Garcibracgluinols A-C, Structurally Intriguing Polycyclic Polyprenylated Acylphloroglucinols from Garcinia Bracteata Alleviate Hepatocyte Lipid Accumulation and Insulin Resistance

Abstract:Garcibracgluinols A-C (1-3), three unprecedented polyprenylated polycyclic acylphloroglucinols (PPAPs) with new carbon skeletons, were isolated from the fruits of Garcinia bracteata. Compound 1 is the first example of degraded seco-PPAPs featuring a 9-oxabicyclo [5.3.0] decane skeleton. Compound 2 represents the first instance of macrolide PPAPs, exhibiting an unprecedented 6-oxatricyclic [7.4.1.11,11] pentadecane motif with a unique 10/6/5 ring system. Compound 3 is obtained from 2 through retro-Claisen reaction, methanolysis, and esterification. Their structures containing absolute configurations were elucidated by spectroscopic analysis and quantum chemical calculations. Palmitic acid (PA)-induced HepG2 cells were utilized as the cell model to investigate the effects of PPAPs on non-alcoholic fatty liver disease (NAFLD). The findings demonstrated that garcibracgluinol A could mitigate lipid accumulation and enhance glucose consumption in PA-induced cells. Western blotting analysis revealed that garcibracgluinol A could activate AMPK phosphorylation and suppress the expression of SREBP-1, FAS, and SCD1, thereby inhibiting lipogenesis. Furthermore, garcibracgluinol A treatment activated the canonical PI3K/Akt pathway, leading to the alleviation of insulin resistance induced by PA. These results highlight the potential of garcibracgluinol A as a promising candidate for the treatment of NAFLD.
