Abstract:The regulation of DNA methylation plays crucial roles in response to various abiotic stress in plants. However, the underlying regulatory mechanisms remain unclear. Here, we reported that OsDML4, a DNA demethylase gene, is required for regulating rice salt tolerance. The osdml4 mutants were sensitive to salt stress, with abnormal accumulated levels of reactive oxygen species (ROS) and increased Na+/K+ ratio. By using RNA-Seq for the identification of candidate targets of OsDML4, we found that the expression of the ROS-scavenging-related gene OsCATA (fold change > 1.7, p-value < 0.05) and JA-related gene OsJAZ13 (fold change > 2.8, p-value < 0.05) was significantly reduced in the osdml4 mutant under salt stress. Moreover, the CHH methylation in the promoter region of OsCATA was increased under salt stress, and it was much more significant in the osdml4 mutant (from 3.24% to 6.25%) than in the Nip (from 3.12% to 4.16%). Together, OsDML4 may directly or indirectly regulate ROS homeostasis and the JA signaling to improve salt tolerance in rice, which would provide new insight for further research on the role of the demethylase gene in response to abiotic stress.

Abstract:Plant productivity is greatly affected by environmental stresses such as drought, salt, and freezing. We previously described a C-repeat binding transcription factor from maize (ZmCBF3) that was upregulated by both abscisic acid and low-temperature and actively expressed during embryogenesis. To understand the stress response in rice, transgenic ZmCBF3 rice with ubiquitin promoter was developed. T3 generation was planted and analyzed. The results showed that overexpression of ZmCBF3 in rice did not cause growth retardation under normal growth conditions with improved tolerance to drought, high-salt, and low-temperature stresses. Moreover, the transgenic rice grain yield was similar to wild type plants under normal conditions. The transgenic plants showed enhanced survival rate and reduced malondialdehyde content and relative conductivity under drought, salt, and low-temperature stresses. ZmCBF3 overexpression in transgenic rice increased the transcript levels of stress-induced genes and enhanced the tolerance to drought, salt, and low-temperature stresses.

Abstract:Calmodulin-like (CML) genes function in regulating plant responses to different abiotic stresses, such as high salt and drought. Using japonica type rice seedlings (‘Nipponbare’), we induced the expression of OsCML4 (Accession No. NM_001057768), which encodes a rice CML protein. Under drought conditions, transgenic plants over-expressing that gene exhibited obviously improved growth performance and higher survival rates than the wild type (WT). Activity by reactive oxygen species (ROS)- scavenging enzymes superoxide dismutase (SOD) and catalase (CAT), as well as proline concentrations, were significantly increased in transgenic plants. Expression by ROS scavenging-related genes APXI and Cat-B and the stress-related gene OsP5CS1 was also enhanced in the transgenics under drought conditions. However, the sensitivity to abscisic acid (ABA) was similar between over-expressing and WT plants. These results suggest that OsCML4 confers drought tolerance partially through ROS-scavenging and also by inducing other stress-related genes in an ABAindependent manner.

Abstract:Calmodulin-like (CML) genes regulate plant growth, development, and responses to abiotic stresses such as salinity and drought. Many genes encoding CML proteins have been identified from rice (Oryza sativa), but their functions remain largely unknown. Our characterization of one putative CML gene, OsDSR-1 (O. sativa Drought Stress Response-1), showed that its protein binds Ca2+ and displays Ca2+-dependent conformational changes. In contrast to wild-type (WT) and OsDSR-1-RNA interference (OsDSR-1-Ri) plants, transgenic rice plants that overexpress OsDSR-1 were significantly more drought tolerant and had increased sensitivity to abscisic acid. Furthermore, their concentrations of free proline and soluble sugars and the activities of reactive oxygen species-scavenging enzymes as well as the transcript levels of many ROS-scavenging and stress-related genes were significantly enhanced under drought stress. Much less hydrogen peroxide and malondialdehyde accumulated in OsDSR-1-overexpressing (OsDSR-1-OE) plants than in either the Ri or WT plants. All of these results suggest that OsDSR-1 plays important roles in conferring tolerance to drought in rice by decreasing the occurrence of oxidative damage.

Abstract:Drought is a major adverse environmental factor that plants face in nature but the molecular mechanism by which plants transduce stress signals and further endow themselves with tolerance remains unclear. Malectin/malectin-like domains containing receptor-like kinases (MRLKs) have been proposed to act as receptors in multiple biological signaling pathways, but limited studies show their roles in drought-stress signaling and tolerance. In this study, we demonstrate OsMRLK63 in rice (Oryza sativa L.) functions in drought tolerance by acting as the receptor of 2 rapid alkalization factors, OsRALF45 and OsRALF46. We show OsMRLK63 is a typical receptor-like kinase that positively regulates drought tolerance and reactive oxygen species (ROS) production. OsMRLK63 interacts with and phosphorylates several nicotinamide adenine dinucleotide phosphate (NADPH) oxidases with the primarily phosphorylated site at Ser26 in the N-terminal of RESPIRATORY BURST OXIDASE HOMOLOGUE A (OsRbohA). The application of the 2 small signal peptides (OsRALF45/46) on rice can greatly alleviate the dehydration of plants induced by mimic drought. This function depends on the existence of OsMRLK63 and the NADPH oxidase-dependent ROS production. The 2 RALFs interact with OsMRLK63 by binding to its extracellular domain, suggesting they may act as drought/dehydration signal sensors for the OsMRLK63-mediated process. Our study reveals a OsRALF45/46-OsMRLK63-OsRbohs module which contributes to drought-stress signaling and tolerance in rice.

Abstract:Abstract Background The NAC (NAM, AFAT, and CUC) transcription factors play critical roles in rice ( Oryza sativa ) development and stress regulation. Overexpressing a microRNA (miR164b)-resistant OsNAC2 mutant gene, which generates transcripts that cannot be targeted by miR164b, improves rice plant architecture and yield; however, the performance of these mOsNAC2 -overexpressing lines, named ZUOErN3 and ZUOErN4, under abiotic stress conditions such as drought have not yet been fully characterized. Results In this study, we showed that the germination of ZUOErN3 and ZUOErN4 seeds was delayed in comparison with the wild-type (WT) seeds, although the final germination rates of all lines were over 95%. The quantification of the endogenous ABA levels revealed that the germinating mOsNAC2 -overexpressing seeds had elevated ABA levels, which resulted in their slower germination. The mOsNAC2 -overexpressing plants were significantly more drought tolerance than the WT plants, with the survival rate increasing from 11.2% in the WT to nearly 70% in ZUOErN3 and ZUOErN4 plants after a drought treatment. Salt (NaCl) tolerance was also increased in the ZUOErN3 and ZUOErN4 plants due to significantly increased ABA levels. A reverse transcription quantitative PCR (RT-qPCR) analysis showed a significant increase in the expression of the ABA biosynthesis genes OsNCED1 and OsNCED3 in the mOsNAC2 -overexpressing lines, and the expression levels of the stress-responsive genes OsP5CS1 , OsLEA3 , and OsRab16 were significantly increased in these plants. Moreover, OsNAC2 directly interacted with the promoters of OsLEA3 and OsNCED3 in yeast one-hybrid assays. Conclusions Taken together, our results show that OsNAC2 plays a positive regulatory role in drought and salt tolerance in rice through ABA-mediated pathways.

Abstract:Myo-inositol oxygenase (MIOX), a unique monooxygenase, catalyzes the oxidation of myo-inositol to d-glucuronic acid. However, the protective role of MIOX in plants against oxidative stress or drought stress remains unknown. In this study, the functional characterization of MIOX obtained from the cDNA library of upland rice (Oryza sativa L. cv. IRAT109), was performed. OsMIOX was expressed predominantly in the roots and induced by drought, H2O2, salt, cold and abscisic acid. The transgenic rice lines overexpressing OsMIOX showed obviously improved growth performance in the medium containing 200mM mannitol. Further, the survival rate of leaves from the transgenic rice lines was significantly higher than that of the wild type plants under polyethylene glycol treatment. It was discovered that the activity of ROS-scavenging enzymes and proline content, as well as the transcript levels of many ROS scavenging genes were significantly increased in transgenic plants compared to the wild type plants under drought stress conditions. Together, these data suggest that OsMIOX has a specific function in drought stress tolerance by decreasing oxidative damage.
Involvement of the Metallothionein gene OsMT2b in Drought and Cadmium Ions Stress in Rice

Abstract:Abiotic stress is one of the major factors restricting the production of rice (Oryza sativa L.). Developing rice varieties with dual abiotic stress tolerance is essential to ensure sustained rice production, which is necessary to illustrate the regulation mechanisms underlying dual stress tolerance. At present, only a few genes that regulate dual abiotic stress tolerance have been reported. In this study, we determined that the expression of OsMT2b was induced by both drought and Cd2+ stress. After stress treatment, OsMT2b-overexpression lines exhibited enhanced drought tolerance and better physiological performance in terms of relative water content and electrolyte leakage compared with wild-type (WT). Further analysis indicated that ROS levels were lower in OsMT2b-overexpression lines than in WT following stress treatment, suggesting that OsMT2b-overexpression lines had a stronger ability to scavenge ROS under stress. Reverse transcription-quantitative PCR (RT-qPCR) results demonstrated that under drought stress, OsMT2b influenced the expression of genes involved in ROS scavenging to enhance drought tolerance in rice. In addition, OsMT2b-overexpression plants displayed increased tolerance to Cd2+ stress, and physiological assessment results were consistent with the observed phenotypic improvements. Thus, enhancing ROS scavenging ability through OsMT2b overexpression is a novel strategy to boost rice tolerance to both drought and Cd2+ stress, offering a promising approach for developing rice germplasm with enhanced resistance to the abiotic stressors.

Abstract:Rice (Oryza sativa) responds to various abiotic stresses during growth. Plant-specific NAM, ATAF1/2, and CUC2 (NAC) transcription factors (TFs) play an important role in controlling numerous vital growth and developmental processes. To date, 170 NAC TFs have been reported in rice, but their roles remain largely unknown. Herein, we discovered that the TF OsNAC006 is constitutively expressed in rice, and regulated by H2O2, cold, heat, abscisic acid (ABA), indole-3-acetic acid (IAA), gibberellin (GA), NaCl, and polyethylene glycol (PEG) 6000 treatments. Furthermore, knockout of OsNAC006 using the CRISPR-Cas9 system resulted in drought and heat sensitivity. RNA sequencing (RNA-seq) transcriptome analysis revealed that OsNAC006 regulates the expression of genes mainly involved in response to stimuli, oxidoreductase activity, cofactor binding, and membrane-related pathways. Our findings elucidate the important role of OsNAC006 in drought responses, and provide valuable information for genetic manipulation to enhance stress tolerance in future plant breeding programs.
