A versatile biosensing platform coupling CRISPR–Cas12a and aptamers for detection of diverse analytes

Abstract:<p>Rapid and sensitive detection of various analytes is in high demand. Apart from its application in genome editing, CRISPR–Cas also shows promises in nucleic acid detection applications. To further exploit the potential of CRISPR–Cas for detection of diverse analytes, we present a versatile biosensing platform that couples the excellent affinity of aptamers for broad-range analytes with the collateral single-strand DNA cleavage activity of CRISPR–Cas12a. We demonstrated that the biosensors developed by this platform can be used to detect protein and small molecule in human serum with a complicated background, i.e., the tumor marker alpha fetoprotein and cocaine with the detection limits of 0.07 fM and 0.34 μM, respectively, highlighting the advantages of simplicity, sensitivity, short detection time, and low cost compared with the state-of-the-art biosensing approaches. Altogether, this biosensing platform with plug-and-play design show great potential in the detection of diverse analytes.</p>

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to develop a universal and sensitive biosensing platform for detecting various analytes. Currently available biosensors do not always meet the increasing detection requirements, especially in fields such as scientific research, environmental monitoring, food safety, and clinical diagnosis. To meet this challenge, the authors proposed a multifunctional biosensing platform that combines CRISPR - Cas12a and aptamers. This platform utilizes the non - specific single - stranded DNA cleavage activity of CRISPR - Cas12a and the high affinity of aptamers for a wide range of target molecules, thereby achieving rapid and sensitive detection of different analytes such as proteins and small molecules. Specifically, the paper mentions that by combining the collateral single - stranded DNA cleavage activity of CRISPR - Cas12a with the excellent affinity of aptamers, a multifunctional biosensing platform was constructed. This platform can be used to detect the tumor marker alpha - fetoprotein (AFP) and cocaine in the complex background of human serum, with detection limits of 0.07 fM and 0.34 μM respectively. This indicates that the platform has significant advantages in terms of simplicity, sensitivity, short detection time, and low cost, and shows great potential compared to the existing state - of - the - art biosensing technologies. In addition, the design of this platform is flexible and easy to be extended to the detection of other analytes, providing a new strategy for future biosensor development.
