Transcriptomic and Biochemical Analyses Reveal the Cell Membrane Defense Regulated by LiaFSR-LiaX System in Lacticaseibacillus Paracasei L9 under Bile Salt Stress

Abstract:Tolerance to bile salt stress is crucial for probiotics to survive in the intestinal tract. Our previous study identified a gene (LPL9_0968) encoding the surface protein LiaX in Lacticaseibacillus paracasei L9, which was shown to participate in the bile resistance mechanism. In this study, transcriptomic analysis under 0.05% glycine deoxycholate revealed the down-regulation of 87 genes when LPL9_0968 were insertionally inactivated. Among these, three genes encoding the LiaFSR three-component system have been identified, which have been reported to regulate cell envelope remodeling under daptomycin stress and have a regulatory relationship with liaX. RT-qPCR verified that the absence of LiaX impaired the activation of liaFSR genes under bile stress, resulting in the decreased expression of surface proteins, such as ABC transporters and cell wall-binding proteins. Transmission electron microscopy and antibiotic resistance experiments showed that the mutant strain had a more fragile cell membrane. In addition, genes rex and adhE related to oxidative stress were down-regualted without LiaX, and oxidative resistance of the mutant strain was decreased, indicating that LiaFSR-LiaX may regulate a more extensive range of genes. These findings provide new insights into the LiaFSR-LiaX system in the bile resistance mechanism and reveal possible target genes of LiaR.
