Abstract:We describe herein an iridium-catalyzed highly diastereo- and enantioselective hydrogenation of 1,2-azaborines ac- cessing δ-aminoboronic esters of potential biological importance. This method represents the first enantioselective hydrogenation of boron-containing heteroarene and features a diverse substitution pattern and wide scope. The synthetic utility of our method was demonstrated by the synthesis of (–)-Phenibut and the formal synthesis of (+)-3-PPP and Fluvirucinine A1.

What problem does this paper attempt to address?
The paper aims to address the following issues: 1. **Developing new synthetic methods**: The paper proposes a highly enantioselective and stereoselective hydrogenation reaction of 1,2-azaborolines catalyzed by iridium to prepare δ-aminoboronic esters with potential biological activity. This is the first reported enantioselective hydrogenation reaction of boron-containing heterocycles. 2. **Expanding the substrate scope**: This method demonstrates broad substrate applicability and diverse substitution patterns, including various C3 and C5 substituted 1,2-azaboroline compounds. 3. **Improving synthetic efficiency**: Through this method, researchers can synthesize a series of δ-aminoboronic esters with high stereoselectivity, and these products have potential applications in medicinal chemistry. For example, the method was used to synthesize precursors of (–)-Phenibut, (+)-3-PPP, and Fluvirucinine A1. 4. **Addressing the limitations of existing synthetic methods**: Compared to previous methods, this approach avoids cumbersome functional group manipulations and the use of large amounts of chiral reagents, thereby simplifying the synthetic steps and improving yield and stereoselectivity. In summary, this research aims to provide an efficient, modular synthetic strategy for the preparation of δ-aminoboronic ester compounds with broad application prospects.
