Abstract:Abstract The platelet fibrinogen receptor α IIbβ 3 exists in an equilibrium between inactive and active conformations. In its inactive conformation, the transmembrane (TM) domains of α IIb and β 3 interact, but they separate when α IIbβ 3 assumes its active conformation. Peptides corresponding to the α IIb TM domain form homodimers in vitro and in bacterial membranes and the interface that mediates this interaction overlaps with the interface that mediates the heteromeric association of the α IIb TM domain with that of β 3. Because the homomeric association of α IIb TM domain is relatively strong, we expected that peptides spanning the α IIb TM domain might activate α IIbβ 3 by binding to the α IIb TM domain, thereby disrupting the α IIb/β 3 TM domain heterodimer. We synthesized a 26 residue peptide corresponding to the α IIb TM domain, flanked by pairs of lysine residues to increase its solubility in water, and assessed its ability to activate washed or gel-filtered platelets in an aggregometer. Addition of 3 μM peptide induced platelet aggregation after a short lag, but unlike aggregation stimulated by ADP, peptide-induced aggregation was not preceded by platelet shape change. Nonetheless, like ADP-induced aggregation, peptide-induced aggregation was inhibited by EDTA and the α IIbβ 3-specific monoclonal antibody A2A9. On the other hand, pre-incubating platelets with PGE1 or apyrase caused only a small decrease in the rate and extent of peptide-induced aggregation, suggesting that the peptide induced aggregation by binding directly to α IIbβ 3. A peptide corresponding the β 3 TM domain behaved in an identical manner, whereas an unrelated peptide, the model TM domain MS-1, had no effect. Similarly, there was little response to an α IIb TM peptide in which glycines 972 and 976, the first and last residues of a GxxxG motif, were changed to leucine. Confirming that the α IIb TM peptide binds to α IIbβ 3, we found that a FITC-labeled α IIb TM peptide co-eluted with purified α IIbβ 3 during size exclusion gel filtration. To determine whether the latter interaction involved the α IIb TM domain, we used the TOXCAT assay. In TOXCAT, the α IIb TM domain-mediated dimerization of a fusion protein containing the α IIb TM domain interposed between maltose binding protein and the ToxR’ transcriptional activation domain in the inner membrane of E. coli drives the activation of the chloramphenicol acetyl transferase (CAT) gene. We found that adding the α IIb TM peptide to the bacterial growth medium consistently decreased CAT synthesis, implying that its presence impaired α IIb-mediated fusion protein dimerization. These results provide evidence for the presence of an α IIb/β 3 TM domain heterodimer in unstimulated human platelets and suggest that the GxxxG motif in the α IIb TM domain is involved in its formation. Because the homomeric interaction of α IIb and β 3 TM domains is substantially stronger than their heteromeric interaction, our data are also consistent with the hypothesis that the TM peptides disrupt the heterodimer which functions to maintain α IIbβ 3 in an inactive state. Lastly, the data suggest that stabilizing the α IIb/β 3 TM heterodimer may represent a new approach for the development of novel anti-platelet agents.

Abstract:The aggregation of human platelets is an important physiological hemostatic event contingent upon receptor-dependent activation of the surface integrin alpha(IIb)beta(3) and subsequent binding of fabrinogen. Aggregating platelets form phosphatidylinositol 3,4-bisphosphate (PtdIns(3,4)P-2), which has been reported to stimulate in vitro the activity of the proto-oncogenic protein kinase PKB/Akt, as has phosphatidylinositol 3,4,5-trisphosphate (PtdIns(3,4,5)P-3. It has been assumed that PtdIns(3,4)P-2 is synthesized by either 5-phosphatase-catalyzed hydrolysis of PtdIns(3,4,5)P-3 produced by phosphoinositide 3-kinase (PI3K) or phosphorylation by PI3K. of PtdIns4P. We investigated the route(s) by which PtdIns(3,4)P, is formed after directly activating alpha(IIb)beta(3) with anti-ligand-induced binding site Fab fragment and report that aggregation does not lead to the generation of PtdIns(3,4,5)P-3, but to transient formation of PtdIns3P and generation of PMIns(3,4)P-2, the latter primarily by PMIns3P 4-kinase. Both this novel pathway and the activation of PKB/Akt are inhibited by the PI3K inhibitor, wort-mannin, and the calpain inhibitor, calpeptin, constituting the first evidence that PMIns(3,4)P-2 can stimulate PKB/Akt in vivo in the absence of PtdIns(3,4,5)P-3, Integrin-activated generation of the second messenger PMIns(3,4)P-2 thus depends upon a route distinct from that known to be utilized initially by growth factors. This pathway is of potential general relevance to the function of integrins.

Also, PP2 and wortmannin, and the palmitoylated β3 peptide R(724)KEFAKFEEER(734), each inhibited the phosphorylation of Akt residue Ser473 and prevented TxA2 production and storage granule secretion. Similarly, Akt phosphorylation in mouse platelets stimulated by the PAR4 agonist peptide AYPGKF was αIIbβ3-dependent, and blocked by PP2, wortmannin and the palmitoylated peptide p-RKEFAKFEEER. Abstract:Following vascular injury, the adenosine nucleotide ATP is released from the dense granules of adhering and activated platelets, as well as from injured endothelial cells and damaged red blood cells. ATP instantaneously interacts with the ion channel P2X(1) in the platelet membrane, through autocrine and paracrine mechanisms, amplifying the initial phase of a platelet activation event. A multitude of platelet activation studies in vitro and in vivo have identified P2X(1) as a receptor with a distinct pharmacological profile, capable of regulating various intracellular signaling cascades, implicated in platelet function. This review discusses findings on the function of the platelet P2X(1) receptor and its downstream signaling pathways, collected over the last two decades, against more recent in vivo observations in thrombosis models in P2X(1) gene-deficient and transgenic mice. Our present understanding of its physiology has identified platelet P2X(1) as a target in the management of thrombosis, its inhibition potentially capable of platelet function modulation. In view of the availability of specific agonists and antagonists, P2X(1) is also discussed as a therapeutic target for antithrombotic therapy.</.

Abstract:Agonist-stimulated platelet activation triggers conformational changes of integrin αIIbβ3, allowing fibrinogen binding and platelet aggregation. We have previously shown that an octapeptide, p1YMESRADR8, corresponding to amino acids 313-320 of the β-ribbon extending from the β-propeller domain of αIIb, acts as a potent inhibitor of platelet aggregation. Here we have performed in silico modelling analysis of the interaction of this peptide with αIIbβ3 in its bent and closed (not swing-out) conformation and show that the peptide is able to act as a substitute for the β-ribbon by forming a clasp restraining the β3 hybrid and βI domains in a closed conformation. The involvement of species-specific residues of the β3 hybrid domain (E356 and K384) and the β1 domain (E297) as well as an intrapeptide bond (pE315-pR317) were confirmed as important for this interaction by mutagenesis studies of αIIbβ3 expressed in CHO cells and native or substituted peptide inhibitory studies on platelet functions. Furthermore, NMR data corroborate the above results. Our findings provide insight into the important functional role of the αIIb β-ribbon in preventing integrin αIIbβ3 head piece opening, and highlight a potential new therapeutic approach to prevent integrin ligand binding.
Regulation of the Function of Αvβ3 in Platelets by a Designed Peptide Targeting the Αv Transmembrane Domain.

Abstract:Integrins reside on cell surfaces in an equilibrium between inactive and active conformations. When inactive, the transmembrane (TM) domains of integrin α and β subunits interact, but the domains separate when integrins assume their active conformation. Although this conformational change has not been shown for αvβ3, we hypothesized that a peptide designed to bind to the αv TM domain might activate αvβ3 in platelets by disrupting the TM domain heterodimer of the inactive molecule. To design such a peptide, we used CHAMP (Computed Helical Anti-Membrane Protein) methodology. In the CHAMP method, the αv TM helix was scanned for motifs likely to mediate αvβ3 TM domain interactions. Next, the backbone conformation for an αv-binding peptide was selected based on known structural preferences for the motifs identified in the αv helix. Finally, the sequence for the peptide was designed computationally using a side-chain repacking algorithm. The CHAMP peptide, anti-αv, and its TM helix target, αv-TM, were synthesized by solid phase synthesis. We also synthesized anti-αvmut in which the putative anti-αv-TM binding motif, GXXXG, was mutated to LXXXL. Lys2 dipeptides and short polyethylene glycol sequences were appended to the C- and N-termini of the peptides, respectively, to facilitate their solubility and insertion into membranes. CD spectroscopy revealed that both anti-αv and αv-TM were helical in micelles and phospholipid vesicles. Measurement of Trp fluorescence intensity revealed that anti-αv rapidly inserted into unilamellar POPC/POPG vesicles. Analytical ultracentrifuge and fluorescence resonance energy transfer experiments demonstrated that anti-αv bound to the αv-TM, but not to a homologous αIIb-TM domain peptide. In addition, there was negligible interaction between αv-TM and anti-αvmut. Anti-αv also formed heteromeric complexes with the αv TM domain in bacterial membranes but not with the TM domains of αIIb, α2, β1, or β3. αvβ3 mediates the adhesion of agonist-stimulated platelets to the matrix protein osteopontin (OPN). We found that anti-αV at μM concentrations induced platelet adhesion to OPN. Adhesion was prevented by the divalent cation chelator EDTA, consistent with an integrin-mediated process, but was only minimally affected by pre-incubating the platelets with PGE1, implying that anti-αv-induced adhesion by interacting directly with the αv TM domain. Force spectroscopy using laser tweezers confirmed the specificity of the interaction of anti-αv with platelets: anti-αv induced specific rupture forces between platelets and OPN-coated beads, but not between platelets and fibrinogen-coated beads. Moreover, only non-specific rupture forces were detected between OPN-coated beads and platelets incubated with anti-αvmut. These results demonstrate the successful application of computational methods to design a soluble peptide that specifically recognizes the TM domain of αv in platelets membranes, even when a 400-fold excess of the homologous integrin subunit αIIb is present. Further, because the peptide binds to the site on the αv TM domain that interacts with the β3 TM helix and activates αvβ3, these results strongly support the hypothesis that separation of the αv and β3 TM domains regulates the function of this integrin.

Abstract:Glycoprotein (GP) Ib, the ligand-binding subunit of platelet GPIb-IX complex, interacts with von Willebrand factor (VWF) exposed at the injured vessel wall, initiating platelet adhesion, activation, hemostasis, and thrombus formation. The cytoplasmic tail of GPIb interacts with 14-3-3, regulate ng the VWF-GPIb-elicited signal transduction and VWF binding function of GPIb. However, we unexpectedly found that the GPIb-14-3-3 association, beyond VWF-dependent function, is essential for general platelet activation. We found that the GPIb cytoplasmic tail peptide MPC, a potential GPIb inhibitor, by itself induced platelet aggregation, integrin αIIbβ3 activation, granule secretion, and phosphatidylserine (PS) exposure. Conversely, the deletion of the cytoplasmic tail of GPIb in mouse platelets (10aa-/-) decreased platelet aggregation, integrin IIb3 activation, granule secretion, and PS exposure induced by various physiological agonists. Phosphoproteome-based kinase activity profiling revealed significantly upregulated protein kinase C (PKC) activity in MPC-treated platelets. MPC-induced platelet activation was abolished by the pan-PKC inhibitor and PKC deletion. Decreased PKC activity was observed in both resting and agonist-stimulated 10aa-/- platelets. GPIb regulates PKC activity by sequestering 14-3-3 from PKC. In vivo, the deletion of the GPIb cytoplasmic tail impaired mouse hemostasis and thrombus formation and protected against platelet-dependent pulmonary thromboembolism. Therefore, our findings demonstrate an essential role for the GPIb cytoplasmic tail in regulating platelet general activation and thrombus formation beyond the VWF-GPIb axis.

Abstract:Class B G protein-coupled receptors (GPCRs) are important therapeutic targets for major diseases. Here, we present structures of peptide and Gs-bound pituitary adenylate cyclase-activating peptide, PAC1 receptor, and corticotropin-releasing factor (CRF), (CRF1) receptor. Together with recently solved structures, these provide coverage of the major class B GPCR subfamilies. Diverse orientations of the extracellular domain to the receptor core in different receptors are at least partially dependent on evolutionary conservation in the structure and nature of peptide interactions. Differences in peptide interactions to the receptor core also influence the interlinked TM2-TM1-TM6/ECL3/TM7 domain, and this is likely important in their diverse signaling. However, common conformational reorganization of ECL2, linked to reorganization of ICL2, modulates G protein contacts. Comparison between receptors reveals ICL2 as a key domain forming dynamic G protein interactions in a receptor- and ligand-specific manner. This work advances our understanding of class B GPCR activation and Gs coupling.

Abstract:Platelets play a key role in the physiological hemostasis or pathological process of thrombosis. Rhodocytin, an agonist of the C-type lectin-like receptor-2 (CLEC-2), elicits powerful platelet activation signals in conjunction with Src family kinases (SFKs), spleen tyrosine kinase (Syk), and phospholipase γ2 (PLCγ2). Previous reports have shown that rhodocytin-induced platelet aggregation depends on secondary mediators such as thromboxane A2 (TxA2) and ADP, which are agonists for G-protein-coupled receptors (GPCRs) on platelets. How the secondary mediators regulate CLEC-2-mediated platelet activation in terms of signaling is not clearly defined. In this study, we report that CLEC-2-induced Syk and PLCγ2 phosphorylation is potentiated by TxA2 and that TxA2 plays a critical role in the most proximal event of CLEC-2 signaling, i.e. the CLEC-2 receptor tyrosine phosphorylation. We show that the activation of other GPCRs, such as the ADP receptors and protease-activated receptors, can also potentiate CLEC-2 signaling. By using the specific Gq inhibitor, UBO-QIC, or Gq knock-out murine platelets, we demonstrate that Gq signaling, but not other G-proteins, is essential for GPCR-induced potentiation of Syk phosphorylation downstream of CLEC-2. We further elucidated the signaling downstream of Gq and identified an important role for the PLCβ-PKCα pathway, possibly regulating activation of SFKs, which are crucial for initiation of CLEC-2 signaling. Together, these results provide evidence for novel Gq-PLCβ-PKCα-mediated regulation of proximal CLEC-2 signaling by Gq-coupled receptors.
Distinct platelet packaging, release, and surface expression of proangiogenic and antiangiogenic factors on different platelet stimuli

Abstract:The present study characterized platelet secretion and surface expression of proangiogenic stromal cell-derived factor-1 alpha (SDF-1 alpha) and vascular endothelial growth factor (VEGF) and antiangiogenic PF4 and endostatin on activation. The angiogenic factors presented in randomly distributed granules in resting platelets, which were peripherized on activation. Confocal and immunogold electron microscopy demonstrated that SDF1 alpha/CXCL12 and PF4/CXCL4 mostly present in different granules. Platelet activation induced marked SDF-1 alpha and endostatin but mild PF4 or no VEGF surface expression. PAR1-activating peptide (PAR1-AP), adenosine diphosphate (via P2Y1/P2Y12), and glycoprotein VI-targeting collagen-related peptide induced massive SDF-1 alpha and VEGF but modest PF4 or no endostatin release. In contrast, PAR4-AP triggered marked PF4 and sole endostatin release but limited SDF-1 alpha or VEGF secretion. Distinct platelet release of SDF-1 alpha and endostatin involved different engagements of intracellular signaling pathways. In conclusion, different platelet stimuli evoke distinct secretion and surface expression of proangiogenic and antiangiogenic factors. Abstract:Platelets play a central role in hemostasis and thrombosis, initiating clot formation in response to vessel wall damage. Platelet aggregates are formed at sites of injury by the binding and crosslinking of the integrin IIb3 to fibrinogen, von Willebrand factor and other soluble ligands. Platelets also form pathological thrombi, and the resulting arterial occlusion can lead to myocardial infarction or stroke. Inhibition of IIb3 binding can decrease the formation of pathologic thrombi, and IIb3 has become an important pharmacological target. Anti-IIb3 therapies have been highly successful in preventing death following myocardial infarction and percutaneous arterial stent placement.(Topol, Lincoff et al. 2002; De Luca, Ucci et al. 2009) However, attempts to design novel, orally available anti-IIb3 agents have been hampered by what appears to be paradoxical activation of IIb3 by the drug, in some cases leading to an increased risk of mortality in patients who received the drug.(Quinn, Plow et al. 2002)

Abstract:Platelets have a crucial role in the maintenance of normal haemostasis, and perturbations of this system can lead to pathological thrombus formation and vascular occlusion, resulting in stroke, myocardial infarction and unstable angina. ADP released from damaged vessels and red blood cells induces platelet aggregation through activation of the integrin GPIIb-IIIa and subsequent binding of fibrinogen. ADP is also secreted from platelets on activation, providing positive feedback that potentiates the actions of many platelet activators. ADP mediates platelet aggregation through its action on two G-protein-coupled receptor subtypes. The P2Y1 receptor couples to Gq and mobilizes intracellular calcium ions to mediate platelet shape change and aggregation. The second ADP receptor required for aggregation (variously called P2Y(ADP), P2Y(AC), P2Ycyc or P2T(AC)) is coupled to the inhibition of adenylyl cyclase through Gi. The molecular identity of the Gi-linked receptor is still elusive, even though it is the target of efficacious antithrombotic agents, such as ticlopidine and clopidogrel and AR-C66096 (ref. 9). Here we describe the cloning of this receptor, designated P2Y12, and provide evidence that a patient with a bleeding disorder has a defect in this gene. Cloning of the P2Y12 receptor should facilitate the development of better antiplatelet agents to treat cardiovascular diseases.

Abstract:The impact of flow disturbances on platelet adhesion is complex and incompletely understood. At the molecular scale, platelet glycoprotein Ibα (GPIbα) must associate with the von Willebrand factor A1 domain (VWF-A1) with a rapid on-rate under high hemodynamic forces, as occurs in arterial thrombosis, where various transport mechanisms are at work. Here, we theoretically modeled the coupled transport-reaction process of the two-dimensional (2D) receptor-ligand association kinetics in a biomembrane force probe to explicitly account for the effects of molecular length, confinement stiffness, medium viscosity, surface curvature, and separation distance. We experimentally verified the theoretical approach by visualizing association and dissociation of individual VWF-A1-GPIbα bonds in a real-time thermal fluctuation assay. The apparent on-rate, reciprocal of the average time intervals between sequential bonds, decreased with the increasing gap distance between A1- and GPIbα-bearing surfaces with an 80-nm threshold (beyond which bond formation became prohibitive) identified as the combined contour length of the receptor and ligand molecules. The biomembrane force probe spring constant and diffusivity of the protein-bearing beads also significantly influenced the apparent on-rate, in accordance with the proposed transport mechanisms. The global agreement between the experimental data and the model predictions supports the hypothesis that receptor-ligand association behaves distinctly in the transport- and reaction-limited scenarios. To our knowledge, our results represent the first detailed quantification of physical regulation of the 2D on-rate that allows platelets to sense and respond to local changes in their hemodynamic environment. In addition, they provide an approach for determining the intrinsic kinetic parameters that employs simultaneous experimental measurements and theoretical modeling of bond association in a single assay. The 2D intrinsic forward rate for VWF-A1-GPIbα association was determined from the measurements to be (3.5 ± 0.67) × 10(-4)μm(2) s(-1).
Role of G Protein-Gated Inwardly Rectifying Potassium Channels in P2y(12) Receptor-Mediated Platelet Functional Responses

Abstract:The role of the G(i)-coupled platelet P2Y(12) receptor in platelet function has been well established. However, the functional effector or effectors contributing directly to alphaIIbbeta3 activation in human platelets has not been delineated. As the P2Y(12) receptor has been shown to activate G protein-gated, inwardly rectifying potassium (GIRK) channels, we investigated whether GIRK channels mediate any of the functional responses of the platelet P2Y(12) receptor. Western blot analysis revealed that platelets express GIRK1, GIRK2, and GIRK4. In aspirin-treated and washed human platelets, 2 structurally distinct GIRK inhibitors, SCH23390 (R(+)-7-chloro-8-hydroxy-3-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine hydrochloride) and U50488H (trans-(+/-)-3,4-dichloro-N-methyl-N-[2-(pyrrolidinyl)cyclohexyl] benzeneacetamide methanesulfonate), inhibited adenosine diphosphate (ADP)-, 2-methylthioADP (2-MeSADP)-, U46619-, and low-dose thrombin-mediated platelet aggregation. However, the GIRK channel inhibitors did not affect platelet aggregation induced by high concentrations of thrombin, AYPGKF, or convulxin. Furthermore, the GIRK channel inhibitors reversed SFLLRN-induced platelet aggregation, inhibited the P2Y(12)-mediated potentiation of dense granule secretion and Akt phosphorylation, and did not affect the agonist-induced G(q)-mediated platelet shape change and intracellular calcium mobilization. Unlike AR-C 69931MX, a P2Y(12) receptor-selective antagonist, the GIRK channel blockers did not affect the ADP-induced adenlylyl cyclase inhibition, indicating that they do not directly antagonize the P2Y(12) receptor. We conclude that GIRK channels are important functional effectors of the P2Y(12) receptor in human platelets.

Abstract:Integrins are heterodimeric receptors comprising α and β subunits. They are expressed on the cell surface and play key roles in cell adhesion, migration, and growth. Several types of integrins are expressed on the platelets, including αvβ3, αIIbβ3, α2β1, α5β1, and α6β1. Among these, physically αIIbβ3 is exclusively expressed on the platelet surface and their precursor cells, megakaryocytes. αIIbβ3 adopts at least three conformations: i) bent-closed, ii) extended-closed, and iii) extended–open. The transition from conformation i) to iii) occurs when αIIbβ3 is activated by stimulants. Conformation iii) possesses a high ligand affinity, which triggers integrin clustering and platelet aggregation. Platelets are indispensable for maintaining vascular system integrity and preventing bleeding. However, excessive platelet activation can result in myocardial infarction (MI) and stroke. Therefore, finding a novel strategy to stop bleeding without accelerating the risk of thrombosis is important. Regulation of αIIbβ3 activation is vital for this strategy. There are a large number of molecules that facilitate or inhibit αIIbβ3 activation. The interference of these molecules can accurately control the balance between hemostasis and thrombosis. This review describes the structure and signal transduction of αIIbβ3, summarizes the molecules that directly or indirectly affect integrin αIIbβ3 activation, and discusses some novel antiαIIbβ3 drugs. This will advance our understanding of the activation of αIIbβ3 and its essential role in platelet function and tumor development.

Abstract:Integrin αIIbβ3 is the key receptor regulating platelet retraction and accumulation and a proven drug-target for antithrombotic therapies. Here we resolve the cryo-EM structures of the full-length αIIbβ3, which covers three distinct states along the activation pathway. Firstly, we obtain the αIIbβ3 structure at 3 Å resolution in the inactive state, revealing the overall topology of the heterodimer with the transmembrane (TM) helices and the ligand-binding domain tucked in a specific angle proximity to the TM region. After the addition of a Mn2+ agonist, we resolve two coexisting structures representing two new states between inactive and active state. Our structures show conformational changes of the αIIbβ3 activating trajectory and a unique twisting of the integrin legs, which is required for platelets accumulation. Our structure provides direct structural evidence for how the lower legs are involved in full-length integrin activation mechanisms and offers a new strategy to target the αIIbβ3 lower leg.
Multi-phased kinetics and interaction of protein kinase signaling in glycoprotein VI-induced platelet αIIbβ3 integrin activation and degranulation

Abstract:Background: Platelet glycoprotein VI (GPVI) stimulation activates the tyrosine kinases Syk and Btk, and the effector proteins phospholipase Cγ 2 (PLCγ2) and protein kinase C (PKC). Here, the activation sequence, crosstalk and downstream effects of this Syk-Btk-PKC signalosome in human platelets was analyzed. Methods and Results: Using immunoblotting, we quantified 14 regulated phospho-sites in platelets stimulated by convulxin with and without inhibition of Syk, Btk or PKC. Convulxin induced fast, reversible tyrosine phosphorylation (pY) of Syk, Btk, LAT and PLCγ2, followed by reversible serine/threonine phosphorylation (pS/T) of Syk, Btk and downstream kinases MEK1/2, Erk1/2, p38 and Akt. Syk inhibition by PRT-060318 abolished all phosphorylations, except Syk pY352. Btk inhibition by acalabrutinib strongly decreased Btk pY223/pS180, Syk pS297, PLCγ2 pY759/Y1217, MEK1/2 pS217/221, Erk1/2 pT202/Y204, p38 pT180/Y182 and Akt pT308/S473. PKC inhibition by GF109203X abolished most pS/T phosphorylations except p38 pT180/Y182 and Akt pT308, but enhanced most Y-phosphorylations. Acalabrutinib,but not GF109203X, suppressed convulxin-induced intracellular Ca2+ mobilization, whereas all three protein kinase inhibitors abolished degranulation and αIIbβ3 integrin activation assessed by flow cytometry. Inhibition of autocrine ADP effects by AR-C669931 partly diminished convulxin-triggered degranulation. Conclusion: Kinetic analysis of GPVI-initiated multisite protein phosphorylation in human platelets demonstrates multiple phases and interactions of tyrosine and serine/threonine kinases with activation-altering feedforward and feedback loops partly involving PKC. The protein kinase inhibitor effects on multisite protein phosphorylation and functional readouts reveal that the signaling network of Syk, Btk and PKC controls platelet granule exocytosis and αIIbβ3 integrin activation.

Modeling Platelet P2Y$_1$/$_{12}$ Pathway to Integrin Activation

Abstract:Through experimental studies, many details of the pathway of integrin $\alpha_{\rm IIb}\beta_3$ activation by ADP during the platelet aggregation process have been mapped out. ADP binds to two separate G protein coupled receptors on platelet surfaces, leading to alterations in the regulation of the small GTPase RAP1. We seek to (1) gain insights into the relative contributions of both pathways to RAP1-mediated integrin activation and to (2) predict cell behavior in response to a continuous range of external agonist concentrations. To this end, we develop a dynamical systems model detailing the action of each protein in the two pathways up to the regulation of RAP1. We perform a parameter estimation using flow cytometry data to determine a number of unknown rate constants. We then validate with already published data; in particular, the model confirmed the effect of impaired P2Y$_1$ receptor desensitization or reduced RASA3 expression on RAP1 activation. We then predict the effect of protein expression levels on integrin activation and show that components of the P2Y$_{12}$ pathway are critical to the regulation of integrin. This model aids in our understanding of interindividual variability in platelet response to ADP and therapeutic P2Y$_{12}$ inhibition. It also provides a more detailed view of platelet activation in the ongoing mathematical study of platelet aggregation.

Abstract:Summary Human platelets express two distinct G protein-coupled ADP receptors, one coupled to phospholipase C through Gq, P2Y1, and the other to inhibition of adenylyl cyclase through Gi, P2TAC. We have recently shown that concomitant intracellular signaling from both the P2TAC and P2Y1 receptors is essential for ADP-induced platelet aggregation. Previous studies have tested whether ADP causes a decrease in the basal cAMP level and this reduction promotes platelet aggregation, but did not study the effect of decreased cAMP levels when the Gq pathway is selectively activated. Since we are now aware that platelet aggregation requires activation of two receptors, we investigated whether the function of P2TAC receptor activation, leading to inhibition of platelet adenylyl cyclase, could be replaced by direct inhibition of adenylyl cyclase, when Gq pathway is also activated, a possibility that has not been addressed to date. In the present study, we supplemented the P2Y1 mediated Gq signaling pathway with inhibition of the platelet adenylyl cyclase by using SQ22536 or dideoxyadenosine, or by selective activation of the α2A adrenoceptors with epinephrine. Although SQ22536, dideoxyadenosine, and epinephrine reduced the cAMP levels, only epinephrine could mimic the P2TAC receptor mediated signaling events, suggesting that reduction in basal cAMP levels does not directly contribute to ADP-induced platelet activation. Adenosine-5’-phosphate-3’-phosphosulfate, a P2Y1 receptor antagonist, completely blocked ADP-induced inositol 1,4,5-trisphosphate and inositol 1,3,4-trisphosphate formation suggesting that P2TAC-mediated activation of Gi (or other G proteins) does not activate phospholipase C. These results suggest that a signaling event downstream from Gi, independent of the inhibition of platelet adenylyl cyclase, contributes to αIIbβ3 activation.
