Abstract:
Alzheimer's disease is a progressive neurodegenerative disorder characterized by extracellular amyloid beta peptides and neurofibrillary tangles consisted of intracellular hyperphosphorylated Tau in the hippocampus and cerebral cortex. Most of the mutations in key genes that code for amyloid precursor protein can lead to significant accumulation of these peptides in the brain and cause Alzheimer's disease. Moreover, some point mutations in amyloid precursor protein can cause familial Alzheimer's disease, such as Swedish mutation (KM670/671NL) and A673V mutation. However, recent studies have found that the A673T mutation in amyloid precursor protein gene can protect against Alzheimer's disease, even if it is located next to the Swedish mutation (KM670/671NL) and at the same site as A673V mutation, which are pathogenic. It makes us curious about the protective A673T mutation. Here, we summarize the most recent insights of A673T mutation, focus on their roles in protective mechanisms against Alzheimer's disease, and discuss their involvement in future treatment.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore the mechanism of action of the protective A673T mutation in Amyloid Precursor Protein (APP) in Alzheimer's Disease (AD) and its potential therapeutic significance. Specifically, the paper focuses on the following points: 1. **Protection mechanism of A673T mutation**: The paper discusses in detail how the A673T mutation provides protection against AD by altering the metabolic pathway of APP, reducing the production and aggregation of Aβ, and affecting the clearance of Aβ by microglia. 2. **Population frequency of A673T mutation**: The paper summarizes the frequency distribution of the A673T mutation in different countries and regions, especially the differences between Nordic countries and other non - Nordic populations. 3. **Structural and functional changes of A673T mutation**: Through structural prediction and experimental verification, the paper reveals how the A673T mutation affects the secondary and tertiary structures of APP, and further affects the efficiency of its cleavage by BACE1 enzyme and the production of Aβ. 4. **Relationship between A673T mutation and TGFβ2 signaling pathway**: The paper explores how the A673T mutation protects neurons from damage by interfering with the TGFβ2 - induced intracellular death signaling pathway. ### Specific content summary #### 1. **Protection mechanism of A673T mutation** - **Reducing Aβ production**: The A673T mutation reduces the production of C99 and Aβ by increasing the activity of α - secretase and decreasing the activity of β - secretase. - **Reducing Aβ aggregation**: The A673T mutation changes the aggregation tendency of Aβ, making it more difficult to form toxic oligomers. - **Affecting microglial clearance**: Aβ with the A673T mutation is more easily internalized and cleared by microglia, thereby reducing the accumulation of Aβ in the brain. #### 2. **Population frequency of A673T mutation** - **Nordic countries**: In Nordic countries such as Iceland, Denmark, Norway, and Sweden, the frequency of the A673T mutation is relatively high. - **Non - Nordic countries**: In non - Nordic populations such as Asians and North American whites, the A673T mutation is extremely rare. #### 3. **Structural and functional changes of A673T mutation** - **Secondary structure changes**: Through the Jpred 4 prediction tool, it was found that the A673T mutation causes significant changes in the secondary structure of APP, especially in the α - helix and β - sheet regions. - **Tertiary structure changes**: Through the RaptorX prediction tool, it was found that the A673T mutation causes changes in the tertiary structure of APP, especially conformational changes near the mutation region. #### 4. **Relationship between A673T mutation and TGFβ2 signaling pathway** - **Interfering with TGFβ2 signaling pathway**: The A673T mutation can block the TGFβ2 - induced intracellular death signaling pathway, reduce the phosphorylation of JNK, and thus protect neurons from damage. ### Conclusion The paper summarizes the protection mechanism of the A673T mutation in AD and points out that this mutation may provide new ideas for the treatment of AD. Future research can further explore the molecular mechanism of the A673T mutation to develop new drug targets and treatment methods.
