Ultrasound-triggered Cascade Reaction Via MnO2-CpG Nanoparticles for Boosting Pyroptosis-Mediated Cancer Immunotherapy

Abstract:Nanoparticles is promising for enhancing cancer immunotherapy efficacy via the regulation of tumor microenvironment (TME), but the efficiency of regulation remains challenging. Herein, we proposed a simple strategy to construct a MnO2-CpG nanoparticles that is composed of MnO2 and immunostimulatory nucleic acid cytosine-phosphate-guanine oligonucleotides (CpG ODNs). We discovered that MnO2-CpG nanoparticles alleviated the hypoxic and immunosuppressive TME while enhancing systematic antitumor immune responses. Notably, combined with local ultrasound (US) irradiation, the MnO2-CpG nanoparticles exhibited numerous US-amplified cascade effects, including increased cellular uptake, enhanced lysosome escape, improved chemodynamic therapy and sonodynamic therapy, ultimately activating the Caspase-1 mediated pyroptosis signaling pathway to trigger abundant neoantigen exposure. The cascade-amplified release of immunostimulatory agents Mn2+ and CpG ODNs further enhanced antitumor immune responses. Overall, our study provides guidance for the rational design of nanoplatforms based on high-performance US-triggered cascade amplification, serving as cancer immunotherapy boosters for triple-negative breast cancer patients.
