Insight into Mesoporous Silica Nanoparticles for Embedding and Improving Environmental and Digestive Stability of Cannabidiol

Abstract:Cannabidiol (CBD) has low water-solubility and chemical instability, which limits its widespread use in food, cosmetics and healthcare products. In this experiment, mesoporous silica nanoparticles (MSN) were used to encapsulate CBD. Additionally, its storage and digestion stability were extensively investigated. The embedding efficiency and loading rate of the MSN-CBD microcapsules reached 76.36% and 19.67%, respectively, in 15 mg/ mL CBD solution after loading time for 24 h with centrifugation impregnation method. Furthermore, the main infrared spectroscopic characteristic absorption peaks and the X-ray diffraction patterns of MSN-CBD microcapsules qualitatively verified that CBD was embedded in the micro pores of MSN at amorphous state. The retention rates of CBD in MSN-CBD microcapsules were significantly higher than that of unembedded CBD at different conditions (high temperatures, strong acids and alkalis, air, incandescent and ultraviolet light). The cumulative release rates of CBD from MSN-CBD microcapsules after simulated gastrointestinal fluid digestion were consistently significantly lower than that of unembedded CBD (P < 0.05), and more than 80% of CBD was conserved in MSN. This work presented CBD delivery system with high encapsulation efficiency and loading rate based on MSN, effectively improving storage stability of CBD and reducing degradation of CBD in gastrointestinal fluids. It provided important guidance on the construction of CBD delivery system with high embedding efficiency, loading rate, stability and bioavailability of oral CBD, which might broaden the application of CBD in the fields of food and pharmaceuticals.
