Abstract:The design of biomimetic electronic nose and electronic tongue is presented. First, the smell and taste sensors mimicking mammalian olfaction and gustation are described; then, some mimetic signal processing methods for the recognition of odorants and tastants are also developed. Finally, olfactory and gustatory cell-based biosensors are presented, which are based on the mimetic bioelectronic nose and bioelectronic tongue research, trying to culture living olfactory and taste cells on the surface of chips, that can detect odorants and tastants using microelectronic chips such as those based on field effect transistors (FETs), microelectrode arrays (MEAs), and light-addres sable potentiometric sensors (LAPSs) to record action potential and identify the extracellular chemical and biological substances.

Abstract:We discussed the biological mechanism of taste, studied the artificial taste system through mimicking of biological taste structure as well as the sensitive characteristics and selectivity of artificial lipid membranes. We designed a multi-channel taste sensor array based on biological lipid materials. Abstract:As two of the basic senses of human beings, olfaction and gustation play a very important role in daily life. These two types of chemical sensors are important for recognizing environmental conditions. Electronic nose and electronic tongue, which mimics animals' olfaction and gustation to detect odors and chemical components, have been carried out due to their potential commercial applications for biomedicine, food industry and environmental protection. In this report, the biomimetic artificial nose and tongue is presented. Firstly, the smell and taste sensors mimicking the mammalian olfaction and gustation was described, and then, some mimetic design of electronic nose and tongue for odorants and tastants detection are developed. Finally, olfactory and gustatory biosensors are presented as the developing trends of this field.

Abstract:Taste and smell are very important chemical senses that provide indispensable information on food quality, potential mates and potential danger. In recent decades, much progress has been achieved regarding the underlying molecular and cellular mechanisms of taste and odor senses. Recently, biosensors have been developed for detecting odorants and tastants as well as for studying ligand-receptor interactions. This review summarizes the currently available biosensing approaches, which can be classified into two main categories: in vitro and in vivo approaches. The former is based on utilizing biological components such as taste and olfactory tissues, cells and receptors, as sensitive elements. The latter is dependent on signals recorded from animals' signaling pathways using implanted microelectrodes into living animals. Advantages and disadvantages of these two approaches, as well as differences in terms of sensing principles and applications are highlighted. The main current challenges, future trends and prospects of research in biomimetic taste and odor sensors are discussed.

Bioanalytical and Chemical Sensors Using Living Taste, Olfactory, and Neural Cells and Tissues: a Short Review

Abstract:Biosensors utilizing living tissues and cells have recently gained significant attention as functional devices for chemical sensing and biochemical analysis. These devices integrate biological components (i.e. single cells, cell networks, tissues) with micro-electro-mechanical systems (MEMS)-based sensors and transducers. Various types of cells and tissues derived from natural and bioengineered sources have been used as recognition and sensing elements, which are generally characterized by high sensitivity and specificity. This review summarizes the state of the art in tissue- and cell-based biosensing platforms with an emphasis on those using taste, olfactory, and neural cells and tissues. Many of these devices employ unique integration strategies and sensing schemes based on sensitive transducers including microelectrode arrays (MEAs), field effect transistors (FETs), and light-addressable potentiometric sensors (LAPSs). Several groups have coupled these hybrid biosensors with microfluidics which offers added benefits of small sample volumes and enhanced automation. While this technology is currently limited to lab settings due to the limited stability of living biological components, further research to enhance their robustness will enable these devices to be employed in field and clinical settings.

Abstract:The intrinsic perception process of taste is obviously far less known than those of vision, audition, touch and olfaction. Despite that taste cells utilize a variety of sensory mechanisms to translate plenty of gustatory sensations such as sour, sweet, bitter, salty and umami into cellular signals, gustatory perception mechanisms are still under exploration due to the lack of effective methods on cellular and molecular level. Recently the development of molecular biological and electrophysiological studies has promoted exploration of olfactory and gustatory transduction and coding mechanisms dramatically. Based on the studies of artificial olfaction, artificial taste and cell-based biosensor in our laboratory, this paper reviews the current research on taste transduction mechanism. We introduce the recent advances in cell chip that combined biology with microelectronics, discuss taste cell chip as well as its potential of prospective application in taste transduction mechanism in detail and propose the research trends of taste chip in future.
A Biomimetic Taste Biosensor Based on Bitter Receptors Synthesized and Purified on Chip from a Cell-Free Expression System

Abstract:The conventional approaches for the preparation of receptor proteins towards chemical sensing are usually based on the expression of receptors in a heterologous cell system, which suffers from time-consuming, low efficiency, and complicated purification process. To address these limitations, in this study, a bitter receptor, T2R4, was synthesized and purified on chip from a cell-free protein expression system, which can be realized within a few hours. The bitter receptors were utilized as sensitive elements of a chemical biosensor for the detection of specific bitter substances. Quartz crystal microbalance (QCM) devices were used as transducers to monitor the process of sensor preparation and responses to different bitter substances by recoding the crystal resonant frequency shifts. The obtained results show that the dose-response curves of denatonium and crystal resonant frequency shifts are established in a wide linear range of 10 nM to 0.1 mM with a lower detection limit of 5 nM, which is superior to previous reports. Nearly 3 times of enhancement in sensitivity were achieved, which is calculated to be 4.96 kHz/mM. Such on-chip rapid synthesis and purification of receptors serve to achieve increased coupling efficiency and improved sensing performances. The combination of simple and rapid cell-free expression technique provides a very promising approach for the rapid and high-efficient development of receptor-based biosensors for chemical sensing.

Abstract:Olfaction is a very important sensation for all animals. Recently great progress has been made in the research of olfactory transduction. Especially the novel finding of the gene superfamily encoding olfactory receptors has led to rapid advances in olfactory transduction. These advances also promoted the research of biomimetic olfactory-based biosensors and some obvious achievements have been obtained due to their potential commercial prospects and promising industrial applications. This paper briefly introduces the biological basis of olfaction, summarizes the progress of olfactory signal transduction in the olfactory neuron, the olfactory bulb and the olfactory cortex, outlines the latest developments and applications of biomimetic olfactory-based biosensors. Finally, the olfactory biosensor based on light addressable potentiometric sensor (LAPS) is addressed in detail based on our recent work and the research trends of olfactory biosensors in future are discussed.

Abstract:In recent years, taste cell- and receptor-based biosensors have gained considerable attention as an emerging technology for chemical sensing as well as research on taste signal transduction mechanisms. Taste cells and receptors can recognize the specific chemical signals presented by various taste substances and compounds offering unique performance characteristics which cannot be achieved by current artificial devices. As a result, taste cells and receptors have been increasingly recognized as promising candidates for the development of chemical sensors. Currently, one of the main challenges of this technology is the preparation of functional taste cells and receptors to satisfy the need of building sensing components for chemical sensors. Additionally, the coupling of taste cells and receptors with transducers is a key step to develop chemical sensors which can meet the specific performance requirements for biomedical applications. Therefore, current efforts are mainly focused on the preparation of functional taste cells and receptors and their coupling with transducers. This review will summarize the most recent advances in taste cell- and receptor-based biosensors. We will describe the basis of biological taste sensing systems, current approaches for preparing functional taste cells and receptors, and basic design principles and approaches for coupling taste cells and receptors with transducers. Furthermore, several major applications of taste cell- and receptor-based biosensors will be presented. We will conclude by discussing the current and future challenges of taste cell- and receptor-based biosensors and offer possible strategies to overcome these limitations.

Abstract:This review paper introduces the research progress of bioelectronic nose and bioelectronic tongue technology in vitro and in vivo in our group recent years. Bioelectronic nose and bioelectronic tongue is one kind of biomimetic sensors system or a smart instrument combined electronic sensors with biological olfactory and gustatory organ and pattern recognition techniques etc. In this research work we presented some novel ideas in series including culturing living olfactory and gustatory tissue and cells and cell network on the surface of devices and the implanted electrode in olfactory bulb, and while the paper also gives some practiccal applications in the fields of detection of odorants and tastants in food.

Abstract:A review of an olfactory and taste cell sensor is presented in this article. Olfactory and taste cell sensor is one kind of cell-based on biosensors which are based on the electronic nose and electronic tongue research, trying to culture olfactory and taste living cell on the surface of chips. This is a really mimicking bioelectronic nose and tongue technology, which can detect odors and taste by using chips such as light-addressable potentiometric sensor to record action potential representing the odorants and taste in cell membrane. Now, several kinds of cells from olfactory bulb and taste bud on chip have been cultured, while the experiments and analysis about the detection of olfactory and taste and its action potential under the stimulus of different odors and taste also are in progress.

Abstract:Taste receptor cell, the taste sensation organ with its intrinsic advantage of high sensitivity, specificity, fast response and powerful ability of information processing from taste system, can recognize taste substances. In order to investigate the electrophysiological signals of taste receptor cells and realize acidic sensation in a biomimetic manner, a cell-based biosensor platform of neuron–silicon interface was designed and characterized. Acid-sensing taste receptor cells were cultured on light-addressable potentiometric sensor (LAPS) to recognize sour stimuli as compared with non-acid solutions. A computational model of acid-sensing taste receptor cells was constructed to simulate the action potentials of taste receptor cells and to help decode the extracellular signals recorded by LAPS. The temporal firing and characteristic features from LAPS recordings upon acidic solutions demonstrate that this kind of hybrid biosensor based on taste receptor cells and LAPS can realize acidic sensation. This study also provides a prototype of a novel biosensor which deploys the sensory transduction and decoding mechanism from biology to realize chemosensing, potentially in a non-invasive and long-term manner.
Integrated Olfaction, Gustation and Toxicity Detection by a Versatile Bioengineered Cell-Based Biomimetic Sensor

Abstract:The biological olfactory and gustation system can discriminate thousands of odor and taste substances with high sensitivity and specificity, specific receptor proteins play an important role in this process. This study used the human neuroblastoma SH-SY5Y cell line endogenously expressing the human bitter receptor, T2R16. Meanwhile, an olfactory receptor, ODR-10, was transfected on the plasma membrane of SH-SY5Y cells. T2R16 could specifically respond to bitter compounds with the structure of β-glucopyranosides by activation of G protein coupled receptors (GPCRs) causing cell morphologic changes, which could be monitored using a cell-impedance sensor. ODR-10 could specifically respond to diacetyl by changing the extracellular potential of the cells, the resopnse was recorded by a microelectrode array (MEA). The cell index (CI) value and firing rates were extracted from the signals as the biosensor response characteristics. The results with the sensors indicated a dose-dependent response within a defined concentration range. Moreover, this cell-impedance biosensor enabled quick toxicity detection of salicin when the concentration was ≥6 mM. In conclusion, the biomimetic sensors integrated olfaction, gustation and toxicity detection using the same cell, and has showed great potential for use in both basic research and practical applications.

Abstract:Biological olfactory and taste systems are natural chemical sensing systems with unique performances for the detection of environmental chemical signals. With the advances in olfactory and taste transduction mechanisms, biomimetic chemical sensors have achieved significant progress due to their promising prospects and potential applications. Biomimetic chemical sensors exploit the unique capability of biological functional components for chemical sensing, which are often sourced from sensing units of biological olfactory or taste systems at the tissue level, cellular level, or molecular level. Specifically, at the cellular level, there are mainly two categories of cells have been employed for the development of biomimetic chemical sensors, which are natural cells and bioengineered cells, respectively. Natural cells are directly isolated from biological olfactory and taste systems, which are convenient to achieve. However, natural cells often suffer from the undefined sensing properties and limited amount of identical cells. On the other hand, bioengineered cells have shown decisive advantages to be applied in the development of biomimetic chemical sensors due to the powerful biotechnology for the reconstruction of the cell sensing properties. Here, we briefly summarized the most recent advances of biomimetic chemical sensors using bioengineered olfactory and taste cells. The development challenges and future trends are discussed as well.
Biomimetic sensor for sweet taste detection based on graphene composite materials

Abstract:Sweet taste detection is of great significance in fields of disease prevention, food safety inspection, and drug regulation. In this study, a novel biomimetic sensor was constructed for sweet taste detection by compositing graphene oxide, hemin, and 3-aminophenylboronic acid (APBA). First, optimal fabrication of the sensor was investigated by comparing different composition proportions of graphene oxide and APBA functionalized hemin in saccharide measurements. Then, typical substances of natural sugars (i.e., glucose and fructose) and artificial sweeteners (i.e., saccharin and aspartame) were measured with the sensor through optical absorption spectroscopy. The spectra displayed apparent but different absorbance changes in the presence of those two kinds of saccharides, showing different sensing mechanisms of the biomimetic sensor for sugars and sweeteners like sweet receptors. Finally, dose-dependent and specific responses of the sensor were observed by comparing with other taste substances. The results suggested that the sensor can detect and distinguish natural sugars and artificial sweeteners in a biomimetic way. The research provided a low-cost, long-lifetime, and efficient approach for sweet taste detection, indicating its promising applications in fields such as food and beverage analysis. Abstract:The detection of olfaction, gustation and toxicity has played an important role in industries and research. The key factor to realize this function is to detect different substances with the same sensitive element. Abstract:Taste, as one of the natural chemical sensations, has attracted worldwide attention due to their great significance to organismal survival in the area of seeking advantages and avoiding disadvantages. Therefore, it is highly essential and worthy to develop taste detection methods with higher performances. Fast advancements of the gustatory biosensing technologies, especially in the life sciences and micro-/nano-technologies, are providing unprecedented opportunities for bioelectronic tongue (BioET). This review summarizes the most recent advances in BioETs for gustation detection. First, we briefly present the fundamental of mammalian gustation system (MGS). Then, BioETs are introduced from two aspects of in vitro and in vivo according to their technical architectures. Strategies, target tastes, analytes, sensing elements, transducers, coupling methods and practical performance are highlighted and discussed to assess the taste detection applications comprehensively. Finally, the current challenges and future development trends of BioETs are discussed.

Abstract:With the continuous research breakthroughs on the gustatory system, special biosensors have been developed for taste-related analyses, such as taste perception, transduction, signaling pathways, and taste detection. Taste biosensors provide high sensitivity and specificity for taste analysis, which can achieve real-time and efficient acquisition of taste information in the target object. Moreover, partial-taste biosensors have been integrated based on mimic or natural taste components of mammals (e.g. taste epithelium, taste cells, taste receptors or whole animals). Thus, they have become a main research component of gustatory system correlations. At present, taste biosensors are still under development, the limitation of their application mainly caused by long-term storage/performance stability, high cost, and low production yield. Overcoming these shortcomings will drive the research for practical application of taste biosensors. The review aims to show the great potential of taste biosensors for gustatory system-related analysis and taste detection, and to provide an update on advances in taste-based biosensors. In addition, the challenges and future perspectives of these devices in performance improvement, development and application are discussed.

Abstract:Thanks to the gustatory system, humans can experience the flavors in foods and drinks while avoiding the intake of some harmful substances. Although great advances in the fields of biotechnology, microfluidics, and nanotechnologies have been made in recent years, this astonishing recognition system can hardly be replaced by any artificial sensors designed so far. Here, taste organoids are coupled with an extracellular potential sensor array to form a novel bioelectronic organoid and developed a taste organoids-on-a-chip system (TOS) for highly mimicking the biological sense of taste ex vivo with high stability and repeatability. The taste organoids maintain key taste receptors expression after the third passage and high cell viability during 7 days of on-chip culture. Most importantly, the TOS not only distinguishs sour, sweet, bitter, and salt stimuli with great specificity, but also recognizes varying concentrations of the stimuli through an analytical method based on the extraction of signal features and principal component analysis. It is hoped that this bioelectronic tongue can facilitate studies in food quality controls, disease modelling, and drug screening.
