Abstract:The Scholander-Hammel pressure chamber has been used in thousands of papers to measure osmotic pressure, c, turgor pressure, Pt, and bulk modulus of elasticity, epsilon, of leaf cells by pressure-volume (PV) curve analysis. PV analysis has been questioned in the past. In this paper we use micromechanical analysis of leaf cells to examine the impact on PV curve analysis of negative turgor in living cells (Pt). Models predict negative Pt (-0.1 to -1.8MPa) depending on leaf cell size and shape in agreement with experimental values reported by J. J. Oertli. Modeled PV curves have linear regions even when Pt is quite negative, contrary to the arguments of M.T. Tyree. Negative Pt is totally missed by PV curve analysis and results in large errors in derived c and Pt but smaller errors in epsilon. A survey of leaf cell sizes vs habitat (arid, temperate, and rainforest), suggests that the majority of published PV curves result in errors of 0.1-1.8MPa in derived c and Pt, whereby the error increases with decreasing cell size. We propose that small cell size in leaves is an ecological adaptation that permits plants to endure negative values of water potential with relatively little water loss.
Morpho-mechanics of pressurized cellular sheets

Abstract:Everyday experience shows that cellular sheets are stiffened by the presence of a pressurized gas: from bicycle inner tubes to bubble wrap, the presence of an internal pressure increases the stiffness of otherwise floppy structures. The same is true of plants, with turgor pressure (due to the presence of water) taking the place of gas pressure; indeed, in the absence of water, many plants wilt. However, the mechanical basis of this stiffening is somewhat opaque: simple attempts to rationalize it suggest that the stiffness should be independent of the pressure, at odds with everyday experience. Here, we study the mechanics of sheets that are a single cell thick and show how a pressure-dependent bending stiffness may arise. Our model rationalizes observations of turgor-driven shrinkage in plant cells and also suggests that turgor is unlikely to provide significant structural support in many monolayer leaves, such as those found in mosses. However, for such systems, turgor does provide a way to control leaf shape, in accordance with observations of curling upon drying of moss leaves. Guided by our results, we also present a biomimetic actuator that un-curls upon pressurization.

Abstract:Plant cell growth depends on turgor pressure, the cell hydrodynamic pressure, which drives expansion of the extracellular matrix (the cell wall). Turgor pressure regulation depends on several physical, chemical and biological factors, including: vacuolar invertases, which modulate osmotic pressure of the cell, aquaporins, which determine the permeability of the plasma membrane to water, cell wall remodeling factors, which determine cell wall extensibility (inverse of effective viscosity), and plasmodesmata, which are membrane-lined channels that allow free movement of water and solutes between cytoplasms of neighbouring cells, like gap junctions in animals. Plasmodesmata permeability varies during plant development and experimental studies have correlated changes in the permeability of plasmodesmal channels to turgor pressure variations. Here we study the role of plasmodesmal permeability in cotton fiber growth, a type of cell that increases in length by at least 3 orders of magnitude in a few weeks. We incorporated plasmodesma-dependent movement of water and solutes into a classical model of plant cell expansion. We performed a sensitivity analysis to changes in values of model parameters and found that plasmodesmal permeability is among the most important factors for building up turgor pressure and expanding cotton fibers. Moreover, we found that non-monotonic behaviors of turgor pressure that have been reported previously in cotton fibers cannot be recovered without accounting for dynamic changes of the parameters used in the model. Altogether, our results suggest an important role for plasmodesmal permeability in the regulation of turgor pressure.

Simulating Turgor-Induced Stress Patterns in Multilayered Plant Tissues

Abstract:The intertwining between mechanics and developmental biology is extensively studied at the shoot apical meristem of land plants. Indeed, plant morphogenesis heavily relies on mechanics; tissue deformations are fueled by turgor-induced forces, and cell mechanosensitivity plays a major regulatory role in this dynamics. Since measurements of forces in growing meristems are still out of reach, our current knowledge relies mainly on theoretical and numerical models. So far, these modeling efforts have been mostly focusing on the epidermis, where aerial organs are initiated. In many models, the epidermis is assimilated to its outermost cell walls and described as a thin continuous shell under pressure, thereby neglecting the inner walls. There is, however, growing experimental evidence suggesting a significant mechanical role of these inner walls. The aim of this work is to investigate the influence of inner walls on the mechanical homeostasis of meristematic tissues. To this end, we simulated numerically the effect of turgor-induced loading, both in realistic flower buds and in more abstract structures. These simulations were performed using finite element meshes with subcellular resolution. Our analysis sheds light on the mechanics of growing plants by revealing the strong influence of inner walls on the epidermis mechanical stress pattern especially in negatively curved regions. Our simulations also display some strong and unsuspected features, such as a correlation between stress intensity and cell size, as well as differential response to loading between epidermal and inner cells. Finally, we monitored the time evolution of the mechanical stresses felt by each cell and its descendants during the early steps of flower morphogenesis.

Abstract:Plant morphogenesis involves multiple biochemical and physical processes inside the cell wall. With the continuous progress in biomechanics field, extensive studies have elucidated that mechanical forces may be the most direct physical signals that control the morphology of cells and organs. The extensibility of the cell wall is the main restrictive parameter of cell expansion. The control of cell wall mechanical properties largely determines plant cell morphogenesis. Here, we summarize how cell wall modifying proteins modulate the mechanical properties of cell walls and consequently influence plant morphogenesis.

Abstract:Filamentous pathogens need to overcome plant barriers for successful infection. To this end, special structures, most commonly appressoria, are used for penetration. In differentiated appressoria, the generation of high turgor pressure is mandatory to breach plant cell wall and cuticle. However, quantitative description of turgor pressure and resulting invasive forces are only described for a handful of plant pathogens. Recent advances in methodology allowed determination of surprisingly high pressures and corresponding forces in oomycetes and a necrotrophic fungus. Here, we describe turgor generation in appressoria as essential function for host penetration. We summarize the known experimentally determined turgor pressure as well as invasive forces and discuss their universal role in plant pathogen infection.

Abstract:Turgor is the driving force of plant growth, making possible for roots to overcome soil resistance or for stems to counteract gravity. Maintaining a constant growth rate while avoiding the cell content dilution, which would progressively stop the inward water flux, imposes the production or import of osmolytes in proportion to the increase of volume. We coin this phenomenon stationary osmoregulation. The article explores the quantitative consequences of this hypothesis on the interaction of a cylindrical cell growing axially against an obstacle. An instantaneous axial compression of a pressurized cylindrical cell generates a force and a pressure jump which both decrease toward a lower value once water has flowed out of the cell to reach the water potential equilibrium. In a first part, the article derives analytical formula for these force and over-pressure both before and after relaxation. In a second part, we describe how the coupling of the Lockhart's growth law with the stationary osmoregulation hypothesis predicts a transient slowdown in growth due to contact before a re-acceleration in growth. We finally compare these predictions with the output of an elastic growth model which ignores the osmotic origin of growth: models only match in the early phase of contact for high stiffness obstacle.

Abstract:Turgor pressure and envelope elasticity of bacterial cells are two mechanical parameters that play a dominant role in cellular deformation, division, and motility. However, a clear understanding of these two properties is lacking because of their strongly interconnected mechanisms. This study established a nanoindentation method to precisely measure the turgor pressure and envelope elasticity of live bacteria. The indentation force-depth curves of Klebsiella pneumoniae bacteria were recorded with atomic force microscopy. Through combination of dimensional analysis and numerical simulations, an explicit expression was derived to decouple the two properties of individual bacteria from the nanoindentation curves. We show that the Young's modulus of bacterial envelope is sensitive to the external osmotic environment, and the turgor pressure is significantly dependent on the external osmotic stress. This method can not only quantify the turgor pressure and envelope elasticity of bacteria, but also help resolve the mechanical behaviors of bacteria in different environments.
A Mechanical Model to Interpret Cell-Scale Indentation Experiments on Plant Tissues in Terms of Cell Wall Elasticity and Turgor Pressure

Abstract:Morphogenesis in plants is directly linked to the mechanical elements of growing tissues, namely cell wall and inner cell pressure. Studies of these structural elements are now often performed using indentation methods such as atomic force microscopy. In these methods, a probe applies a force to the tissue surface at a subcellular scale and its displacement is monitored, yielding force-displacement curves that reflect tissue mechanics. However, the interpretation of these curves is challenging as they may depend not only on the cell probed, but also on neighboring cells, or even on the whole tissue. Here, we build a realistic three-dimensional model of the indentation of a flower bud using SOFA (Simulation Open Framework Architecture), in order to provide a framework for the analysis of force-displacement curves obtained experimentally. We find that the shape of indentation curves mostly depends on the ratio between cell pressure and wall modulus. Hysteresis in force-displacement curves can be accounted for by a viscoelastic behavior of the cell wall. We consider differences in elastic modulus between cell layers and we show that, according to the location of indentation and to the size of the probe, force-displacement curves are sensitive with different weights to the mechanical components of the two most external cell layers. Our results confirm most of the interpretations of previous experiments and provide a guide to future experimental work.

Abstract:Gaining insight into osmotic pressure and its biological implications is pivotal for revealing mechanisms underlying numerous fundamental biological processes across scales and will contribute to the biomedical and pharmaceutical fields. This review aims to provide an overview of the current understanding, focusing on two central issues: (i) how to determine theoretically osmotic pressure and (ii) how osmotic pressure affects important biological activities. More specifically, we discuss the representative theoretical equations and models for different solutions, emphasizing their applicability and limitations, and summarize the effect of osmotic pressure on lipid phase separation, cell division, and differentiation, focusing on the mechanisms underlying the osmotic pressure dependence of these biological processes. We highlight that new theory of osmotic pressure applicable for all experimentally feasible temperatures and solute concentrations needs to be developed, and further studies regarding the role of osmotic pressure in other biological processes should also be carried out to improve our comprehensive and in-depth understanding. Moreover, we point out the importance and challenges of developing techniques for the in vivo measurement of osmotic pressure.

Abstract:Global climate change‐caused drought stress, high temperatures and other extreme weather profoundly impact plant growth and development, restricting sustainable crop production. To cope with various environmental stimuli, plants can optimize the opening and closing of stomata to balance CO2 uptake for photosynthesis and water loss from leaves. Guard cells perceive and integrate various signals to adjust stomatal pores through turgor pressure regulation. Molecular mechanisms and signaling networks underlying the stomatal movements in response to environmental stresses have been extensively studied and elucidated. This review focuses on the molecular mechanisms of stomatal movements mediated by abscisic acid, light, CO2, reactive oxygen species, pathogens, temperature, and other phytohormones. We discussed the significance of elucidating the integrative mechanisms that regulate stomatal movements in helping design smart crops with enhanced water use efficiency and resilience in a climate‐changing world.

Abstract:Stomata function as osmotically tunable pores that facilitate gas exchange at the surface of plants. Stomatal opening and closure are regulated by turgor changes in guard cells that result in mechanically regulated deformations of guard cell walls. However, how the molecular, architectural, and mechanical heterogeneities that exist in guard cell walls affect stomatal dynamics is unclear. In this work, stomata of wild type Arabidopsis thaliana plants or of mutants lacking normal cellulose, hemicellulose, or pectins were experimentally induced to close or open. Three-dimensional images of these stomatal complexes were collected using confocal microscopy, images were landmarked, and three-dimensional finite element models (FEMs) were constructed for each complex. Stomatal opening was simulated with a 5 MPa turgor increase. By comparing experimentally measured and computationally modeled changes in stomatal geometry across genotypes, anisotropic mechanical properties of guard cell walls were determined and mapped to cell wall components. Deficiencies in cellulose or hemicellulose were both predicted to stiffen guard cell walls, but differentially affected stomatal pore area and the degree of stomatal opening. Additionally, reducing pectin molecular mass altered the anisotropy of calculated shear moduli in guard cell walls and enhanced stomatal opening. Based on the unique architecture of guard cell walls and our modeled changes in their mechanical properties in cell wall mutants, we discuss how each polysaccharide class contributes to wall architecture and mechanics in guard cells. This study provides new insights into how the walls of guard cells are constructed to meet the mechanical requirements of stomatal dynamics.

Coupling water fluxes with cell wall mechanics in a multicellular model of plant development

Abstract:The growth of plant organs is a complex process powered by osmosis that attracts water inside the cells; this influx induces simultaneously an elastic extension of the walls and pressure in the cells, called turgor pressure; above a threshold, the walls yield and the cells grow. Based on Lockhart’s seminal work, various models of plant morphogenesis have been proposed, either for single cells, or focusing on the wall mechanical properties. However, the synergistic coupling of fluxes and wall mechanics has not yet been fully addressed in a multicellular model. This work lays the foundations of such a model, by simplifying as much as possible each process and putting emphasis on the coupling itself. Its emergent properties are rich and can help to understand plant morphogenesis. In particular, we show that the model can display a new type of lateral inhibitory mechanism that could contribute to the amplification of growth heterogeneities, essential for shape differentiation. Significance Statement Plant cells are surrounded by a rigid wall that prevents cell displacements and rearrangements as in animal tissues. Therefore, plant morphogenesis relies only on cell divisions, shape changes, and local modulation of growth rate. It has long been recognized that cell growth relies on the competition between osmosis that tends to attract water into the cells and wall mechanics that resists to it, but this interplay has never been fully explored in a multicellular model. The goal of this work is to analyze the theoretical consequences of this coupling. We show that the emergent behavior is rich and complex: among other findings, pressure and growth rate heterogeneities are predicted without any ad-hoc assumption; furthermore the model can display a new type of lateral inhibition based on fluxes that could complement and strengthen the efficiency of already known mechanisms.

Abstract:Abstract Studies on the mechanics of plant cells usually focus on understanding the effects of turgor pressure and properties of the cell wall (CW). While the functional roles of the underlying cytoskeleton have been studied, the extent to which it contributes to the mechanical properties of cells is not elucidated. Here, we study the contributions of the CW, microtubules (MTs) and actin filaments (AFs), in the mechanical properties of Nicotiana tabacum cells. We use a multiscale biomechanical assay comprised of atomic force microscopy and micro-indentation in solutions that (i) remove MTs and AFs and (ii) alter osmotic pressures in the cells. To compare measurements obtained by the two mechanical tests, we develop two generative statistical models to describe the cell’s behaviour using one or both datasets. Our results illustrate that MTs and AFs contribute significantly to cell stiffness and dissipated energy, while confirming the dominant role of turgor pressure.

Abstract:Abstract Osmotic pressure is vital to many physiological activities, such as cell proliferation, wound healing and disease treatment. However, how cells interact with the extracellular matrix (ECM) when subjected to osmotic shock remains unclear. Here, we visualize the mechanical interactions between cells and the ECM during osmotic shock by quantifying the dynamic evolution of the cell traction force. We show that both hypertonic and hypotonic shocks induce continuous and large changes in cell traction force. Moreover, the traction force varies with cell volume: the traction force increases as cells shrink and decreases as cells swell. However, the direction of the traction force is independent of cell volume changes and is always toward the center of the cell-substrate interface. Furthermore, we reveal a mechanical mechanism in which the change in cortical tension caused by osmotic shock leads to the variation in traction force, which suggests a simple method for measuring changes in cell cortical tension. These findings provide new insights into the mechanical force response of cells to the external environment and may provide a deeper understanding of how the ECM regulates cell structure and function.

Turgor pressure affects transverse stiffness and resonant frequencies of buzz-pollinated poricidal anther

Abstract:Several agriculturally valuable plants store their pollen in tube-like poricidal anthers, which release pollen through buzz pollination. In this process, bees rapidly vibrate the anther using their indirect flight muscles. The stiffness and resonant frequency of the anther are crucial for effective pollen release, yet the impact of turgor pressure on these properties is not well understood. Here, we perform three-point flexure tests and experimental modal analysis to determine anther transverse stiffness and resonant frequency, respectively. Dynamic nanoindentation is used to identify the anther's storage modulus as a function of excitation frequency. We subsequently develop mathematical models to estimate how turgor pressure changes after the anther is removed from a flower, thereby emulating zero water availability. We find that 30 minutes post-ablation, anther stiffness decreases 60%. Anther resonant frequency decreases 20% 60 minutes post-ablation. Models indicate turgor pressure in the fresh anther is about 0.2 - 0.3 MPa. Our findings suggest that natural fluctuations in turgor pressure due to environmental factors like temperature and light intensity may require bees to adjust their foraging behaviors. Interestingly, anther storage modulus increased with excitation frequency, underscoring the need for more sophisticated mechanical models that consider viscous fluid transport through plant tissue moving forward.

Abstract:In plants, the robust maintenance of tissue structure is crucial to supporting its functionality. The multi-layered shoot apical meristem (SAM) ofArabidopsis,containing stem cells,is an approximately radially symmetric tissue whose shape and structure is maintained throughout the life of the plant. In this paper, a new biologically calibrated pseudo-three-dimensional (P3D) computational model of a longitudinal section of the SAM is developed. It includes anisotropic expansion and division of cells out of the cross-section plane, as well as representation of tension experienced by the SAM epidermis. Results from the experimentally calibrated P3D model provide new insights into maintenance of the structure of the SAM epidermal cell monolayer under tension and quantify dependence of epidermal and subepidermal cell anisotropy on the amount of tension. Moreover, the model simulations revealed that out-of-plane cell growth is important in offsetting cell crowding and regulating mechanical stresses experienced by tunica cells. Predictive model simulations show that tension-determined cell division plane orientation in the apical corpus may be regulating cell and tissue shape distributions needed for maintaining structure of the wild-type SAM. This suggests that cells' responses to local mechanical cues may serve as a mechanism to regulate cell- and tissue-scale patterning.

Abstract:The growth of plants is a hydromechanical phenomenon in which cells enlarge by absorbing water, while their walls expand and remodel under turgor-induced tension. In multicellular tissues, where cells are mechanically interconnected, morphogenesis results from the combined effect of local cell growths, which reflects the action of heterogeneous mechanical, physical, and chemical fields, each exerting varying degrees of nonlocal influence within the tissue. To describe this process, we propose a physical field theory of plant growth. This theory treats the tissue as a poromorphoelastic body, namely a growing poroelastic medium, where growth arises from pressure-induced deformations and osmotically-driven imbibition of the tissue. From this perspective, growing regions correspond to hydraulic sinks, leading to the possibility of complex non-local regulations, such as water competition and growth-induced water potential gradients. More in general, this work aims to establish foundations for a mechanistic, mechanical field theory of morphogenesis in plants, where growth arises from the interplay of multiple physical fields, and where biochemical regulations are integrated through specific physical parameters.
