory (DFT) methods. Furthermore, the excited state intramolecular proton transfer (ESIPT) mechanisms of T were systematically investigated. The constructed potential energy surfaces (PESs) of T at ground (S0) and first exited (S1) states indicate that, after photo-excitation, the intramolecular single-proton transfer reaction occurs more readily with the H atom removing from O to neighboring N. Furthermore, thermodynamic analysis of the T-Zn(II) complex shows that mononuclear complex (C1) is the most thermodynamically stable structure, and the intramolecular single proton transfer process of C1 is thermodynamically unfavorable at the S1 state. Time-dependent DFT calculations show that the fluorescence emission is from the S1 states of the single-proton transfer tautomer of T, as predicted from Kasha's rule, in comparison, the fluorescence emission of Zn(II) complex is from the S1 states of T-Zn(II) rather than its single-proton transfer tautomer. The inhibitor of Zn(II) on the single proton transfer reaction plays an important role on fluorescence enhancement.

Correlated Single Quantum Dot Blinking and Interfacial Electron Transfer Dynamics

Abstract:The electron transfer (ET) dynamics from core/multi-shell (CdSe/CdS(3ML)ZnCdS(2ML)ZnS(2ML)) quantum dots (QDs) to adsorbed Fluorescein (F27) molecules have been studied by single particle spectroscopy to probe the relationship between single QD interfacial electron transfer and blinking dynamics. Electron transfer from the QD to F27 and the subsequent recombination were directly observed by ensemble-averaged transient absorption spectroscopy. Single QD-F27 complexes show correlated fluctuation of fluorescence intensity and lifetime, similar to those observed in free QDs. With increasing ET rate (controlled by F27-to-QD ratio), the lifetime of on states decreases and relative contribution of off states increases. It was shown that ET is active for QDs in on states, the excited state lifetime of which reflects the ET rate, whereas in the off state QD excitons decay by Auger relaxation and ET is not a competitive quenching pathway. Thus, the blinking dynamics of single QDs modulate their interfacial ET activity. Furthermore, interfacial ET provides an additional pathway for generating off states, leading to correlated single QD interfacial ET and blinking dynamics in QD-acceptor complexes. Because blinking is a general phenomenon of single QDs, it appears that the correlated interfacial ET and blinking and the resulting intermittent ET activity are general phenomena for single QDs.

Abstract:A fluorophore consisting of two fluorescent moieties could display unusual optical behaviors that are unattainable in a single-fluorophore compound. Herein we reported two "dual-fluorophore" dyes: DNS-linked tetraphenylethene demonstrates bright aggregation-induced emission, while NBD-linked tetraphenylethene exhibits aggregation-caused quenching. Our results have important implications for engineering emission behaviors of molecular aggregates for practical applications.

Abstract:In terms of the molecular design for thermally activated delayed fluorescence (TADF), the high degree of spatial separation of frontier molecular orbitals can be achieved through constructing cofacial donor-acceptor alignment in a through-space charge transfer (TSCT) framework. When extremely low Delta E-ST is realized, TSCT-TADF molecules still suffer a low radiative transition rate (k(r)) of the emissive TSCT state. Compact TSCT-TADF emitters, mCz-Xo-TRZ and dCz-Xo-TRZ, have been reported, showing a high photoluminescence quantum yield (PLQY) with large k(r). To explore the physical mechanism of high PLQY and large k(r) in the TSCT conformation, comprehensive theoretical calculations on geometries, excitation energies, transition character, strong and weak electronic coupling, and vibration analysis on structural reorganization accompanied by time-resolved spectroscopy were applied to illustrate the electronic coupling mechanism in the compact and rigid TSCT framework and reveal the limited structural relaxation contributing to the nonradiative dissipation in excited states. This work contributes a deep understanding of the electronic interaction mechanism in TSCT molecular frameworks and provides theoretical guidance for the rational design of TSCT luminescent materials.

Abstract:Stable luminescent pi-radicals with doublet emission have aroused a growing interest for functional molecular materials. We have demonstrated a neutral pi-radical dye (4-N-carbazolyl-2,6-dichlorophenyl)bis(2,4,6-trichlorophenyl)-methyl (TTM-1Cz) with remarkable doublet emission, which could be used as triplet sensitizer to initiate the photophysical process of triplet-triplet annihilation photon upconversion (TTA-UC). Dexter-like excited doublet-triplet energy transfer (DTET) was confirmed by theoretical calculation. A mixed solution of TTM-1Cz and aromatic emitters could ambidextrous upconvert red light to cyan light or green light = 532 nm) to blue light. This finding of DTET phenomena provides a new perspective in designing new triplet sensitizer for TTA-UC.

Abstract:Photoinduced electron transfer (PET) between organic fluorophores and suitable electron donating moieties, for example, the amino acid tryptophan or the nucleobase guanine, can quench fluorescence upon van der Waals contact and thus report on molecular contact. PET-quenching has been used as reporter for monitoring conformational dynamics in polypeptides, proteins, and oligonucleotides. Whereas dynamic quenching transiently influences quantum yield and fluorescence lifetime of the fluorophore, static quenching in pi-stacked complexes efficiently suppresses fluorescence emission over time scales longer than the fluorescence lifetime. Static quenching therefore provides sufficient contrast to be observed at the single-molecule level. Here, we review complex formation and static quenching of different fluorophores by various molecular compounds, discuss applications as reporter system for macromolecular dynamics, and give illustrating examples.

Donor-Acceptor Fluorophores for Energy-Transfer-Mediated Photocatalysis

Abstract:Triplet-triplet energy transfer (EnT) is a fundamental activation pathway in photocatalysis. In this work, we report the mechanistic origins of the triplet excited state of carbazole-cyanobenzene donor-acceptor (D-A) fluorophores in EnT-based photocatalytic reactions and demonstrate the key factors that control the accessibility of the 3LE (locally excited triplet state) and 3CT (charge-transfer triplet state) via a combined photochemical and transient absorption spectroscopic study. We found that the energy order between 1CT (charge transfer singlet state) and 3LE dictates the accessibility of 3LE/3CT for EnT, which can be effectively engineered by varying solvent polarity and D-A character to depopulate 3LE and facilitate EnT from the chemically more tunable 3CT state for photosensitization. Following the above design principle, a new D-A fluorophore with strong D-A character and weak redox potential is identified, which exhibits high efficiency for Ni(II)-catalyzed cross-coupling of carboxylic acids and aryl halides with a wide substrate scope and high selectivity. Our results not only provide key fundamental insight on the EnT mechanism of D-A fluorophores but also establish its wide utility in EnT-mediated photocatalytic reactions.

Abstract:In view of the application prospects in biomedicine of (E)-1-(4-(diethyla-mino)-2-hydroxybenzylidene)-4,4-dimethylthiosemicarbazide (DAHTS), the behavior of excited-state dynamics and photophysical properties were studied using the density functional theory/time-dependent density functional theory method. A series of studies indicated that the intramolecular hydrogen-bond (IHB) intensity of DAHTS was enhanced after photoexcitation. This was conducive to promoting the excited-state intramolecular proton-transfer (ESIPT) process. Combining the analysis of the IHB and hole-electron, it revealed that the molecule underwent both the ESIPT process and the twisted charge-transfer (TICT) process. Relying on exploration of the potential energy surface, it was proposed that the different competitive mechanisms between the ESIPT and TICT processes were regulated by solvent polarity. In acetonitrile (ACN) solvent, the ESIPT process occurred first, and the TICT process occurred later. In contrast, in the CYH solvent, the molecule first underwent the TICT process and then the ESIPT process. Furthermore, we raised the possibility that the TICT behavior was the cause of weak fluorescence emission for the DAHTS in CYH and ACN solvents. By the dimer correlation analysis, the corresponding components of triple fluorescence emission were clearly assigned, corresponding to the monomer, dimer, and ESIPT isomer in turn. Our work precisely elucidated the photophysical mechanism of DAHTS and the attribution of the triple fluorescence emission components, which provided valuable guidance for the development and regulation of bioactive fluorescence probes with multiband and multicolor emission characteristics.

Abstract:In an energy transfer (ET) process, it is the optical responses of donor and acceptor materials on the single-particle level that ultimately determine its overall performance. Here we conduct time-tagged, time-resolved optical measurements to correlate the photoluminescence (PL) intensities and lifetimes of a donor semiconductor nanocrystal (NC) and acceptor dye molecules linked to its surface. We reveal that the PL intensity of dye molecules follows exactly the blinking behavior of the donor NC and shows a step-like quenching behavior due to the photobleaching effect. The corresponding recovery of the NC PL intensity has allowed us to realize the textbook definition of PL quantum efficiency measurement in dye molecules upon absorbing a single exciton. Our theoretical fitting of the lifetime data demonstrates that the buildup time of acceptor PL could be solely determined by the radiative lifetime of dye molecules when it is any shorter than the NC lifetime, thus confirming the long-existing Förster theory on ET dynamics.

Abstract:Molecular design of dual-fluorescent probes requires precise adjustment of the energy levels of two excited states and the energy barrier between them. While the hybridized local and charge-transfer (HLCT) state has been recently focused as an important excited state for high radiation efficiency with a tunable energy level, a dual emission involving the HLCT state has been only achieved with the excited-state intramolecular proton transfer (ESIPT) system. Here, a series of dual-fluorescent molecules involving a HLCT excited state with the excited-state conjugation enhancement (ESCE) motif is presented as the first case. The energy level of the HLCT state has been adjusted by modulating substituents and solvents, separately from the ESCE energy level. The HLCT-ESCE molecular design with tunable fluorescence properties proposes a new strategy for the development of advanced fluorescent probes.
