Topological Abnormalities of Brain Functional Networks Based on Microstates in Early Parkinson’s Disease

Abstract:Early diagnosis of Parkinson’s disease (PD) remains challenging in clinical practice. Utilizing brain network properties aids in understanding and diagnosing the disease. Many studies use traditional sliding window methods for brain functional network construction, lacking research on personalized brain state-based network construction algorithms. In this work, we introduced a method for dynamic functional connectivity network construction based on electroencephalography microstates. Graph theory analysis was used to assess network information processing and transmission efficiency, revealing abnormal topological features of brain networks in early PD. Results showed significantly lower functional connectivity strength of microstate C networks in early PD, primarily in frontal, parietal, and temporal lobes. Furthermore, abnormalities in internal information integration and transmission functions varied across microstate networks. Compared to the healthy level, microstate B networks showed higher clustering coefficients and local efficiency in the parietal and occipital lobes, while microstate C networks exhibited significantly lower metrics in the same regions. These findings indicate that microstate-based brain functional networks capture significant pathological alterations in early PD, encompassing changes in connectivity strength and graph theory parameters. Our study not only advances methods for brain network construction but also provides deeper insights into the pathological changes in early PD brain function, offering valuable reference for clinical diagnosis.
