Abstract:Near-infrared (NIR) responsive nanoparticles are an important platform for multimodal phototherapy. Importantly, the simultaneous NIR-triggered photodynamic (PDT) and photothermal (PTT) therapy is a powerful approach to increase the antitumor efficiency of phototherapic nanoparticles due to the synergistic effect. Herein, a boron dipyrromethene (BODIPY)-based amphiphilic dye with enhanced electron donor-acceptor-donor (D-A-D) structure (BDP-AP) was designed and synthesized, which could self-assemble into stable nanoparticles (BDP-AP NPs) for the synergistic NIR-triggered PDT/PTT therapy. BDP-AP NPs synchronously generated singlet oxygen (1O2) and achieved preeminent photothermal conversion efficiency (61.42%). The in vitro and in vivo experiments showed that BDP-AP NPs possessed negligible dark cytotoxicity and infusive anticancer performance. BDP-AP NPs provide valuable guidance for the construction of PDT/PTT-synergistic NIR nanoagents to improve the efficiency of photoinduced cancer therapy in the future.

Abstract:Flower‐like and mesoporous sphere‐like PDA@Pt nanozymes are synthesized and compared for their enzyme‐mimicking activity and photothermal conversion efficiency. The unique morphology of the flower‐like PDA@Pt results in an enhanced ability to kill tumor cells. This variant highlights the importance of considering the physicochemical properties of nanozymes to optimize their therapeutic potential in antitumor strategies . Recently, nanozyme‐based photothermal‐catalytic therapy has emerged as a promising strategy for antitumor treatment. Extensive research has focused on optimizing the catalytic activity and photothermal conversion performance of nanozymes through size, morphology, and surface property regulations. However, the biological effects of nanozymes, such as cellular uptake and cytotoxicity, resulting from their physicochemical properties, remain largely unexplored. In this study, two types of polydopamine/platinum (PDA@Pt) nanozymes, flower‐like (FPDA@Pt) and mesoporous spherical‐like (MPDA@Pt), to comprehensively compare their enzyme‐mimicking activity, photothermal conversion capacity, and antitumor efficiency are designed. These findings revealed that FPDA@Pt exhibited superior peroxidase‐like activity and higher photothermal conversion efficiency compared to MPDA@Pt. This led to enhanced production of reactive oxygen species (ROS) and increased heat generation at tumor sites. Importantly, it is observed thatthe flower‐like structure of FPDA@Pt facilitated enhanced cellular uptake, leading to an increased accumulation of nanozymes within tumor cells. Furthermore, the light irradiation on tumors also triggered a series of anti‐tumor immune responses, further enhancing the therapeutic efficacy. This work provides a possible design orientation for nanozyme‐based photothermal‐catalytic tumor therapy, highlighting the importance of considering the physicochemical properties of nanozymes to optimize their therapeutic potential in antitumor strategies.

A Photosensitizer-Loaded Polydopamine Nanomedicine Agent for Synergistic Photodynamic and Photothermal Therapy

Abstract:Photodynamic therapy (PDT) and photothermal therapy (PTT) have emerged as promising non-invasive approaches to cancer treatment. However, the development of multifunctional nanomedicines is necessary to enhance these approaches' effectiveness and safety. In this study, we investigated a polydopamine-based nanoparticle (PDA-ZnPc+ Nps) loaded with the efficient photosensitizer ZnPc(4TAP)12+ (ZnPc+) through in vitro and in vivo experiments to achieve synergistic PDT and PTT. Our results demonstrated that PDA-ZnPc+ Nps exhibited remarkable efficacy due to its ability to generate reactive oxygen species (ROS), induce photothermal effects, and promote apoptosis in cancer cells. Moreover, in both MCF-7 cells and MCF-7 tumor-bearing mice, the combined PDT/PTT treatment with PDA-ZnPc+ Nps led to synergistic effects. Subcellular localization analysis revealed a high accumulation of ZnPc+ in the cytoplasm of cancer cells, resulting in cellular disruption and vacuolation following synergistic PDT/PTT. Furthermore, PDA-ZnPc+ Nps exhibited significant antitumor effects without causing evident systemic damage in vivo, enabling the use of lower doses of photosensitizer and ensuring safer treatment. Our study not only highlights the potential of PDA-ZnPc+ Nps as a dual-functional anticancer agent combining PDA and PTT but also offers a strategy for mitigating the side effects associated with clinical photosensitizers, particularly dark toxicity.

Facile Synthesis of ZnPc‐Polydopamine Co‐loaded Nanoparticles for Synergetic Photodynamic‐Photothermal Therapy

Abstract:A major obstacle to clinical photodynamic therapy (PDT) is tissue hypoxia in a solid tumor, which substantially reduces therapeutic efficacy by insufficient oxygen supply. To tackle this issue, herein, we reported a type of facilely prepared multifunctional nanoparticles (NPs) for combined treatment of PDT and photothermal therapy (PTT). A novel dopamine-modified epoxysilane (GSDA) was firstly synthesized from 3-glycidoxypropyl(trimethoxy) silane and dopamine via ring-opening reaction, which was subsequently incorporated into NPs along with zinc(II) phthalocyanine (ZnPc) via a reprecipitation-encapsulation method. Interestingly, polydopamine (PDA) was found to be automatically developed inside NPs by oxidative polymerization of dopamine monomers. Owing to strong-light-absorption PDA and ZnPc, the resultant biocompatible NPs exhibited high photothermal conversion efficiency (eta=46.4%) and a decent singlet oxygen quantum yield (phi Delta=0.28), respectively, under the irradiation of 808 nm and 660 nm light. Synergetic treatment was then performed on tumor cells using the multifunctional NPs, i. e., PDT followed by PTT, and enhanced phototherapeutic efficacy was achieved, particularly in the case of hypoxia. This work demonstrates that (i) the newly-synthesized GSDA might function as a bi-functional coupling agent for silica and PDA coating, and (ii) the simply-prepared NPs are very promising for synergetic PDT/PTT of solid tumors.
Dual-response Oxygen-Generating MnO2 Nanoparticles with Polydopamine Modification for Combined Photothermal-Photodynamic Therapy

Abstract:Photonic therapy (e.g., photodynamic therapy (PDT) and photothermal therapy (PTT)), features easy handling, good targeting, high curative effect and lower side effects compared to other treatments, thus showing considerable potential in cancer treatment. However, adverse tumor microenvironment, low light conversion efficiency, and inherent toxicity of photonic agents limit their further application in clinic. In this work, hollow mesoporous MnO2 nanoparticles (NPs) loaded with photosensitizer chlorin e6 and coated with folic acid-functionalized polydopamine were fabricated, and used as a platform for photonic therapy. The NPs enabled accurately controlled drug release, and sustainable and extensive O-2 production by reacting with endogenous H2O2 in vitro. In addition, the NPs allowed efficient NIR light-to-heat conversion, owing to the core-shell MnO2/polydopamine structure. With the synergy of folic acid mediated active targeting and enhanced permeability and retention effect mediated passive targeting, a pronounced NPs accumulation at the tumor sites of mouse model was observed. The NPs also exhibited excellent biodegradation properties, alleviating the possible concerns about the long-term safety of photonic agents. Finally, a combination of O-2-strengthened PDT and PTT resulted in effective tumor growth inhibition upon irradiation with 660 nm and 808 nm laser in vivo treatment. The above features of the devised NPs highlight their potential for improved cancer therapy and imaging.

Abstract:The unsatisfactory efficacy of conventional theranostic agents in ablating tumor poses urgent demands on the development of high-performance integrated theranostic agents utilizing rising nanotechnology. To cope with the existing limitations, here we presented an intelligent nanoplatform based on yolk-shell Fe3O4@polydopamine prepared by mussel-inspired polydopamine chemistry and sacrificial template method as well as subsequent incorporation of Pt nanoparticles and chlorine 6 (Ce6) by in situ reduction and electrostatic adsorption for photodynamic therapy (PDT) and photothermal (PTT). The resultant nanoplatform could effectively deliver photosensitizer Ce6 to tumor sites, then promoting the decomposition of endogenous H2O2 to oxygen, finally achieving enhanced PDT therapy, which is demonstrated by in vitro and in vivo evaluations. Importantly, the generated oxygen bubbles could improve the echogenicity signal of yolk-shell microspheres and thereby provide enhanced ultrasonic (US) signal for imaging solid tumors. Overall, the synergistic combination of magnetic Fe3O4, green polydopamine, catalytic Pt nanoparticles, photosensitive Ce6 enabled the hybrid nanoplatform to have good biocompatibility, efficient tumor accumulation, excellent phototherapy efficiency, high T2-weighted magnetic resonance imaging (MRI) and fluorescence imaging ability (FL). Our study integrating the merits of PDT/PTT and US/MRI/FL into a single nanoplatform will open an avenue of therapeutic strategy toward biomedical applications.

Abstract:Nanocatalytic therapy (NCT) has made great achievements in tumor treatments due to its remarkable enzyme-like activities and high specificity. Nevertheless, the limited types of nanozymes and undesirable tumor microenvironments (TME) greatly weaken the therapeutic efficiency. Developing a combination therapy integrating NCT and other strategies is of great significance for optimal treatment outcomes. Herein, a AuPt-loaded Cu-doped polydopamine nanocomposite (AuPt@Cu-PDA) with multiple enzyme-like activities was rationally designed, which integrated photothermal therapy (PTT) and NCT. The peroxidase (POD)-like activity of AuPt@Cu-PDA can catalyze hydrogen peroxide (H2O2) into ·OH, and the catalase (CAT)-mimic activity can decompose H2O2 into O2 to alleviate hypoxia of TME, and O2 can be further converted into toxic ·O2- by its oxidase (OXD)-mimic activity. In addition, Cu2+ in AuPt@Cu-PDA can effectively consume GSH overexpressed in tumor cells. The boosting of reactive oxygen species (ROS) and glutathione (GSH) depletion can lead to severe oxidative stress, which can be enhanced by its excellent photothermal performance. Most importantly, the accumulation of Cu2+ can disrupt copper homeostasis, promote the aggregation of lipoylated dihydrolipoamide S-acetyltransferase (DLAT), disrupt the mitochondrial tricarboxylic acid (TCA) cycle, and finally result in cuproptosis. Collectively, photothermal and photoacoustic imaging (PTI/PAI)-guided cuproptosis-enhanced NCT/PTT can be achieved. This work may expand the application of nanozymes in synergistic therapy and provide new insights into cuproptosis-related therapeutic strategies.
Oxygen-evolving hollow polydopamine alleviates tumour hypoxia for enhancing photodynamic therapy in cancer treatment

Abstract:Hypoxia, a characteristic hallmark of solid tumours, restricts the therapeutic effect of photodynamic therapy (PDT) for cancer treatment. To address this issue, a facile and nanosized oxygen (O2) bubbles template is established by mixing oxygenated water and water-soluble solvents for guiding hollow polydopamine (HPDA) synthesis, and O2 is encapsulated in the cavity of HPDA. HPDA with abundant catechol is designed as carrier for Zinc phthalocyanine (ZnPc, a boronic acid modified photosensitizer) via borate ester bonds to fabricate nanomedicine (denoted HZNPs). The results in vitro and in vivo indicate O2-evolving HZNPs could alleviate tumour hypoxia and enhance PDT-anticancer efficiency. Melanin-like HPDA with photothermal conversion rate (η) of 38.2% shows excellent synergistic photothermal therapy (PTT) efficiency in cancer treatment.

Abstract:Chemodynamic therapy (CDT) and photothermal therapy (PTT) have both demonstrated considerable efficacy in the tumor treatment individually, owing to their non-invasive nature and excellent selectivity. However, due to the propensity of tumors for metastasis and recurrence, a singular therapeutic approach falls short of achieving optimal treatment outcomes. Polydopamine (PDA) has excellent photothermal conversion ability and polyoxometalates (POMs) possess diverse enzymatic activities. Here, we synthesized PDA@POM nanospheres comprising polydopamine-coated Tungsten-based polyoxometalate (W-POM). These nanospheres leverage dual enzymatic activities that synergistically enhance both chemodynamic and photothermal therapies for tumor treatment. The PDA-mediated PTT effect enables precise tumor cell destruction, while the W-POM nanozymes catalyzes the generation of highly toxic reactive oxygen species (ROS) from hydrogen peroxide within tumor cells through a Fenton-like reaction, which mitigates tumor hypoxia and induces tumor cell death. This synergistic photothermal catalytic therapy shows enhanced efficacy in tumor suppression, providing a promising new approach for tumor treatment.
Polymeric Nanoparticles with ROS‐Responsive Prodrug and Platinum Nanozyme for Enhanced Chemophotodynamic Therapy of Colon Cancer

Abstract:The combination of chemotherapy and photodynamic therapy (PDT) has promising potential in the synergistic treatment of cancer. However, chemotherapy and photodynamic synergistic therapy are impeded by uncontrolled chemotherapeutics release behavior, targeting deficiencies, and hypoxia‐associated poor PDT efficacy in solid tumors. Here, a platinum nanozyme (PtNP) loaded reactive oxygen species (ROS)‐responsive prodrug nanoparticle (CPT‐TK‐HPPH/Pt NP) is created to overcome these limitations. The ROS‐responsive prodrug consists of a thioketal bond linked with camptothecin (CPT) and photosensitizer‐2‐(1‐hexyloxyethyl)‐2‐devinyl pyropheophorbide‐a (HPPH). The PtNP in CPT‐TK‐HPPH/Pt NP can efficiently catalyze the decomposition of hydrogen peroxide (H2O2) into oxygen to relieve hypoxia. The production of oxygen can satisfy the consumption of HPPH under 660 nm laser irradiation to attain the on‐demand release of CPT and ensure enhanced photodynamic therapy. As a tumor diagnosis agent, the results of photoacoustic imaging and fluorescence imaging for CPT‐TK‐HPPH/Pt NP exhibit desirable long circulation and enhanced in vivo targeting. CPT‐TK‐HPPH/Pt NPs effectively inhibit tumor proliferation and growth in vitro and in vivo. CPT‐TK‐HPPH/Pt NP, with its excellent ROS‐responsive drug release behavior and enhanced PDT efficiency can serve as a new cancer theranostic agent, and will further promote the research of chemophotodynamic synergistic cancer therapy.

Abstract:Photothermal therapy (PTT) that utilizes phSUPPotothermal conversion agents (PTC) to ablate tumor under NIR light irradiation has attracted increasing attention due to its excellent therapeutic efficacy and improved target selectivity. Herein, a novel core-shell nanoparticle based on disulfide-crosslinked poly(methacrylic acid) (PMAA) layer coated polydopamine (PDA) particle has been successfully synthesized by precipitation polymerization. For these PDA@PMAA composite nanoparticles, PDA core exhibits high photothermal efficacy, meanwhile, the redox-labile PMAA shell serves as carriers to encapsulate anticancer drugs and selectively release them. Due to the characteristic of the disulfide bond, PMAA shell occurs at selective degradation as well as controlled drug release upon entering cancer cells. Moreover, the DOX-loaded PDA@PMAA nanoparticles demonstrated a synergistic effect, which shows a significantly improved inhibition effect against cancer cells by the combination of photothermal therapy and traditional chemotherapy with low drug dosage and short laser irradiation in an in vitro study.

Abstract:Abstract Polyamine detection and depletion have been extensively investigated for cancer prevention and treatment. However, the therapeutic efficacy is far from satisfactory, mainly due to a polyamine compensation mechanism from the systemic circulation in the tumor environment. Herein, we explore a new solution for improving polyamine detection as well as a possible consumption therapy based on a new photosensitizer that can efficiently consume polyamines via an irreversible chemical reaction. The new photosensitizer is pyrrolopyrrole aza -BODIPY pyridinium salt ( PPAB-PyS ) nanoparticles that can react with the over-expressed polyamine in cancer cells and produce two photosensitizers with enhanced phototoxicity on cancer destruction. Meanwhile, PPAB-PyS nanoparticles provide a simultaneous ratiometric fluorescence imaging of intracellular polyamine. This combination polyamine consumption with a chemical reaction provides a new modality to enable polyamine detection along with photodynamic therapy as well as a putative depletion of polyamines for cancer treatment and prevention.

Polydopamine Coated Au-Pt Nanorods: Enhanced Photothermal Properties and Efficient Reactive Oxygen Scavengers

Abstract:As an emerging cancer treatment strategy, photothermal therapy (PTT) is precise, controllable, minimally invasive, low cost and less toxic side effects, thus photothermal transduction agents have been extensively investigated in recent years. Noble metal nanomaterials with unique localized surface plasmon resonance (LSPR) effects are particularly suitable as photothermal transduction agent, but the currently developed precious noble metal nano photothermal transduction agents face serious problems such as complex synthesis process, poor photothermal performance and high biotoxicity. Moreover, the large amount of reactive oxygen species (ROS) produced during PTT treatment could cause irreversible damage to the healthy tissues surrounding the tumor. In this work, we deposited platinum (Pt) on the tips of gold nanorods (AuNRs) to form dumbbell-shaped Au-Pt bimetallic nanorods (AuPtNRs), and functionalized AuPtNRs with biocompatible polydopamine (PDA) to obtain AuPt@PDA. With 808 nm laser irradiation, the prepared AuPt@PDA exhibited excellent photothermal stability, and its photothermal conversion efficiency (PCE) reached 81.78%, which was significantly higher than that of AuNRs (52.32%) and AuPtNRs (78.76%). With low cytotoxicity, AuPt@PDA decreased cell viability from 91.12% to 39.36% after PTT on cancer cells in vitro, while significantly reducing intracellular ROS levels generated by heat stress. Therefore, the excellent photothermal properties, high cancer cell killing and ROS scavenging activity of AuPt@PDA in PTT could be an ideal candidate for improving therapeutic efficacy while reducing the risk of toxic side effects due to heat stress-induced ROS formation.
Polydopamine-encapsulated zinc peroxide nanoparticles to target the metabolism-redox circuit against tumor adaptability for mild photothermal therapy

Abstract:Regulating the metabolism-redox circuit of cancer cells has emerged as an attractive strategy to improve the therapeutic outcome, while often confronting the glaring issue of resistance due to the multiple adaptive responses of tumor cells. This study presents a simple yet efficient approach to regulate this circuit simultaneously against tumor adaptability by utilizing polydopamine-encapsulated zinc peroxide nanoparticles (ZnO2@PDA NPs). The nanoparticles could deliver large amounts of Zn2+ and H2O2 into tumor cells to unfold an intracellular self-amplifying loop for breaking the balance in zinc and redox homeostasis by H2O2-mediated endogenous Zn2+ release from metallothioneins due to its oxidation by H2O2 and Zn2+-induced in situ H2O2 production by disturbing mitochondrial respiration, ultimately disrupting tumor adaptability to exogenous stimuli. The elevated levels of Zn2+ and H2O2 also inhibited adenosine triphosphate (ATP) generation from glycolysis and mitochondrial respiration to disrupt energy adaptability. Furthermore, insufficient ATP supply could reduce glutathione and heat shock protein expression, thereby sensitizing oxidative stress and enabling PDA-mediated mild photothermal therapy (PTT). Consequently, this trinity nanoplatform, which integrated dual-starvation therapy, amplified oxidative stress, and mild PTT, demonstrated outstanding therapeutic effects and a facile strategy.
Polydopamine Nanoparticle As A Multifunctional Nanocarrier for Combined Radiophotodynamic Therapy of Cancer

Abstract:Combination of different therapeutic strategies to treat cancer has attracted tremendous attention in recent years. Herein, the authors develop polydopamine (PDA) nanoparticles with polyethylene glycol (PEG) modification as a multifunctional nanocarrier for coloading photosensitizer chlorine6 (Ce6) and curcumin (Cur) for combined photodynamic therapy (PDT) and radiotherapy (RT) of cancer. PEGylated PDA nanoparticles (PDA‐PEG) exhibit well water soluble and biocompatible in different physiological solutions and cause no obvious toxicity to cancer cells. In this nanoparticle, the loaded Ce6 can trigger the generation of single oxygen under near‐infrared laser irradiation for PDT, while the loaded Cur can act as an excellent radiosensitizer under X‐ray irradiation for enhanced external RT. As demonstrated by in vitro and in vivo therapeutic efficiency, combined PDT and RT based on PDA‐PEG/Cur/Ce6 nanoparticles exhibits significant inhibition the growth of cancer cells, revealing perfect performance in cancer treatment. Therefore, the study not only presents a polymer‐based theranostic platform for cancer treatment but also demonstrates the potential applications of combined RT and PDT for the future clinic cancer therapy.
Polydopamine-drug conjugate nanocomposites based on ZIF-8 for targeted cancer photothermal-chemotherapy

Abstract:Stimuli-responsive prodrug-based nanoplatform with synergistic antitumor activity is of central importance to the development of promising nanomedicines for cancer therapy. Here, we describe a polydopamine-drug conjugate nanocomposite (ZP-PDA-DOX) with targeted cancer photothermal-chemotherapy (PTT-CT), which constructed by a gradual copolymerization of dopamine (DA) and pH-sensitive dopamine-derived prodrug (DA-DOX) into the porous channels of zeolite imidazolate frameworks-8 (ZIF-8), followed by PEGylation with amino-terminated folic acid-polyethylene glycol (NH2-PEG-FA) to acquire the high biocompatibility, specificity, and excellent tumor-targeting property. The incorporation of polydopamine strengthened the stability and dispersion of ZIF-8, and also conferred photothermal conversion effect. In the tumor acidic microenvironment, the acid-labile hydrazone linker of DA-DOX and ZIF-8 promptly degraded to release activated DOX. Moreover, the generated hyperthermia due to the high photothermal conversion efficiency of PDA component could accelerate drug release, and simultaneously thermally ablate tumor tissue to maximize the DOX-induced CT, which could also assist PTT to eradicate tumor cells. This study provides a promising strategy for targeted cancer PTT-CT with synergistic anti-tumor effect.
Amplified Photoacoustic Signal and Enhanced Photothermal Conversion of Polydopamine-Coated Gold Nanobipyramids for Phototheranostics and Synergistic Chemotherapy.

Abstract:Light-responsive nanoprobes were suffering from the threat of high-dose laser irradiation, and it was important for constructing new nanoprobes for safe and efficient photo-theranostics. Here, polydopamine (PDA)-coated gold nano-bipyramids (AuNBPs@PDA) were synthesized for amplified photoacoustic (PA) signal and enhanced photothermal conversion with low-dose laser irradiation and then doxorubicin (DOX)-loaded AuNBPs@PDA-DOX nanoprobes were constructed for PA imaging-guided synergistic photothermal therapy (PTT) and chemotherapy. The AuNBPs@PDA nanoparticles possessed higher photothermal conversion efficiency (42.07%) and stronger PA signal than those of AuNBP nanoparticles, and the AuNBPsg PDA-DOX nanoprobes showed dual-responsive DOX release of pH and photothermal stimulation. With low-dose laser irradiation (1.0 W/cm(2)) and low-concentration AuNBPs@PDA-DOX (60 mu g/mL), the 4T1 cell viability was reduced to about 5%, owing to the combination of PTT and chemotherapy, compared with 42.3% of single chemotherapy and 25.3% of single PTT. Moreover, by modeling 4T1 tumor-bearing nude mice, in vivo PA imaging was achieved and the tumors were completely inhibited, demonstrating the excellent synergistic effect of PTT/chemotherapy. Therefore, the developed AuNBPs@PDA-DOX nanoprobes can be used for phototheranostics and synergistic chemotherapy, achieving low-dose laser irradiation and high-efficient visualized theranostics.

Abstract:Photodynamic therapy (PDT) has significant advantages in treating primary tumors. However, the hypoxic tumor microenvironment hinders the generation of sufficient reactive oxygen species during PDT to effectively kill tumor cells, further greatly limiting the applications of PDT in cancer treatment. Herein, we reported a temperature/pH dual controlled drug delivery system LPC@PCN@PDA/Fe3+-AS1411 based on a porous coordination network (PCN (Mn)) coated with polydopamine (PDA) and modified with an aptamer AS1411. β-lapachone (LPC) was loaded inside the PCN (Mn) framework, and Fe3+ was attached to the surface of the PDA coating. These nanoparticles (NPs) exhibited excellent multimodal cancer therapeutic effects and tumor targeting ability with their photo- and chemodynamic properties. The therapeutic effect can be enhanced by the production of sufficient oxygen by the internal hydrogen peroxide, which improves the photodynamic effect of the photosensitizer PCN (Mn) and the chemotherapy effect of β-lapachone. Notably, the conversion of Fe2+ to Fe3+ in the tumor cells exerts the Fenton effect, which generates hydroxyl radicals that cause lipid peroxidation in tumor cells and induce apoptosis, thus enhancing the chemodynamic therapeutic effect. In vitro and in vivo experiments revealed that the NPs demonstrated specific tumor targeting, excellent inhibition effect on tumor growth, and biocompatibility. Together, our findings can help develop an intelligent multifunctional therapeutic nanoplatform for cancer therapy.
Targeted Polydopamine Nanoparticles Enable Photoacoustic Imaging Guided Chemo-Photothermal Synergistic Therapy of Tumor

Abstract:Near infrared light responsive nanoparticles can transfer the absorbed NIR optical energy into heat, offering a desirable platform for photoacoustic (PA) imaging guided photothermal therapy (PTT) of tumor. However, a key issue in exploiting this platform is to achieve optimal combination of PA imaging and PTT therapy in single nanoparticle. Here, we demonstrate that the biodegradable polydopamine nanoparticles (PDAs) are excellent PA imaging agent and highly efficient for PTT therapy, thus enabling the optimal combination of PA imaging and PTT therapy in single nanoparticle. Upon modification with argi nine-glycine-aspartic-cysteine acid (RGDC) peptide, PDA-RGDC can successfully target tumor site. Moreover, PDA-RGDC can load a chemotherapy drug, doxorubicin (DOX), whose release can be triggered by near-infrared (NIR) light and pH dual-stimuli. The in vitro and in vivo experiments show that this platform can deliver anti-cancer drugs to target cells, release them intracellular upon NIR irradiation, and effectively eliminate tumors through chemo-photothermal synergistic therapeutic effect. Our results offer a way to harness PDA-based theranostic agents to achieve PA imaging-guided cancer therapy. Statement of Significance NIR-light adsorbed nanoparticles combing the advantage of PAI and PIT (TNP-PAI/PTT) are expected to play a significant role in the dawning era of personalized medicine. However, the reported Au-, Ag-, Cu-, Co-, and other metal based, carbon-based TNP-PAI/PTT suffer from complex multicomponent system and poor biocompatibility and biodegradability. To overcome this limitation, biocompatible polydopamine nanoparticles (PDAs), structurally similar to naturally occurring melanin, were designed as both PA imaging contrast agent and a chemo-thermotherapy therapy agent for tumor. RGDC peptide modified PDAs can improve the PA imaging and VI I efficiency and specific targeted deliver doxorubicin (DOX) to perinuclear region of tumor cells. Abstract:Traditional anticancer treatments fail to significantly improve prognoses, and exploration of novel promising therapeutic modalities is urgently needed. In this study, multifunctional mesoporous polydopamine nanoparticles (Pt@MPDA/GOx/Fe3+ NPs) loaded with glucose oxidase (GOx), Fe ions and ultrasmall Pt nanoparticles (NPs) were prepared for magnetic resonance imaging (MRI)-guided photothermal therapy (PTT)-enhanced chemodynamic therapy (CDT). The oxidation of intratumoral glucose to H2O2 and GOx induced an H2O2-rich microenvironment, and then elevated H2O2 was catalyzed into highly cytotoxic·OH by Fe3+ via a Fenton reaction for CDT to induce cancer cell death efficiently. Notably, the heat generated by MPDA NPs under laser irradiation offered a moderate PTT to cascade the CDT effect. Moreover, Pt NPs can oxidize H2O2 to yield O2, which in turn accelerates the catalytic process of GOx to increase the efficiency of CDT. Meanwhile, in the high oxidation environment of tumor cells, Pt NPs are oxidized into Pt2+ to achieve a tumor chemotherapy effect. In addition, chelated Fe3+ endows the system with an MRI-visible function to monitor the treatment efficacy. In conclusion, this study provides a novel MRI-guided PTT-enhanced CDT synergistic nanomedicine platform for cancer therapy.
