Abstract:A novel pH-sensitive chiral amphiphile, N-(4-decyloxy-2-hydroxybenzylidene)-L-leucine, was synthesized. Two kinds of vesicles were found at different pH ranges, 6.5-8.0 and 11.5-12.5, and a helical structure was observed at pH = 11.0. This unique behavior can be attributed to the formation of a dimeric structure. The interaction between this amphiphile and beta-cyclodextrin was investigated by fluorescence emission spectra, H-1 NMR, and molecular modeling.

Abstract:A novel Gemini amphiphile, N,N′-bis-(4-decyloxy-2-hydroxybenzylidene)-ethane-1,2-diamine, was synthesized and characterized. This amphiphile is capable of forming stable monolayer at air–water interface. The limiting area has a minimum when the pH value of subphase is 12. The unique behavior is attributed to the formation of intramolecular hydrogen bond between hydroxyl groups. The existence of this hydrogen bond was supported by the FTIR results. With the addition of copper ion into subphase, the limiting area showed a decreasing value, which is quite different from that of the conventional neutral amphiphiles. XPS and UV–vis were also carried out to investigate the effect of copper ion on molecular area.

Abstract:The aggregation behavior of amphiphile bearing azobenzene surfactant (C(12)AzoC(6)N(+)) was investigated using both in-situ and ex-situ AFM techniques. For the four substrates used (mica, HOPG and hydrophilic and hydrophobic silicon), the surfactant exhibits different structures in each case. In addition, the effect of adding electrolyte and ethanol on the aggregation behavior Of C(12)AzoC(6)N(+) was studied.

Abstract:We report a novel strategy to study the chain dynamics of poly(acrylic acid) (PAA) in a relative concentrated solution (1.0 g/L). The strategy is based on the fluorescent probe (DCTPE) with unique aggregation-induced emission (AIE) characteristics. Free DCTPE molecules are non-emissive in aqueous solution, but they become highly emissive when trapped in polymer coils. The fluorescence intensity is proportional to the efficiency of trapping DCTPE molecules in polymer coils. By correlation the change of fluorescence intensity with the variation of pH value (from 1.78 to 12.06), the PAA chain’s dynamics in the relatively concentrated solution have been elucidated into three processes. In the pH range from 12.06 to 6.0, PAA chains take an extended and non-folding conformation. Changing pH from 6.0 to 3.86, PAA chains are partially protonated and loosely packed polymer coils are formed. Further lowering the pH value of the solution (from 3.86 to 1.78), protonated segments dominate the PAA chains, and at the same time, the intermolecular hydrogen bonding takes effect, thus the polymer chains posses in the conformation of more compact coils.

Abstract:A pair of optical isomerism amphiphlic molecules L-C10HL(N-(4-decaoxygen-2-hydroxybenzene methylene) -L-leucine) and D-C10HL(N-(4-decaoxygen-2-hydroxybenzene methylene) -D-leucine) were synthesized. It was found that these two compounds showed superior surface activity and aggregation capability at higher pH value, which is so different from the traditional carboxylate surfactants. Furthermore, the interaction of L-C10HL and D-C10HL with cyclodextrin was also investigated by fluorescence excitation spectrum and circular dichroism spectrum.
Tetraphenylethene Cross-Linked Thermosensitive Microgels Via Acylhydrazone Bonds: Aggregation-Induced Emission in Nanoconfined Environments and the Cononsolvency Effect

Abstract:We studied the aggregation-induced emission (AIE) phenomenon in a nanoconfined environment, where the AIE-active molecule, namely, 1,1,2,2-tetrakis(4-methanoylphenyl)ethane (TPE-4ALD), was held in space via four acylhydrazone bonds within the thermosensitive microgel networks. The thermosensitive microgels, namely N-AH-TPE, were synthesized via the copolymerization of N-isopropylacrylamide (NIPAM) and 4-acylhydrazine-(2-hydroxy-3-(methacryloxypropyl)pyridine hydrochloride (AH monomer) with TPE-4ALD as cross-linker via surfactant free emulsion polymerization (SFEP) in aqueous solution at 70 degrees C. Acylhydrazone-bonded tetraphenylethene (TPE-4AH) moieties were thus constructed and worked as the fluorophore in N-AH-TPE microgels. The aqueous suspensions of N-AH-TPE microgels exhibit strongly bluish-green fluorescence under ultraviolet excitation because the four arms of TPE-4AH moieties were held and their intramolecular motions are strongly restricted. It is estimated that there is one TPE-4AH moiety per about 394 nm3 for the swollen N-AH-TPE microgels. The fluorescent properties of N-AH-TPE microgels can be modulated via the change of hydrophilic and hydrophobic environments of TPE-4AH moieties exerted by external stimuli, like addition of various good solvents for TPE-based structures, i.e., N,N-dimethylformamide (DMF), methanol, ethanol, tetrahydrofuran (THF), and N,N-dimethyl sulfoxide (DMSO), varying the solution temperature as well as the counteranions of the microgels. An unusual enhancement in the fluorescent intensity is observed when specific amounts of organic solvent are added into the aqueous suspensions of N-AH-TPE microgels, which can be attributed to the cononsolvency of the polyNIPAM network chains. The shrinkage of N-AH-TPE microgels caused by the cononsolvency effect further strengthens the confinement of TPE-4AH moieties and hence enhances the fluorescent emission of the microgels even though the organic solvents added are good solvents for TPE-4AH. Increasing the solution temperature of N-AH-TPE microgels or introducing hydrophobic counteranions into the microgels also significantly enhances the fluorescent emission of the microgels.
Cationic Side Chain Identity Directs the Hydrophobically Driven Self-Assembly of Amphiphilic β-Peptides in Aqueous Solution

Abstract:Hydrophobic interactions mediated by nonpolar molecular fragments in water are influenced by local chemical and physical contexts in ways that are not yet fully understood. Here, we use globally amphiphilic (GA) β-peptides (GA-Lys and GA-Arg) with stable conformations to explore if replacement of β3-homolysine (βLys) with β3-homoarginine (βArg) influences the hydrophobically driven assembly of these peptides in bulk aqueous solution. The studies were conducted in 10 mM triethanolamine buffer at pH 7, where both βLys (ammonium) and βArg (guanidinium) side chains are substantially protonated. Comparisons of light scattering measurements and cryo-electron micrographs before and after the addition of 60 vol% MeOH indicate very different outcomes of the hydrophobically driven assembly of AcY-GA-Lys versus AcY-GA-Arg (AcY denotes an N-acetylated-β3-homotyrosine (βTyr) at each N-terminus). Nuclear magnetic resonance and analytical ultracentrifugation confirm that AcY-GA-Lys assembles into large aggregates in aqueous buffer, whereas AcY-GA-Arg at comparable concentrations forms only small oligomers. Titration of AcY-GA-Arg into aqueous solutions of AcY-GA-Lys reveals that the driving force for AcY-GA-Lys association is far stronger than that for AcY-GA-Arg association. We discuss these results in the light of past experimental observations involving single molecule force measurements with GA β-peptides and hydrophobically driven dimerization of conventional peptides that form a GA α-helix upon dimerization (but do not display the Lys versus Arg trend predicted by extrapolating from the earlier AFM studies with β-peptides). Abstract:The membranes of a double-chain amphiphile dioctadecyl-N-[4-[[p-(N,N-dimethylamino)phenyl]azo]- pyridiniumbutyryl]-L-aspartate bromide (DDPA) were investigated by atomic force microscopy (AFM) and UV-vis absorption spectroscopy. The line-shaped aggregates of the molecules were found in the Langmuir-Blodgett (LB) monolayers of DDPA. The widths of these lines were about twice the length of the molecule. A semicylindrical structure was proposed to account for the observation. It was found that the DDPA molecules self-organized into bilayers and vesicles when the LB film was immersed in aqueous solutions. The bilayer showed good compressibility. The mixed DDPA and alpha-L-dipalmitoylphosphatidylcholine LB films were also examined, and the phase separation was observed. From the in situ AFM observation, the self-organization process was found to occur in the DDPA region. The mechanism of self-organization in aqueous solutions was also discussed.
Energy Landscapes Associated with the Self-Aggregation of an Alanine-Based Oligopeptide (Aaka)(4)

Abstract:The alanine-based 16-mer peptide (AAKA)(4) has recently been shown to aggregate into a hydrogel at centimolar concentrations and high ionic strength (Measey, T. J.; Schweitzer-Stenner, R. J. Ani. Chem. Soc. 2006, 128, 13324.). This is a surprising result since the current understanding of peptide self-aggregation would lead one to expect that the positively charged lysine residues inhibit the formation of beta-sheet structures. The present study was aimed at shedding some light on the mechanism governing the initial phase of the self-aggregation process. To this end we measured CD and FTIR spectra of the (AAKA)(n) at millimolar concentration and low ionic strength and found that the peptide forms soluble aggregates rather than a hydrogel under these conditions. To analyze the initial phase of this aggregation process, we carried out simulations on the dimerization and trimerization of (AAKA)(4) Using replica exchange molecular dynamics (MD) methods based on an implicit solvent model. The results indicate that the peptide aggregates into stable antiparallel beta-sheet structures under conditions comparable to experiments. Furthermore, the existence of trimers is a very sensitive function of the peptide concentration and temperature; no significant amount of trimers is formed at low concentration and the trimer population drops sharply as temperature increases above about 320 K. These results on oligomers are consistent with the trend observed in our experiments (reported elsewhere). We have also obtained the free energy landscapes of the trimers as a function of reaction coordinates, which reveal a few basins of minimal free energy. Associated with them, six major types of stable conformers were identified and the structures of transition states between them were also determined. We suggest a possible transition mechanism between these conformers based on the landscape and structural analysis of the aggregates.
Synthesis and aggregation behaviors of tail-branched surfactant Guerbet-cetyl trimethyl ammonium chloride

Abstract:A novel cationic surfactant Guerbet-cetyl trimethyl ammonium chloride (G-CTAC) was synthesized by combining several traditional synthetic processes in laboratory. The molecular structure of G-CTAC was characterized by Fourier transform infrared (FT-IR) spectra and proton nuclear magnetic resonance (1HNMR). This new amphiphile exhibited excellent performances in aqueous solution, which was investigated by surface tension measurement, contact angle on parafilm, particle size of aggregates, transmission electron microscopy (TEM), emulsifying experiments, and antistatic tests. It was found that the new quaternary ammonium compound could effectively decrease the surface tension of aqueous solution to 26.2 mN m−1 with the critical micelle concentration (CMC) being 5.9 mmol L−1. In addition, the cationic surfactant synthesized could efficiently decrease the contact angle of aqueous solution on paraffin surface. We suppose that these unique properties are attributed to the presence of the branching Guerbet-cetyl being hydrophobic group in the molecule. Moreover, according to TEM observations, this quaternary ammonium compound was inclined to aggregate into vesicles spontaneously without induction of any other additives in aqueous solution. Antistatic tests and emulsifying experiments were also conducted to explore the potential applications of this new material.

Abstract:Through continuous efforts in developing a new class of foldamers, homogeneous sulfono‐γ‐AApeptides have been designed and synthesized using tetraphenylethylene (TPE) moieties attached to the backbone as luminogenic sidechains. Based on previous crystal structures, it is found that these foldamers adopted a left‐handed 414‐helix. Due to the constraint of the helical scaffold, the rotation of the TPE moieties is restricted, leading to fluorescent emissive properties with high quantum yields not only at the aggregate state but also in solution. Investigation of the relationship between the structure and fluorescence behavior reveals that emission is induced by the combined effect of the aggregation‐induced emission and the rotated restriction from the backbone. Furthermore, as the packing mode of the luminogens can be precisely adjusted by the helical backbone, these foldamers are found to be circularly polarizable with relatively large luminescence dissymmetry factor. Interestingly, possessing cationic amphipathic structures similar to that of host‐defense peptides, these sulfono‐γ‐AApeptides are able to inhibit the growth of Gram‐positive bacteria, methicillin‐resistant Staphylococcus aureus through membrane interactions.

Abstract:A bolaform amphiphile(BP-15) bearing biphenyl group and long spacers of alkyl chains was synthesized. In-situ AFM studies indicate that the amphiphile can form two kinds of surface aggregates with different morphologies depending on solution concentration: short-range ordered cylindrical surface micelles at the lower concentration and flat domains at the higher concentration. These results agree well with the two critical micelle concentrations in bulk solution as determined by the concentration-dependent conductivity.

Abstract:The self-assembly characteristics in aqueous solutions of cationic bolaamphiphiles with systematic changes in their chemical structure is described with respect to their interfacial properties within water and at the air/water interface. Six cationic bolaamphiphiles were synthesized from multifunctional vernonia oil with the following variations: (a) two different alkyl chain lengths connecting the head groups, (b) polar ester or hydrogen bonding amide groups within the hydrophobic domain, and (c) an acetylcholine cationic head group with different conjugation sites to the alkyl chain. Surface tension measurements were used for determining critical aggregation concentration (CAC) values and air/water interfacial parameters such as 'effectiveness', surface excess concentration and area occupied by one molecule in the air/water interface. Fluorescent studies with pyrene were used to characterize CAC properties within the aqueous volume and transmission electron microscopy (TEM) for determining the aggregate structure's size, homogeneity and morphology. A bolaamphiphile molecular structure vs. interfacial property relationship was derived from this data which could be used to determine the molecular structure properties needed to generate interfacial forces to form either spherical vesicles or fibrous networks. The effects of the aliphatic chain length, head group orientation and functional groups within the hydrophobic domain on CAC, surface tension properties and self-aggregate morphology are described. Most bolaamphiphiles studied had CAC values in the 10-190 μM range, while two out of the six were found to assemble into MLM spherical vesicles with diameters ranging up to 120 nm suitable for drug delivery applications. Others formed a gelatinous network of fibers or multi-lamellar vesicles.

Abstract:A novel jellyfish-shaped triazine hexamer quaternary ammonium chloride surfactant (TH12QC) was synthesized, which consisted of one triazine spacer group and six long flexible hydrophobic chains. The molecular structure and aggregation behavior of TH12QC was investigated by nuclear magnetic resonance (NMR), surface tension, electrical conductivity, dynamic light scattering (DLS), transmission electron microscope (TEM), etc. The results show that the jellyfish-shaped TH12QC has better surface activity and lower surface tension than traditional ionic and Gemini surfactants in aqueous solution. There are two inflection points in the curve of conductivity versus concentration of the TH12QC aqueous solution, which correspond to the critical aggregation concentration (CAC) and the critical micelle concentration (CMC) respectively. The existence of CAC indicates that there is a pre-aggregation process before TH12QC forms micelles. The results of DLS and TEM show that network pre-aggregation, spherical aggregation and dense spherical aggregation were observed in different concentration of TH12QC aqueous solution, and the electrostatic equilibrium of the system subtly depends on the concentration of the solution. In addition, intramolecular and intermolecular hydrogen bonding is also an important factor. This study provides a method for studying the aggregation behavior and morphology of oligomeric surfactants with rigid spacer groups.
Helical Sulfono-γ-AApeptides with Aggregation-Induced Emission and Circularly Polarized Luminescence

Abstract:Aggregation-induced emission (AIE) was intensively studied because of packing of small molecules and polymers; however, mid-molecular-weight (1000-3000) molecular scaffold containing a precise number of AIE luminogens is rare. Herein, we report the investigation of three tetraphenylethylene (TPE)-modified sulfono-gamma-AApeptides in which multiple TPE moieties are conjugated to the chiral right-handed helical peptidomimetic backbone as functional side chains. The crystal structure of the TPE-alpha/sulfono-gamma-AA peptide 1 demonstrates that because of the rigid helical scaffold of the TPE-alpha/sulfono-gamma-AA peptides, the intramolecular rotations of the TPE with short linker are restricted, therefore leading to the boosted fluorescent emission in solution. Peptides 2 and 3 exhibit aggregation-induced emission enhancement (AIEE), possibly because of the combination of both AIE and rotation restriction. Moreover, because of their preoriented assembly induced by the right-handed helical scaffold, these emissive chiral luminogens show effective circularly polarized luminescence signals with high dissymmetry factor g(lum). Finally, the amphiphilic nature of TPE-alpha/sulfono-gamma-AA peptides could enable them to penetrate the bacterial membranes and exhibit strong fluorescence. Their antimicrobial activity and labeling-free character could further augment their potential applications in both materials and biomedical sciences.

Abstract:A series of pyridazine coupled tetrathiafulvalene T-shaped derivatives with varying amino acid moieties have been synthesized and their gelation properties were studied in various organic solvents. Among these derivatives, two gelators bearing glycine or phenylalanine units display efficient gelation in aromatic and polar solvents. Interestingly, these gelators, except for the gelator containing two tryptophan units, are able to gel DMF via a solution-to-gel transformation when triggered with sonication for less than 20 s or cooled below zero. A number of experiments revealed that these gelator molecules self-assembled into elastically interpenetrating three-dimensional chiral fibrillar aggregates. Importantly, all of the resulting gels result in a dramatic enhancement of the fluorescence intensity compared with their hot solution in spite of the absence of a conventional fluorophore unit and the fluorescence was effectively quenched by the introduction of C60. Moreover, the gelators can be utilized for the removal of different types of toxic molecules, such as aromatic solvents and cationic dyes, from wastewater.

Abstract:A series of amphiphilic poly(amidoamine) dendrimers (PAMAM, G2-G5) composed of a hydrophilic core and a hydrophobic shell of aniline pentamer (AP) were synthesized and characterized. The modified dendrimers self-assembled to vesicular aggregates in water with the critical aggregation concentration (CAC) decreased in the order of G2 > G3 > G4 > G5. It was found that the modified dendrimers self-organized into spherical aggregates with a bilayer vesicular structures and that the dendrimers in higher generation have more order structure, which may be attributed to the crystallization induced by the compacted effect of AP segments. In addition, larger spherical vesicles were observed under acidic and alkaline conditions, as compared with sizes of aggregates in neutral medium. At low pH, the tertiary amine groups of PAMAM-AP were transformed to ammonium salts; the polarons were formed from AP units by doping with strong acids, thereby leading to the stability of vesicular aggregates being better than that in double distilled water. Interfacial Films Formed by A Biosurfactant Modularized with A Silken Tail

Abstract:This paper reports the dynamic interfacial behavior of a new interfacially active peptide AM-S, which was designed based on a peptide surfactant AM1 modularized with an additional silk-derived hydrophobic tail to enhance anchoring to air water interfaces. AM-S peptide shows a random coil conformation in bulk solution similar to AM1 as determined by circular dichroism spectroscopy, which facilitates rapid adsorption at the air water interface, reducing interfacial tension from 72 to 52 mN/m within 300 s at a low concentration of 10 mu M. Although the interfacial films formed by AM-S demonstrated low tensile stress as compared to AM1, the AM-S films in the presence of Zn(II), but not in its absence, show significant resistance against compression, as peptides were unable to desorb quickly under the compression conditions imposed by the Cambridge interfacial tensiometer (CIT). These results indicate that AM-S peptides tend to undergo a multilayer adsorption at the interfaces, in contrast to AM1 peptide that only forms an interfacial monolayer, demonstrating a distinct physical effect of the silk tail. The multilayer structure of AM-S in the presence of Zn(II) was also apparent on a thin-film pressure balance experiment. The thin films formed by AM-S peptide were thicker than the films formed by AM1 peptide, thus enabling stabilization of the films against increased critical air pressure as well as the self-assembly of the AM-S peptides. Also, AM-S peptide was shown to be capable of forming dense foams with small bubble size and maintained foam stability comparable to AM1 peptide. This study demonstrated that addition of a silk tail peptide to a biosurfactant can significantly modify interfacial adsorption behavior at a fluid-fluid interface, which may guide further molecular design strategies.
Self-assembling Amphipathic Alpha-Helical Peptides Induce the Formation of Active Protein Aggregates in Vivo

Abstract:We recently found that several self-assembling alpha, beta, or surfactant-like peptides, when terminally attached to proteins, can promote the in vivo assembly of active protein aggregates (or active inclusion bodies, AIBs) in Escherichia coil. In this work, we systematically examined the AIBs induced by an amphipathic alpha-helical peptide 18Awt (EWLKAFYEKVLEKLKELF) and its variants with altered ion pairs. Transmission electron microscopic and Fourier transform infrared spectroscopic analyses suggested that the AIBs appeared to adopt an amorphous mesh-like structure, and were likely induced by helical structures formed by the assembly of the 18A peptides. Confocal fluorescent micrographic analysis revealed that the AIBs resided around the periphery of the cell membrane or in the cytoplasm, depending on the distribution of ion pairs on the 18A peptides, which suggested that the association between the aggregates and the cell membrane was mediated by the lipid-18A interaction. Two of these 18A peptide variants were further used in constructing cleavable self-aggregating tags (cSAT) in conjunction with an intein molecule for protein purification, and verified using two model proteins. This extends the cSAT approach for laboratory and potentially industrial uses. Our study might also provide new insights into aggregation-related diseases.

Abstract:Abstract Novel sugar-based gemini surfactants with a 1,3-propan-2-ol spacer (1, 3-(N- alkyl-2-D-glucosaminyl acetyl) propan-2-ol, Glu(n)-3(OH)-Glu(n), n = 12, 14) were synthesized with D-(+)-glucono-1,5-lactone as starting material in two steps, whose structures were confirmed using proton nuclear magnetic resonance spectroscopy ( 1 H NMR) and carbon nuclear magnetic resonance carbon spectroscopy ( 13 C NMR). The micellization of Glu(n)-3(OH)-Glu(n) (n = 12, 14) in aqueous solution at 25.0°C was investigated by using surface tension measurement. The results show that the critical micelle concentration (CMC) of Glu(12)-3(OH)-Glu(12) is around 10 –5 mol × L –1 , and is one order of magnitude smaller than that of Glu(14)-3(OH)-Glu(14), indicating that the surface activity of Glu(12)-3(OH)-Glu(12) is superior to that of Glu(14)-3(OH)-Glu(14). Moreover, the aggregation behavior of Glu(12)-3(OH)-Glu(12) in aqueous solution at different pH values was investigated by surface tension, dynamic light scattering (DLS), and cryogenic transmission electron microscopic (Cryo-TEM) measurements. The results indicate that the CMC slightly increases with the decrease of the solution pH. The microstructure of Glu(12)-3(OH)-Glu(12) aggregates transitions from micelle to vesicle with the solution pH from acidic to neutral and alkaline. The microstructural transformation of Glu(12)-3(OH)-Glu(12) with the solution pH is mainly determined by the protonation of the two tertiary amine nitrogen atoms in its hydrophilic headgroups.
