Abstract:Objective: To prepare an inclusion complex of daidzein and hydropropyl-β-cyclodextrin to enhance the solubility of daidzein. Method: The inclusion complex of daidzein was prepared by the solution stirring method. The binary system of daidzein and HP-β-CD was confirmed by differential thermal, thermogravimetry analysis, infrared spectroscopy and X-ray diffractometry. Result: The drug content in the inclusion complex was 6.76% and the solubility was 13.68 mg·mL-1. The identification results showed that the inclusion complex was formed. Conclusion: The preparation method of the inclusion complex of daidzein and hydropropyl-β-cyclodextrin is simple and available, with a increased solubility of daidzein.

Abstract:The ginger essential oil/β-cyclodextrin (GEO/β-CD) composite, ginger essential oil/β-cyclodextrin/chitosan (GEO/β-CD/CTS) particles and ginger essential oil/β-cyclodextrin/chitosan (GEO/β-CD/CTS) microsphere were prepared with the methods of inclusion, ionic gelation and spray drying. Their properties were studied by using scanning electron microscopy (SEM), differential scanning calorimetry (DSC), thermo-gravimetry analysis (TGA), Fourier transform infrared spectroscopy (FT-IR) and X-ray diffraction (XRD). The results showed that the particle size of GEO/β-CD composite was smaller than that of β-CD and GEO/β-CD/CTS particles were loose and porous, while the microsphere obtained by spray drying had certain cohesiveness and small particle size. Besides, results also indicated that β-CD/CTS could modify properties and improve the thermal stability of GEO, which would improve its application value in food and medical industries.
[Preparation and in vitro property evaluation of β-cyclodextrin-daidzein/PEG_(20000)/Carbomer_(940) nanocrystals]

Abstract:This study aims to improve the solubility and bioavailability of daidzein by preparing the β-cyclodextrin-daidzein/PEG_(20000)/Carbomer_(940) nanocrystals. Specifically, the nanocrystals were prepared with daidzein as a model drug, PEG_(20000), Carbomer_(940), and NaOH as a plasticizer, a gelling agent, and a crosslinking agent, respectively. A two-step method was employed to prepare the β-cyclodextrin-daidzein/PEG_(20000)/Carbomer_(940) nanocystals. First, the insoluble drug daidzein was embedded in β-cyclodextrin to form inclusion complexes, which were then encapsulated in the PEG_(20000)/Carbomer_(940) nanocrystals. The optimal mass fraction of NaOH was determined as 0.8% by the drug release rate, redispersability, SEM morphology, encapsulation rate, and drug loading. The inclusion status of daidzein nanocrystals was determined by Fourier transform infrared spectroscopy(FTIR), thermogravimetric analysis(TGA), and X-ray diffraction(XRD) analysis to verify the feasibility of the preparation. The prepared nanocrystals showed the average Zeta potential of(-30.77±0.15)mV and(-37.47±0.64)mV and the particle sizes of(333.60±3.81)nm and(544.60±7.66)nm before and after daidzein loading, respectively. The irregular distribution of nanocrystals before and after daidzein loading was observed under SEM. The redispersability experiment showed high dispersion efficiency of the nanocrystals. The in vitro dissolution rate of nanocrystals in intestinal fluid was significantly faster than that of daidzein, and followed the first-order drug release kinetic model. XRD, FTIR, and TGA were employed to determine the polycrystalline properties, drug loading, and thermal stability of the nanocrystals before and after drug loading. The nanocrystals loaded with daidzein demonstrated obvious antibacterial effect. The nanocrystals had more significant inhibitory effects on Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa than daidzein because of the improved solubility of daidzein. The prepared nanocrystals can significantly increase the dissolution rate and oral bioavailability of the insoluble drug daidzein.

Abstract:Abstract Dihydroquercetin (DHQ) is a natural occurring dihydroflavonol that has strong antioxidant and antibacterial activities. However, its application is limited due to its poor solubility. This study aims to improve the aqueous solubility of DHQ by complexing DHQ with β-cyclodextrin (β-CD) to boost its biological activity. DHQ was encapsulated with β-CD by freeze drying at a 1:1-M ratio. The structure of DHQ/β-CD complex prepared was elucidated by using Fourier transform infrared spectroscopy, differential scanning calorimetry, X-ray diffraction, scanning electron microscopy, and 1 H nuclear magnetic resonance ( 1 H NMR). In addition, molecular docking further revealed two energetically favorable conformations of the DHQ/β-CD complex, in which DHQ interacted with β-CD via hydrogen bonds. Experimental results showed that the solubility of the DHQ increased 22.63-fold by encapsulating with β-CD. Also the dissolution rate, antioxidant activity and antibacterial activity of the DHQ were significantly improved by encapsulating. The encapsulating with β-CD solves the problem of the poor aqueous solubility of DHQ, and broadens the path for a more optimal use of the health promoting effect of DHQ in pharmaceutical and food products.

Abstract:Abstract In our work, the inclusion ability and binding pattern of flavonoids (catechin, iso‐liquiritigenin, luteolin, puerarin, rutin, and curcumin) with β‐cyclodextrin (β‐CD) and curcumin (CUR) with CDs (α‐CD, β‐CD, γ‐CD, dimethyl‐β‐CD, Hydroxypropyl‐β‐CD, and glucosyl‐β‐CD) were studied by molecular docking. The results showed that CUR among the six flavonoids was more likely to form stable inclusion complexes with β‐CD, and the HP‐β‐CD is the most suitable for CUR inclusion among the 6 types of CD. Phase solubility studies showed that CUR with β‐CD or HP‐β‐CD, respectively, could form stable inclusion complexes in a stoichiometric ratio of 1 : 1. The CUR/β‐CD and CUR/HP‐β‐CD inclusion complexes (ICs) were prepared by freeze‐drying method. The successful preparation of IC was confirmed by SEM images, FT‐IR spectra, and TG‐DSC. The results of in vitro release showed that the release of CUR from IC prepared with β‐CD or HP‐β‐CD could be improved significantly, and the release effect of HP‐β‐CD was better than that of β‐CD. The inclusion with CD could be used to improve the solubility and bioavailability of CUR.

Abstract:Cannabidiol (CBD), which has a wide variety of biological activities, is one of the phytocannabinoids from Cannabis sativa L. However, due to the poor aqueous solubility, its applications in food, cosmetic, and pharmaceutical fields are limited. Moreover, the previous researches on the improvement in the solubility of CBD are rare. Cyclodextrins (CDs) are useful materials to improve the solubility of poorly water-soluble molecules. Therefore, this work was aimed to study the physicochemical properties, water solubility, and dissolution rate of the inclusion complexes of CBD with β-cyclodextrin (β-CD) and 2, 6-di-O-methyl-β-cyclodextrin (DM-β-CD), and to determine the influence of complexation on the antioxidant activity of CBD. Job's plot and phase solubility study revealed that CBD formed inclusion complexes with β-CD and DM-β-CD at a 1:1 stoichiometric ratio. The thermodynamics analysis demonstrated that the inclusion process was spontaneous and endothermic. Then, the inclusion complexes of CBD with β-CD or DM-β-CD were prepared and characterized by PXRD, FT-IR, DSC, SEM, and 1H NMR. The loading efficiency (LE) of the two inclusion complexes was 20.4% and 17.7% respectively, while the complexation efficiency (CE) was 92.4% and 90.8% respectively. Moreover, the results of molecular docking further showed that CBD was encapsulated successfully. The results also displayed that the water solubility of CBD in CBD/β-CD inclusion complex (IC) and CBD/DM-β-CD IC was significantly increased to 0.395 and 14.118 μg/mL, which was enhanced by 17-fold and 614-fold respectively at such a high LE, and the in vitro dissolution rate of CBD was also promoted after complexation. Besides, the antioxidant activity of CBD was improved after encapsulation with β-CD and DM-β-CD. Significantly, the ABTS free radical scavenging ability of the two inclusion complexes was better than that of Vitamin C (Vc).

Abstract:Quercetin (QCT) has a variety of pharmacological effects, such as antioxidant, antibacterial, anticancer, anticardiovascular and antiaging effects. However, its poor water solubility, stability and bioavailability limit its applications. The special structure of cyclodextrins and their derivatives with a hydrophobic inner cavity and hydrophilic outer wall can load a variety of hydrophobic drugs of a suitable size and shape, thereby improving the stability and solubility of these molecules. In this study, an inclusion complex of quercetin and sulfobutylether-β-cyclodextrin was prepared. It was characterized via FT-IR, UV, 1 H NMR, XRD, DSC, and SEM analysis, which revealed the successful formation of the inclusion complex. In vitro biological activity estimations were carried out and the results indicated that the inclusion complex displayed higher antioxidative and antibacterial properties compared with free QCT. In addition, the mechanisms of inclusion were explored using 1 H NMR analysis and docking calculations, thus providing a theoretical basis for obtaining an inclusion complex.

Abstract:Taking the inclusion rate of β-carotene as a criterion, inclusion compound of β-carotene with β-cyclodextrin via ultrasonic method was prepared. The optimum conditions determined by single factor and orthogonal design were as follows: the ultrasonic power was 300W with duration time of 40 min; n(β-carotene)∶n(β-cyclodextrin) =1∶4. The inclusion compound was confirmed by micrograph and X-RD. The experiment results showed that the retaining rate of β- carotene increased by 25.5% in inclusion compound, compared with a controlled sample.

Abstract:The inclusion complexation behavior of azadirachtin with several cyclodextrins and their methylated derivatives has been investigated in both solution and the solid state by means of XRD, TG–DTA, DSC, NMR, and UV–vis spectroscopy. The results show that the water solubility of azadirachtin was obviously increased after resulting inclusion complex with cyclodextrins. Typically, β-cyclodextrin (β-CD), dimethyl-β-cyclodextrin (DMβCD), permethyl-β-cyclodextrin (TMβCD), and hydroxypropyl-β-cyclodextrin (HPβCD) are found to be able to solubilize azadirachtin to high levels up to 2.7, 1.3, 3.5, and 1.6mg/mL (calculated as azadirachtin), respectively. This satisfactory water solubility and high thermal stability of the cyclodextrin–azadirachtin complexes, will be potentially useful for their application as herbal medicine or healthcare products.

Abstract:An inclusion complex between 2,6-di-O-methyl-β-cyclodextrin (DM-β-CD) and chlorzoxazone was prepared and investigated. Freeze-drying method was used to synthetize the complex; Fourier transform infrared spectroscopy, laboratory powder X-ray diffraction, 1H nuclear magnetic resonance spectroscopy, Rotating-frame overhauser effect spectroscopy, and Differential scanning calorimetry were utilized to confirm the formation of the complex. Furthermore, the solubilization and dissolution study results showed that the water solubility and dissolution rate of CZX were obviously improved through complexation with DM-β-CD. Finally, the cytotoxicity of the complex was determined by MTT assay. As expected, the complex expressed lower toxicities for normal liver cells than free CZX. This study provides a promising idea to develop novel CZX solid dosage form with good dissolution rate and low liver toxicity.

Abstract:The inclusion complex of 2-hydroxypropyl-β-cyclodextrin(2-HP-β-CD) with lycopene was prepared.The solubility and stability of the inclusion complex were tested and the difference between inclusion complex and raw lycopene was compared using UV-visible spectra DSC.The results showed that after kneading lycopene and 2-HP-β-CD(molar ratio 1∶4 respectively), lycopene solubility increased to 0.5mg/ml.The inclusion process increased the lycopene stability and reduced the loss during 2 months storage time at 4 ℃.

Abstract:Vitamin E (VE) and β-cyclodextrin (β-CD) can form an inclusion complex; however, the inclusion rate is low because of the weak interaction between VE and β-CD. The results of a molecular docking study showed that the oxygen atom in the five-membered ring of octenyl succinic anhydride (OSA) formed a strong hydrogen bond interaction (1.89 Å) with the hydrogen atom in the hydroxyl group of C-6. Therefore, β-CD was modified using OSA to produce octenyl succinic-β-cyclodextrin (OCD). The inclusion complexes were then prepared using OCD with VE. The properties of the inclusion complex were investigated by Fourier-transform infrared spectroscopy (FT-IR), 13C CP/MAS NMR, scanning electron microscopy (SEM), and atomic force microscopy (AFM). The results demonstrated that VE had been embedded into the cavity of OCD. Furthermore, the emulsifying properties (particle size distribution, ζ-potential, and creaming index) of the OCD/VE inclusion-complex-stabilized emulsion were compared with that stabilized by β-CD, OCD, and an OCD/VE physical mixture. The results showed that the introduction of the OS group and VE could improve the physical stability of the emulsion. In addition, the OCD/VE inclusion complex showed the strongest ability to protect the oil in the emulsion from oxidation. OCD/VE inclusion complex was able to improve the physical and oxidative stability of the emulsion, which is of great significance to the food industry.

Abstract:Calculations have been done on β-cyclodextrin 6-O-monobenzoate and its complexes using molecular mechanics MM2 force field. The guest molecules involved were n-hexane, n-heptane, n-octane, cyclohexane, methylcyclohexane, cyclooctane and methylcyclooctane. Owing to lack of dipole–dipole and hydrogen-bonding interactions in the inclusion complexation of the modified cyclodextrin, the calculations of the geometry of the complexes and the inclusion process indicated that the complex formation is stabilized by van der Waals interaction. The energy differences of the modified cyclodextrins by calculation show that the size, shape, conformation of the guest molecules play significant roles in the inclusion complexation.

Abstract:Rutin is a natural flavonoid that carries out a variety of biological activities, but its application in medicine and food is limited by its water solubility. One of the classical methods used to enhance drug solubility is encapsulation with cyclodextrins. In this paper, the encapsulation of different cyclodextrins with rutin was investigated using a combination of experimental and simulation methods. Three inclusions of rutin/beta-cyclodextrin (β-CD), rutin/2-hydroxypropyl beta-cyclodextrin (HP-β-CD) and rutin/2,6-dimethyl beta-cyclodextrin (DM-β-CD) were prepared by the freeze-drying method, and the inclusions were analyzed using Fourier infrared spectroscopy (FTIR), X-ray diffraction analysis (XRD), differential scanning calorimetry (DSC) and ultraviolet–visible spectroscopy (UV) to characterize and demonstrate the formation of the inclusion complexes. Phase solubility studies showed that rutin formed a 1:1 stoichiometric inclusion complex and significantly increased its solubility. β-CD, HP-β-CD, DM-β-CD, rutin and the three inclusion complexes were modeled by using MS2018 and AutoDock 4.0, and molecular dynamics simulations were performed to calculate the solubility parameters, binding energies, mean square displacement (MSD), hydrogen bonding and radial distribution functions (RDF) after the equilibration of the systems. The results of simulation and experiment showed that rutin/DM-β-CD had the best encapsulation effect among the three cyclodextrin inclusion complexes.

Abstract:In recent years, the research and discovery of antioxidants of natural origin, such as those found plants, have increased dramatically. Ellagic acid is a bioactive compound found in many fruits and vegetables, which carries many biological activities, such as antioxidant, anti-inflammatory, anti-cancer and antibacterial activities. However, the low solubility of ellagic acid in water decreases its practical application. In this study, a complex of ellagic acid with hydroxypropyl-cyclodextrin was synthesized in the water-ethanol solvent. The results showed that the solvent with a volume content of EtOH of 20% was the most suitable for complex formation, with complexation yield of 46%. The complex was characterized by FTIR, DSC methods. The infrared spectrum of the complex is similar to that of HP-β-CD, however, the intensity and position of some oscillations in the complex have changed significantly, compared to spectra of EA and HP-β-CD. The sharp adsorbance band at 3475 cm-1 of the O-H bond of EA was not observed in the spectrum of the complex, indicating that the O-H group participated in the bonding and covered by the hollow cavity of the HP-β-CD molecules. The DSC curve of the complex shows that the melting points of EA and HPβCD in the complex are declined in terms of temperature and intensity. This is evidence that there is a complex interaction between EA and HPβCD. The complexation improved the solubility and antioxidant activity of EA. Especifically, the solubility of EA was increased by 3.2 times compared to raw EA; EC50 value of EA was reduced from 5.3x10-5 to 4.9x10-5mol. L-1 after complexation.

Abstract:Isoliquiritigenin (ISL) possesses a wide variety of pharmacological properties, however, its poor solubility and oral bioavailability pose a significant barrier to its application. In present studies, the ISL inclusion complex was prepared with sulfobutyl ether-β-cyclodextrin (SBE-β-CD). The physicochemical characterizations of ISL-SBE-β-CD were performed with Fourier transform infrared (FT-IR) spectroscopy and X-ray powder diffraction (XRD). Phase solubility study suggested a 1:1 formation of ISL-SBE-β-CD complexes. The water solubility of ISL rose from 13.6 μM to 4.05 mM by the inclusion of SBE-β-CD. The antioxidant activities (IC50) of ISL-SBE-β-CD reached 42.2 μg/ml, which was significantly lower than that of ISL (60.5 μg/ml). Its stability in biological environments was also enhanced.

Abstract:The inclusion complex formation of irisquinone with hydroxypropyl-beta-cyclodextrin(HP-beta-CD) and the physicochemical properties were studied. Complex of host molecules with irisquinoe was studied by x-ray diffraction (XRD) and differential scanning calorimetry(DSC). The quantitative analysis with uv-vis indicated that HP-beta-CD could increase the solubility of irisquinone, where the best results was given with HP-beta-CD :irisquinone 2:1 molar ratio. The binding constants of inclusion complex formulation were obtained with relative Higuchi Principle and thermodynamics
