Abstract:Nanoscale carbon particles have emerged as versatile precursors for a new class of highly fluorescent nanomaterials that resemble semiconductor quantum dots. The surface-passivated fluorescent carbon nanoparticles, dubbed ‘carbon dots’, were already demonstrated for their potential optical bioimaging applications in vitro and in vivo. In this study, we conducted a systematic cytotoxicity evaluation on the carbon dots prepared by various combinations of precursor carbon nanoparticles and molecules for the particle surface functionalization. The results suggested that the cytotoxicity of carbon dots was dependent on the selection of surface passivation molecules. Those dots showing more significant cytotoxicity at higher concentrations were also evaluated for their effects on the fluorescence imaging of live cells. The implications of the results on the eventual use of carbon dots as cell imaging agents are discussed.

Abstract:Water dispersible carbon-dots (CDs) with tunable photoluminescence were synthesized via one-pot hydrothermal oxidation of nanodiamond and subsequently utilized for cell imaging applications. The CDs were characterized by the following techniques including transmission electron microscopy, atomic force microscopy, Fourier transform infrared spectroscopy, UV–Visible spectroscopy, and fluorescent spectroscopy. Results showed that the size of CDs is mainly distributed at 3–7nm. Many functional groups were introduced on the surface of CDs during hydrothermal oxidation procedure. Cell morphology observation and cell viability measurement demonstrated the good biocompatibility of CDs, suggesting their potential bioimaging applications.

Abstract:Fluorescent nanodiamond (FND) has received increasing research interests for bioimaging and drug delivery owing its outstanding physicochemical properties and low toxicity. However, synthesis of FND in large scale with desirable optical property is still a big challenge. In this work, we reported for the first time that non-fluorescent ND can be facilely converted into FND through surface modified with aggregation-induced emission fluorophore containing luminescent polymers through the microwave-assisted typical Diels-Alder (D-A) reaction. The final FND (named as ND-poly(Phe-PEGMA-IA)) displayed extensive fluorescence, high water dispersibility and desirable biocompatibility. Cell imaging results demonstrated ND-poly(Phe-PEGMA-IA) exhibited great stain performance. Furthermore, anticancer drug (cisplatin, DDP) can be effectively loaded onto ND-poly(Phe-PEGMA-IA) and controlled release it under acidic environment. All of these favorable properties endow ND-poly(Phe-PEGMA-IA) great potential for biological tracing and controlled drug delivery. Taken together, we have developed a microwave-assisted D-A reaction for preparation of multifunctional FND with great potential for biological imaging and controlled drug delivery applications.
Reduced background autofluorescence for cell imaging using nanodiamonds and lanthanide chelates

Abstract:Bio-imaging is a key technique in tracking and monitoring important biological processes and fundamental biomolecular interactions, however the interference of background autofluorescence with targeted fluorophores is problematic for many bio-imaging applications. This study reports on two novel methods for reducing interference with cellular autofluorescence for bio-imaging. The first method uses fluorescent nanodiamonds (FNDs), containing nitrogen vacancy centers. FNDs emit at near-infrared wavelengths typically higher than most cellular autofluorescence; and when appropriately functionalized, can be used for background-free imaging of targeted biomolecules. The second method uses europium-chelating tags with long fluorescence lifetimes. These europium-chelating tags enhance background-free imaging due to the short fluorescent lifetimes of cellular autofluorescence. In this study, we used both methods to target E-selectin, a transmembrane glycoprotein that is activated by inflammation, to demonstrate background-free fluorescent staining in fixed endothelial cells. Our findings indicate that both FND and Europium based staining can improve fluorescent bio-imaging capabilities by reducing competition with cellular autofluorescence. 30 nm nanodiamonds coated with the E-selectin antibody was found to enable the most sensitive detective of E-selectin in inflamed cells, with a 40-fold increase in intensity detected.

Abstract:Fluorescent nanodiamonds (fNDs) are unique carbon‐based nanomaterials due to their outstanding optical and magnetic properties. However, realization of the full potential of fNDs is often limited by their processability because fNDs aggregate strongly in both organic and aqueous solutions. Therefore, robust and potentially universal coating strategies are urgently needed to address these limitations. Derived from mussel foot proteins, the polymerization of l‐3,4‐dihydroxyphenylalanine (l‐DOPA) provides important surface functional groups including amines, carboxylic acid, alcohols, and conjugated Michael acceptors. Herein, l‐DOPA is polymerized on fNDs with a high control over the shell thickness. Photoluminescence and optically detected magnetic resonance studies reveal that the unique photophysical properties of fNDs are preserved after thin poly(l‐DOPA) film coating. Subsequently, conjugation of transferrin, a heme protein that provides efficient receptor‐specific cellular transport, improves the colloidal stability and cellular uptake of the poly(l‐DOPA)‐coated fNDs. The loading of FDA‐approved indocyanine green as a photothermal agent yields an integrated biohybrid material exhibiting an amplified photothermal effect in cells at very low energy intake (≈90 mW cm−2). A photothermal nanodiamond bioconjugate is synthesized stepwise mediated by a coating platform derived from l‐dihydroxyphenylalanine. The coating technology provides an avenue to functionalize fluorescent nanodiamonds with both proteins (transferrin) and small molecules (indocyanine green). The combination results in a cell‐permeable nanodiamond conjugate that exhibits a highly efficient and light/dark‐selective photothermal effect with an excitation energy of only 90 mW cm−2.

Abstract:Nanodiamonds (ND) are chemically inert and stable owing to their sp 3 covalent bonding structure, but their surface sp 2 graphitic carbons can be easily homogenized with diverse functional groups via oxidation, reduction, hydrogenation, amination, and halogenation. Further surface conjugation of NDs with hydrophilic ligands can boost their colloidal stability and functionality. In addition, NDs are non-toxic as they are made of carbons. They exhibit stable fluorescence without photobleaching. They also possess paramagnetic and ferromagnetic properties, making them suitable for use as a new type of fluorescence imaging (FI) and magnetic resonance imaging (MRI) probe. In this review, we focused on recently developed ND production methods, surface homogenization and functionalization methods, biocompatibilities, and biomedical imaging applications as FI and MRI probes. Finally, we discussed future perspectives.

Nanodiamond Landmarks for Subcellular Multimodal Optical and Electron Imaging

Abstract:There is a growing need for biolabels that can be used in both optical and electron microscopies, are non-cytotoxic, and do not photobleach. Such biolabels could enable targeted nanoscale imaging of sub-cellular structures, and help to establish correlations between conjugation-delivered biomolecules and function. Here we demonstrate a subcellular multi-modal imaging methodology that enables localization of inert particulate probes, consisting of nanodiamonds having fluorescent nitrogen-vacancy centers. These are functionalized to target specific structures, and are observable by both optical and electron microscopies. Nanodiamonds targeted to the nuclear pore complex are rapidly localized in electron-microscopy diffraction mode to enable "zooming-in" to regions of interest for detailed structural investigations. Optical microscopies reveal nanodiamonds for in-vitro tracking or uptake-confirmation. The approach is general, works down to the single nanodiamond level, and can leverage the unique capabilities of nanodiamonds, such as biocompatibility, sensitive magnetometry, and gene and drug delivery.

Luminescence of Nanodiamond Driven by Atomic Functionalization: Towards Novel Biomolecular Detection Principles

Abstract:High biocompatibility, variable size ranging from ~ 5 nm, stable luminescence from its color centers and simple carbon chemistry for biomolecule grafting make nanodiamond (ND) particles an attractive alternative to molecular dyes for drug-delivery. Here we present a novel method for remote monitoring of chemical processes in biological environment based on color changes from photo-luminescent NV centers in ND. We propose to drive the NV luminescence chemically, by alternating the surface electric field developed by interacting atoms and molecules with the diamond surface. Due to the small ND size, the developed electric field penetrates into the bulk of the nanoparticle and intermingles with the electronic NV states. This allows construction of optical chemo-biosensors operating in cells, visible in classical confocal microscopes. We demonstrate this phenomenon on oxidized and hydrogenated ND as well as single crystal diamond containing engineered NV centers. Hydrogenation of NDs leads to quenching of luminescence related to negatively charged (NV-) centers and by this way produces color shifts from NV- (636 nm) to neutral NV0 (575nm) luminescence. We model how the reduction of diamond size increases the magnitude of NV color shift phenomena.

Fe Doped Magnetic Nanodiamonds Made by Ion Implantation as Contrast Agent for MRI

Abstract:We report in this paper a new MRI contrast agent based on magnetic nanodiamonds fabricated by Fe ion implantation. The Fe atoms that are implanted into the nanodiamonds are not in direct contact with the outside world, enabling this new contrast agent to be free from cell toxicity. The image enhancement was shown clearly through T2 weighted images. The concentration dependence of the T2 relaxation time gives a relaxivity value that is about seven times that of the regular non-magnetic nanodiamonds. Cell viability study has also been performed. It was shown that they were nearly free from cytotoxicity independent of the particle concentration used. The imaging capability demonstrated here adds a new dimension to the medical application of nanodiamonds. In the future one will be able to combine this capability of magnetic nanodiamonds with other functions through surface modifications to perform drug delivery, targeted therapy, localized thermal treatment and diagnostic imaging at the same time.

Abstract:We directly observe non-fluorescent nanodiamonds in living cells using transient absorption microscopy. This label-free technology provides a novel modality to study the dynamic behavior of nanodiamonds inside the cells with intrinsic three-dimensional imaging capability. We apply this method to capture the cellular uptake of nanodiamonds under various conditions, confirming the endocytosis mechanism.
Bioorthogonal Fluorescent Nanodiamonds for Continuous Long-Term Imaging and Tracking of Membrane Proteins.

Abstract:Real-time tracking of membrane proteins is essential to gain an in-depth understanding of their dynamics on the cell surface. However, conventional fluorescence imaging with molecular probes like organic dyes and fluorescent proteins often suffers from photobleaching of the fluorophores, thus hindering their use for continuous long-term observations. With the availability of fluorescent nanodiamonds (FNDs), which have superb biocompatibility and excellent photostability, it is now possible to conduct the imaging in both short and long terms with high temporal and spatial resolution. To realize the concept, we have developed a facile method (e.g., one-pot preparation) to produce alkyne-functionalized hyperbranched-polyglycerol-coated FNDs for bioorthogonal labeling of azide-modified membrane proteins and azide-modified antibodies of membrane proteins. The high specificity of this labeling method has allowed us to continuously monitor the movements of the proteins of interest (such as integrin α5) on/in living cells over 2 h. The results open a new horizon for live cell imaging with functional nanoparticles and fluorescence microscopy.

One-Pot Synthesis of Highly Dispersible Fluorescent Nanodiamonds for Bioconjugation

Abstract:Fluorescent nanodiamonds (FNDs) have been attracting much attention as promising therapeutic agents and probes for bioimaging and nanosensing. For their biological applications, several hydrophilizing methods to enhance FND colloidal stability have been developed to suppress their aggregation and the nonspecific adsorption to biomolecules in complex biomedical environments. However, these methods involve several complicated synthetic and purification steps, which prohibit the use of FNDs for bioapplications by biologists. In this study, we describe a simple one-pot FND hydrophilization method that comprises coating of the surface of the nanoparticles with COOH-terminated hyperbranched polyglycerol (HPG-COOH). HPG-COOH-coated FNDs (FND-HPG-COOHs) were found to exhibit excellent dispersibility under physiological conditions despite the thinness of the 5 nm HPG-COOH layer. Biotinylated FND-HPG-COOHs specifically captured avidin molecules in the absence of nonspecific protein adsorption. Moreover, we demonstrated that FND-HPG-COOHs conjugated with antibodies can be used to selectively target integrins in fixed HeLa cells. In addition, intracellular temperature changes were measured via optically detected magnetic resonance using FND-HPG-COOHs conjugated with mitochondrial localization signal peptides. Our one-pot synthetic method will encourage the broad use of FNDs among molecular and cellular biologists and pave the way for extensive biological and biomedical applications of FNDs.
Targeted Nanodiamonds for Identification of Subcellular Protein Assemblies in Mammalian Cells

Abstract:Transmission electron microscopy (TEM) can be used to successfully determine the structures of proteins. However, such studies are typically done ex situ after extraction of the protein from the cellular environment. Here we describe an application for nanodiamonds as targeted intensity contrast labels in biological TEM, using the nuclear pore complex (NPC) as a model macroassembly. We demonstrate that delivery of antibody-conjugated nanodiamonds to live mammalian cells using maltotriose-conjugated polypropylenimine dendrimers results in efficient localization of nanodiamonds to the intended cellular target. We further identify signatures of nanodiamonds under TEM that allow for unambiguous identification of individual nanodiamonds from a resin-embedded, OsO4-stained environment. This is the first demonstration of nanodiamonds as labels for nanoscale TEM-based identification of subcellular protein assemblies. These results, combined with the unique fluorescence properties and biocompatibility of nanodiamonds, represent an important step toward the use of nanodiamonds as markers for correlated optical/electron bioimaging.

Abstract:Due to its small particle size, high specific surface area, functional capability and long-term stable fluorescence, nanodiamond (ND) has been widely used in biomedical applications including fluorescent probes, drug delivery and biological imaging. Nevertheless, the metabolism of ND and its derivatives is not yet well understood for diverse biomedical applications. In this study, to evaluate the cytotoxicity and biodistribution of the ND derivative, pure ND was functionalized with ethylenediamine (EDA) as a linker (ND-EDA) and then covalently labeled with an organic fluorescent dye called calcein. The modified ND is referred to as ND-EDA-calcein. Even at the maximum dose of 0.1 mg·mL −1 , the in vitro results for ND-EDA-calcein against HepG2 and LO2 cell lines showed negligible cytotoxicity, which was in line with the in vivo observations. In addition, it was found that following an intraperitoneal injection, ND-EDA-calcein could be accumulated mostly in the liver and kidney of the mice but not in the spleen. It is interesting to note that after 5 days following intraperitoneal injection, mice can totally metabolize the elimination of ND-EDA-calcein outside of their bodies. Therefore, our findings suggested that NDs can be considered for various nanomedicine applications including drug delivery and biomarkers.

Abstract:There is a renewed interest in nanodiamonds and their applications in biology and medicine, especially in bioimaging and photothermal therapy. This is due to their small size, chemical inertness and unique photo-properties such as bright and robust fluorescence, resistant to photobleaching and photothermal response under near infrared (NIR) irradiation. However, the biggest challenge limiting the wide-spread use of nanodiamonds is the high-energy consuming, dangerous and sophisticated synthetic methods currently adopted by industry named higher temperature high pressure approach, and detonation method. Despite over a decade of research towards the development of new synthetic approaches, most of the methods developed to date require sophisticated instrumentations and have high energy demand. To circumvent the reliance on high energy demanding sophisticated experimental setups, here we present a simple synthetic approach using solar energy as a sustainable sole energy source. Using low-grade coal as carbon precursor, we used high power magnifying glasses to concentrate and focus sunlight to induce synthesis of nanodiamonds. The synthesized nanodiamonds exhibit similar physicochemical and photo-properties as nanodiamonds synthesized using other synthetic approaches.In vitrostudies using macrophage Raw 264.7 cells demonstrated rapid uptake and bright fluorescence of the synthesized nanodiamonds with superior biocompatibility (≥95% cell viability). The synthesized nanodiamonds also exhibited dose dependent photothermal response under NIR irradiation.
Nanoparticles and nanocomposites for fluorescence sensing and imaging

Abstract:The assortment of fluorescence reporters is changing dramatically. Traditionally explored intrinsic fluorescence of biological macromolecules and cellular pigments and of externally introduced organic dyes are presently in strong competition with new nanomaterials. Among them are conjugated polymers, semiconductor nanocrystals (quantum dots), up-converting nanocrystals, magic-size clusters of silver and gold, nanodiamonds and carbon dots. They demonstrate diverse photophysical behavior and allow one to obtain diverse information when used in analytical tools or when they form images in biological systems. Based on them, functional nanocomposites displaying a variety of useful features, thus extending dramatically the information content of output data, can be constructed. We describe their properties and compare them with those of small-molecular emitters, such as organic dyes. With their aid, one can modulate over a wide range the wavelengths of excitation and emission, the lifetimes and anisotropies and design the systems with 'superenhancement' and 'superquenching'. Such unlimited possibilities are offered by combining different types of luminophores based on electronic conjugation, plasmonic effects or excited-state resonance energy transfer. This tutorial review provides a comparative analysis of the properties of new nanoscale materials and of their hybrid nanocomposites for applications in fluorescence sensing and imaging.
High-resolution two-photon fluorescence microscope imaging of nanodiamonds containing NV color centers

Abstract:Fluorescent nanodiamonds containing nitrogen-vacancy (NV) color centers play an important role in nanoscale sensing for their unique properties, including extraordinary photon stability, bio-compatibility, and chemical inertness at room temperature. Two-photon fluorescence imaging of fluorescent nanodiamonds (FNDs) has been studied with a picosecond pulsed laser operating at 1032 nm. We obtained an imaging resolution of 318 nm at nearly a third of the excitation wavelength with 70 nm FNDs. In this case, the resolution was improved by 2.2-fold, compared with the diffraction-limited resolution of the near-infrared (NIR) single-photon confocal microscopic imaging. The axial resolution of the imaging is about 750 nm, which is about 1.4 times higher than the Rayleigh length under the same excitation. The fluorescence spectra of visible and near-infrared excitation of FNDs are similar, suggesting that the energy levels involved in the two processes are identical. With the unique advantages of high spatial resolution, high signal-to-noise ratio (SNR), and infrared window matching, the study of two-photon excitation microscope imaging of FNDs is conducive to its broader application in biology, physics, and materials sciences.

Effects of Silver Nanoparticles on Raman Spectrum and Fluorescence Enhancement of Nano-Diamond

Abstract:The nano-diamond has been a hot topic in the field of nano-science and nanotechnology for its optical properties. Much effort has been devoted to improving the fluorescence and Raman scattering intensity of nitrogen-vacancy (NV) center in nano-diamond by using plasmon resonance effect in sensing area. A combination of Ag nanoparticle and diamond can not only take advantage of the stability and biocompatibility of diamond, but also enhance the local electric field around NV center through the Ag nanoparticles, thereby speeding up the radiation of the fluorescent near the surface of the substrate, improving the strength and stability of the fluorescence, and greatly broadening the application areas of Raman spectroscopy. In this paper, we mix the nano-diamonds with Ag nanoparticles to improve the fluorescence and Raman scattering intensity on the basis of the localized surface plasmon resonance effect. The influences of Ag mass concentration on the Raman spectrum and fluorescence intensity are investigated. The results show that when the concentration of nano-Ag nanoparticles reaches up to 5 wt%, the light intensity becomes saturated, but the concentration further increases up to a value more than 7 wt% the light intensity begins to decline. Then the corresponding radiative transition rate and the fluorescence quantum efficiency are investigated, and based on these researches, influences and mechanism of surface plasmon resonance (SPR) enhancement are discussed thoroughly. We deduced that the fluorescence enhancement is mainly due to the enhanced surface plasmon field caused by transfer of surface plasmon resonance energy and the energy transfer between surface plasmon and excited state of NV centers. When the concentration of Ag nanoparticles reaches an appropriate value, a suitable distance between metal nanoparticles and diamond is obtained, thereby ensuring the strong local electric field forming on the metal surface, accelerating the emitting photons of diamond in the excited state, and also suppressing the transfer of non-radiative energy, eventually leading to the increase of diamond fluorescence emission intensity.

Abstract:Modified fluorescent nanoparticles continue to emerge as promising candidates for drug delivery, bioimaging, and labeling tools for various biomedical applications. The ability of nanomaterials to fluorescently label cells allow for the enhanced detection and understanding of diseases. Silica nanoparticles have a variety of unique properties that can be harnessed for many different applications, causing their increased popularity. In combination with an organic dye, fluorescent nanoparticles demonstrate a vast range of advantageous properties including long photostability, surface modification, and signal amplification, thus allowing ease of manipulation to best suit bioimaging purposes. In this study, the Stöber method with tetraethyl orthosilicate (TEOS) and a fluorescent dye sulfo-Cy5-amine was used to synthesize fluorescent silica nanoparticles. The fluorescence spectra, zeta potential, quantum yield, cytotoxicity, and photostability were evaluated. The increased intracellular uptake and photostability of the dye-silica nanoparticles show their potential for bioimaging.
