Abstract:Brownian motion allows microscopically dispersed nanoparticles to be stable in ferrofluids, as well as causes magnetization relaxation and prohibits permanent magnetism. Here we decoupled the particle Brownian motion from colloidal stability to achieve a permanent fluidic magnet with high magnetization, flowability and reconfigurability. The key to create such permanent fluidic magnets is to maintain a stable magnetic colloidal fluid by using non-Brownian magnetic particles to self-assemble a three-dimensional oriented and ramified magnetic network structure in the carrier fluid. This structure has high coercivity and permanent magnetization, with long-term magnetization stability. We establish a scaling theory model to decipher the permanent fluid magnet formation criteria and formulate a general assembly guideline. Further, we develop injectable and retrievable permanent-fluidic-magnet-based liquid bioelectronics for highly sensitive, self-powered wireless cardiovascular monitoring. Overall, our findings highlight the potential of permanent fluidic magnets as an ultrasoft material for liquid devices and systems, from bioelectronics to robotics.

Abstract:The purpose of this study was to develop an instant MR cell labeling technique, called magnetosonoporation. First, a magnetosonoporation apparatus was successfully established for MR labeling of stem cells. Then, the safety of this new cell labeling approach was confirmed by evaluation of cell viability, proliferation, and differentiation of magnetosonoporation-labeled and unlabeled C17.2 neural stem cells. Subsequently, the feasibility of using in vivo MRI to detect magnetosonoporation/Feridex-labeled stem cells was validated in living animals and confirmed by histologic correlation. The magnetosonoporation technique is expected to be convenient, efficient, and safe for future clinical application of MRI-guided cell therapies.

Abstract:We report the fabrication and characterisation of magnetic liquid beads with a solid magnetic shell and liquid core using microfluidic techniques. The liquid beads consist of a fluorinated oil core and a polymer shell with magnetite particles. The beads are generated in a flow-focusing polydimethylsiloxane (PDMS) device and cured by photo polymerisation. We investigated the response of the liquid beads to an external magnetic field by characterising their motion towards a permanent magnet. Magnetic sorting of liquid beads in a channel was achieved with 90% efficiency. The results show that the liquid beads can be controlled magnetically and have potential applications in digital microfluidics including nucleic acid amplification, drug delivery, cell culture, sensing, and tissue engineering. The present paper also discusses the magnetophoretic behaviour of the liquid bead by varying its mass and magnetite concentration in the shell. We also demonstrated the two-dimensional self-assembly of magnetic liquid beads for potential use in digital polymerase chain reaction and digital loop mediated isothermal amplification.

A Hand-Held Magnetic Acoustic Device With Integrated Real-Time Monitoring for Targeted Drug Delivery

Abstract:Advances in magnetic materials have enabled the development of new therapeutic agents that can be localized by external magnetic fields. These agents offer a potential means of improving treatment targeting and reducing the toxicity-related side effects associated with systemic delivery. Achieving sufficiently high magnetic fields at clinically relevant depths in vivo, however, remains a challenge. Similarly, there is a need for techniques for real-time monitoring that do not rely on magnetic resonance imaging (MRI). Here, we present a hand-held device to meet these requirements, combining an array of permanent magnets and a thin 64-element capacitive micromachined ultrasonic transducer (CMUT) interfaced to a real-time imaging system. Drug carrier localization was assessed by measuring the terminal velocity of magnetic microbubbles in a column of fluid above the magnetic array. It was found that the magnetic pull force was sufficient to overcome buoyancy at equivalent tissue depths of at least 35 mm and that the median terminal velocity ranged from 0.7 to 20 [Formula: see text]/s over the distances measured. A Monte Carlo study was performed to estimate capture effectiveness in tumor microvessels over a range of different tissue depths and flow rates. Finally, B-mode and contrast-enhanced ultrasound (CEUS) imaging were demonstrated using a gel flow phantom containing a 1.6-mm diameter vessel. Real-time monitoring provided visual confirmation of retention of magnetic microbubbles along the vessel wall at a flow rate of 0.5 mL/min. These results indicate that the system can successfully retain and image magnetic microbubbles at tissue depths and flow rates relevant for clinical applications such as molecular ultrasound imaging of atherosclerosis, sonodynamic and antimetabolite cancer therapy, and clot dissolution via sonothrombolysis.

Abstract:Superparamagnetic nanoparticles can be attached in great numbers to pathogenic cells using specific antibodies so that the magnetically-labeled cells themselves become superparamagnets. The cells can then be manipulated and drawn out of biological fluids, as in a biopsy, very selectively using a magnetic needle. We examine the origins and uncertainties in the forces exerted on magnetic nanoparticles by static magnetic fields, leading to a model for trajectories and collection times of dilute superparamagnetic cells in biological fluids. We discuss the design and application of such magnetic needles and the theory of collection times. We compare the mathematical model to measurements in a variety of media including blood. Abstract:Molecular monitoring in the dermal interstitial fluid (ISF) is an attractive approach to painlessly screen markers of health and disease status on the go. One promising strategy for accessing ISF involves the use of wearable patches containing microneedle sensor arrays. To date, such microneedle sensors have been fabricated via various manufacturing strategies based on injection molding, machining, and advanced lithography to name a few. Our groups previously reported 3D-printed microneedles as a convenient and scalable approach to sensor fabrication that, when combined with aptamer-based molecular measurements, can support continuous molecular monitoring in ISF. However, the original platform suffered from poor patch stability when deployed on the skin of rodents in vivo. We identified that this problem was due to the rheological properties of the rodent skin, which can contract post microneedle placement, physically pushing the microneedles out of the skin. This sensor retraction caused a loss of electrical contact between working and reference needles, irreversibly damaging the sensors. To address this problem, we report here an innovative approach that allows magnetic placement of microneedle sensor arrays on the skin of live rodents, affixing the patches under light pressure that prevents needle retraction. Using this strategy, we achieved sensor signaling baselines that drift at rates comparable to those seen with other in vivo deployments of electrochemical, aptamer-based sensors. We illustrate real-time pharmacokinetic measurements in live Sprague-Dawley rats using SLA-printed, aptamer-functionalized microneedles and demonstrate their ability to support drift correction via kinetic differential measurements. We also discuss future prospects and challenges.

Abstract:Magnetic nanoparticles (MNPs) have garnered significant attention in biomedical applications. Due to their large surface area and tunable properties, MNPs are used in microfluidic systems, which allow for the manipulation and control of fluids at micro- or nanoscale. Using microfluidic systems allows for a faster, less expensive, and more efficient approach to applications like bioanalysis. MNPs in microfluidics can precisely identify and detect bioanalytes on a single chip by controlling analytes in conjunction with magnetic particles (MPs) and separating various particles for analytical functions at the micro- and nanoscales. Numerous uses for these instruments, including cell-based research, proteomics, and diagnostics, have been reported. The successful reduction in the size of analytical assays and the creation of compact LOC platforms have been made possible with the assistance of microfluidics. Microfluidics is a highly effective method for manipulating fluids as a continuous flow or discrete droplets. Since the implementation of the LOC technology, various microfluidic methods have been developed to improve the efficiency and precision of sorting, separating, or isolating cells or microparticles from their original samples. These techniques aim to surpass traditional laboratory procedures. This review focuses on the recent progress in utilizing microfluidic systems that incorporate MNPs for biological applications.

Abstract:Reliable, time-resolved detection of biochemical markers is critical for providing actionable healthcare data. Biosensors can track various clinically relevant biomarkers to inform clinicians to make data-driven decisions. However, the notorious effects of complex fluids limit the lifetime of biosensors by fouling the active sensing sites, hindering their long-term functionality. Biomaterials with passive antifouling capabilities have been developed to minimize the effects of biofouling. However, these attempts can only delay the unavoidable outcome of fouling accumulation, which still limits long-term device functionality. Here, we present a novel class of biosensor capable of self-clearing their sensing surface that can avoid and actively remove biofoulants from the electrode surfaces. The active anti-biofouling modality consists of an electrochemical sensing platform embedded with magnetic microactuators previously proven to disrupt protein attachment and blood clot formation. In this study, we characterize the static and dynamic mechanical responses of the torsional thin-film magnetic microactuators. We confirm its protein-clearing capabilities using fluorescent bovine serum albumin. To quantify the advantages of magnetic actuation using electrochemical techniques, we conducted cyclic voltammetry and electrochemical impedance spectroscopy experiments. We believe this work presents a pivotal step towards enabling a truly longitudinal and continuous monitoring of multiple analytes in a complex in vivo environment for a truly personalized medicine.

Abstract:Microfluidics is assumed to be one of the leading and most promising areas of research since the early 1990s. In microfluidic systems, small spherical magnetic particles with superparamagnetic properties, called magnetic beads, play an important role in the design of innovative methods and tools, especially in bioanalysis and medical sciences. The intention of this review paper is to address main aspects from the state-of-the-art in the area of magnetic bead research, while demonstrating the broad variety of applications and the huge potential to solve fundamental biological and medical problems in the fields of diagnostics and therapy. Basic issues and demands related to the fabrication of magnetic particles and physical properties of nanosize magnets are discussed in Section 2. Of main interest are the control and adjustment of the nanoparticles' properties and the availability of adequate approaches for particle detection via their magnetic field. Section 3 presents an overview of magnetic bead applications in nanomedicine. In Section 4, practical aspects of sample manipulation and separation employing magnetic beads are described. Finally, the benefits related to the use of magnetic bead-based microfluidic systems are summarized, illustrating ongoing questions and open tasks to be solved on the way to an approaching microfluidic age.

Abstract:The detection of magnetic nanoparticles in a liquid medium and the quantification of their concentration have the potential to improve the efficiency of several relevant applications in different fields, including medicine, environmental remediation, and mechanical engineering. To this end, sensors based on the magneto-impedance effect have attracted much attention due to their high sensitivity to the stray magnetic field generated by magnetic nanoparticles, their simple fabrication process, and their relatively low cost. To improve the sensitivity of these sensors, a multidisciplinary approach is required to study a wide range of soft magnetic materials as sensing elements and to customize the magnetic properties of nanoparticles. The combination of magneto-impedance sensors with ad hoc microfluidic systems favors the design of integrated portable devices with high specificity towards magnetic ferrofluids, allowing the use of very small sample volumes and making measurements faster and more reliable. In this work, a magneto-impedance sensor based on an amorphous Fe73.5Nb3Cu1Si13.5B9 wire as the sensing element is integrated into a customized millifluidic chip. The sensor detects the presence of magnetic nanoparticles in the ferrofluid and distinguishes the different stray fields generated by single-domain superparamagnetic iron oxide nanoparticles or magnetically blocked Co-ferrite nanoparticles.
Polyacrylamide Ferrogels with Magnetite or Strontium Hexaferrite: Next Step in the Development of Soft Biomimetic Matter for Biosensor Applications

Abstract:Magnetic biosensors are an important part of biomedical applications of magnetic materials. As the living tissue is basically a "soft matter." this study addresses the development of ferrogels (FG) with micron sized magnetic particles of magnetite and strontium hexaferrite mimicking the living tissue. The basic composition of the FG comprised the polymeric network of polyacrylamide, synthesized by free radical polymerization of monomeric acrylamide (AAm) in water solution at three levels of concentration (1.1 M, 0.85 M and 0.58 M) to provide the FG with varying elasticity. To improve FG biocompatibility and to prevent the precipitation of the particles, polysaccharide thickeners-guar gum or xanthan gum were used. The content of magnetic particles in FG varied up to 5.2 wt % depending on the FG composition. The mechanical properties of FG and their deformation in a uniform magnetic field were comparatively analyzed. FG filled with strontium hexaferrite particles have larger Young's modulus value than FG filled with magnetite particles, most likely due to the specific features of the adhesion of the network's polymeric subchains on the surface of the particles. FG networks with xanthan are stronger and have higher modulus than the FG with guar. FG based on magnetite, contract in a magnetic field 0.42 T, whereas some FG based on strontium hexaferrite swell. Weak FG with the lowest concentration of AAm shows a much stronger response to a field, as the concentration of AAm governs the Young's modulus of ferrogel. A small magnetic field magnetoimpedance sensor prototype with Co68.6Fe3.9Mo3.0Si12.0B12.5 rapidly quenched amorphous ribbon based element was designed aiming to develop a sensor working with a disposable stripe sensitive element. The proposed protocol allowed measurements of the concentration dependence of magnetic particles in gels using magnetoimpedance responses in the presence of magnetite and strontium hexaferrite ferrogels with xanthan. We have discussed the importance of magnetic history for the detection process and demonstrated the importance of remnant magnetization in the case of the gels with large magnetic particles.

Abstract:Needle-free injector technology (NFIT) has drawn attention due to their advantages. By using NFIT, it can be used several time unlike a conventional needle. Also, it makes patients free from pain. Since NFIT by jet injection is achieved by ejecting a liquid drug through a narrow orifice at high pressure, thereby creating a fine high-speed fluid jet that can readily penetrate skin and tissue. Until very recently, all jet injectors utilized force-and pressure-generating principles that progress injection in anuncontrolled mannerwith limited ability to regulate delivery volume and injection depth. In order to address these shortcomings, we have developed a controllable jet injection device, based on a custom high-stroke linear Lorentz-force motor. Using this device, we are able to monitor and modulate continuously the speed of the drug jet, and regulate precisely the volume of drug delivered during the injection process.

Abstract:Superparamagnetic nanoparticles are attracting significant attention. Therefore, being explored in microsystems for a wide range of applications. Typical examples include lab-on-a-chip and microfluidics for synthesis, detection, separation, and transportation of different bioanalytes, such as biomolecules, cells, and viruses to develop portable, sensitive, and cost-effective biosensing systems. Particularly, microfluidic systems incorporated with magnetic nanoparticles and, in combination with magnetoresistive sensors, shift diagnostic and analytical methods to a microscale level. In this context, nanotechnology enables the miniaturization and integration of a variety of analytical functions in a single chip for manipulation, detection, and recognition of bioanalytes reliably and flexibly. In consideration of the above, recent development and benefits are elaborated herein to discuss the role of magnetic nanoparticles inside the microchannels to design highly efficient disposable point-of-care applications from transportation to the detection of bioanalytes.
Development of a Microfluidic Chip System with Giant Magnetoresistance Sensor for High-Sensitivity Detection of Magnetic Nanoparticles in Biomedical Applications

Abstract:Magnetic nanoparticles (MNPs) have been widely utilized in the biomedical field for numerous years, offering several advantages such as exceptional biocompatibility and diverse applications in biology. However, the existing methods for quantifying magnetic labeled sample assays are scarce. This research presents a novel approach by developing a microfluidic chip system embedded with a giant magnetoresistance (GMR) sensor. The system successfully detects low concentrations of MNPs with magnetic particle velocities of 20 mm/s. The stray field generated by the magnetic subject flowing through the microchannel above the GMR sensor causes variations in the signals. The sensor's output signals are appropriately amplified, filtered, and processed to provide valuable indications. The integration of the GMR microfluidic chip system demonstrates notable attributes, including affordability, speed, and user-friendly operation. Moreover, it exhibits a high detection sensitivity of 10 μg/μL for MNPs, achieved through optimizing the vertical magnetic field to 100 Oe and the horizontal magnetic field to 2 Oe. Additionally, the study examines magnetic labeled RAW264.7 cells. This quantitative detection of magnetic nanoparticles can have applications in DNA concentration detection, protein concentration detection, and other promising areas of research.

Abstract:The physical properties of magnetic liquids are theoretically analyzed in this paper, such as the effect of the magnetic volume force on the free surface of magnetic liquids, the density spectrum of the magnetic liquids based on horizontal component of the magnetic volume force, the first order levitation principle and the second order levitation principle, the magneto-electric principle of magnetic liquids, the fluidity of magnetic liquids used as magnetic core and the fluxgate effect of magnetic liquids. Some methods to study the properties of magnetic liquids were provided by the authors, and the effects of the magnetic volume force on the free surface of magnetic liquids and the second order levitation effect were experimentally studied in this paper. The analysis methods that provided in this paper may be applied to the design of new sensors.

Some characteristics of the sensor are determined by theoretical predictions based on the state equation of ideal gas. The variation of external pressure will lead to the compression of a chamber between the body and mobile part, and a corresponding signal occurs.
