Abstract:It remains a tough task for designing a microscopy system to approach the diffraction limit over a wide field of view due to its restricted space bandwidth product (SBP). Fourier ptychographic microscopy (FPM) has been developed to broaden the SBP of a conventional microscope platform without major hardware modifications. In this paper, we propose a novel FPM system based on the phase-coded speckle illumination, termed PSI-FPM, which employs a laser source with a speckle illumination control optical system. A phase-only spatial light modulator (SLM) with a condenser lens is utilized in the PSI-FPM system to project the phase-coded speckle illumination patterns onto the sample. The convolution between the sample spectrum and the illumination spectrum in the Fourier domain extend the numerical aperture of the objective that acts as a low-pass filter with cut-off frequency. The SLM is introduced in the PSI-FPM system to produce the specific illumination patterns so that the number of the captured images used for reconstruction are greatly reduced. In addition, the generated pattern is scanned to different directions without mechanical movement, which improves the robustness of the system. The PSI-FPM system is validated against simulations and experiments, and as a result reconstructs high-resolution images once the phase retrieval algorithm is employed. We envision that the PSI-FPM system will be suitable for many other speckle imaging applications.

Abstract:Ghost imaging is an indirect system that allows the imaging of an object without directly seeing the object. The speckle pattern that contains the information about light and objects has increasingly become a popular topic in pseudothermal light ghost imaging. However, existing research still has encountered problems of poor imaging quality and slow sampling speeds. We propose a ghost imaging method based on N-order speckle patterns to recover the object (NSGI). The N-order speckle patterns combine N independent laser speckles individually produced by passing an expanded and collimated He-Ne laser through a digital micro-mirror device (DMD). The sampling frequency can be improved by controlling the trigger signals of different DMDs. The results of the simulation and experiment have verified that our method can increase sampling speed and reconstruction accuracy. In addition, NSGI can be applied to more applications by designing multiple independent speckles with different properties. (C) 2018 Society of Photo-Optical Instrumentation Engineers (SPIE).

Abstract:High spatial resolution is the goal of many imaging systems. While designing a high-resolution lens with diffraction-limited performance over a large field of view remains a difficult task, creating a complex speckle pattern with wavelength-limited spatial features is easily accomplished with a simple random diffuser. With this observation and the concept of near-field ptychography, we report a new imaging modality, termed near-field Fourier ptychography, to address high-resolution imaging challenges in both microscopic and macroscopic imaging settings. 'Near-field' refers to placing the object at a short defocus distance with a large Fresnel number. We project a speckle pattern with fine spatial features on the object instead of directly resolving the spatial features via a high-resolution lens. We then translate the object (or speckle) to different positions and acquire the corresponding images by using a low-resolution lens. A ptychographic phase retrieval process is used to recover the complex object, the unknown speckle pattern, and the coherent transfer function at the same time. In a microscopic imaging setup, we use a 0.12 numerical aperture (NA) lens to achieve an NA of 0.85 in the reconstruction process. In a macroscale photographic imaging setup, we achieve ~7-fold resolution gain by using a photographic lens. The collection optics do not determine the final achievable resolution; rather, the speckle pattern's feature size does. This is similar to our recent demonstration in fluorescence imaging settings (Guo et al., Biomed. Opt. Express, 9(1), 2018). The reported imaging modality can be employed in light, coherent X-ray, and transmission electron imaging systems to increase resolution and provide quantitative absorption and object phase contrast.

Abstract:We present a novel approach to compensate coherence effect via combining the transport of intensity equation (TIE) with look-up table phase compensation (LUT-PC) method. It is the better version of the Speckle-TIE method we demonstrated before on the basis of the quantitative phase imaging camera with a weak diffuser (QPICWD). With the phase gradient ratio theory and the look-up table method, the phase blurring caused by underestimation of phase gradient will be compensated correctly by reasonable rescaling. The LUT-PC Speckle-TIE method has the evident predominance of speediness since it only needs one slightly defocused speckle image in one time owing to that the reference speckle image can be captured beforehand. The deblurring achieved by this method improves the imaging resolution to the theoretical partial coherence limit with good robustness, reducing artifacts and improving the accuracy and contrast. The experimental results show the effectiveness of the technique.

Abstract:Optical coherence tomography (OCT) has been an important and powerful tool for biological research and clinical applications. However, speckle noise significantly degrades the image quality of OCT and has a negative impact on the clinical diagnosis accuracy. In this paper, we propose a novel speckle noise suppression technique which changes the spatial distribution of sample beam using a special optical chopper. Then a series of OCT images with uncorrelated speckle patterns could be captured and compounded to improve the image quality without degradation of resolution. Typical signal-to-noise ratio improvement of ∼6.4 dB is experimentally achieved in tissue phantom imaging with average number n = 100. Furthermore, compared with conventional OCT, the proposed technique is demonstrated to view finer and clearer biological structures in human skin in vivo, such as sweat glands and blood vessels. The advantages of low cost, simple structure and compact integration will benefit the future design of handheld or endoscopic probe for biomedical imaging in research and clinical applications.

Abstract:We describe an angular compounding method by full-channel B-scan modulation encoding for speckle reduction in optical coherence tomography. The complex-valued spectral interferogram (SI) is reconstructed by removing one of the conjugate terms in the depth space. Fourier transform of the complex SI along the lateral direction enables a full-channel (with both negative and positive Fourier space) for B-scan modulation in the spatial frequency (ν) domain. A full-size probe beam, determined by the scanning mirror size, is centered on the mirror pivot, which allows the negative and positive half-channels working in parallel. Compared with the existing method, where only a half-channel (negative or positive) works alone, the proposed full-channel method offers more than twice the performance in speckle reduction. The feasibility of the proposed full-space approach is validated through both the phantom and in vivo human thumbnail experiments.

Abstract:Speckle patterns have been widely used in imaging techniques such as ghost imaging, dynamic speckle illumination microscopy, structured illumination microscopy, and photoacoustic fluctuation imaging. Recent advances in the ability to control the statistical properties of speckles has enabled the customization of speckle patterns for specific imaging applications. In this work, we design and create special speckle patterns for parallelized nonlinear pattern-illumination microscopy based on fluorescence photoswitching. We present a proof-of-principle experimental demonstration where we obtain a spatial resolution three times higher than the diffraction limit of the illumination optics in our setup. Furthermore, we show that tailored speckles vastly outperform standard speckles. Our work establishes that customized speckles are a potent tool in parallelized super-resolution microscopy.

Abstract:Plane-wave ultrasound imaging has been widely used in many frontier research areas in medical ultrasound due to its ultra-high frame rate. To improve its image quality, many novel beamforming methods have been proposed and in-depth studied, such as coherent compounding method and methods based on compressive sensing. However, the influence of the structure and working mode of the ultrasound probe on the image's lateral resolution and contrast is still rarely studied. Convenient ultrasound probe is always self-focused with an acoustic len in the elevation direction, and thus the acoustic energy distributed in the y-z plane is not uniform. We hypothesized that, if the probe is not focused in the elevation direction, the quality of the image obtained with a kind of "pure plane-wave" will be increased. To verify our hypothesis, the non-elevation-focused probe (NEFP) was fabricated, and the plane-wave ultrasound images obtained with NEFP were compared with those obtained with the traditional elevation-focused probe (EFP). The experiments were conducted with a ultrasound driven/acquire system and a general purpose ultrasound imaging phantom. The two images were obtained with coherent compounding method by 9 plane-wave images. The image obtained using NEFP visually has a lower speckle noise background than the one obtained using EFP in the shallow region. It is shown that, in the deeper region, the contrast of NEFP image are much better than the EFP image. However, the difference of their lateral resolution is unapparent. The results of this study indicates that, the NEFP probe is actually better than EFP probe in some respects, and this may provide us an additional way to enhance the image quality of plane-wave ultrasound imaging technique.

Abstract:Speckle noise is problematic in optical coherence tomography (OCT). With the fast scan rate, swept source OCT scans the same position in the retina for multiple times rapidly and computes an average image from the multiple scans for speckle reduction. However, the eye movement poses some challenges. In this paper, we propose a new method for speckle reduction from multiply-scanned OCT slices. The proposed method applies a preliminary speckle reduction on the OCT slices and then registers them using a global alignment followed by a local alignment based on fast iterative diamond search. After that, low rank matrix completion using bilateral random projection is utilized to iteratively estimate the noise and recover the underlying clean image. Experimental results show that the proposed method achieves average contrast to noise ratio 15.65, better than 13.78 by the baseline method used currently in swept source OCT devices. The technology can be embedded into current OCT machines to enhance the image quality for subsequent analysis.

Abstract:Memory-effect-based speckle-correlated imaging through opacity has been exploited widely in recent years. Generally, spatially incoherent light sources are required to provide effective illumination that can eliminate the interference noise. Here, the wavelength-dependent speckle multiplexing (WDSM) technique is first proposed and demonstrated as an effective method to suppress the interference noise, thereby facilitating the use of partially spatially coherent light as an alternative promising illuminating source. In contrast to the conventional decoherent approach which often employs a rapidly rotating diffuser to reduce the spatial coherence of a spatially coherent light, the WDSM shows greatly enhanced structural robustness by combining a tunable laser and the speckle illumination using a multimode fibre. The proposed WDSM not only expands the scope of candidate light sources for speckle-correlated imaging based on the classical phase-retrieval algorithm, but also enables the multi-channel detection determined by the speckle spectral decorrelation, which would further facilitate the waveband selective light-matter detection through opacity. This work also combines the advantages of fibre optics and laser technology and enriches the potential applications of speckle-correlated imaging.

Abstract:Abstract. We present a technique to reduce speckle in visible-light optical coherence tomography (vis-OCT) that preserves fine structural details and is robust against sample motion. Specifically, we locally modulate B-scans orthogonally to their axis of acquisition. Such modulation enables acquisition of uncorrelated speckle patterns from similar anatomical locations, which can be averaged to reduce speckle. To verify the effectiveness of speckle reduction, we performed in-vivo retinal imaging using modulated raster and circular scans in both mice and humans. We compared speckle-reduced vis-OCT images with the images acquired with unmodulated B-scans from the same anatomical locations. We compared contrast-to-noise ratio (CNR) and equivalent number of looks (ENL) to quantify the image quality enhancement. Speckle-reduced images showed up to a 2.35-dB improvement in CNR and up to a 3.1-fold improvement in ENL with more discernable anatomical features using eight modulated A-line averages at a 25-kHz A-line rate.

Abstract:In recent years, X-ray speckle-tracking techniques have emerged as viable tools for wavefront metrology and sample imaging applications. These methods are based on the measurement of near-field images. Thanks to their simple experimental setup, high angular sensitivity and compatibility with low-coherence sources, these methods have been actively developed for use with synchrotron and laboratory light sources. Not only do speckle-tracking techniques give the potential for high-resolution imaging, but they also provide rapid and robust characterization of aberrations of X-ray optical elements, focal spot profiles, and sample position and transmission properties. In order to realize these capabilities, software implementations are required that are equally rapid and robust. To address this need, a software suite has been developed for the ptychographic X-ray speckle-tracking technique, an X-ray speckle-based method suitable for highly divergent wavefields. The software suite is written in Python 3, with an OpenCL back end for GPU and multi-CPU core processing. It is accessible as a Python module, through the command line or through a graphical user interface, and is available as source code under Version 3 or later of the GNU General Public License.

Speckle Space-Time Covariance in High-Contrast Imaging

Abstract:We introduce a new framework for point-spread function (PSF) subtraction based on the spatio-temporal variation of speckle noise in high-contrast imaging data where the sampling timescale is faster than the speckle evolution timescale. One way that space-time covariance arises in the pupil is as atmospheric layers translate across the telescope aperture and create small, time-varying perturbations in the phase of the incoming wavefront. The propagation of this field to the focal plane preserves some of that space-time covariance. To utilize this covariance, our new approach uses a Karhunen-Loéve transform on an image sequence, as opposed to a set of single reference images as in previous applications of Karhunen-Loéve Image Processing (KLIP) for high-contrast imaging. With the recent development of photon-counting detectors, such as microwave kinetic inductance detectors (MKIDs), this technique now has the potential to improve contrast when used as a post-processing step. Preliminary testing on simulated data shows this technique can improve contrast by at least 10-20% from the original image, with significant potential for further improvement. For certain choices of parameters, this algorithm may provide larger contrast gains than spatial-only KLIP.

Abstract:Speckle projection profilometry (SPP), which establishes the global correspondences between stereo images by projecting only a single speckle pattern, has the advantage of single-shot 3D reconstruction. Nevertheless, SPP suffers from the low matching accuracy of traditional stereo matching algorithms, which fundamentally limits its 3D measurement accuracy. In this work, we propose a single-shot 3D shape measurement method using an end-to-end stereo matching network for SPP. To build a high-quality SPP dataset for training the network, by combining phase-shifting profilometry (PSP) and temporal phase unwrapping techniques, high-precision absolute phase maps can be obtained to generate accurate and dense disparity maps with high completeness as the ground truth by phase matching. For the architecture of the network, a multi-scale residual subnetwork is first leveraged to synchronously extract compact feature tensors with 1/4 resolution from speckle images for constructing the 4D cost volume. Considering that the cost filtering based on 3D convolution is computationally costly, a lightweight 3D U-net network is proposed to implement efficient 4D cost aggregation. In addition, because the disparity maps in the SPP dataset should have valid values only in the foreground, a simple and fast saliency detection network is integrated to avoid predicting the invalid pixels in the occlusions and background regions, thereby implicitly enhancing the matching accuracy for valid pixels. Experiment results demonstrated that the proposed method improves the matching accuracy by about 50% significantly compared with traditional stereo matching methods. Consequently, our method achieves fast and absolute 3D shape measurement with an accuracy of about 100µm through a single speckle pattern.

Abstract:The enhancement of a laser speckle photography technique by means of a modification of the film development procedure is presented. Using a holographic film, an amplitude specklegram can be changed to a phase specklegram. This conversion is achieved during the film development procedure by a controlled bleaching of the exposed film. The contrast and visibility of Young’s fringes reconstructed from a phase specklegram are better than those from an amplitude specklegram. The diffraction efficiency of a phase specklegram is greater than that of an amplitude specklegram. The experimental arrangement and the method to get a phase specklegram are described. The phase specklegram characteristics are presented in detail and experimental results are discussed.

Abstract:As a high-speed 3D surface imaging technique, the single-shot speckle projection profilometry (SPP) is widely applied to various fields, where the design of speckle pattern plays a critical role in the fundamental issue of correspondence matching. However, the assessment of speckle patterns in the real imaging system is a time consuming and tedious procedure and their performance might vary with the operators and testing environment. To solve this problem, a visualised simulation platform based on 3ds Max engine is proposed in the paper. Furthermore, the performance of digital speckle patterns (DSPs) on the SPP technique is comprehensively evaluated in terms of 3D reconstruction precision, algorithm efficiency and pattern uniqueness. Firstly, the influence of the noise levels of the projection and imaging processes is considered. Then, the 3D profile reconstruction using five types of speckle pattern is performed in a real experiment. Afterward, a comparative analysis with the optimal DSP is conducted. In addition, the effectiveness of the typical metric of the painted speckle pattern in the field of digital image correlation (DIC) is investigated; results demonstrate its insufficient capability in the SPP field. Moreover, the scaling model of speckle size, influence of speckle deformation and material surface are also discussed to guide the design of DSP. This study can potentially facilitate the selection of high-quality DSPs and the standardisation of the SPP technique. The virtual experiment platform executable file of 3ds Max and the recommended DSP are uploaded as reference.
