Abstract:We report the successful synthesis of well-defined, hydrophilic thioester-containing polyacrylamide copolymers by the RAFT copolymerisation of dibenzo[c,e]oxepane-5-thione with either acrylamide or N-isopropylacrylamide. These copolymers efficiently degrade in aqueous solutions of sodium hydroxide, isopropylamine, L-cysteine, and household bleach, reducing the weight-average molecular weight by up to ~80%.
What problem does this paper attempt to address?
The paper aims to address the issues of degradability and environmental friendliness faced by current water-soluble polymers (such as polyacrylamide) in applications like biomedicine, agriculture, and wastewater treatment. Specifically, the authors synthesized a water-soluble polyacrylamide copolymer containing thioester bonds through radical ring-opening copolymerization (rROP). This copolymer exhibits UCST (upper critical solution temperature) behavior in salt solutions and can efficiently degrade under various conditions, including alkaline conditions, amino alcohol conditions, physiological conditions, and household bleach conditions.
### Main Research Questions:
1. **Develop degradable water-soluble polymers**: Many current water-soluble polymers (such as polyacrylamide) are difficult to degrade after use, leading to environmental pollution issues. Therefore, researchers are committed to developing new polymers with controllable degradability to reduce environmental impact.
2. **Introduce thioester bonds to enhance degradability**: By introducing thioester bonds, the polymers can rapidly degrade under specific conditions (such as alkaline, amino alcohol, physiological conditions, etc.), thereby reducing their molecular weight.
3. **Explore UCST behavior**: Investigate the UCST behavior of these copolymers in salt solutions, which helps to understand their solubility and aggregation behavior under different conditions, and is significant for applications such as drug delivery.
### Research Methods:
- **Synthesis Method**: A series of water-soluble copolymers were prepared by RAFT (reversible addition-fragmentation chain transfer) copolymerization of dibenzo[c,e]oxepane-5-thione (DOT) with acrylamide or N-isopropylacrylamide.
- **Degradation Performance Testing**: The degradation performance of these copolymers under different conditions, including alkaline conditions, amino alcohol conditions, physiological conditions (such as L-cysteine), and household bleach conditions, was evaluated by SEC (size exclusion chromatography) analysis.
- **UCST Behavior Study**: The UCST behavior of these copolymers in salt solutions was studied using DLS (dynamic light scattering) technology, exploring their solubility and aggregation behavior at different temperatures.
### Main Findings:
- **High Degradability**: These copolymers containing thioester bonds can efficiently degrade under various conditions, especially under alkaline and amino alcohol conditions, with molecular weight reductions of over 80%.
- **UCST Behavior**: These copolymers exhibit UCST behavior in salt solutions, meaning their solubility decreases at low temperatures, forming larger aggregates. This characteristic has potential value for applications such as drug delivery.
- **Environmental Friendliness**: These copolymers not only have good water solubility but can also rapidly degrade under the action of commercial products (such as household bleach), demonstrating excellent environmental friendliness.
In summary, the paper successfully addresses the shortcomings of existing water-soluble polymers in terms of degradability and environmental friendliness by synthesizing water-soluble copolymers with specific structures, providing new material options for future applications in biomedicine, agriculture, and wastewater treatment.
