Abstract:Waterborne pathogens, in particular, emerging antibiotic-resistant bacteria (ARB), can cause serious infectious diseases, posing a huge threat to public health. However, existing water disinfection technologies are often not only energy and chemical intensive but also inefficient at eliminating antibiotic resistance genes (ARGs). Here we show a sequential electrochemical process of ‘chemical (H2O2 pre-treatment)–physical (nanotip electroporation)–chemical (•OH injection)’ that is highly effective in inactivating ARB and removing ARGs. The bacteria are first pre-treated by H2O2 generated via two-electron water oxidation in the SnO2−x/TiO2 anode region to reduce the defences of bacterial outer walls against electroporation. Then, the ‘softened’ and ‘weakened’ bacteria are easily punctured by electroporation in the Pd-Au/TiO2 cathode region and by synchronously injected abundant •OH generated via three-electron oxygen reduction. Bacterial inclusions, including nuclear body and cytoplasm, are effectively decomposed by •OH oxidation, resulting in the destruction of the entire cell structure from the inside out. This bactericidal mechanism of synergetic physical damage and chemical oxidation inactivated >99.9999% of ARB and removed ~99% of ARGs at short retention time (~16 s), high flux (~4.5 m3 h−1 m−2) and low energy consumption (~42.4 Wh m−3) over 15 days of continuous operation. This approach may act as an alternative to meet the urgent need for efficient and residue-free water disinfection. Zhang et al. present a sequential electrochemical process that enables synergetic physical damage and chemical oxidation of antibiotic-resistant bacteria and antibiotic resistance genes, providing an efficient and residue-free water disinfection approach.

Abstract:Electrochemical disinfection (ED) is effective in removal of antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARGs) during wastewater treatment. However, the inner relationship of antibiotic-resistant phenotypes with their genotypes of ARB is still unclear in the ED process. This study explored the disinfection mechanism at the microcosmic level using four Escherichia coli (E. coli) strains with different antibiotic-resistant phenotypes. Results showed that bacteria with multiple antibiotic resistance tended to have stronger viability in disinfection compared with those resistant to fewer antibiotics. Interestingly, antibiotic-resistant phenotypes changed little in the disinfection process, while genotypes closely responded to bacterial resistance of different E. coli. In detail, beta-lactamase resistance genes played a leading role in the cross-resistance between antibiotics and electrochemical oxidation. They were proved to reduce oxidative destruction by increasing the strength of the cell wall and membrane. In addition, diaminopyrimidine and macrolide-lincosamide-streptogramin B resistance genes are also closely related to the bacterial resistance to ED. Combined with the quantitative determination of ARGs, the abundance of IntI1 and aminoglycoside resistance genes can be regarded as indicators for the risk of cross-resistance to antibiotics and electrochemical oxidation. This study provided important references for in depth understanding and accurate control of antibiotic resistance with advanced oxidation processes.

Abstract:Antibiotic resistance genes (ARGs) are a new environmental contaminant that poses a major hazard to humans and the environment. This research discusses the methods and drawbacks of two ARG removal approaches, constructed wetlands (CWs) and photocatalysis. CWs primarily rely on the synergistic effects of substrate adsorption, plant uptake, and microbial processes to remove ARGs. The removal of ARGs can be influenced by wetland plants, substrate type, wetland type, and hydraulic conditions. The absolute abundance of ARGs in effluent decreased, but their relative abundance increased. Photocatalysis deactivates ARGs predominantly through reactive oxygen species, with removal effectiveness determined by catalyst type, radiation type, and radiation intensity. The drawback is that it exposes intracellular resistance genes, perhaps increasing the risk of ARG spread. To address the current shortcomings, this paper proposes the feasibility of combining a constructed wetland with photocatalysis technology, which provides a novel strategy for ARG removal.

Abstract:The rising challenge of antibiotic resistance, accelerated by mobile genetic elements carrying antibiotic resistance genes, necessitates effective and environmentally friendly control strategies. Here we present an approach employing Shewanella oneidensis, transformed into an efficient whole-cell DNA scavenger by expressing a highly active nuclease. This approach aimed at enzymatically eliminating cell-free mobile genetic elements in wastewater treatment plants. The DNA scavenger demonstrated high efficiency and ultrafast degradation with a model multiple antibiotic resistance plasmid. Experiments across various dosages and hydraulic retention times showed substantial antibiotic resistance gene reduction, even at minimal dosages. Effectiveness was confirmed in practical scenarios involving return activated sludge and secondary clarifier effluent, where a low dose of 0.08 U ml−1 achieved over 99.92% removal in 4 h and almost complete inactivation in 6 h. Simulations showed consistent, high efficiency across various wastewater treatment plant reactors. These findings establish enzymatic scavenging as a new strategy for managing the antibiotic resistance spread. The development of antibiotic resistance in wastewater is accelerated by the wide presence of mobile genetic elements. An engineered DNA scavenger based on Shewanella oneidensis is shown to be an efficient tool for eliminating mobile genetic elements in wastewater treatment plants, thus decreasing the rise and dissemination of antibiotic resistance.

Abstract:Antibiotic resistance genes (ARGs) in the environment have gradually engendered extensive concern recently due to the harm caused to environment and human beings.In this study,two sulfonamides resistant bacteria isolated from secondary effluents of a municipal wastewater treatment plant were used to compare the efficacy of three different disinfection processes (chlorination,ultraviolet and ozone) in reducing sulfonamides resistance pollution using serial dilution plate counting method and qPCR.The behavior characteristics of sulfonamides resistance genes during the disinfection process were elucidated using disinfection + DNase Ⅰ treatment.The results showed that all three disinfection techniques could reduce the concentration of target bacteria,but the abundance of sulfonamides resistance genes was still at a high level.Furthermore,chlorination disinfection might reduce sulfonamides resistance genes by reducing microbial biomass.Ultraviolet disinfection could damage genes directly.However,during ozone disinfection,sulfonamides resistance genes would escape to environment,and its potential environmental risks could not be ignored.
Mechanism and potential risk of antibiotic resistant bacteria carrying last resort antibiotic resistance genes under electrochemical treatment

Abstract:The significant rise in the number of antibiotic resistance genes (ARGs) that resulted from our abuse of antibiotics could do severe harm to public health as well as to the environment. We investigated removal efficiency and removal mechanism of electrochemical (EC) treatment based on 6 different bacteria isolated from hospital wastewater carrying 3 last resort ARGs including NDM-1, mcr-1 and tetX respectively. We found that the removal efficiency of ARGs increased with the increase of both voltage and electrolysis time while the maximum removal efficiency can reach 90%. The optimal treatment voltage and treatment time were 3 V and 120 min, respectively. Temperature, pH and other factors had little influence on the EC treatment process. The mechanism of EC treatment was explored from the macroscopic and microscopic levels by scanning electron microscopy (SEM) and flow cytometry. Our results showed that EC treatment significantly changed the permeability of cell membrane and caused cells successively experience early cell apoptosis, late cell apoptosis and cell necrosis. Moreover, compared with traditional disinfection methods, EC treatment had less potential risks. The conjugative transfer frequencies of cells were significantly reduced after treatment. Less than 1% of bacteria entered the viable but nonculturable (VBNC) state and less than 5% of intracellular ARGs (iARGs) turned into extracellular ARGs (eARGs). Our findings provide new insights into as well as important reference for future electrochemical treatment in removing ARB from hospital wastewater.

Abstract:Conventional water disinfection methods such as chlorination typically involve the generation of harmful disinfection byproducts and intensive chemical consumption. Emerging electroporation disinfection techniques using nanowire-enhanced local electric fields inactivate microbes by damaging their outer structures without byproduct formation or chemical dosing. However, this physical-based method suffers from a limited inactivation efficiency under high water flux due to an insufficient contact time. Herein, we integrate electrochlorination with nanowire-enhanced electroporation to achieve a synergistic flow through process for efficient water disinfection targeting bacteria and viruses. Electroporation at the cathode induces sub-lethal damages on the microbial outer structures. Subsequently, electrogenerated active chlorine at the anode aggravates these electroporation-induced injuries to the level of lethal damage. This sequential flow-through disinfection system achieves complete disinfection (> 6.0-log) under a very high water flux of 2.4 x 10(4) L/(m(2) h) with an applied voltage of 2.0 V. This disinfection efficiency is 8 times faster than that of electroporation alone. Further, the specific energy consumption for the disinfection by this novel process is extremely low (8 x 10(-4) kW h/m(3)). Our results demonstrate a promising method for rapid and energy-efficient water disinfection by coupling electroporation with electrochlorination to meet vital needs for pathogen elimination.

Abstract:Developed control of chemical disinfection techniques is beginning to be pro-gressively significant in order to equilibrate under-treatment (minimal pathogen demobilization) and over-treatment (immoderate consumption of disinfectant and disinfection by-products generation) that way giving great ecological and economic advantages. This work reviews the most recent and pertinent re-searches in this field of eliminating Antibiotic-resistant Bacteria (ARB) carrying genes (ARGs) during wastewater treatment especially disinfection. Traditional disinfection techniques may not be efficient in demobilizing ARB and the simultaneou s liberation of ARB and antibiotics at sub-lethal concentrations into municipal wastewater treatment plant effluent may promote the develop-ment of resistance among bacteria in receiving water. The pathway of the in-fluences of diverse disinfection techniques in water and wastewater (chlorina-tion, UV irradiation, Fenton reaction, ozonation, and photocatalytic oxidation) deserves more attention. The impacts of constructed wetlands and nanotech-nology on ARB and ARG have to be more explored. As the best available technology, membranes processes should be widely adopted through the world for removing ARB and ARGs from the perspective of reusing treated wastewater as drinking water. These safe barriers against pollutants diffusion in nature merit more technical and economic expansion for their larger industrial application especially in developing countries.

Abstract:More than 10% of the people in the world still suffer from inadequate access to clean water. Traditional water disinfection methods (e.g., chlorination and ultraviolet radiation) include concerns about the formation of carcinogenic disinfection byproducts (DBPs), pathogen reactivation, and/or excessive energy consumption. Recently, a nanowire-assisted electroporation-disinfection method was introduced as an alternative. Here, we develop a new copper oxide nanowire (CuONW)-modified three-dimensional copper foam electrode using a facile thermal oxidation approach. An electroporation-disinfection cell (EDC) equipped with two such electrodes has achieved superior disinfection performance (>7 log removal and no detectable bacteria in the effluent). The disinfection mechanism of electroporation guarantees an exceedingly low operation voltage (1 V) and level of energy consumption (25 J L(-1)) with a short contact time (7 s). The low operation voltage avoids chlorine generation and thus reduces the potential of DBP formation. Because of irreversible electroporation damage on cell membranes, no regrowth and/or reactivation of bacteria occurs during storage after EDC treatment. Water disinfection using EDCs has great potential for practical applications.

Abstract:An increasing amount of attention has been given to antimicrobial resistance in the environment because of its substantial threat to human health. The effluent from municipal wastewater treatment plants has been regarded as one of the important sources for the spread of antibiotic resistance genes (ARGs). However, conventional disinfection techniques fail to effectively remove ARGs from effluents. In this work, in situ synthesized hydrated manganese oxide (HMO) coupled with permanganate was applied for the first time in ARG removal from the effluent of wastewater treatment plants. The results show that five ARGs (sulI, sulII, tetQ, tetO, and tetW) as well as the intI1 and 16S rRNA genes had removal efficiencies of 2.46-4.23 logs, which were significantly higher than those obtained by using these reagents individually. This implied that there was a synergistic effect between permanganate and HMO toward the removal of ARGs. Moreover, the contributions of HMO coagulation and permanganate oxidation to ARG removal were semiquantitatively studied, which demonstrated that destruction of the microbial cells by oxidation and removal of the extracellular ARGs released by coagulation were the two main processes in this system. The results of this study provide an alternative method for ARG removal from the effluent of wastewater treatment plants with high efficiencies to control the spreading of ARGs.
Localized heating coupling with radical oxidation eliminating antibiotic resistance genes (ARGs) in interfacial photothermal Fenton-like disinfection process

Abstract:Conventional oxidative disinfection processes are inefficient in eliminating intracellular antibiotic resistance genes (iARGs) due to the barrier of the cell membrane and the competitive reaction of cellular constituents within antibiotic-resistant bacteria (ARB), resulting in the widespread prevalence of ARGs in recycled water. This study presented the first application of localized heating coupling with advanced oxidation to destroy the resistant Escherichia coli cells and improved subsequent iARGs (blaTEM-1) degradation in a novel photothermal Fenton-like disinfection process. The Fe-Mn@CNT microfiltration membrane, comprising carbon nanotubes wrapped with Fe and Mn nanoparticles (Fe-Mn@CNT), was employed as a nanomaterial for photothermal conversion and H2O2 activation. The highly efficient absorption of full-spectrum photons by CNTs enabled the Fe-Mn@CNT membrane to concentrate light to generate localized intense heat, resulting in the destruction of ARB nearby, and the subsequent release of iARGs. Interfacial heat favored Fe-Mn-induced H2O2 activation, leading to the production of more ·OH, which in turn promoted the oxidation for ARG degradation and ARB cell damage. The results of the acetylcysteine quenching experiments indicated that interfacial heating and radical oxidation-induced accumulation of intracellular reactive oxygen species contributed to the elimination of about 1-log iARGs through direct attack. The integrity of the cell membrane, the morphology of ARB and the variation of i/e ARG copy numbers were observed to reveal that the introduction of interfacial heating aggravated the cell lysis and accelerated the iARGs release, resulting in the inactivation of 7.27-log ARB and the elimination of 4.64-log iARGs and 2.23-log eARGs. Localized heating coupling with ·OH oxidation achieved a 143 % increase in iARGs removal compared to the conventional Fenton-like oxidation. The interfacial photothermal Fenton-like disinfection process exhibited remarkable material stability, robust disinfection performance, and effective suppression of horizontal gene transfer, underscoring its immense potential to mitigate the risk of ARG dissemination in reclaimed water systems.

Abstract:Wastewater reuse may serve as an important solution to the emerging issue of water scarcity. However, the potential public and ecological risks of antimicrobial resistance (AMR) may hinder the wide application of treated wastewater reclamation. Wastewater treatment plants have been recognized as hotspots of antibiotic resistance genes (ARGs) and key nodes of their spreading. Owing to their characterization of bioactive and penetrating ability from the treatment processes, it is important to design the tertiary treatment to enhance removal efficiencies of intracellular- and extracellular ARGs. Herein, we critically reviewed the available technologies classified into physicochemical, eco-biological, and integrated processes for reclaimed water generation and ARG removal from recently published articles. We summarized the performance, mechanisms, and bottlenecks of each process. For future prospects, we proposed a framework for multimode reuse to achieve the goal of sustainable social water cycle with low risk posed by AMR.

Abstract:In this study, the simultaneous removal mechanism of antibiotics and antibiotic resistance genes (ARGs) was investigated using the novel ultraviolet/peracetic acid (UV/PAA) combination disinfection process and conventional disinfection processes were also applied for comparison. The results showed that UV/PAA disinfection with a high UV dosage (UV/PAA-H) was most effective for the removal of tetracyclines, quinolones, macrolides and β-lactams; their average removal efficiencies ranged from 25.7% to 100%, while NaClO disinfection was effective for the removal of sulfonamides (∼81.6%). The majority of ARGs were well removed after the UV/PAA-H disinfection, while specific genes including tetB, tetC, ermA and bla<sub>TEM</sub> significantly increased after NaClO disinfection. In addition, β-lactam resistance genes (-35.9%) and macrolides resistance genes (-12.0%) remarkably augmented after UV/NaClO disinfection. The highly reactive oxidation species generated from UV/PAA process including hydroxyl radicals (•OH) and carbon-centered organic radicals (R-C•), were responsible for the elimination of antibiotics and ARGs. Correlation analysis showed that tetracycline, sulfonamide and macrolide antibiotics removal showed a positive correlation with the corresponding ARGs, and a low dose of antibiotic residues played an important role in the distribution of ARGs. Metagenomic sequencing analysis showed that UV/PAA disinfection could not only greatly decrease the abundance of resistant bacteria but also downregulate the expression of key functional genes involved in ARGs propagation and inhibit the signal transduction of the host bacteria, underlying that its removal mechanism was quite different from that of NaClO-based disinfection processes. Our study provides valuable information for understanding the simultaneous removal mechanism of antibiotics and ARGs in wastewater during the disinfection processes, especially for the novel UV/PAA combination process.

Abstract:Constructed wetland (CW) is considered a promising technology for the removal of emerging contaminants. However, its removal performance for antibiotic resistance genes (ARGs) is not efficient and influence of virulence factor genes (VFGs) have not been elucidated. Here, removal of intracellular and extracellular ARGs as well as VFGs by electricity-intensified CWs was comprehensively evaluated. The two electrolysis-intensified CWs can improve the removal of intracellular ARGs and MGEs to 0.96- and 0.85-logs, respectively. But cell-free extracellular ARGs (CF-eARGs) were significantly enriched with 1.8-logs in the electrolysis-intensified CW. Interestingly, adding Fe-C microelectrolysis to the electrolysis-intensified CW is conducive to the reduction of CF-eARGs. However, the detected number and relative abundances of intracellular and extracellular VFGs were increased in all of the three CWs. The biofilms attached onto the substrates and rhizosphere are also hotspots of both intracellular and particle-associated extracellular ARGs and VFGs. Structural equation models and correlation analysis indicated that ARGs and VFGs were significantly cooccurred, suggesting that VFGs may affect the dynamics of ARGs. The phenotypes of VFGs, such as biofilm, may act as protective matrix for ARGs, hindering the removal of resistance genes. Our results provide novel insights into the ecological remediation technologies to enhance the removal of ARGs.

Abstract:The electro-peroxone (EP) process is an electricity-based oxidation process enabled by electrochemically generating hydrogen peroxide (H2O2) from cathodic oxygen (O2) reduction during ozonation. In this study, the removal of antibiotic resistant bacteria (ARB) and plasmid-encoded antibiotic resistance genes (ARGs) during groundwater treatment by ozonation alone and the EP process was compared. Owing to the H2O2-promoted ozone (O3) conversion to hydroxyl radicals (•OH), higher •OH exposures, but lower O3 exposures were obtained during the EP process than ozonation alone. This opposite change of O3 and •OH exposures decreases the efficiency of ARB inactivation and ARG degradation moderately during the EP process compared with ozonation alone. These results suggest that regarding ARB inactivation and ARG degradation, the reduction of O3 exposures may not be fully counterbalanced by the rise of •OH exposures when changing ozonation to the EP process. However, due to the rise of •OH exposure, plasmid DNA was more effectively cleaved to shorter fragments during the EP process than ozonation alone, which may decrease the risks of natural transformation of ARGs. These findings highlight that the influence of the EP process on ARB and ARG inactivation needs to be considered when implementing this process in water treatment.
Performance evaluation for the inactivation of multidrug-resistant bacteria in wastewater effluent by different disinfection technologies

Abstract:The escalating presence of antibiotic-resistant bacteria (ARB) in aquatic ecosystems underscores the critical role of wastewater treatment plants (WWTPs) in mitigating antibiotic resistance. Disinfection is the final, pivotal step in WWTPs, and it is essential to control the dissemination of ARB before water discharge. This study utilized both phenotypic analysis and transcriptome (RNA-seq) approach to investigate the efficiency and mechanisms of disinfection using chlorination, ultraviolet (UV), and peracetic acid (PAA) on multidrug-resistant bacteria (MRB). Our results demonstrated that the use of 100 mg min L −1 of chlorine, 8.19 mJ cm −2 of UV irradiation or 50 min mg L −1 of PAA significantly reduced the abundance of MRB. Intriguingly, RNA-seq clarified distinct mechanisms of chlorination and UV disinfection. UV radiation triggered the SOS response to cope with DNA damage, induced the expression of multi-drug resistance genes by increasing the expression of efflux pump transporters. UV radiation also promoted the absorption of iron through chelation and transportation to participate in various cell life processes. Chlorination, on the other hand, significantly up-regulated osmotic response elements, including the synthesis of glycine betaine, iron-sulfur clusters, and related transporters. Both chlorination and UV significantly down-regulated key metabolic pathways ( P < 0.05), inhibiting the process of amino acid synthesis and energy metabolism. Imbalance in energy homeostasis was the most important factor leading to cytotoxicity. These results provide useful insights into optimizing the wastewater disinfection process in order to prevent the dissemination of ARB in aquatic environment.

Abstract:Water disinfection is the most important process to control the risks caused by the microbes within water. Conventional water disinfection processes, including chlorination, ozonation, and ultraviolet, show intrinsic disadvantages such as low disinfection efficiency, harmful byproduct formation, and microbial reactivation/regrowth during extensive applications. In order to overcome these limitations, electrochemical membrane technology is explored as an emerging efficient water disinfection approach, with novel disinfection mechanisms achieved by the specific physical, electrochemical, and biological characteristics of electrochemical membranes. This chapter gives an overview into the main mechanisms involved in the disinfection process by electrochemical membranes, as well as the development progress of the reactor configuration designs. Different water disinfection mechanisms and reactor configurations are comprehensively compared with the aspect of disinfection efficiency and energy consumption. Furthermore, the limitations and future perspectives of electrochemical membrane technology for water disinfection are also proposed.

Abstract:Antibiotic-resistant bacteria (ARB) and their resistance genes (ARGs), which are commonly detected in waters, are emerging environmental contaminants with strong pathogenicity. Simultaneous removal of ARB and ARGs in water by mesoporous plasma was explored in this study, focusing on evaluating the impacts of common inorganic ions on ARB inactivation, antibiotic resistance reduction, and change in ARGs profiles. The antibiotic-resistant Escherichia coli (AR E. coli) was selected as the model ARB. The experimental results demonstrated that the presence of NO3 −, Cu2+, and Fe2+ all promoted AR E. coli inactivation and ARGs elimination, reduced its antibiotic resistance, and also inhibited horizontal gene transfer of ARGs; whereas CO3 2− inhibited these processes, and SO4 2− did not have distinct effect. During the plasma treatment process, Fe2+ significant enhancement on OH formation via reactions with H2O, the highest performance was observed in the presence of Fe2+. The membrane structure, disruption of biological processes, dissolution of intracellular components, and DNA. These finding shed light on the potential effects of inorganic ions on ARB and ARGs elimination induced by the mesoporous plasma.

Abstract:Global consumption and discharge of antibiotics have led to the rapid development and spread of bacterial antibiotic resistance. Among treatment strategies, electrochemical advanced oxidation processes (EAOPs) arc gaining popularity for treating water/wastewater containing antibiotics due to their high efficiency and easiness of operation. In this review, we summarize various forms of EAOPs that contribute to antibiotic degradation, including common electro-chemical oxidation (EO), electrolyte enhanced EO, electro-Fenton (EF) processes, EF-like process, and EAOPs coupling with other processes. Then we assess the performance of various EAOPs in antibiotic degradation and discuss the influence of key factors, including electrode, initial concentration and type of antibiotic, operation conditions, electrolyte, and water quality. We also review mechanisms and degradation pathways of various antibiotics degradation by EAOPs, and address the species and toxicity of intermediates produced during antibiotics treatment. Finally, we highlight challenges and critical research needs to facilitate the application of EAOPs in antibiotic treatment.
Synergistic singlet oxygen and UV irradiation for efficient intracellular ARGs removal via peroxymonosulfate/catalytic membrane-UV system

Abstract:The eliminate of antibiotic resistance genes (ARGs) is pivotal in mitigating the proliferation of antibiotic resistance. In this study, a PMS/CM-UV system was engineered, combining a Co 3 O 4 -modified carbon nanotubes catalytic membrane with LED-UV lamps, to effectively eliminate intracellular ARGs (iARGs). Leveraging the synergistic effect of singlet oxygen ( 1 O 2 ) and UV irradiation, this process requires only a brief hydraulic retention time of a few minutes and standard UV disinfection irradiation intensity. The cellular physiological function and transcriptomic analysis indicated that reactive oxygen species (ROS) and UV irradiation compromised the cell membrane integrity of E. coli MG1655-SD, as indicated by the down-regulation of the feoB gene, leading to an increased concentration of 1 O 2 within the intracellular environment. The synergistic effect of 1 O 2 and UV irradiation resulted in the down-regulation of btuE , thereby curtailing the SOS and oxidative stress responses. Additionally, UV irradiation down-regulated ftsK , uvrB , and uvrA genes, involved in DNA replication, damage site recognition, and self-repair. These processes collectively contribute to the oxidative damage of iARGs by 1 O 2 before their release into the extracellular environment. This work provided a strategy to develop advanced oxidation disinfection technology aimed at ARGs removal.
