Multi-enzyme Mimic Nanozyme with Enhanced Catalysis for Methylene Blue Probe Promotes the One-Step Detection of Tau Protein

Abstract:The rapid development of nanostructured electrochemical biosensing has ushered in a new era of precision diagnosis, providing an improved sensitivity and timeliness for detection of clinical biomarkers. In this work, a multi-enzyme mimic nanozyme (MWCNTs/MnO2/Au)-enhanced catalysis for methylene blue (MB) based strategy was firstly developed for high-sensitivity, one-step chronocoulometric detection of low-abundance AD-related biomarker (tau protein) in clinical samples. The core point of this study is based on the highly efficient catalysis of MB in the hairpin probe by numerous reactive oxygen species producing by the nanozyme (MWCNTs/MnO2/Au) catalysis of hydrogen peroxide and oxygen, which significantly enhanced the electrochemical signal. Both MnO2 and gold nanoparticles (AuNPs) presented dual enzyme activities of oxidase and peroxidase, while the excellent electrical conductivity of MWCNTs and Au is very conducive to electron transport and reduces charge transfer impedance. The efficient catalytic effect of MWCNTs/MnO2/Au on MB makes the detection limit of tau protein was obtained as low as similar to 0.3 pg/mL. Meanwhile, a good accuracy in the analysis of tau-spiked plasma samples was also observed with the recovery ranging from 88.90 % to 105.51 %. In conclusion, the MWCNTs/MnO2/Au composite enhanced catalysis for MB probe promoted the one-step detection of tau protein. It performed well in practical applications, and presented lower inspection costs and higher detection timeliness.
