Abstract:Introduction: The characteristic of ABO blood subgroup is crucial for elucidating the mechanisms of such variant phenotypes and offering useful information in blood transfusion. Methods: In total, 211 ABO variants including part of available family members were investigated in this study. The phenotypes of these individuals were typed with serologic methods. The full coding regions of ABO gene and the erythroid cell-specific regulatory elements in intron 1 of them were amplified with polymerase chain reaction and then directly sequenced. The novel alleles were confirmed by cloning and sequencing. Phylogenetic tree was made using CLUSTAL W software. 3D structural analyses of the glycosyltransferases (GTs) with some typical mutations were performed by PyMOL software. Results: Forty-eight distinctly rare ABO alleles were identified in 211 Chinese variant individuals, including 16 novel ABO alleles. All of the alleles were categorized as 5 groups: 16 ABO*A alleles, 23 ABO*B alleles, 4 ABO*BA alleles, 4 ABO*cisAB alleles, and 1 ABO*O alleles. ABO*A2.08 and ABO*BA.02 were the relatively predominant A and B subgroup alleles, respectively. According to the phylogenetic tree, 28 alleles (5 common alleles and 23 alleles identified in our laboratory) were classified into 3 major allelic lineages. The structural analysis of 3D homology modeling predicted reduced protein stability of the mutant GTs and may explain the reduced ABO antigen expression. Conclusions: The molecular basis of ABO variants was analyzed, and 16 novel ABO alleles were identified. The results extended the information of ABO variants and provided a basis for better transfusion strategies and helped to improve blood transfusion safety.

Abstract:ABO blood group system is the most important system in the context of blood transfusion. A allele and B allele encode glycosyltransferase (GT) A and GT B, which can add UDP-N-acetylgalactosamine and UDP-galactose to the common precursor H antigen to form A antigen and B antigen, respectively. If there is a variant in the gene, insertion, deletion, or replacement, the transferase function may be impaired. In the present work, we found a Chinese individual was proved to have a weak A blood phenotype by serological testing. By polymerase chain reaction (PCR) sequencing-based typing (Sanger method) and gene cloning sequencing, we describe a novel variant, c.27delC on the ABO*A1.02 allele.

Here, we describe the identification of the novel allele HLA-A*11:01:10, which has been detected in a registered donor of the China Marrow Donor Program. Abstract:Background and objectives: The presence of blood subtypes may lead to difficulties in blood group identification; however, third-generation sequencing (TGS) can help in accurately identifying difficult blood groups, and study the serological characteristics and molecular mechanism of Ael subtypes. Materials and methods: ABO blood group was identified by the standard serological technique, weak blood group antigen was identified by adsorption-elution experiments, ABH substance in the saliva was determined and glycosyltransferase activity of A and B was detected. The ABO gene full-length sequence and promoter region were amplified by specific primers using single-molecule real-time sequencing, with the amplified products being sequenced directly and analysed in real time. Results: The patient was serologically identified as Ael subtype, and TGS analysis revealed new intron mutations in Ael patients (c.467C>T; c.29-10T>A). Conclusion: The discovery of the new allele and the identification of ABO subtypes can be combined with serological characterization and molecular biological methods.

Abstract:A novel HLA-A allele, HLA-A*0279, was identified using PCR-SSP and PCR-SBT methods. It is inheritable. HLA-A*0279 differs from HLA-A*020601 by a single nucleotide at position 497 in exon 3, leading to an amino acid change from Threonine to Isoleucine at the alpha2 helix of HLA molecule. To investigate whether the altered amino acid residue could affect its peptide-binding repertoire, we compared the predicted crystal structure of HLA-A*020601 and HLA-A*0279 by Swiss-PdbViewer software analysis. We found that the crystal structure of the two molecules is very similar except for a difference in the number of hydrogen bonds they can possibly form, which in turn could affect their structural stability. To test whether HLA-A*0279 has the ability to cross-present A*0201 - restricted peptides to T cells, the full lenght cDNA of HLA-A*0201, -A* 020601 and -A*0279 were respectively transfected into COS-7 cells, which were then used as targets in IFN-gamma release Elispot assay. A*2079 was found to be able to present A*0201- restricted peptides to and induce the response of CTL, thus it can be classified as member of the HLA-A2 functional supertype family. This finding would benefit the design of peptide vaccines to be applied in broader populations.

Abstract:Background and ObjectivesABO antigens are produced from H antigen by the activity of glycosyltransferase enzyme encoded by the ABO gene. Variants in the ABO gene can produce a weak ABO phenotype. In this study, we identify a novel ABO*BW allele and investigate the underlying mechanism leading to the B-weak phenotype. Materials and MethodsThe ABO phenotype and genotype of the sample were determined using serological and direct DNA sequencing methods. We assessed the impact of the novel variant by three-dimensional modelling to predict protein stability changes (& UDelta;& UDelta;G), and carried out an in vitro expression assay. The total glycosyltransferase transfer capacity in the supernatant of transfected cells was also examined. ResultsSerological analysis confirmed the B-weak phenotype in the subject, and gene sequencing identified a novel variant c.761C>T (p.A254V) on the ABO*B.01 allele, resulting in a BW-var/O.01.02 genotype. In silico analysis suggested that the p.A254V variant on the B allele may reduce the stability of glycosyltransferase B (GTB), as indicated by the & UDelta;& UDelta;G values. In vitro expression studies showed that the variant p.A254V impaired H to B antigen conversion, although it did not affect the expression of GTB. ConclusionWe identified a novel BW allele and demonstrated that the variant c.761C>T (p.A254V) can cause the B-weak phenotype by reducing the stability of GTB.

Abstract:The ABO gene is mainly composed of A, B, and O alleles. The most common O alleles share one nucleotide deletion of G at position 261 in the ABO gene. This report found a novel nonsense mutation in an ABO allele which led to group O in a Chinese individual. Forward and reverse typing tests were carried out first using the manual tube method. Weak expression of A or B antigens on red blood cells (RBCs) was confirmed by absorption and elution test. Exons 6 and 7 in the ABO gene were amplified and direct-sequenced. The haplotypes of the ABO gene were identified by clone-sequencing. Serologic results showed that the phenotype of proband was group O. Based on direct sequencing results, the proband was heterozygous for the ABO*A1.01 allele and ABO*O.01.01 allele, except for a heterozygous c.618C>G mutation. The clone sequencing demonstrated that a nonsense C>G mutation at position 618 was identified in exon 7 of the ABO*A1.01-like allele, which caused a p.Tyr206Ter exchange in the ABO glycosyltransferase. Thus, the novel ABO allele was classified as O allele. The study suggested that a novel nonsense mutation in the ABO gene was identified in a Chinese individual with group O, resulting in the truncated, 205-amino-acid ABO glycosyltransferase, which lost its transferase activity.

Abstract:AbstractBackgroundThe Am phenotype, which arises from mutations of the α‐1,3‐N‐acetylgalactosaminyltransferase gene, is rare in the Chinese population. The present study focuses on a novel mutation with the Am phenotype in a Chinese individual.Study design and methodsThe sample with ABO blood group discrepancy was analyzed by serologic techniques. The full coding and flanking regions of the ABO gene, including the Intron 1 transcription factor–binding site, were identified through direct sequencing of polymerase chain reaction (PCR)‐amplified products. PCR products of Exons 6 and 7 were validated to isolate the ABO gene haplotypes by cloning and sequencing individual colonies. The impact of the novel mutation on enzyme function was predicted with Polymorphism Phenotyping algorithm V2 and bioinformatic software programs.ResultsThe serologic characteristics of ABO blood typing showed the rare Am phenotype. The c.467C/T and c.912C/A heterozygous sites in Exon 7 were identified by direct sequencing analysis. Further TA cloning and sequencing revealed that the patient carried an ABO*O.01.01 allele and a novel ABO*A allele. The new allele sequence had one nucleotide alteration (C>A) at position 912 on the background of the ABO*A1.02 allele. The c.912C>A mutation was predicted to be “probably damaging” and “deleterious” by PolyPhen‐2 and PROVEAN algorithms, respectively.ConclusionA novel mutation c.912C>A in α‐1,3‐N‐acetylgalactosaminyltransferase gene resulting in Am phenotype was identified in a Chinese individual.

Abstract:Background and objectives: Discrepancy between forward and reverse ABO grouping could be due to several reasons including genetic mutations of the alleles encoding group specific transferase. The healthy donors found with weak A antigen were investigated to ascertain the allele responsible for variation. Materials and methods: Standard serological methods were employed using commercial antisera. The molecular sequencing was performed on DNA with enrichment library prep kit and a custom designed overlapping probe panel. Binary alignment mapping files, generated on board the Illumina MiSeq instrument and aligned to the GRCh37/Hg19 reference genome, were uploaded to the QIAGEN CLC genomics workbench software (version. 20) where variant call files were generated and analyzed. Results: Red blood cells (RBCs) of six healthy donors, showing weak mix-field agglutination by anti-A and anti-A, B and plasma with absence or weakly reacting anti-A, were investigated serologically. The RBCs incubated with anti-A yield positive elution and their saliva lacked A but possessed H antigen thereby classifying as a historical known phenotype Aend. Family study on 4 probands showed inheritance of the trait. Molecular studies revealed presence of ABO*A allele carrying rare novel variant referred to as c.106delinsGG in line with HGVS recommendation that was thought to be responsible for the variant of A. Conclusion: Six cases serologically defined as Aweak were found to be associated with novel allele ABO*A (c.106delinsGG). The Aweak phenotype with the novel allele has not been displayed on International Society of Blood Transfusion database, though c.106delinsGG is listed in the UCSC genome browser under rs782544248.

Abstract:Objective: Serological and molecular biology methods were used to identify the blood type of a patient with forward and reverse ABO typing inconsistency, and to explore the genetic characteristics of this blood type. Methods: The ABO phenotype of the proband was identified by tube method, and the ABO blood group genotype of the proband and her parents was determined by fluorescent PCR. The 7 exons of the ABO gene were directly sequenced and analyzed. Results: According to preliminary serological identification, the ABO phenotype of this patient was Bel subtype. Genotyping tests showed that the ABO genotype of the proband and her father was B/O1 , and her mother was O1/O1. Sequencing of exons revealed novel heterozygous variations in exon 1: c.16_17delinsTGTTGCA. Conclusion: The Novel variations in exon 1 led to Bel subtype in the ABO blood group of the proband, and these variations are heritable.
