Overexpression of BnaC8.NRT2.1b Improves Nitrogen Uptake Efficiency in Brassica Napus L

Abstract:Rapeseed (Brassica napus L.) is a worldwide important oil crop that requires more nitrogen (N) than cereal crops. However, overuse of N fertilizer has negative environmental impacts. Therefore, it is imperative to investigate the rapeseed’s potential for enhancing N use efficiency (NUE). Nitrate transporters play a pivotal role in the N absorption and utilization by plants, yet limited information is available regarding their functions in rapeseed. Here, we characterized the biological function of BnaC8.NRT2.1b, a nitrate transporter gene that is mainly expressed in roots, leaves, and stamens, and induced in roots after exposure to low nitrate stress. BnaC8.NRT2.1b localizes to the plasma membrane and interacts with chaperonin BnaC2.NAR2.1. Overexpression of BnaC8.NRT2.1b complemented the phenotype of Arabidopsis (Arabidopsis thaliana L.) mutant atnrt2.1 under low N conditions, significantly improving grain yield, seed/plant N accumulation, N uptake efficiency (NUpE), and biomass in Arabidopsis as well as rapeseed biomass and NUpE. These results indicate that BnaC8.NRT2.1b positively regulates NUE and has great potential for improving rapeseed N uptake/utilization. This study lays the groundwork for the biotechnology-driven cultivation of N-efficient rapeseed varieties, which are essential for sustainable agricultural development.
