Abstract:The methods for synthesizing 1,8-naphthalimide and its derivatives were narrated in brief, and their application progresses in the fields of fluorescent brightening agent, fluorescent dye and other functional materials in recent years were emphatically introduced, especially the application in the fields of biomedical material and fluorescent sensor. The applications in the biomedical materials included mainly anticancer drugs, DNA fluorescence probe and fluorescence molecular switch. And the anion sensor and cation sensor were included in the application fields of fluorescent sensor. Finally, the development trend of these functional materials was forecasted.

Abstract:Abstract Naphthalimide derivatives are attracting increasing attention as fluorescent probes due to their excellent modifiability and photophysical properties. Their large, rigid coplanar structures and conjugated systems enable easy generation of fluorescence. The imide units and bromonaphthalene functionality offer facile and concise modification pathways in preparing amenable probes for different metal cations, anions, small molecules, and organelles. Therefore, naphthalimide probes have great potential for application in research fields, such as analytical chemistry, cell biology, and clinical diagnosis.

Abstract:In recent years, more and more chemists and biologists pay attention to the research of fluorescent probes, and great progress has been made in the design and application. In this review, we first discuss the excellent photophysical properties of 4-hydroxy-1,8-naphthalimide (HNI) structure, including excellent photostability, high fluorescence quantum yield, and easy modification of the structure. Then, by discussing the fluorescence behavior of the probe in the corresponding recognition process, the contributions of fluorescent probes built on the platform of HNI in the fields of chemical sensing, biological imaging, pharmaceutical chemistry, environment and food safety in the past ten years are systematically introduced (173 references). At the same time, we hope to develop more powerful fluorescent chemical sensors, which can play a more extensive and exciting role in the future.

This can be a major breakthrough when combined with magnetic resonance imaging and nanotechnology. It is not meant to be a comprehensive review of the field but rather to give some flavour of this fast growing area of research and offer new ideas for the design and application of fluorescent chemosensors.

This account aims at the first-year graduate students as the primary audience and showcases the versatile design principles applicable to the naphthalimide fluorophore when designing a probe or label, with focused examples from the Qian research laboratory. We also provide a general synthetic scheme to naphthalimides of various substitution patterns.

Abstract:Life are governed by cellular complex metabolic processes. One of the on-going endeavours in the scientific community is to study and understand these dynamic biochemical reactions and the roles of biomolecules in sustaining life. Fluorescent probes have been widely used in the visualization of analytes in physiological and pathological processes, due to their advantages such as easy operation, low cost, high sensitivity, capability of multi-channel and real-time visualization in vivo. However, most fluorescent probes can only respond to single analyte, which is not suitable to mornitor multiple analytes in complex biological system. In recent five years, significantly more multi-analyte fluorescent probes based on 1, 8-naphthimide fluorophore have been designed in biological or environmental field. Categorized by the fluorescent mechanisms, such as single fluorescent mechanism (e. g. photoinduced electron transfer (PET), intramolecular charge transfer (ICT) and fluorescence resonance energy transfer (FRET)), dual fluorescent mechanisms (e. g. PET-ICT, PET-FRET, and ICT-FRET) and so on, this review highlights the recent progress in designing strategies, corresponding recognition process, optical property and cell imaging of 1, 8-naphthalimide-based multi-analyte fluorescent probes. Finally, the development trend and possible challenges of such fluorescent probes are prospected.

Abstract:Research into next‐generation fluorescent functional molecules in biomedicine has gained prominence recently. This review mainly summarizes the structures and performances of the 1,8‐naphthalimide‐based functional molecules, including fluorescence sensors for monitoring metal ions and biological small molecules in organisms, and multifunctional photosensitizers for disease therapy. 1,8‐Naphthalimide fluorophore, as the classical skeleton for fluorescent functional molecules, has gained much attention in the chemistry and biomedical field due to its excellent fluorescent properties, photostability, and structural flexibility. The rational design of the substituents on the C‐4 position and N‐position of 1,8‐naphthalimide chromophore can provide specific functions for the molecules, such as fluorescence sensing, energy‐trapping, and targeting. This review presents the design progress of 1,8‐naphthalimide‐based fluorescence sensors for metal ions and biological molecules, and 1,8‐naphthalimide‐functionalized photosensitive dyes in the biological field over the past years. We summarize the general fluorescent molecular design strategies, recent developments of 1,8‐naphthalimide in innovative photosensitizers, and the challenges, wishing to promote new ideas in developing smart and functional molecules from 1,8‐naphthalimide derivatives.

Abstract:A series of novel mono- and di-substituted N-n-butyl-1,8-naphthalimide derivatives were synthesized simultaneously via a three-step reaction. The single crystal structure of N-n-butyl-4-[N',N'-bis(2',4'-dichlorobenzoyl)ethylamino]-1,8-naphthalimide (3f) was determined. The UV-vis and fluorescence properties of compound 3f were investigated. The 3f showed highly selective and sensitive fluorescence changes response towards Pb2+. A titration of monomer with Pb2+ ion was performed. When Pb2+ ion concentration increased from 0 to 10 eq., the fluorescent intensity of 3f decreased from 199.97 to 48.21. The pH effect on 3f showed that it is stable in a wide range of pH. The results indicated that 3f might be a probe molecule for Pb2+.

Abstract:In this feature article, we report our recent progresses in fluorescent sensors of biological dyes from the viewpoint of supramolecular and bioorganic chemistry. For signalling fluorophores, we extended or created naphthalene-based ICT systems, e.g. amino-1,8-naphthalimides, amino-1,8-dicyanonaphthalenes and acenaphthopyrrol-9-carbonitriles. We also developed BODIPY derivatives with large Stokes shifts and high fluorescence quantum yields in polar solvents, and a rhodamine analogue working in strong competitive aqueous solution as well as its silaanthracene analogue with a bathochromic shift as large as 90 nm. For sensing mechanisms, we extended or developed the following methods to improve sensing: e.g. PET in a photogenerated electronic field, TICT promoted PET derived from aminoalkyl or piperazino aminonaphthalimides, and the translation/amplification effect of surfactant micelles or aggregation on fluorescent sensing. We also successfully designed deprotonation strengthened ICT, FRET-chemodosimeter sensing systems. For non-cyclic recognition receptors, naphthalimides with two or more side chains at their 4,5- or 3,4-positions, as a convenient and simple platform for ratiometric sensors, were created for the recognition of heavy and transition metallic cations; multi-armed polyamides with more side chains were innovated as a versatile platform for the sensing of metal ions with high affinity, selectivity and positive homotropic allosteric effects. We designed V-shape sensors of the bis(aminomethyl)pyridine receptor with two fluorophores to show high performance. Finally, the intracellular applications of the above sensors and dyes, e.g. imaging heavy and transition metal ions in cells, fluorescent marking of hypoxia of tumour cells, are also reviewed.

Abstract:The accurate determination of specific biomolecules and their monitoring in complex biological systems will help us understand their role in the body. In comparison with cumulative fluorescent probes, responsive fluorescent probes exhibit a higher level of sensitivity for the detection of biomolecules. Recently, responsive fluorescent probes based on 1,8-naphthalimide have been used widely in the biomedical field because of their excellent luminescence performance. This review focuses on the progress in research on responsive fluorescent probes obtained by modifying the C4-position of 1,8-naphthalimide to achieve biological molecule detection over the last five years. Based on the differences in species, the corresponding recognition groups and responsive probes were classified systematically and thoroughly, and the significance of the probes used specifically for biological molecule detection was discussed. Finally, we discussed the prospect of developing this type of probe, to make it a powerful diagnostic tool in clinical research.

Abstract:A new fluorescent chemosensor 3 based upon bis-4-piperidine-1,8-naphthalimide linked by the diethylenetriamine was synthesized, and its fluorescent properties were also investigated. Different from other bis-1,8-naphthalimides, no changes in the fluorescence spectra of 3 in the presence of metal cations was observed, and it displayed an unexpectedly strong fluorescence quench in weakly acidic aqueous solutions. Its pKa value (7.25) indicated that it would be suited as a new pH fluorescent chemosensor to monitor changes at the physiological pH range.

Abstract:A novel 4-amino-1,8-naphthalimide-based fluorescent sensor with iminodiacetic acid as receptor, was developed and applied successfully to image Zn2+ in living cells. Under physiological pH conditions, it demonstrates high selectivity and sensitivity for sensing Zn2+ with about 34-fold enhancement in aqueous solution, with a characteristic emission band of 4-amino-1,8-naphthalimide with a green color centered at 550 nm.

Abstract:The fluorophores based on xanthene scaffolds, mainly containing rhodamine and fluorescein dyes, have attracted considerable interest from chemists due to their excellent photophysical properties such as high absorption coefficient, high fluorescence quantum yield, high photostability and relatively long wavelengths of fluorescence emission spectra. Abstract:A fluorescent chemosensor N-n-butyl-3,4-diamino-1,8-naphthalimide (DAN) for detecting NO was developed on the basis of 1,8-naphthalimide with a similar structure of o-phenylenediamine as a NO reaction site. Due to the interaction between the photoinduced electron transfer (PET) and the intramolecular charge transfer (ICT), the chemosensor exhibits a remarkable enhancement in the emission intensity that is ca. 160-fold increase and a blue shift in the emission wavelength after the addition of NO. Meanwhile, it displays a colorimetric response accompanied with a color change from yellow to colorless. The chemosensor DAN shows a high selectivity for NO in the presence of various reactive nitrogen species (RNS) and reactive oxygen species (ROS). Furthermore, DAN can be used for bioimaging of NO in living cells.
