Abstract:Given increasing use of copper-based nanomaterials, particularly in applications with direct release, it is imperative to understand their human and ecological risks. A comprehensive and systematic approach was used to determine toxicity and fate of several Cu nanoparticles (Cu NPs). When used as pesticides in agriculture, Cu NPs effectively control pests. However, even at low (5–20mg Cu/plant) doses, there are metabolic effects due to the accumulation of Cu and generation of reactive oxygen species (ROS). Embedded in antifouling paints, Cu NPs are released as dissolved Cu+2 and in nano- and micron-scale particles. Once released, Cu NPs can rapidly (hours to weeks) oxidize, dissolve, and form CuS and other insoluble Cu compounds, depending on water chemistry (e.g. salinity, alkalinity, organic matter content, presence of sulfide and other complexing ions). More than 95% of Cu released into the environment will enter soil and aquatic sediments, where it may accumulate to potentially toxic levels (>50–500μg/L). Toxicity of Cu compounds was generally ranked by high throughput assays as: Cu+2>nano Cu(0)>nano Cu(OH)2>nano CuO>micron-scale Cu compounds. In addition to ROS generation, Cu NPs can damage DNA plasmids and affect embryo hatching enzymes. Toxic effects are observed at much lower concentrations for aquatic organisms, particularly freshwater daphnids and marine amphipods, than for terrestrial organisms. This knowledge will serve to predict environmental risks, assess impacts, and develop approaches to mitigate harm while promoting beneficial uses of Cu NPs.
Toxicity of Copper Nanoparticles to Daphnia Magna under Different Exposure Conditions

Abstract:Although the risks of metallic nanoparticles (NPs) to aquatic organisms have already been studied for >10years, our understanding of the link between the fate of particles in exposure medium and their toxicity is still in its infancy. Moreover, most of the earlier studies did not distinguish the contribution of particles and soluble ions to the toxic effects caused by suspensions of metallic NPs. In this study, the toxicity of CuNPs to Daphnia magna upon modification of the exposure conditions, achieved by aging the suspensions of CuNPs and by altering water chemistry parameters like the pH and levels of dissolved organic carbon (DOC), was investigated. The LC50 values for CuNPs exposure decreased by about 30% after 7days of aging. The LC50 values increased >12-fold upon addition of DOC at concentrations ranging from 0 to 10mg/L to the exposure medium. Changing the pH from 6.5 to 8.5 resulted in a 3-fold higher LC50 value. Furthermore, it was found that during 7days of aging of the exposure medium (without addition of DOC and at pH7.8), the toxicity could be mostly ascribed to the particles present in the suspension (around 70%). However, adding DOC or decreasing the pH of the exposure medium reduced the contribution of the particles to the observed toxicity. We thus found that the effective concentration regarding the toxicity was mainly driven by the contribution of the soluble ions in the presence of DOC or at pH6.5. Our results suggest that the toxicity results of CuNPs obtained from laboratory tests may overestimate the risk of the particles in polluted waters due to the common absence of DOC in laboratory test solutions. Moreover, the role of the ions shedding from CuNPs is very important in explaining the toxicity in natural waters.

Abstract:There is increasing recognition that the wide use of nanoparticles, such as Cu (CuNPs) and ZnO nanoparticles (ZnONPs), may pose risks to the environment. Currently there is insufficient insight in the contribution of metal-based nanoparticles and their dissolved ions to the overall toxicity and accumulation. To fill in this gap, we combined the fate assessment of CuNPs and ZnONPs in aquatic test media with the assessment of toxicity and accumulation of ions and particles present in the suspensions. It was found that at the LC50 level of Daphnia magna exposed to the nanoparticle suspensions, the relative contributions of ions released from CuNPs and ZnONPs to toxicity were around 26% and 31%, respectively, indicating that particles rather than the dissolved ions were the major source of toxicity. It was additionally found that at the low exposure concentrations of CuNPs and ZnONPs (below 0.05 and 0.5 mg/L, respectively) the dissolved ions were predominantly accumulated, whereas at the high exposure concentrations (above 0.1 mg/L and 1 mg/L, respectively), particles rather than the released ions played a dominant role in the accumulation process. Our results thus suggest that consideration on the contribution of dissolved ions to nanoparticle toxicity needs to be interpreted with care.
Transformation of CuO Nanoparticles in the Aquatic Environment: Influence of Ph, Electrolytes and Natural Organic Matter

Abstract:Many studies have shown the effect of solution chemistry on the environmental behavior of metal-based nanoparticles (NPs), except CuO NPs. Here, we investigated the agglomeration, sedimentation, dissolution, and speciation of CuO NPs by varying pH, ionic strength, ionic valence, and natural organic matter (NOM). The results showed that as the pH moved away from 6, the size of CuO agglomerates decreased, along with the enhanced NP suspension stabilization, due to the increase of electrostatic repulsive force. Increasing ionic strength and valence intensified the agglomeration and sedimentation of CuO NPs because of the compression of electrical double layers. The presence of humic acid and citric acid enhanced the dispersion and stabilization of CuO NP suspension, but l-cysteine showed a different impact. Decreasing pH, increasing ionic strength and all NOM improved the dissolution of CuO NPs, but the divalent electrolyte (CaCl2) inhibited the Cu2+ release from CuO NPs compared to the monovalent electrolyte (NaCl). In addition, X-ray absorption near edge structure (XANES) analysis demonstrated that the presence of l-cysteine transformed more than 30% of CuO NPs to Cu(I)-cysteine by coordinating with thiol group. This study can give us an in-depth understanding on the environmental behavior and fate of CuO NPs in the aquatic environment.

Abstract:A full-factorial test design was applied to systematically investigate the contribution and significance of water chemistry parameters (pH, divalent cations and dissolved organic carbon (DOC) concentration) and their interactions on the behavior and fate of copper nanoparticles (CuNPs). The total amount of Cu remaining in the water column after 48 h of incubation was mostly influenced by divalent cation content, DOC concentration and the interaction of divalent cations and DOC. DOC concentration was the predominant factor influencing the dissolution of CuNPs, which was far more important than the effect of pH in the range from 6 to 9 on the dissolution of the CuNPs. The addition of DOC at concentrations ranging from 5 to 50 mg C/L resulted in a 3-5 fold reduction of dissolution of CuNPs after 48 h of incubation, as compared to the case without addition of DOC. Divalent cation content was found to be the most influential factor regarding aggregation behavior of the particles, followed by DOC concentration and the interaction of divalent cations and DOC. In addition, the aggregation behavior of CuNPs rather than particulate dissolution explained most of the variance in the sedimentation profiles of CuNPs. These results are meaningful for improved understanding and prediction of the behavior and fate of metallic NPs in aqueous environments. (C) 2017 Elsevier Ltd. All rights reserved.

Abstract:Toxicity of metallic nanoparticle suspensions (NP(total)) is generally assumed to result from the combined effect of the particles present in suspensions (NP(particle)) and their released ions (NP(ion)). Evaluation and consideration of how water chemistry affects the particle-specific toxicity of NP(total) are critical for environmental risk assessment of nanoparticles. In this study, it was found that the toxicity of Cu NP(particle) to Daphnia magna, in line with the trends in toxicity for Cu NP(ion), decreased with increasing pH and with increasing concentrations of divalent cations and dissolved organic carbon (DOC). Without the addition of DOC, the toxicity of Cu NP(total) to D. magna at the LC50 was driven mainly by Cu NP(ion) (accounting for ≥53% of the observed toxicity). However, toxicity of Cu NP(total) in the presence of DOC at a concentration ranging from 5 to 50mg C/L largely resulted from the NP(particle) (57%–85%), which could be attributable to the large reduction of the concentration of Cu NP(ion) and the enhancement of the stability of Cu NP(particle) when DOC was added. Our results indicate that water chemistry needs to be explicitly taken into consideration when evaluating the role of NP(particle) and NP(ion) in the observed toxicity of NP(total).

Abstract:Nanoparticles are used for several commercial applications, especially medical applications, energy-based research, and environmental applications. The usage of nanoparticles is increasing worldwide due to their unique properties, so all livings are exposed to these materials deliberately or unknowingly. The toxic nanoparticles disrupt the ecological balance of environment. The accumulation, surface area, particle size or shape of nanoparticles, exposure route, exposure time and dissolution contribute to the toxicity. The production of copper (II) oxide nanoparticles has expanded speedily in the last years so; it is important to know the risks that may be caused by their toxicological risks in ecosystems. This review emphasizes the environmental fate of copper (II) oxide nanoparticles. The environmental fate relates to their shapes, sizes, and physicochemical properties. The updated information on the toxicity of copper (II) oxide nanoparticles to aquatic organisms is given. Copper (II) oxide nanoparticles penetrate directly or indirectly into the organs of aquatic organisms through skin absorption, and/or nutrition. After the exposure, protein denaturation, protein oxidation, DNA damage, oxidative stress, generation of reactive oxygen species, hemolysis, and even cell death occurs in aquatic organisms. This review may assist in interdisciplinary studies to evaluate potential toxicity of copper (II) oxide nanoparticles in aquatic ecosystems.

Effects Of Copper Oxide Nanoparticles On Paddy Soil Properties And Components

Abstract:The wide use of metal-based nanoparticles (MNPs) will inevitably lead to their release into soil, and consequently affect the quality and ecological functions of soil environments. In this study, two paddy soils with different properties were exposed to CuO NPs to evaluate the transformation of CuO NPs and their effects on soil properties and components. The results of single chemical extraction and X-ray absorption fine structure analysis showed that CuO NPs could release Cu ions once being applied into the flooding paddy soil and then progress toward the more stable forms (Cu2S and Cu(OH)(2) ). CuO NPs could change the soil properties by increasing the pH and Eh of the lower organic matter-soil rather than those of the higher organic matter-soil. Furthermore, we found that the 1000 mg/kg CuO NPs could accelerate the degradation or mineralization of the organic matter, as well as the Fe reduction process, by increasing the Fe(II) content by 293% after flooding for 60 days in the lower organic matter soil. The microbial biomass in both soils was severely inhibited by CuO NPs and the organic matter could partly mitigate the negative effects of CuO NPs.

Abstract:Nanoparticles (NPs), heavy metals and natural organic matter may co-exist in the water bodies. Currently, knowledge on their interaction effects on the behaviors and fates of NPs and heavy metal ions is rather limited, which is critical to comprehensively understand their environmental risk. In this study, the aggregation, solubility and Cd-adsorption of CuO NPs co-existing with humic acid (HA) and Cd2+ upon different solution pH and contact sequences were determined. In the ternary systems of CuO NPs, HA and Cd2+, pH was more important than the contact sequence of the components in affecting the NP aggregation, while the contact sequence was a predominant factor in determining the NP solubility. Pre-equilibration of CuO NPs and HA before addition of Cd2+ resulted in the highest solubility and lowest aggregation of the NPs, relative to other sequences of addition of the components. The adsorption capacity of CuO NPs for Cd-ions increased with an increasing pH value from 5 to 9. HA significantly enhanced the Cd-adsorption capacity of CuO NPs at pH 7 and 9, while at pH 5 a non-significant effect was observed. The results are helpful to better estimate the behaviors and fates of CuO NPs and Cd2+ when they coexisting in natural waters.

Abstract:Copper nanoparticles (Cu-NPs) were widely used in various industrial and commercial applications. In this study the effects of Cu-NPs and soluble Cu were investigated on juvenile Epinephelus coioides. The fish were exposed in triplicate to control, 20 or 100 mu g Cu L-1 as either copper sulphate (CuSO4) or Cu-NPs in a semi-static aqueous culture for 25 days. The growth parameters were significantly lower at 100 mu g Cu L-1 as CuSO4 or Cu-NPs treatment compared to control. Time-dependent Cu accumulation in all tissues increased with increasing the Cu dose. The percentage of total Cu found in remaining tissues (head, bones, fins, etc.) decreased more in the CuSO4 than Cu-NPs treatment after 25 days, but increased in all other tissues (especially in liver). Compared with the control, either Cu-NPs or CuSO4 induced higher malonaldehyde concentration in tissues by overwhelming total superoxide dismutase activity, total glutathione concentration and Na+/K+-ATPase activity, but the opposite results were recorded for the brain. With increasing the CuSO4 or Cu-NPs dose, apoptosis was exacerbated in the liver and gills, more so by CuSO4 than Cu-NPs. Overall, these findings showed that Cu-NPs had the toxic effects similar to dissolved Cu; hence, Cu-NPs need to be included in the assessment of toxicological impacts in the aquatic environment. Abstract:Recently, studies on the biological effects of nanomaterials show signs that some of the manufactured nanoparticles exhibit unexpected toxicity to living organisms. It has previously been reported that the copper particles possess size-depended toxicity. In this paper, we propose that the ultrahigh chemical reactivity of nano-copper results in the specific nanotoxicity which is fully proved by in vitro and in vivo experiment. Using chemical kinetics study (in vitro) and blood gas and plasma electrolytes analysis (in vivo), we found that high reactivity cause the big toxicological difference between small size (23.5nm) and big size (17μm). The result is also consistent with biochemistry assay, pathological examination and copper content measurement in renal tissue in vivo. For chemical reactive nanoparticles, metallic nano-copper for instance, both the particles themselves and the resulting product (copper ion) should be fully explored. The nano-copper particles may not compromise the mice directly, however, they lead to the accumulation of excessive alkalescent substance and heavy metal ions (copper ions) culminating the metabolic alkalosis and copper ion overload.

Abstract:To assess the toxicity of copper nanoparticles (23.5 nm) in vivo, LD(50), morphological changes, pathological examinations and blood biochemical indexes of experimental mice are studied comparatively with micro-copper particles (17 microm) and cupric ions (CuCl(2).2H(2)O). The LD(50) for the nano-, micro-copper particles and cupric ions exposed to mice via oral gavage are 413, >5000 and 110 mg/kg body weight, respectively. The toxicity classes of nano and ionic copper particles both are class 3 (moderately toxic), and micro-copper is class 5 (practically non-toxic) of Hodge and Sterner Scale. Kidney, liver and spleen are found to be target organs of nano-copper particles. Nanoparticles induce gravely toxicological effects and heavy injuries on kidney, liver and spleen of experimental mice, but micro-copper particles do not, on mass basis. Results indicate a gender dependent feature of nanotoxicity. Several factors such as huge specific surface area, ultrahigh reactivity, exceeding consumption of H(+), etc. that likely cause the grave nanotoxicity observed in vivo are discussed.
Effect of Titanium Dioxide Nanoparticles on Copper Toxicity to Daphnia Magna in Water: Role of Organic Matter.

Abstract:Inevitably released into natural water, titanium dioxide nanoparticles (nano-TiO2) may affect the toxicity of other contaminants. Ubiquitous organic matter (OM) may influence their combined toxicity, which has been rarely reported. This study investigated the effect of nano-TiO2 on Cu toxicity to Daphnia magna and the role of OM (dissolved or particle surface bound) in inducing combined effects. The effect of nano-TiO2 on heavy metal accumulation depended on the adsorption capacity for heavy metals of nano TiO2 and the uptake of nano-TiO2 metal complexes by organisms. Nano-TiO2 significantly decreased Cu accumulation in D. magna, but the reducing effect of nano-TiO2 was eliminated in the presence of humic acid (HA, a model OM). In the Cu and HA solution, nano-TiO2 slightly affected the bioavailability of Cu2+ and Cu HA complexes and thus slightly influenced Cu toxicity. The nanoparticle surface-bound HA reduced the effect of nano TiO2 on the speciation of the accumulated Cu; therefore, the combined effects of nano-TiO2 and Cu on biomarkers similarly weakened. HA-altered Cu speciation may be the main factor responsible for the influence of HA on the combined effects of nano-TiO2 and Cu. Abstract:Nanomaterials released into the environment will interact with many materials including other contaminants. This may influence bioavailability and fate of both the nanoparticles and the other contaminants. The present study examined the effect of a combination of soluble copper and surface‐modified single‐walled carbon nanotubes (SWNTs) on Daphnia magna. Lysophosphatidylcholine (LPC) was used to modify the surface of SWNTs, reducing the surface hydrophobicity of the tubes and thereby producing a stable aqueous nanoparticle suspension. The toxicity of the nanoparticle–copper (Cu) mixture was determined to be additive. The addition of nontoxic concentration of LPC‐SWNTs enhanced the uptake and toxicity of copper. Abstract:The environmental behavior and the potential toxicity of copper nanoparticles (nano-Cu) in water are major concerns for assessing their environmental safety. The present study was undertaken to characterize the properties of nano-Cu in E3 medium, such as size changes, solubility, zeta-potential and pH, and to test the toxicity of nano-Cu suspension to zebrafish embryos. Dynamic light scattering and solubility experiments showed that three components coexisted in the nano-Cu exposure system, including small nano-Cu aggregates still suspended in E3 medium, large nano-Cu aggregates deposited on the container bottom and dissolved copper species (Cu(dis)). Both the zeta-potential of nano-Cu particles in E3 medium and the pH of the nano-Cu suspension showed no change during a 24 hour period. It is found that nano-Cu retarded the hatching of zebrafish embryos and caused morphological malformation of the larvae, and high concentrations (>0.1 mg/L) of nano-Cu even killed the gastrula-stage zebrafish embryos. Cu2+ ions were used to study the toxicity caused by nano-Cu dissolution. The embryo toxicity of nano-Cu at 0.01 and 0.05 mg/L showed no significant difference from Cu2+ at the corresponding concentrations (0.006 and 0.03 mg/L), but 0.1 mg/L nano-Cu had a greater toxicity than 0.06 mg/L Cu2+.

Abstract:This review summarizes the present knowledge on the toxicity of copper and copper nanoparticles (CuNPs) to various fish species. In previous decades, the excessive usage of metal and metallic nanoparticles has increased significantly, increasing the probability of the accumulation and discharge of metals in various trophic levels of the environment. Due to these concerns, it is important to understand the toxicity mechanisms of metals and metallic nanoparticles before they lead to unhealthy effects on human health. In this review paper, we specifically focus on the effect of metal copper and CuNPs on different fish organs under different physiochemical parameters of various water bodies. Nowadays, different forms of copper have distinctive and specific usages, e.g., copper sulfate is a well-established pesticide which is used to control the growth of algae in lakes and ponds. Deactivating the fungi enzymes prevents fungal spores from germinating. This process of deactivation is achieved via the free cupric ions, which are established as the most toxic forms of copper. Complexes of copper with other ligands may or may not be bioavailable for use in aquatic organisms. On the other hand, CuNPs have shown cost-effectiveness and numerous promising uses, but the toxicity and availability of copper in a nanoparticle form is largely unknown, Additionally, physiochemical factors such as the hardness of the water, alkalinity, presence of inorganic and organic ligands, levels of pH, and temperature in various different water bodies affect the toxicity caused by copper and CuNPs. However, comprehensive knowledge and data regarding the pattern of toxicity for copper metal ions and CuNPs in marine organisms is still limited. In this review, we carry out a critical analysis of the availability of the toxicological profiles of copper metal ions and CuNPs for different fishes in order to understand the toxicity mechanisms of copper and CuNPs. We believe that this review will provide valuable information on the toxicological profile of copper, which will further help in devising safe guidelines for the usage of copper and CuNPs in a sustainable manner.

Abstract:Metallic nanoparticles are among the most widely used types of engineered nanomaterials; however, little is known about their environmental fate and effects. To assess potential environmental effects of engineered nanometals, it is important to determine which species are sensitive to adverse effects of various nanomaterials. In the present study, zebrafish, daphnids, and an algal species were used as models of various trophic levels and feeding strategies. To understand whether observed effects are caused by dissolution, particles were characterized before testing, and particle concentration and dissolution were determined during exposures. Organisms were exposed to silver, copper, aluminum, nickel, and cobalt as both nanoparticles and soluble salts as well as to titanium dioxide nanoparticles. Our results indicate that nanosilver and nanocopper cause toxicity in all organisms tested, with 48-h median lethal concentrations as low as 40 and 60 microg/L, respectively, in Daphnia pulex adults, whereas titanium dioxide did not cause toxicity in any of the tests. Susceptibility to nanometal toxicity differed among species, with filter-feeding invertebrates being markedly more susceptible to nanometal exposure compared with larger organisms (i.e., zebrafish). The role of dissolution in observed toxicity also varied, being minor for silver and copper but, apparently, accounting for most of the toxicity with nickel. Nanoparticulate forms of metals were less toxic than soluble forms based on mass added, but other dose metrics should be developed to accurately assess concentration-response relationships for nanoparticle exposures.

Abstract:Titanium dioxide (TiO2) nanoparticles (NPs) encounter heavy metals in the environment under different scenarios. However, the mechanism of their joint toxicity effects on Daphnia magna remains vague. This study assessed the effects of hydrophobicity of TiO2 NPs (TDONPs) and exposure scenarios on copper uptake and toxicity in Daphnia magna. In the individual exposure scenario, hydrophilic and hydrophobic TDONPs both showed no acute toxicity to Daphnia magna, whereas individual Cu2+ exposure resulted in a 30% mortality rate. Co-exposure and sequential exposure to the two types of TDONP and Cu2+ resulted in mortality rates of 40%-50%. The mechanisms of the increased Cu2+ toxicity caused by hydrophilic and hydrophobic TDONP were different. In the presence of hydrophobic TDONPs, the Cu toxicity could be attributed to the increased bioaccumulation of Cu and Ti, leading to high oxidative stress injury. The Cu toxicity due to hydrophilic TDONPs could be induced by intensified intestinal membrane damage. The obtained data suggest that the hydrophobicity of the TDONPs plays a critical role in regulating the toxicity of Cu2+.

Abstract:The single-celled eukaryote Euplotes aediculatus was chosen to test and compare the toxic effects of Cu and CuO nanoparticles (NPs). The antioxidant enzymatic activity, morphological changes, and functional groups on the membrane were determined using spectrophotometry, microscopy, and Fourier transform infrared spectroscopy after NPs treatment. The toxicity of the NPs to cells was dose-dependent, and the 24 h-LC 50 values of the CuNPs and CuONPs were 0.46 µg/L and 1.24 × 10 3 µg/L, respectively. These NPs increased the activities of superoxide dismutase, glutathione peroxidase, and catalase and destroyed the cell structure; moreover, the CuNPs were more toxic than the CuONPs. In addition to the higher enzymatic activity, CuNPs also caused nucleoli disappearance, chromatin condensation, and mitochondrial and pellicle damage. The oxidization of the functional groups of the membrane (PO 2 − , C–O–C, and δ(COH) of carbohydrates) also confirmed the severe damage caused by CuNPs. Our study showed that oxidative stress and organelle destruction played important roles in the toxic effects of these NPs on this protozoan. Compared with other aquatic organisms, E. aediculatus can be considered a potential indicator at the preliminary stage of environmental pollution.

Abstract:This review summarizes the underlying toxicity mechanisms of CuO nanoparticles (NPs) on the basis of various environmentally relevant test species and cells. The toxicity effects related to CuO NPs have been analyzed in bacteria, algae, drosophila, plants, aquatic animals, mammals and other models in vitro and in vivo experiments. In addition, the combined toxicity evaluation of CuO NPs is also discussed. This review reveals the following major factors driving the toxicity of CuO NPs: (i) the dissolution of NP metal ions, (ii) the size, shape and coating of NPs, and (iii) oxidative stress and nonoxidative stress mechanisms.
