Cross-Linking Proteomics Strategy to Provide Insights into the Direct Corona Bound to Amino-Free Ligands on Gold Nanoparticles

Abstract:Gold nanoparticles (GNPs) are commonly functionalized using different biological ligands for various medical applications. In biological environment, the corona enveloped on GNP ligands is composed of proteins that interact directly and indirectly with the ligands, which can spatially hinder their functions. However, there is currently no effective method for capturing and distinguishing the ligand-binding corona to elucidate the blocking mechanisms. Herein, we propose a proteomics strategy based on the use of two cross-linkers-formaldehyde and bis(sulfosuccinimidyl) suberate-and mass spectrometry (MS). Cysteine and glutathione were used as models for amino-free ligands and connected to GNPs. After the formation of the plasma protein corona, the interactions of the GNP ligands and coronal proteins were fixed through chemical cross-linking. The ligand-binding proteins were isolated and obtained through stringent washing, and they were then subjected to proteomic analysis. The proteins having the MS-mapped cross-linked sites were considered to belong to the corona that was bound directly to the GNP ligands. The proteomic analysis revealed the features of the proteins in the ligand-binding corona, and the direct corona displayed distinctive physicochemical properties and functional profiles. Further, protein-protein interaction network analysis of the ligand-binding corona demonstrated that the architectures and behaviors of the direct and indirect corona are potentially affected by the properties of the ligand such as its size. Our strategy has the capability to specifically characterize the protein composition of the direct corona bound to amino-free GNP ligands, providing an effective way to investigate the impacts on the functions of the ligand.
