Abstract:We theoretically demonstrate the creation of sub-wavelength focal spot with ultra long depth of focus (DOF) non-diffracting. To achieve a sub-half-wavelength focal spot (0.4λ) with modulation, extended DOF (over 9.5λ) in vacuum, an integration of amplitude phase encoding, cylindrical polarization and the combination of dual-beams is utilized. Further analysis also predicts that a flattop intensity distribution along z axis can be observed around the ideal focus point.

Abstract:Holographic techniques enable precise laser manipulation, but suffer from two considerable limitations: speckle and deterioration of axial distribution. Here, we propose a cylindrical quadratic phase (CQP) method with temporal focusing (TF) to generate speckle-free holographic illumination with high axial resolution. TF-CQP utilizes a superposed cylindrical phase as the initial guess to iteratively optimize phase hologram, realizing speckle-free holographic reconstruction on the target focal plane and eliminating secondary focus on the defocused planes. TF-CQP further disperses defocused beams symmetrically by a blazed grating, placed conjugate to the focal plane, which enhances axial confinement. Simulation and experimental results show that TF-CQP reconstructs speckle-free illumination with arbitrary shapes and <10 µm axial resolution. Compared to TF-GS (Gerchberg-Saxton algorithm), widely used in holographic optogenetics, TF-CQP shows increased uniformity of 200% and improved modulation efficiency of 32.33% for parallel holographic illumination, as well as a 10% increment in axial resolution.

Abstract:Random phase masks can transform the defocus noise into a speckle-like pattern in optical scanning holography (OSH). In this Letter, we presented a speckle reduction based on combined frame difference and connected component method in a random phase-coded OSH system. The image quality of the reconstructed sections is improved with better visibility.

Abstract:In holographic reconstruction, the two-order and twin image noise produced by the phase-conjugate wave arising from the symmetry of the complex field, will affect the accuracy and the image quality of the reconstructed image. The acquisition of the holographic focus distance parameter for judging the best focal plane of object is also the key to holographic imaging. In this work, a numerical focusing imaging algorithm based on compressive sensing is proposed. By choosing a stack of reconstructed images in the axial direction with fixed interval, every image is processed by the total variation regularization. These images are used to calculate and construct the focus curve of the digital holography according to the focus metrics such as the tamura coefficient and the absolute gradient operator. Then the best focus plane is evaluated. The proposed method suppresses the noise of in -line digital reconstruction, improves the axial accuracy of reconstructing, and realizes a larger range of high-resolution imaging around the focus plane.

Abstract:In this Letter, an autofocusing method in optical scanning holography(OSH) system is proposed. By introducing Lissajous scanning into multiple signal classification(MUSIC) method in time-reversal(TR) OSH, the axial locations of the targets can be retrieved with better resolution and the peak prominence increases from 0.21 to 0.34. The feasibility of this method is confirmed by simulation as well as experiment.

Abstract:Holography is widely used for imaging plankton because of its large depth of field. Autofocusing is a critical step in holographic imaging, where the optimal focal plane is determined by locating the extremum of the evaluation function curve. However, most of these methods perform auto-focusing on single focus holograms. To apply these methods to multi-focus holograms, the hologram must be subjected to object segmentation. It is difficult to achieve good results when focusing on holograms with small objects and large depth of field. Therefore, we propose a multi-focus autofocus method based on connected domain analysis (CDA), which allows focusing on multiple objects at different focal lengths simultaneously without image segmentation. Each object generates a focus evaluation curve, resulting in high focusing accuracy. We set up an in-line holographic optical system with a 30 mm depth of field and recorded holograms of Nauplius, Cladocera, Rotifera, Copepods, Noctiluca scintillans and real seawater samples. We demonstrated the robustness of the CDA algorithm for different target types and its effectiveness in focusing multi-focus images by applying it to holograms of six types of samples.
Versatile optimization-based speed-up method for autofocusing in digital holographic microscopy

Abstract:We propose a speed-up method for the in-focus plane detection in digital holographic microscopy that can be applied to a broad class of autofocusing algorithms that involve repetitive propagation of an object wave to various axial locations to decide the in-focus position. The classical autofocusing algorithms apply a uniform search strategy, i.e., they probe multiple, uniformly distributed axial locations, which leads to heavy computational overhead. Our method substantially reduces the computational load, without sacrificing the accuracy, by skillfully selecting the next location to investigate, which results in a decreased total number of probed propagation distances. This is achieved by applying the golden selection search with parabolic interpolation, which is the gold standard for tackling single-variable optimization problems. The proposed approach is successfully applied to three diverse autofocusing cases, providing up to 136-fold speed-up.

Coherent Noise Reduction in Digital Holographic Interferometry by Slightly Rotating Object

Abstract:The coherent noise degrades the imaging quality and resolution in digital holographic interferometry. A method to reduce coherent noise by the way of slightly rotating object is proposed. Firstly, a series of digital holograms with different coherent noise patterns were recorded by slightly rotating object. Secondly, the different holograms were reconstructed individually, and the differences between the reconstructed complex amplitudes due to the rotation of the object were removed by using phase compensation and image registration algorithms. Thus, multiple identical retrieved images of object with different coherent noise distributions were obtained. Finally, the coherent noise was reduced by a proper averaging process for amplitude and phase images. Moreover, Non-correlation between two speckle patterns can be achieved by rotating very small angle proven by means of computing the correlation between the circle speckle spots. The experimental results and evaluations are given to confirm the proposed method.

Abstract:Optical scanning holography(OSH)records both the amplitude and phase information of a 3D object by a 2D scan.To recon-struct a 3D volumetric image from an OSH hologram is difficult,as it suffers from the defocus noise from the other sections.The use of a random phase pupil can convert defocus noise into speckle-like noise,which may require further processing in sectional image reconstruction.In this paper,we propose a U-shaped neural network to reduce this speckle haze.Simulation results show that the proposed method works effectively and efficiently both in simple and complex graphics.

Abstract:A method is proposed to suppress speckle noise using only part of the pixels in a single-exposure digital hologram. Different holographic patterns are first generated from a single-exposure digital hologram using specially designed binary masks; then, these holographic patterns are reconstructed according to the Fresnel transform. The reconstructed images are superposed and averaged on the intensity to achieve the suppression of speckle noise. The entire denoising process does not need any additional digital holograms or specific requirements for recording a hologram. Theoretical simulation and experiment verification were carried out and confirm that the proposed method is a very convenient and effective way to suppress speckle noise in digital holography. Abstract:The coherent noise appears in the constructed image of digital holographic microscopy due to the laser source; thus, the imaging quality is degraded. A method of coherent noise reduction using a laterally shifting hologram aperture is presented. An original hologram with coherent noise is captured by a camera first. A series of holograms are sampled by laterally shifting the digital aperture in the original hologram. Instead of extracting the specimen's part information, each sampled hologram, which includes the whole specimen, is reconstructed. The coherent noise is reduced by averaging the different reconstructed images. The experiment demonstrates the feasibility of the approach. The presented approach with a single recorded hologram realizes the coherent noise reduction without loss of spatial resolution, which is useful for real- time measurement. Abstract:A method to reduce coherent noise in digital holographic phase contrast microscopy is proposed. By slightly shifting the specimen, a series of digital holograms with different coherent noise patterns is recorded. Each hologram is reconstructed individually, while the different phase tilts of the reconstructed complex amplitudes due to the specimen shifts are corrected in the hologram plane by using numerical parametric lens method. Afterward, the lateral displacements of the phase maps from different holograms are compensated in the image plane by using digital image registration method. Thus, all phase images have same distribution, but uncorrelated coherent noise patterns. By a proper averaging procedure, the coherent noise of phase contrast image is reduced significantly. The experimental results are given to confirm the proposed method.

Abstract:An auto-focusing method for digital holographic microscopy has been proposed by employing two off-axis illumination beams. When specimens are illuminated by two plane waves in different directions, it is found that the farther the reconstruction plane is from the image plane, the wider the two reconstructed images are separated from each other. Thus, the image plane can be determinated by finding the minimum of the variation between the two reconstructed object waves on both the amplitude and phase distributions. The feasibility of the proposed method is demonstrated by the corresponding simulation and experiment.

Abstract:Due to the limited processing accuracy of the platform and unevenness of the glass substrate itself, a holographic lithography system is prone to out-of-focus imaging problems; therefore, the real-time focusing components are critical for holographic lithography systems. In this paper, a real-time focus monitoring and adjusting system using an electrically tunable lens (ETL) for large-area lithography is introduced. Combined with the ETL, the limited depth of field of the microscopic objective has been effectively expanded, and the automatic focusing evaluation and adjustment are achieved. The development, including simulation using Zemax, optics system design and implementation, experiments, and evaluation are demonstrated in this paper. The results show that the out-of-focus problem in our large-area holographic lithography system has been significantly alleviated.

Abstract:An autofocusing method is proposed that utilizes cosine score of inner angle between the vectors resulted from vectoring the axial adjacent reconstructed images in digital holography. It is based on the fact that the images near the focus contain more regular features of object than in the defocused region, therefore, the neighboring reconstructed images are more similar to each other at the focus position than defocused and a cosine score is employed to evaluate such similarity. However, the cosine scores between the axial adjacent amplitude images are so close that it is difficult to distinguish the extremum. Therefore, a modified cosine algorithm is presented to offset such problem on consideration of the correlation of the elements, by subtracting the inner product term from the denominator of the cosine algorithm. The cosine and modified cosine score based autofocusing method procedure is first introduced, and then it is utilized in simulation and real holographic data. In simulation, it precisely judges out the actual recording distance and the focus curve shows good focus function criteria, which verifies the method as an ideal circumstance. In real experiment, it can easily search out the focus distance from the focus curve, and it shows good focus judgement ability than most traditional focus metrics selected. Therefore, the feasibility and validation of the proposed autofocusing method are proved by simulation and experiment results.
Minimum-Entropy-Based Adaptive Focusing Algorithm for Image Reconstruction of Terahertz Single-Frequency Holography With Improved Depth of Focus

Abstract:In this paper, the defocus of a 2-D image reconstruction of targets illuminated by Gaussian beams in terahertz (THz) single-frequency holography was studied. An analytical point-spread function was derived to quantitatively evaluate the defocus effect, considering the deviation of the restoration distance from the real distance of the target. It is found that the cross-range image defocuses in a similar way to the diffusion of a Gaussian beam from its beam waist, which limits the depth of focus for image restoration. To improve the restored images for targets with varying range distances, an adaptive focusing algorithm based on the minimum-entropy method was proposed for single-frequency holography. Simulation results with fairly good agreement were performed to verify the theoretical results and the proposed algorithm. Proof-of-principle experiments in the 0.2-THz band were also performed based on a monostatic prototype imager with a Gaussian beam transceiver. The experimental results confirm the effectiveness of the adaptive focusing reconstruction algorithm proposed in this paper.

Abstract:Holographic reconstruction is affected by the phase-conjugate wave arising from the symmetry of the complex field. Compressive sensing (CS) has been used in in-line digital holography (DH) to eliminate noise, especially the interference from twin images. Herein, CS with total variation regularization combining autofocusing is presented. It provides an autofocusing function from a single-exposure hologram and obtains reconstructed objects without twin image noise. A series of images at a fixed interval within a reconstruction distance are processed using a two-step iterative shrinkage/thresholding algorithm in CS. It can calculate the focus distance in a larger range around the focal plane using twin-image-free reconstruction, so it can achieve a higher focusing accuracy than traditional focusing methods, including the Laplace operator, absolute gradient operator, and Tamura coefficient. The proposed method is a simple combination of algorithms and a powerful extension that can effectively improve simulated and experimental image quality and handle difficult datasets, which existing algorithms cannot.

Abstract:In this Letter, we propose a new method for auto-focusing and reconstruction without defocus noise in optical scanning holography. By using a connected domain (CD) to calculate the area of different domains, which are labeled by a connected component, the focus distance can be found via the smallest area of each CD. Meanwhile, the sectional images without defocus noise can also be reconstructed based on the labeled domains. The effectiveness of this method has been verified with a simulation and experiments.

Each hologram is reconstructed individually, while the differences between these retrieved complex amplitudes due to the object shifting are corrected with phase compensation and image registration algorithms. So all reconstructed complex amplitudes of object have same distribution, but uncorrelated coherent noise patterns. The speckle noise is well suppressed by averaging these reconstructed images. The theory and experimental results prove that the method is effective and feasible.
