Abstract:Based on the literature on the degradation mechanism and the change of micro-functional groups in the fermentation process of modified rice straw, this study aimed to solve the problems of low biogas production rate and poor stability of the biogas production system. In this work, mathematical equations were developed and combined with duck dung and rice straw mixed raw material to perform a fermentation test. The molecular micro-functional group changes of cellulose, hemicellulose, and lignin were studied to obtain the optimal ratio of mixed raw materials for fermentation and to explore the optimization mechanism of its fermentation biogas production. Experimental results showed that the optimal ratio of mixed raw materials was 2.8:1, and the inclusion of a suitable amount of Mn2+(concentration of 2 mol × L-1) was able to strengthen MnP activity and improve the ability of white-rot fungi to rupture β-O-4 bonds. A modification pre-treatment via activated carbon-based solid acid was performed, and the experimental group generated 15.8% more cumulative biogas than the control group. The biogas yield reached its peak when 300 g of inoculum was added to the pre-treatment at a concentration of 30%.

Abstract:Cellulose, the main structural component of plant cell walls, is the most abundant renewable resource in nature, but it is extremely difficult to be degraded. Cellulosome is a multienzyme complex that can efficiently degrade cellulose and the degradation products can be used by some anaerobic microorganisms to produce ethanol. Recent research status in anaerobic degradation of cellulose for ethanol production by cellulosome-producing bacteria was reviewed in this paper. The latest achievements and research development in cellulosomes’ structure and function, designer cellulosomes, metabolic engineering and co-culture of cellulosome producing microorganisms were also summarized. It is expected that the development of new cellulose degrading microorganisms and relative technology will further cut the cost of cellulosic ethanol in the near future and make it more competitive with gasoline.

Abstract:Anaerobic degradation of lignin in waste straw by ruminal microbes was directly observed using atomic force microscope (AFM). A series of high-resolution AFM images of the straw surface in the biodegradation show that the wax flakelets and lignin granules covering the straw surface were removed by the rumen microorganisms. Such degradation resulted in an exposure of cellulose fibers located inside the straw. The appearance of holes and microfibers; in fermentation reveals that tunneling might be one of the ways for rumen microorganisms to attack the straw. Increases in the atomic ratio of oxygen to carbon (O/C) and the ratio C2/C3 in C1s spectra of X-ray photoelectron spectroscopy confirm that more cellulose was exposed on the surface after the anaerobic fermentation of straw. Gas chromatography/mass spectrometry analytical results demonstrate the decomposition of lignin by rumen microorganisms. Fourier transform infrared spectroscopy spectra and the measurement of degradation efficiency of the main straw components further verify these microscaled observations.

Abstract:In order to investigate the growth and degradation characteristics of a microbial community with high degradation efficiency on lignocelluloses,a batch test was carried out under static condition at 50℃,and 1% rice straw was the only carbon source.The result indicates that dominant bacteria of the microbial community are bacilli,and growth curve shows typical three stages,they are 0~108 h logarithmic stage,108~144 h stable stage and 144~216 h decline stage,respectively.The degradation rate of rice straw may reach 75% when fermentation terminated.Over 90% metabolite is acetic acid,and there is little ethanol,butyric acid and propionic acid in fermentative products.This fermentation type will provide essential material base for stable and effective operation of downstream methanogenic phase in two-phase anaerobic digestion process.

Abstract:In composting, the degradation of lignocellulose in straw is problematic due to its complex structures such as lignin. A common solution to this problem is the addition of exogenous inoculants. AC-1, a stable thermophilic microbial composite, was isolated from high temperature compost samples that can decompose lignocellulose at 50–70 °C. AC-1 had a best degradation efficiency of rice straw at 60 °C (78.92%), of hemicellulose, cellulose and lignin were 82.49%, 97.20% and 20.12%, respectively. It showed degrad-ability on both simple (filter paper, absorbent cotton) and complex (rice straw) cellulose materials. It produced acetic and formic acid during decomposition process and the pH had a trend of first downward then upward. High throughput sequencing revealed the main bacterial components of AC-1 were Tepidimicrobium , Haloplasma, norank-f-Limnochordaceae, Ruminiclostridium and Rhodothermus which provides major theoretical basis for further application of AC-1.

Abstract:The rapid development of agriculture has led to the production of a large amount of crop straw, necessitating effective strategies for its management. Microbial degradation offers a promising method. In this study, a novel microbial consortium composed of Phanerochaete chrysosporium, Aspergillus niger, and Streptomyces griseorubens, known for their robust lignocellulose degradation capabilities, was constructed for rice straw degradation. The establishment of this microbial consortium was based on the growth curve and antagonistic tests. Orthogonal optimization revealed that Streptomyces griseorubens played a predominant role in the degradation of rice straw. The optimal degradation conditions were determined as follows: nitrogen source concentration of 2.5 gL- 1, material-liquid ratio of 40 g L- 1, inoculum size of 3%, and pH value of 9. Under these conditions, the degradation efficiency reached 42% within 15 days. The decomposition of lignocellulosic components in the straw was confirmed through various characterization methods. Additionally, as the degradation process progressed, there was a noticeable decrease in protein-like substances and an increase in humic acid-like substances in the degradation solution.
Properties of cellulose-degrading and hydrogen-producing bacteria flora in rumen fluid at different pH

Abstract:A stable and efficient degradation community was developed based on different initial pH to improve the degradation rate of cellulose and study types and function of bovine rumen microorganisms .The structure and function of bacteria were analyzed by PCR-DGGE .The result showed that the utilization rate of fermentation broth with 5 g/L microcrystalline cellulose as carbon source could reach to the maximum of 81 .3% .The hydrogen production at pH 6 .5 could reach to 178 .2 mL/L .Its degradation rate and hydrogen production were higher than those reported in cellulose degrading bacteria .The community structure analysis showed that it was basically stable after 4 times of domestication . The community is consist of cellulolytic bacteria such as Ruminococcaceae , hydrogen producers such as Anaero f ilum , Enterococcus , Clostridium and Enterobacter cloacae producing H2 from cellulose .

Abstract:Straw cellulose is an abundant renewable resource in nature. In recent years, the conversion of cellulose from waste straw into biofuel by specific microorganisms' fragmentation has attracted extensive attention. Although many bacteria with the ability to degrade cellulose have been identified, comprehensive bioinformatics analyses of these bacteria remain limited, and research exploring optimal fragmentation conditions is scarce. Our study involved the isolation and screening of bacteria from various locations in Yangzhou using carboxymethyl cellulose (CMC) media. Then, the cellulose-degrading bacteria were identified using 16S rRNA and seven candidate bacterial strains with cellulose degrading ability were identified in Yangzhou city for the first time. The cellulase activity was determined by the 3,5-dinitrosalicylic acid (DNS) method in different fragmentation conditions, and finally two bacteria strains with the strongest cellulose degradation ability were selected for whole genome sequencing analysis. Sequencing results revealed that the genome sizes of Rhodococcus wratislaviensis YZ02 and Pseudomonas Xanthosomatis YZ03 were 8.51 Mb and 6.66 Mb, containing 8,466 and 5,745 genes, respectively. A large number of cellulose degradation-related genes were identified and annotated using KEGG, GO and COG analyses. In addition, genomic CAZyme analysis indicated that both R. wratislaviensis YZ02 and P. Xanthosomatis YZ03 harbor a series of glycoside hydrolase family (GH) genes and other genes related to cellulose degradation. Our finding provides new options for the development of cellulose-degrading bacteria and a theoretical basis for improving the cellulose utilization of straw.

Abstract:A microbial consortium, designated LZF-12, was obtained by successive subcultivation in the presence of rice straw under static conditions at 35 ºC.The number of bacilli was dominant in microbial consortium by scanning electron microscopy.The degradation efficiencies of LZF-12 for rice straw, hemicelluloses, cellulose and lignin were 70%、92.9%、82.6%and 21.8%, respectively for 12 days cultivation.There was near 70% of acetic acid in end metabolism products in addition to little butyric acid, propionic acid and ethanol.This fermentation type will provide essential material base for stable and effective operation of methanogenic phase in two-phase anaerobic digestion process.

Abstract:Batch experiments were performed to investigate the anaerobic degradation of crystalline cellulose of 10gl−1 by rumen microorganisms at pH from 4.8 to 7.3. The degradation efficiency increased with pH and the highest value of about 78% was achieved at pH 6.8 and 7.3. Acetate and propionate were the major aqueous products at all pH values. With increasing pH, the molar percentage of acetate decreased, whereas that of propionate increased. A modified Gompertz equation was able to adequately model the fermentation of cellulose by rumen microorganisms. The results from this study might provide useful information for the application of rumen cultures for conversion of cellulosic wastes into value-added products.

Abstract:To evaluate the potential utility of pretreatment of raw biomass with a complex microbial system, we investigated the degradation of rice straw by BMC-9, a lignocellulose decomposition strain obtained from a biogas slurry compost environment. The degradation characteristics and corresponding changes in the bacterial community were assessed. The results showed that rapid degradation occurred from day 0 to day 9, with a peak total biomass bacterium concentration of 3.3 × 10(8) copies/ml on day 1. The pH of the fermentation broth declined initially and then increased, and the mass of rice straw decreased steadily. The highest concentrations of volatile fatty acid contents (0.291 mg/l lactic acid, 0.31 mg/l formic acid, 1.93 mg/l acetic acid, and 0.73 mg/l propionic acid) as well as the highest xylanse activity (1.79 U/ml) and carboxymethyl cellulase activity (0.37 U/ml) occurred on day 9. The greatest diversity among the microbial community also occurred on day 9, with the presence of bacteria belonging to Clostridium sp., Bacillus sp., and Geobacillus sp. Together, our results indicate that BMC-9 has a strong ability to rapidly degrade the lignocelluloses of rice straw under relatively inexpensive conditions, and the optimum fermentation time is 9 days.

Abstract:The degradation of rice straw without any chemical method pretreated by a lignocellulose degradation composite microbial system XDC-2 was studied.After 6 days cultivation,exocellular xylanase activity was 335 U/mL.After 12 days,the rice straw lost 39.71% of its weight,and the weight loss of hemicellulose and cellulose were 78.27% and 14.08% respectively.The total amount of the volatile products came to a maximum in the sixth day.Four major kinds of the volatile products were determined in fermentation broth,they were acetic acid,propionic acid,butanoic acid and glycerin,which could be utilized in industrial production.The composite microbial system XDC-2 became stable in degradation process of rice straw without any chemical method pretreated according to PCR-DGGE analysis.Accordingly,it has the potential for further development and application of composite microbial system XDC-2 due to its effective ability to degrade rice straw without any chemical pretreatment.
[Productions Analyses and Ph Dynamics During Rice Straw Degradation by the Lignocellulose Degradation Bacteria System WSC-6].

Abstract:To detect the metabolic characteristic of rice straw degradation by composite microbial system WSC-6, we cultured WSC-6 in the media used rice straw as the limiting carbon source. The rice straw was added in the style of different quantity once or the same quantity at the different time intervals during 90 days culture. The systems were cultivated under static condition at 50 degrees C. The degradation ratio, absolute degradation quantity,products from degradation and dynamics of pH value of fermentation system were all investigated. The results showed: when 1% rice straw was added once, the pH of fermentation system decreased from initial 7.8 to 6.0 within the first three days inoculation, and after six-day cultivation, it increased to 8.0 and was stable. For dissolved oxygen concentration (DO), the value was maintained at range of 0.01 to 0.12 mg x L(-1) of microaerobic condition. During the rice straw degradation, more than ten kinds of products including ethanol, acetic acid, lactic acid and glycerol and so on were detected using GC-MS. Especially, the highest concentration of lactic acid among all products was 7.381 g x L(-1) at 24 h after inoculation. During 90-day cultivation, for the addition treatments of the different quantity once, the more rice straw added, the quicker and lower the pH decreased, and the longer time intervals returned the pHs were. Especially for 5.0% addition, when 5.0% of rice straw was added once, pH did not increase again after it decreased. Among the addition of the same quantity at the different time intervals, the trend of decrease-increase in pH at 12-day and 15-day intervals could be repeated and high degradation activity well maintained. After 90-day of inoculation, the highest degradation ratio occurred in the treatment at 15-day interval, which was 86.7%. The highest absolute quantity occurred in the treatment at 6-day interval, which was 32.4 g. The trend of pH changes can indicate the activity of lignocellulose degradation and degradation process of the WSC-6.
Construction of a Stable Microbial Community with High Cellulose-Degradation Ability.

Abstract:We bred a microbial community capable of degrading rice straw with high efficiency. The microbial community degraded more than 60% of rice straw within 4 days at 50 degrees C. The high stability of the community's degradation ability was demonstrated by its tolerance of being subcultured several times in medium with/without cellulosic material, being heated to 95 degrees C, and freezing at -80 degrees C. The community degraded both nonsterilized and sterilized substrate; and its degradation ability was not affected by pH changes in the medium (initial pH 5-9). PCR-denaturing gradient gel electrophoresis (DGGE) analyses based on 16S rDNA fragments showed that the community structure remained constant after multiple subcultures extending over 2 years. DNA sequence analyses of DGGE bands indicated the coexistence of both aerobic and anaerobic bacteria in the community.

Abstract:Rumen in the mammalian animals is a natural cellulose-degrading system and the microorganisms inside have been found to be able to effectively digest lignocellulosic biomass. Furthermore, methane or volatile fatty acids, which could be further converted to other biofuels, are the two major products in such a system. This paper offers an overview of recent development in the application of rumen microorganisms for lignocellulosic biomass conversion. Application of recent molecular tools in the analysis of rumen microbial community, progress in the development of artificial rumen reactors, the latest research results about characterizing rumen-dominated anaerobic digestion process and energy products are summarized. Also, the potential application of such a rumen-dominated process is discussed.
Straw Bio-Degradation by Acidogenic Bacteria and Composite Fungi.

Abstract:A composite microbial system, including a strain of Candida tropicalis (W3), a strain of Lactobacillus plantarm(WY3) and three strains of basidiomycete pL104, pL113 and C33, was chosen to degrade corn straw. The final pH was acid owing to the inoculation of acidogenic bacteria, and under this condition the composite fungi system could produce complex enzyme to destroy the compact structure of corn straw. The experimental results showed that the biomass of composite fungi could reach up to maximum when the pH value was 4.5. Through the bio-degradation by combining acidogenic bacteria with the composite fungi system, the cellulose, hemi-cellulose and lignin degradation rates of corn straw powder were 26.36%, 43.30% and 26.96%, respectively. And the gross crude protein content increased 60.41%. This study provided the evidence for the feasibility of developing a composite microbial system with high capability of degrading straw lignocelluloses in order to make reasonable use of straw resource and protect rural eco-environment.

Abstract:In this study, a novel psychrotrophic lignocelluloses degrading microbial consortium LTF-27 was successfully obtained from cold perennial forest soil by successive enrichment culture under facultative anaerobic static conditions. The microbial consortium showed efficient degradation of rice straw, which cellulose, hemicelluloses and lignin lost 71.7%, 65.6% and 12.5% of its weigh, respectively, in 20 days at 15 degrees C. The predominant liquid products were acetic acid and butyric acid during degrading lignocellulose in anaerobic digestion (AD) process inoculated with the LTF-27. The consortium mainly composed of Parabacteroides, Alcaligenes, Lysinibacillus, Sphingobacterium, and Clostridium, along with some unclassified uncultured bacteria, indicating powerful synergistic interaction in AD process. A multi-species lignocellulolytic enzyme system working cooperatingly on lignocelluolse degradation was revealed by proteomics analysis of cellulose bound fraction of the crude extracellular enzyme, which provides key theoretical base for further exploration and application of LTF-27.

Abstract:The biological degradation of plant residues in the soil or on the soil surface is an integral part of the natural life cycle of annual plants and does not have adverse effects on the environment. Crop straw is characterized by a complex structure and exhibits stability and resistance to rapid microbial decomposition. In this study, we conducted a microcosm experiment to investigate the dynamic succession of the soil microbial community and the functional characteristics associated with lignocellulose-degrading pathways. Additionally, we aimed to identify lignocellulose-degrading microorganisms from the straw of three crop species prevalent in Northeast China: soybean (Glycine max Merr.), rice (Oryza sativa L.), and maize (Zea mays L.). Our findings revealed that both the type of straw and the degradation time influenced the bacterial and fungal community structure and composition. Metagenome sequencing results demonstrated that during degradation, different straw types assembled carbohydrate-active enzymes (CAZymes) and KEGG pathways in distinct manners, contributing to lignocellulose and hemicellulose degradation. Furthermore, isolation of lignocellulose-degrading microbes yielded 59 bacterial and 14 fungal strains contributing to straw degradation, with fungi generally exhibiting superior lignocellulose-degrading enzyme production compared to bacteria. Experiments were conducted to assess the potential synergistic effects of synthetic microbial communities (SynComs) comprising both fungi and bacteria. These SynComs resulted in a straw weight loss of 42% at 15 days post-inoculation, representing a 22% increase compared to conditions without any SynComs. In summary, our study provides novel ecological insights into crop straw degradation by microbes.
Isolation and Characterization of an Efficient Cellulose-decomposing Strain YN1

Abstract:To obtain cellulose-decomposing strains, and to use them for decomposition of rice straw and rice hull. A cellulose-decomposing strain YN1 was isolated from cattle and sheep feces compost. Strain YN1 was identified by morphologic and phylogenetic analysis. Moreover, we performed the liquid and solid fermentation of YN1, and the degrading test of cellulose. In addition, scan electron microscope was used to observe the fixed propagation of YN1 on the cellulose and cellulose degradation. The morphologic and ITS gene sequence analysis suggested that YN1 was Aspergillus sp.. Through liquid fermentation, endoglucanases, exoglucanases, beta-glucosidases and the total cellulase activities reached to maximum at the 3th day, and they were 95.7, 14.6, 20.5 and 26.6 U/mL respectively. Though solid fermentation, the 4 kinds of cellulase activities reached to maximum at the 5th day, and they were 1192.2, 100.6, 136.9 and 210.7 U/g respectively. In the degrading experiment, YN1 could degrade 41.87% of rice straw, 31.59% of rice hull in weight in 7 days respectively. The remarkable phenomenon that YN1 degrading crude cellulose could be observed by scan electron microscope. YN1 showed value of practical applications in rice straw and rice hull decomposition.
