Abstract:The purpose of this study is to identify candidate genes that could predict prognosis of early-stage tongue squamous cell carcinoma (TSCC) and its occult cervical lymphatic metastasis by large-scale gene expression profiling. Tumor tissue and matched normal mucosa samples were collected from patients with TSCC and analyzed with Affymetrix HTA2.0 high-density oligonucleotide array. Differentially expressed genes in TSCC with cervical lymph node metastasis (CLNM) were further analyzed with Gene Ontology and Kyoto Encyclopedia of Genes and Genomes for their functions and related pathways. A total of 107 differentially expressed genes (p < 0.05) were identified by microarray in TSCC samples with CLNM (n = 6) compared to those without CLNM (n = 6). Genes involved in the cell-matrix adherens junction and migration function including MFAP5, TNNC1, MGP, FBFBP1 and FBXO32 were selected and validated by RT-PCR in TSCC samples (n = 32). Of the five genes, MFAP5 and TNCC1 expressions were further validated by immohistochemistry (n = 61). The significant positive correlation between MFAP5 and TNNC1 expression (p<0.001) was observed. Notably, over-expression of MFAP5 and TNNC1 were correlated with CLNM, metastasis relapse-free survival and overall survival. Our findings indicated that MFAP5 and TNNC1 may be potential markers for predicting occult cervical lymphatic metastasis and prognosis of oral tongue carcinoma.

Abstract:Tongue squamous cell carcinoma (TSCC) is a rare and aggressive type of cancer, which is associated with a poor prognosis. Identification of patients at high risk of TSCC tumorigenesis may provide information for the early detection of metastases, and for potential treatment strategies. MicroRNA (miRNA; miR) and mRNA expression profiling of TSCC tissue samples and normal control tissue samples were obtained from three Gene Expression Omnibus (GEO) data series. Bioinformatics analyses, including the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes were used to identify genes and pathways specifically associated with miRNA-associated TSCC oncology. A total of 25 miRNAs and 769 mRNAs were differentially expressed in the two groups assessed, and all the differentially expressed miRNA and mRNA target interactions were analyzed. The miRNA target genes were predominantly associated with 38 GO terms and 13 pathways. Of the genes differentially expressed between the two groups, and confirmed in another GEO series, miRNA-494, miRNA-96, miRNA-183, runt-related transcription factor 1, programmed cell death protein 4 and membrane-associated guanylate kinase were the most significantly altered, and may be central in the regulation of TSCC. Bioinformatics may be used to analyze large quantities of data in microarrays through rigorous experimental planning, statistical analysis and the collection of complete data on TSCC. In the present study, a novel differential miRNA-mRNA expression network was constructed, and further investigation may provide novel targets for the diagnosis of TSCC.

Abstract:Objective This study was designed to identify the role of circRNAs in tongue squamous cell carcinoma (TSCC) and discover novel circRNAs as potential diagnostic or therapeutic targets for TSCC. Results The circRNA expression profiles in TSCC were evaluated by high-throughput sequencing in 6 TSCC patients. Our data showed that 69 circRNAs were downregulated and 208 were upregulated significantly (fold change >= 2.0; p < 0.05). GO and KEGG analysis demonstrated that the parental genes of differentially expressed circRNAs were potentially implicated in TSCC pathogenesis. After bioinformatics analysis, 4 circRNAs (hsa_circ_0005035, hsa_circ_0002360, hsa_circ_0066251, and hsa_circ_0003161) were selected and successfully validated by quantitative reverse-transcription polymerase chain reaction (qRT-PCR) in 40 patients and 3 TSCC cell lines. The establishment and analysis of the circRNA-miRNA-mRNA network revealed the potential function and mechanism of these candidate circRNAs. Conclusion Our study provided a comprehensive circRNAs expression profile of TSCC by RNA-seq and discovered 4 novel circRNAs with potential great diagnostic and therapeutic value. These findings provide new insights into the development of potential biomarkers and targets for TSCC treatment.
