Abstract:Life are governed by cellular complex metabolic processes. One of the on-going endeavours in the scientific community is to study and understand these dynamic biochemical reactions and the roles of biomolecules in sustaining life. Fluorescent probes have been widely used in the visualization of analytes in physiological and pathological processes, due to their advantages such as easy operation, low cost, high sensitivity, capability of multi-channel and real-time visualization in vivo. However, most fluorescent probes can only respond to single analyte, which is not suitable to mornitor multiple analytes in complex biological system. In recent five years, significantly more multi-analyte fluorescent probes based on 1, 8-naphthimide fluorophore have been designed in biological or environmental field. Categorized by the fluorescent mechanisms, such as single fluorescent mechanism (e. g. photoinduced electron transfer (PET), intramolecular charge transfer (ICT) and fluorescence resonance energy transfer (FRET)), dual fluorescent mechanisms (e. g. PET-ICT, PET-FRET, and ICT-FRET) and so on, this review highlights the recent progress in designing strategies, corresponding recognition process, optical property and cell imaging of 1, 8-naphthalimide-based multi-analyte fluorescent probes. Finally, the development trend and possible challenges of such fluorescent probes are prospected.

Abstract:The activity levels of key substances (metal ions, reactive oxygen species, reactive nitrogen, biological small molecules, etc.) in organisms are closely related to intracellular redox reactions, disease occurrence and treatment, as well as drug absorption and distribution. Fluorescence imaging technology provides a visual tool for medicine, showing great potential in the fields of molecular biology, cellular immunology and oncology. In recent years, organic fluorescent probes have attracted much attention in the bioanalytical field. Among various organic fluorescent probes, fluorescent organic small molecule probes (FOSMPs) have become a research hotspot due to their excellent physicochemical properties, such as good photostability, high spatial and temporal resolution, as well as excellent biocompatibility. FOSMPs have proved to be suitable for in vivo bioimaging and detection. On the basis of the introduction of several primary fluorescence mechanisms, the latest progress of FOSMPs in the applications of bioimaging and detection is comprehensively reviewed. Following this, the preparation and application of fluorescent organic nanoparticles (FONPs) that are designed with FOSMPs as fluorophores are overviewed. Additionally, the prospects of FOSMPs in bioimaging and detection are discussed.

Abstract:Organic long-lived luminescent probes featuring thermally activated delayed fluorescence (TADF) or room temperature phosphorescence (RTP) have emerged as highly promising tools for time-resolved luminescence imaging (TRLI) and sensing (TRLS). Their unique advantage lies in their ability to effectively eliminate interference from short-lived background signals. By introducing an appropriate delay time between the short-lived autofluorescence and measure of the long-lived luminescence emitted by the probes, a background-free approach emerges, enabling the accurate detection and imaging of diverse biologically relevant species. This review provides a comprehensive overview of metal-free organic compound-based responsive probes utilizing TADF and RTP for TRLI and TRLS. It offers detailed insights into the design strategies and sensing principles employed by these probes. Furthermore, the review discusses the current challenges faced by researchers in this rapidly expanding field and highlights the future prospects for advancements in this area of study.

Abstract:A series of naphthalimide-tetrazines were developed as bioorthogonal fluorogenic probes, which could produce significant fluorescence enhancement, notable aggregation-induced emission (AIE) characters and multicolor emissions after bioorthogonal reaction with strained dienophiles. Manipulating the pi-bridge in the fluorophore skeleton allows fine-tuning of the emission wavelength and influences the AIE-active properties. With these probes, we succeeded in no-wash fluorogenic protein labeling and mitochondria-selective bioorthogonal imaging in live cells.

Abstract:Multiple stimuli-responsive fluorescent materials have gained increasing attention for their fundamental investigation and intelligent applications. In this work, we report design and synthesis of a novel polyamic acid bearing oligoaniline, triphenylamine, and fluorene groups, which served as sensitive units and fluorescence emission unit, respectively. The resulting polymer exhibits multiple stimuli-responsive fluorescence switching behavior triggered by redox species, pH, electrochemical, and pressure stimuli. Every fluorescence switching mechanism upon each stimulus was studied in detail. The interactions and energy transfer between sensitive units and emission unit are largely responsible for this fascinating fluorescent switching behavior. This work provides a deep understanding of the optical switching essence upon these stimuli, opening the way for the development of new fluorescent sensing applications.

Abstract:Fluorescent probes have been widely investigated for their features of rapid response, easy operation and high sensitivity. Among them, reaction-based fluorescent probes, for their unique reaction-based nature, guarantee them with excellent selectivity, effectively avoiding the possible interference from other chemical and biological species in physiological environment. Conventional reaction-based fluorescent probes are aggregation-caused quenching (ACQ) fluorophores. The application of these kinds of probes are limited for their poor photostability and narrow Stokes shifts. Compared with ACQ fluorophores, aggregation-induced emission (AIE) fluorophores become emissive in aggregation states with higher signal-to-noise ratio, better photostability and larger Stokes shifts. In this review, we summarize the latest developed reaction-based AIE-active probes, including the design principle and application in various sensing systems and give an outlook for the future development of this kind of promising fluorescent probes.

Abstract:Optical imaging essentially refers to in vivo fluorescence imaging and bioluminescence imaging. These types of imaging are widely used visualization methods in biomedical research and are important in molecular imaging. A new generation of imaging agents called multimodal probes have emerged in the past few years. These probes can be detected by two or more imaging modalities, which harnesses the strengths of the different modalities and enables researchers to obtain more information than can be achieved using only one modality. Owing to its low cost and the large number of probes available, the optical method plays an important role in multimodality imaging. Abstract:Some important biological species and microenvironments maintain a complex and delicate dynamic balance in life systems, participating in the regulation of various physiological processes and playing indispensable roles in maintaining the healthy development of living bodies. Disruption of their homeostasis in living organisms can cause various diseases and even death. Therefore, real time monitoring of these biological species and microenvironments during different physiological and pathological processes is of great significance. Fluorescent-probe-based techniques have been recognized as one of the most powerful tools for real time imaging in biological samples. In this Account, we introduce the representative works from our group in the field of fluorescent probes for biological imaging capable of detecting metal ions, small bioactive molecules, and the microenvironment. The design strategies of small molecule fluorescent probes and their applications in biological imaging will be discussed. By regulating the design strategy and mechanism (e.g., ICT, PeT, and FRET) of the electronic and spectral characteristics of the fluorescent platforms, these chemical probes show high selectivity and diverse functions, which can be used for imaging of various physiological and pathological processes. Through the exploration of the rational response mechanism and design strategy, combined with a variety of imaging techniques, such as super-resolution imaging, photoacoustic (PA) imaging, etc., we have realized multimode imaging of the important biological analytes from the subcellular level to the in vivo level, which provides powerful means to study the physiological and pathological functions of these species and microenvironments. This Account aims to offer insights and inspiration for the development of novel fluorescent probes for biological imaging, which could provide powerful tools for the study of chemical biology. Overall, we represent a series of turn-on/turn-off/ratiometric fluorescent/PA probes to visually and dynamically trace biological species and microenvironments in cells and even in vivo that seek higher resolution and depth molecular imaging to improve diagnostic methods and clarify new discoveries related to chemical biology. Our future efforts will be devoted to developing multiorganelle targeted fluorescent probes to study the mechanism of subcellular organelle interaction and employing various dual-mode probes of NIR II and PA imaging to investigate the development of related diseases and treat the related diseases at subcellular and in vivo levels.

Abstract:In recent years, with the rise of precision therapy and other frontier fields, the demand of the high quality fluorescent probes with extremely low toxicity, good biocompatibility, strong stability and high brightness is more urgent than ever. Proteins that with abundant active groups and special pockets can bind with different probes to alter the fluoresce properties, and some can even greatly enhance the fluorescence signals. Moreover, some proteins can simultaneously bind with two different molecules to fabricate fluorescence resonance energy transfer (FRET) probes. Recent progress witnessed that the protein-mediated strategies have provided a promising way for enhancing the performance of fluorescent probe. This article will review the up-to-date progress of protein-mediated fluorescent probes. We firstly provide a systemically on the principle of protein fluorescent probes fabrication and the interaction mechanisms between proteins and molecular dyes. Then the bioapplications of these fluorescent probes in bioimaging and biosensing were reviewed. And finally the current challenges and perspectives were discussed in-depth. We hope that this review can provide some new ideas and inspiration for relevant researchers to develop a new generation of fluorescent probes.

Abstract:Biological imaging and biosensing from subcellular/cellular level to whole body have enabled non-invasive visualisation of molecular events during various biological and pathological processes, giving great contributions to the rapid and impressive advances in chemical biology, drug discovery, disease diagnosis and prognosis. Optical imaging features a series of merits, including convenience, high resolution, good sensitivity, low cost and the absence of ionizing radiation. Among different luminescent probes, metal-based molecules offer unique promise in optical bioimaging and biosensing in vitro and in vivo, arising from their small sizes, strong luminescence, large Stokes shifts, long lifetimes, high photostability and tunable toxicity. In this review, we aim to highlight the design of metal-based molecular probes from the standpoint of synthetic chemistry in the last 2 years for optical imaging, covering d block transition metal and lanthanide complexes and multi modal imaging agents.

Abstract:The use of fluorescent probes in visible detection has been developed over the last several decades. Biomolecules are essential in the biological processes of organisms, and their distribution and concentration are largely influenced by environmental factors. Significant advances have occurred in the applications of fluorescent probes for the detection of the dynamic localization and quantity of biomolecules during various environmental stress-induced physiological and pathological processes. Herein, we summarize representative examples of small molecule-based fluorescent probes that provide bimolecular information when the organism is under environmental stress. The discussion includes strategies for the design of smart small-molecule fluorescent probes, in addition to their applications in biomolecule imaging under environmental stresses, such as hypoxia, ischemia-reperfusion, hyperthermia/hypothermia, organic/inorganic chemical exposure, oxidative/reductive stress, high glucose stimulation, and drug treatment-induced toxicity. We believe that comprehensive insight into the beneficial applications of fluorescent probes in biomolecule detection under environmental stress should enable the further development and effective application of fluorescent probes in the biochemical and biomedical fields.

Abstract:In recent years, significant advancements have been made in the research of photoswitchable probes. These probes undergo reversible structural and electronic changes upon light exposure, thus exhibiting vast potential in molecular detection, biological imaging, material science, and information storage. Through precisely engineered molecular structures, the photoswitchable probes can toggle between "on" and "off" states at specific wavelengths, enabling highly sensitive and selective detection of targeted analytes. This review systematically presents photoswitchable fluorescent and colorimetric probes built on various molecular photoswitches, primarily focusing on the types involving photoswitching in their detection and/or signal response processes. It begins with an analysis of various molecular photoswitches, including their photophysical properties, photoisomerization and photochromic mechanisms, and fundamental design concepts for constructing photoswitchable probes. The article then elaborates on the applications of these probes in detecting diverse targets, including cations, anions, small molecules, and biomacromolecules. Finally, it offers perspectives on the current state and future development of photoswitchable probes. This review aims to provide a clear introduction for researchers in the field and guidance for the design and application of new, efficient fluorescent and colorimetric probes.

Abstract:There has been rapid progress in the development of fast, sensitive, cheap and low-cytotoxicity micro-molecule fluorescent probes for application in various fields, including disease diagnosis, food safety and environmental safety. As an analytical tool, dual-function fluorescent probes with dual-emission responses have attracted considerable attention due to their cost-effectiveness and efficiency over single-function sensors. This review primarily describes research progress on multifunctional probes in terms of the reaction type and coordination type, as well as the general design principles of probes. The analytes include reactive oxygen species (ROS), reactive sulfur species (RSS), harmful cations and anions, etc. Multifunctional probes for food, medical and environmental applications are listed for future research. To improve the development of rapid detection methods, trends and strategies in the development of multifunctional fluorescent probes are also discussed.

Abstract:Flavonols, named as 3-hydroxyl-2-phenyl-1-benzopyran-4-one, are important natural botanical functional pigments widely distributing in fruits, vegetables, and flowers. Owing to intramolecular hydrogen bond, the flavonols can undergo the excited-state intramolecular proton transfer (ESIPT) process upon irradiation, leading to unique environmental-sensitive dual emissions, which makes flavonol an ideal fluorophore skeleton for probe design. In this paper, we summarized the recent progress of flavonol-based small-molecule fluorescent probes for diverse sensing applications, including detection of anions, cations, reactive biological species, proteins, DNAs, environmental hazards, and microenvironmental factors. This review highlighted the basic characteristics of flavonol probes in the respect of design principles, photophysical processes, sensing mechanism, sensing performances, and biological applications. The present review article aims to offer a comprehensive summary for researchers to inspire new probe design, and push forward the advancement of flavonol-based fluorescent probes.

Abstract:Bioanalysis based on optical imaging has gained significant progress in the last few decades. Luminescence probes are capable of detecting, monitoring, and tracing particular biomolecules in complex biological systems to figure out the roles of these molecules in organisms. Considering the rapid development of luminescence probes for bio-applications and their promising future, we have attempted to explore the working principles and recent advances in bio-applications of luminescence probes, in the hope of helping readers gain a detailed understanding of luminescence probes developed in recent years. In this review, we first focus on the current widely used luminescence probes, including fluorescence probes, bioluminescence probes, chemiluminescence probes, afterglow probes, photoacoustic probes, and Cerenkov luminescence probes. The working principles for each type of luminescence probe are concisely described and the bio-application of the luminescence probes is summarized by category, including metal ions detection, secretion detection, imaging, and therapy.

Abstract:The emergence of bioorthogonal reactions has provided numerous opportunities for the design and fabrication of innovative fluorescence systems, particularly in the analytical field. These bioorthogonal reactions, including copper-catalyzed azide-alkyne cycloaddition (CuAAC), Staudinger ligation reaction, strain-promoted alkyne-azide cycloaddition (SPAAC), photoclick reactions, strain-promoted alkyne-nitrone cycloaddition (SPANC), and inverse-electron-demand Diels-Alder (IEDDA) reaction, have been extensively employed in the development of fluorescent probes. By combining a wide range of bioorthogonal reaction pairs with diverse fluorophores, fluorescent probes have been created to enable specific and selective sensing of biomolecules, drugs, and other biologically related organic compounds. This review provides a comprehensive overview of the development process of fluorescent probes based on bioorthogonal reactions. It covers the construction strategies, systematic classification, analysis, and applications of these fluorescent probes, while also discussing future prospects in this field.

Abstract:A customizable fluorescent probe platform that can be used to detect various bioactive analytes offers significant potential for engineering a wide range of bioprobes with diverse sensing and imaging functions. Here, we show a facile and innovative strategy for introducing <i>cis</i>-amino-proline as a carrier scaffold, which is appended with three specific functional groups: a target group, a water-soluble group, and fluorophores with triggers. The potency of the designed strategy could be customized to generate variable multifunctional fluorescent probes for detecting bioactive species of interest, including reactive oxygen species (ROS), reactive nitrogen species (RNS), reactive sulfur species (RSS), ROS/RSS, and even enzymes. We designed and synthesized five representative water-soluble and organelle-targeted compounds, PMB, PMN, PMD, PRB, and PME, with emission wavelengths of these fluorescent probes varying from blue to red (465, 480, 535, 550, 565, and 640 nm). Abstract:In recent years, nanomaterials have attracted lots of attention from researchers due to their unique properties. Nanometer fluorescent materials, such as organic dyes, semiconductor quantum dots (QDs), metal nanoclusters (MNCs), carbon dots (CDs), etc., are widely used in biological imaging due to their high sensitivity, short response time, and excellent accuracy. Nanometer fluorescent probes can not only perform in vitro imaging of organisms but also achieve in vivo imaging. This provides medical staff with great convenience in cancer treatment. Combined with contemporary medical methods, faster and more effective treatment of cancer is achievable. This article explains the response mechanism of three-nanometer fluorescent probes: the principle of induced electron transfer (PET), the principle of fluorescence resonance energy transfer (FRET), and the principle of intramolecular charge transfer (ICT), showing the semi-conductor QDs, precious MNCs, and CDs. The excellent performance of the three kinds of nano fluorescent materials in biological imaging is highlighted, and the application of these three kinds of nano fluorescent probes in targeted biological imaging is also introduced. Nanometer fluorescent materials will show their significance in the field of biomedicine.
