Identification of SiEUL Gene Family in Foxtail Millet (setaria Italica L.) and the Drought Tolerance Function of SiEULS3

Abstract:As global climate changes rapidly, drought has emerged as a critical factor influencing crop growth and yield. The C4 crop foxtail millet (Setaria italica L.) is noted for its drought resistance. Investigating its stress response genes and elucidating the molecular mechanisms underlying this tolerance can offer valuable insights for genetic enhancement of drought resistance in other crops. The Euonymus lectin (EUL) family, which consists of unique carbohydrate-binding proteins, is essential for plant defense and stress adaptation. In our study, we identified six members of the SiEUL gene family in foxtail millet. We performed an exhaustive bioinformatics analysis covering their physical and chemical properties, evolutionary relationships, protein structures, chromosomal locations, cis-acting elements in gene promoters, and transcriptional activity across various tissues and developmental stages. Further analysis revealed that SiEULS3 was strongly expressed under osmotic stress and was regulated by different hormones. By expressing SiEULS3 in Arabidopsis, we discovered that over-expression of this gene could enhance plant osmotic and drought tolerance by improving antioxidant capacity and hydration status. Thus, the SiEULS3 gene holds significant potential as a key locus for future genetic engineering studies aimed at improving crop drought resistance.
