Astaxanthin Regulates High-Carbohydrate Diet-Induced Oxidative Stress, Inflammation and Apoptosis in an Organ-Specific Manner in Largemouth Bass (micropterus Salmoides)

Abstract:Largemouth bass ( Micropterus salmoides) ) is widely considered as carbohydrate-intolerant, and immunological disorders would be induced when fed with high-carbohydrate (HC) diets. Astaxanthin (AST) has been exploited as an excellent antioxidant for counteracting oxidative stress, whereas its underlying mechanisms remain to be further unveiled. The present study thus investigated the effects of AST on regulations of oxidative stress, inflammation and apoptosis in largemouth bass subjected to HC diet. Fish were randomly assigned to control group (Con), high-carbohydrate group (HC) and high-carbohydrate plus 0.1% AST group (HCA) for an eight- week rearing trial. Preliminary results showed that AST significantly improved growth performance (increased final body weight and weight gain rate) and mitigated liver damage (decreased hepatosomatic index, improved hepatocyte integrity and decreased serum transaminase levels) of fish fed with HC diet. Further investigations elucidated that: (1) Keap1-Nrf2-ARE axis was crucially involved in solving the oxidative stress caused by HC diet, and NF-kappa B kappa B pathway was induced as well by redox imbalance to exert inflammatory reactions; (2) HC diet remarkably induced oxidative stress, inflammation and apoptosis in liver, which were all effectively attenuated by AST treatment; (3) In head kidney, anti-oxidative and apoptotic changes were caused by HC diet, and AST specifically augmented anti-oxidative activities and alleviated apoptosis; (4) In spleen, the inflammation was prominently mitigated via inhibition of NF-kappa B kappa B pathway under AST treatment, while no remarkable effects were observed on the regulation of anti-oxidative and apoptotic statuses by AST; (5) Oxidative stress, inflammatory and apoptotic responses were all significantly triggered in intestine by HC treatment, whereas AST seemed to exert negligible effects on related biological disorders. Our study mechanistically demonstrated that HC diet caused remarkable immunological disorders and tissue impairment through inducing exorbitant oxidative stress, inflammation and apoptosis, and AST mitigated these damages in an organ-specific manner.
