Abstract:This research is conducted to screen differentially expressed genes (DEGs) between the primary and secondary hair follicle (HF) of Cashmere goat's skin to find candidate genes related to development.To analyze the transcriptome data by bioinformatics methods,the RNA-Seq of six individuals from Inner Mongolia Arbas Cashmere goat was performed.On the basis of quality control of the sequencing result,DEGs were identified as well as functional annotation and enrichment analysis was carried out.In this study,617 DEGs has been selected from primary and secondary HF,and among them 297 are up-regulated while 320 are down-regulated.Additionally,the results of gene ontology (GO) expression elucidate that the main function of up-regulated genes are:electron transport chain,respiratory chain electron transport,mitochondrial electron transport,gene expression and other biological functions.However down-regulated expression of genes is mainly exercised as following:cell autophagy,protein complex,neutrophil aggregation,bacterial fungal defense reaction and other biological functions.Using KEGG database as a reference,these genes are mainly involved in the metabolism of cysteine and methionine,RNA aggregation,shear,Parkinson's disease,substrate phosphorylation,and Alzheimer's disease.It will provide a basis for further study on the related genes and functions of hair follicle development in the future.

Abstract:Background Cashmere growth is a seasonal and cyclic phenomenon under the control of photoperiod and multiple stimulatory and inhibitory signals. Beyond relevant coding genes, microRNA (miRNA) and long non coding RNA (lncRNA) play an indispensable role in hair follicle (HF) development and skin homeostasis. Furthermore, the influence of lncRNA upon miRNA function is also rapidly emerging. However, little is known about miRNAs, lncRNAs and their functions as well as their interactions on cashmere development and cycling. Result Here, based on lncRNA and miRNA high-throughput sequencing and bioinformatics analysis, we have identified 1108 lncRNAs and 541 miRNAs in cashmere goat skin during anagen and telogen. Compared with telogen, 1388 coding genes, 41 lncRNAs and 15 miRNAs were upregulated, while 1104 coding genes, 157 lncRNAs and 8 miRNAs were downregulated in anagen (adjusted P -value ≤0.05 and relative fold-change ≥2). Subsequently, we investigated the impact of lncRNAs on their target genes in cis and trans, indicating that these lncRNAs are functionally conserved during HF development and cycling. Furthermore, miRNA-mRNA and miRNA-lncRNA interaction were identified through the bioinformatics algorithm miRanda, then the ceRNA networks, miR-221-5p-lnc_000679- WNT3 , miR-34a-lnc_000181- GATA3 and miR-214-3p-lnc_000344- SMAD3 , were constructed under defined rules, to illustrate their roles in cashmere goat HF biology. Conclusion The present study provides a resource for lncRNA, miRNA and mRNA studies in cashmere cycling and development. We also demonstrate potential ceRNA regulatory networks in cashmere goat HF cycling for the first time. It expands our knowledge about lncRNA and miRNA biology as well as contributes to the annotation of the goat genome.

Abstract:Long noncoding RNAs (lncRNAs) are a novel class of eukaryotic transcripts. They are thought to act as a critical regulator of protein-coding gene expression. Herein, we identified and characterized 13 putative lncRNAs from the expressed sequence tags from secondary hair follicle of Cashmere goat. Furthermore, we investigated their transcriptional pattern in secondary hair follicle of Liaoning Cashmere goat during telogen and anagen phases. Also, we generated intracellular regulatory networks of upregulated lncRNAs at anagen in Wnt signaling pathway based on bioinformatics analysis. The relative expression of six putative lncRNAs (lncRNA-599618, -599556, -599554, -599547, -599531, and -599509) at the anagen phase is significantly higher than that at telogen. Compared with anagen, the relative expression of four putative lncRNAs (lncRNA-599528, -599518, -599511, and -599497) was found to be significantly upregulated at telogen phase. The network generated showed that a rich and complex regulatory relationship of the putative lncRNAs and related miRNAs with their target genes in Wnt signaling pathway. Our results from the present study provided a foundation for further elucidating the functional and regulatory mechanisms of these putative lncRNAs in the development of secondary hair follicle and cashmere fiber growth of Cashmere goat.
Exploring differentially expressed genes by RNA-Seq in cashmere goat (Capra hircus) skin during hair follicle development and cycling

Abstract:Cashmere goat (Capra hircus) hair follicle development and cycling can be divided into three stages: anagen, catagen and telogen. To elucidate the genes involved in hair follicle development and cycling in cashmere goats, transcriptome profiling of skin was carried out by analysing samples from three hair follicle developmental stages using RNA-Seq. The RNA-Seq analysis generated 8487344, 8142514 and 7345335 clean reads in anagen, catagen and telogen stages, respectively, which provided abundant data for further analysis. A total of 1332 differentially expressed genes (DEGs) were identified, providing evidence that the development of hair follicles among the three distinct stages changed considerably. A total of 683 genes with significant differential expression were detected between anagen and catagen, 530 DEGs were identified between anagen and telogen, and 119 DEGs were identified between catagen and telogen. A large number of DEGs were predominantly related to cellular process, cell & cell part, binding, biological regulation and metabolic process among the different stages of hair follicle development. In addition, the Wnt, Shh, TGF-β and Notch signaling pathways may be involved in hair follicle development and the identified DEGs may play important roles in these signaling pathways. These results will expand our understanding of the complex molecular mechanisms of hair follicle development and cycling in cashmere goats and provide a foundation for future studies.
Screening of microRNA and mRNA related to secondary hair follicle morphogenesis and development and functional analysis in cashmere goats

Abstract:Abstract microRNA (miRNA) is a type of endogenous short-chain non-coding RNA with regulatory function found in eukaryotes, which is involved in the regulation of a variety of cellular and biological processes. However, the research on the development of cashmere goat secondary hair follicles is still relatively scarce. In this study, small RNA libraries and mRNA libraries of 45 days, 55 days, 65 days, and 75 days of fetal skin of cashmere goats were constructed, and the constructed libraries were sequenced using Illumina Hiseq4000, and the expression profiles of miRNA and mRNA in cashmere goat fetal skin were obtained. The differentially expressed miRNAs and mRNAs in six control groups were identified and the qRT-PCR experiment shows that the sequencing results are accurate. Sixty-six miRNAs related to secondary hair follicle development were screened, and used TargetScan and miRanda to predict 33 highly expressed miRNA target genes. At the same time, 664 mRNAs related to the development of secondary hair follicles were screened, and GO enrichment and KEGG pathway analysis were performed. It was found that some miRNA target genes were consistent with the screening results of mRNAs related to secondary hair follicle development and were enriched in Notch signaling pathway, TGF-β signaling pathway. Therefore, miR-145-5p-DLL4, miR-27b-3p-DLL4, miR-30e-5p-DLL4, miR-193b-3p-TGF-β1, miR-181b-5p-NOTCH2, and miR-103-3p-NOTCH2 regulatory network related to the development of secondary hair follicles were constructed and the results of dual-luciferase reporter gene assay indicated that there is a targeted relationship between chi-miR-30e-5p and DLL4, which will provide a basis for molecular mechanism of miRNA-mRNA in the development of the hair follicles in cashmere goats.

Abstract:<h3 class="Heading">Background</h3><p class="Para">Dermal papilla cells (DPCs), the "signaling center" of hair follicle (HF), delicately master continual growth of hair in mammals including cashmere, the fine fiber annually produced by secondary HF embedded in cashmere goat skins. Such unparalleled capacity bases on their exquisite character in instructing the cellular activity of hair-forming keratinocytes via secreting numerous molecular signals. Past studies suggested microRNA (miRNAs) and long non-coding RNAs (lncRNAs) play essential roles in a wide variety of biological process, including HF cycling. However, their roles and related molecular mechanisms in modulating DPCs secretory activities are still poorly understood.</p><h3 class="Heading">Results</h3><p class="Para">Here, we separately cultivated DPCs and their functionally and morphologically distinct dermal fibroblasts (DFs) from cashmere goat skins at anagen. With the advantage of high throughput RNA-seq, we synchronously identified 2540 lncRNAs and 536 miRNAs from two types of cellular samples at 4th passages. Compared with DFs, 1286 mRNAs, 18 lncRNAs, and 42 miRNAs were upregulated, while 1254 mRNAs, 53 lncRNAs and 44 miRNAs were downregulated in DPCs. Through overlapping with mice data, we ultimately defined 25 core signatures of DPCs, including <em class="EmphasisTypeItalic">HOXC8</em> and <em class="EmphasisTypeItalic">RSPO1</em>, two crucial activators for hair follicle stem cells (HFSCs). Subsequently, we emphatically investigated the impacts of miRNAs and lncRNAs (<em class="EmphasisTypeItalic">cis</em>- and <em class="EmphasisTypeItalic">trans</em>- acting) on the genes, indicating that ncRNAs extensively exert negative and positive effects on their expressions. Furthermore, we screened lncRNAs acting as competing endogenous RNAs (ceRNAs) to sponge miRNAs and relief their repressive effects on targeted genes, and constructed related lncRNAs-miRNAs-<em class="EmphasisTypeItalic">HOXC8</em>/<em class="EmphasisTypeItalic">RSPO1</em> interactive lines using bioinformatic tools. As a result, XR_310320.3-chi-miR-144-5p-<em class="EmphasisTypeItalic">HOXC8</em>, XR_311077.2-novel_624-<em class="EmphasisTypeItalic">RSPO1</em> and others lines appeared, displaying that lncRNAs might serve as ceRNAs to indirectly adjust HFSCs status in hair growth.</p><h3 class="Heading">Conclusion</h3><p class="Para">The present study provides an unprecedented inventory of lncRNAs, miRNAs and mRNAs in goat DPCs and DFs. We also exhibit some miRNAs and lncRNAs potentially participate in the modulation of HFSCs activation via delicately adjusting core signatures of DPCs. Our report shines new light on the latent roles and underlying molecular mechanisms of ncRNAs on hair growth.</p>

Abstract:Background: Circular RNAs, a novel class in the eukaryotic transcriptome, are characterized by the 3' and 5' ends that are covalently joined in a covalently closed loop without free ends. Circular RNAs are considerably stable molecules and act as microRNA sponges with regulatory potential to the protein-coding genes. Results: Eight circular RNAs were found to be significantly upregulated at anagen skin tissue of cashmere goat compared with their counterparts at telogen. Rich and complex regulatory patterns were revealed among the eight upregulated circular RNAs at anagen and related miRNAs with their potential regulatory genes. The potential regulatory genes of eight upregulated circular RNAs at anagen were involved in several pathways related to the main physiological process of hair follicle, such as histone acetylation and axon. For chi_circ_1926, chi_circ_3541, chi_circ_0483, chi_circ_3196, and chi_circ_2092, overall, the relative expression in secondary hair follicle exhibited highly similar trends with their corresponding host genes during the different stages of the hair follicle cycle. However, the expression trends of chi_circ_0100, chi_circ_2829, and chi_circ_1967 were found to diverge from their corresponding host genes during the different stages of the hair follicle cycle. Conclusions: A total of eighteen circular RNAs were identified and characterized from skin tissue of cashmere goat. The eight upregulated circular RNAs at anagen might have significant roles in the secondary hair follicle of cashmere goat. Our results would provide a novel regulatory layer to elucidate the molecular mechanisms underlying the development of secondary hair follicle and the growth of cashmere fiber in cashmere goat. Abstract:The development of hair follicle in cashmere goats shows significant periodic change, as with mice and humans. However, for cashmere goat with double-coat, the periodic change may be due to other regulatory molecules and signal pathways. To understand the mechanism of periodic development of hair follicle, we performed a weighted gene coexpression network analysis (WGCNA) to mine key genes and establish an interaction network by utilizing the NCBI public dataset. Ten coexpression modules, including 7689 protein-coding genes, were constructed by WGCNA, six of which are considered to be significantly related to the development of the hair follicle cycle. A functional enrichment analysis for each model showed that they are closely related to ECM- receptor interaction, focal adhesion, PI3K-Akt signaling pathway, estrogen signaling pathway, and so on. Combined with the analysis of differential expressed genes, 12 hub genes from coexpression modules were selected as candidate markers, i.e., COL1A1, C1QTNF6, COL1A2, AQP3, KRTAP3-1, KRTAP11-1, FA2H, NDUFS5, DERL2, MRPL14, ANTKMT and XAB2, which might be applied to improve cashmere production.

Abstract:Hair follicle stem cells (HFSCs) have been shown to be essential in the development and regeneration of hair follicles (HFs). The Inner Mongolia Cashmere goat (<em>Capra hircus</em>) has two types of HFs, primary and secondary, with cashmere being produced from the secondary hair follicle. To identify the genes associated with cashmere growth, transcriptome profiling of anagen and telogen secondary HFSCs was performed by RNA-Seq. The RNA-Seq analysis generated over 58 million clean reads from each group, with 2717 differentially expressed genes (DEGs) detected between anagen and telogen, including 1500 upregulated and 1217 downregulated DEGs. A large number of DEGs were predominantly associated with cell part, cellular process, binding, biological regulation and organelle. In addition, the PI3K-Akt, MAPK, Ras and Rap1 signaling pathways may be involved in the growth of HFSCs cultured <em>in vitro</em>. The RNA-Seq results showed that the well-defined HFSC signature genes and cell cycle-associated genes showed no significant differences between anagen and telogen HFSCs, indicating a relatively quiescent cellular state of the HFSCs cultured <em>in vitro</em>. These results are useful for future studies of complex molecular mechanisms of hair follicle cycling in cashmere goats.

Abstract:Abstract Background: Inner Mongolian cashmere goats have hair of excellent quality and high economic value, and the skin hair follicle traits of cashmere goats have a direct and important effect on cashmere yield and quality.Circular RNA has been studied in a variety of tissues and cells. Result: In this study, high-throughput sequencing was used to obtain the expression profiles of circRNA in the hair follicles of Inner Mongolia cashmere goats at different embryonic stages (45, 55 , 65 and 75 days). A total of 21784 circRNAs were identified. At the same time, the differentially expression circRNA in the six control groups formed in the four stages were: d75vsd45, circRNA up-regulated by 59 and down-regulated by 33; d75vsd55, circRNA up-regulated by 61 and down-regulated by 102; d75vsd65, circRNA up-regulated by 32 and down-regulated by 33; d65vsd55, circRNA up-regulated by 67 and down-regulated by 169; d65vsd45, circRNA up-regulated by 96 and down-regulated by 63; and d55vsd45, circRNA up-regulated by 76 and down-regulated by 42. Six DE circRNA were randomly selected to verify the reliability of the sequencing results by qRT-PCR. Subsequently,the circRNA corresponding host genes were analyzed by GO and the KEGG pathaway. The results showed that the biological processes related to hair follicle growth and development enriched by GO mainly included hair follicle morphogenesis, hair follicle maturation and cell development, and the signal pathways related to hair follicle development included the Notch signaling pathway and NF-kappa B signaling pathway.We combined the DE circRNA of d75vsd45 with miRNA and mRNA databases(unpublished) to construct the co-expression network of circRNA-miRNA-mRNA, and formed a total of 102 pairs of circRNA-miRNA and 126 pairs of miRNA-mRNA interactions. The binding relationship of circRNA3236-chi-miR-27b-3p and circRNA3236-chi-miR-16b-3p was further verified by dual-luciferase reporter assays,and the results showed that circRNA3236 and chi-miR-27b-3p,circRNA3236 and chi-miR-16b-3p have a targeted binding relationship. Conclusion: To summarize, we established the expression profile of circRNA in the fetal skin hair follicles of cashmere goats, and found that the host gene of circRNA may be involved in the development of hair follicles of cashmere goats. The co-expression network of circRNA-miRNA-mRNA that DE circRNA was constructed and preliminary verification was carried out.
Differential Gene Expression Analysis Between Anagen and Telogen of Capra Hircus Skin Based on the De Novo Assembled Transcriptome Sequence.

Abstract:Capra hircus, an economically important livestock, plays an indispensable role in the world animal fiber industry. In the present study, using Illumina/Solexa high throughput sequencing technology, we sequenced and de novo assembled the goat skin transcriptome corresponding to the anagen and telogen of the hair growth cycle. Approximately 53Mb of transcriptome sequences consisting of 57,040 high quality contigs was obtained. More than 8300 contigs were predicted to contain a full length coding sequence. Approximately 43% of the total contigs were identified as harboring homologs of sequences from other organisms in the public database. Based on the assembled transcript-derived contigs, we identified about 7000 transcripts that were differentially expressed between the anagen and telogen libraries. These differentially expressed genes were mainly enriched in signal transduction mechanisms, extracellular structures and cytoskeleton from the KOG database and in ECM receptor interaction, focal adhesion and gap junction from the KEGG pathway database, indicating the essential roles of these genes may play in cell-to-cell and cell-to-matrix communications during the active hair growth phase. In addition, many signaling pathway associated ligands and/or receptors were also identified as up-regulated genes during the anagen phase compared with the telogen stage, suggesting that enhanced cross-talk among signaling transduction pathways may be required for anagen of the hair cycle. These differentially expressed genes, especially those that were over-represented in each of the functional clusters and biochemical pathways, provide valuable resources and opportunities for characterizing the gene functions associated with hair fiber growth as well as for breeding elite Cashmere goat species.

Abstract:BackgroundThe fleece of cashmere goats contains two distinct populations of fibers, a short and fine non-medullated insulating cashmere fiber and a long and coarse medullated guard hair. The former is produced by secondary follicles (SFs) and the later by primary follicles (PFs). Evidence suggests that the induction of PFs and SFs may require different signaling pathways. The regulation of BMP2/4 signaling by noggin and Edar signaling via Downless genes are essential for the induction of SFs and PFs, respectively. However, these differently expressed genes of the signaling pathway cannot directly distinguish between the PFs and SFs.ResultsIn this study, we selected RNA samples from 11 PFs and 7 SFs that included 145,525 exons. The pathway analysis of 4512 differentially expressed exons revealed that the most statistically significant metabolic pathway was related to the ubiquitin-mediated proteolysis pathway (UMPP) (P<3.32x 10(-7)). In addition, the 51 exons of the UMPP that were differentially expressed between the different types of hair follicle (HFs) were compared by cluster analysis. This resulted in the PFs and SFs being divided into two classes. The expression level of two selected exons was analyzed by qRT-PCR, and the results indicated that the expression patterns were consistent with the deep sequencing results obtained by RNASeq.ConclusionsBased on the comparative transcriptome analysis of 18 HFs from cashmere goats, a large number of differentially expressed exons were identified using a high-throughput sequencing approach. This study suggests that UMPP activation is a prominent signaling pathway for distinguishing the PFs and SFs of cashmere goats. It is also a meaningful contribution to the theoretical basis of the biological study of the HFs of cashmere goats and other mammals.
Transcriptome Survey Reveals Hysteresis in Secondary Hair Follicle for Gene Regulation in Cashmere Goat

Abstract:Hair follicle is capable of self-renew and its growth cycle is a dynamic process.In order to investigate the vary of differential expression genes in the Nei Mongol cashmere goat hair folli-cles during anagen,the mRNA expression levels of different genes in skin hair follicles were de-tected from June to October using Illumina technology.Evaluation of transcriptomes was under-taken to understand the time-course gene expression involved in both follicle types.The samples were collected from June to October.Most of differentially expressed genes appeared between July to August in primary hair follicle (PF)and between August to September in secondary hair folli-cle (SF).Seven differentially expressed genes,FAM83C,LOC102185501,FYCO1,CTTNBP2NL, C6orf132,KRTAP29-1 and CRYBG3 were identified.Their expression demonstrated similar trends at different months of cashmere growth cycle and the SF postpone for about one month. These findings indicated the hysteresis in SF gene regulation comparing with PF,and could pro-vide the useful theoretical basis for the molecular mechanism of cashmere regeneration.

Abstract:Cashmere is initiated and develops in the fetal stages and the number and density of secondary hair follicles (SHFs) determine cashmere production and quality. Growing evidence indicates that both microRNA (miRNA) and long non-coding RNA (lncRNA) play an indispensable role in hair follicle (HF) growth and development. However, little is known about miRNAs, lncRNAs, and their functions as well as their interactions during cashmere initiation and development. Here, based on lncRNA and miRNA high-throughput sequencing and bioinformatics analysis, we identified 10,485 lncRNAs, 40,639 mRNAs, and 605 miRNAs in cashmere goat skin during HF induction, organogenesis, and cytodifferentiation stages. Among them, 521 lncRNAs, 5976 genes, and 204 miRNAs were differentially expressed (DE). KEGG analysis of DE genes indicated that ECM–receptor interaction and biosynthesis of amino acids were crucial for HF development. Notch, TGF-beta, and Wnt signaling pathways were also identified, which are conventional pathways associated with HF growth and development. Then, the ceRNA regulatory network was constructed, and the impact of lncRNA H19 was investigated in dermal papilla (DP) cells. The MTT, CCK-8, and EdU assays showed that the viability and proliferation of DP cells were promoted by H19, and mechanistic studies suggested that H19 performed its function through the chi-miR-214-3p/β-catenin axis. The present study created a resource for lncRNA, miRNA, and mRNA studies in cashmere morphogenesis. It could contribute to a better understanding of the molecular mechanism of ncRNAs involved in the regulation of HF growth and development.

Abstract:It is widely accepted that the periodic cycle of hair follicles is controlled by the biological clock, but the molecular regulatory mechanisms of the hair follicle cycle have not been thoroughly studied. The secondary hair follicle of the cashmere goat is characterized by seasonal periodic changes throughout life. In the hair follicle cycle, the initiation of hair follicles is of great significance for hair follicle regeneration. To provide a reference for hair follicle research, our study compared differences in mRNA expression and microRNA expression during the growth and repose stages of cashmere goat skin samples. Through microRNA and mRNA association analysis, we found microRNAs and target genes that play major regulatory roles in hair follicle initiation. We further constructed an mRNA-microRNA interaction network and found that hair follicle initiation and development were related to MiR-195 and the genes CHP1 , SMAD2 , FZD6 and SIAH1 .
Discovery of differentially expressed genes in cashmere goat (Capra hircus) hair follicles by RNA sequencing

Abstract:The mammalian hair follicle (HF) is a unique, highly regenerative organ with a distinct developmental cycle. Cashmere goat (Capra hircus) HFs can be divided into two categories based on structure and development time: primary and secondary follicles. To identify differentially expressed genes (DEGs) in the primary and secondary HFs of cashmere goats, the RNA sequencing of six individuals from Arbas, Inner Mongolia, was performed. A total of 617 DEGs were identified; 297 were upregulated while 320 were downregulated. Gene ontology analysis revealed that the main functions of the upregulated genes were electron transport, respiratory electron transport, mitochondrial electron transport, and gene expression. The downregulated genes were mainly involved in cell autophagy, protein complexes, neutrophil aggregation, and bacterial fungal defense reactions. According to the Kyoto Encyclopedia of Genes and Genomes database, these genes are mainly involved in the metabolism of cysteine and methionine, RNA polymerization, and the MAPK signaling pathway, and were enriched in primary follicles. A microRNA-target network revealed that secondary follicles are involved in several important biological processes, such as the synthesis of keratin-associated proteins and enzymes involved in amino acid biosynthesis. In summary, these findings will increase our understanding of the complex molecular mechanisms of HF development and cycling, and provide a basis for the further study of the genes and functions of HF development.

Abstract:Abstract Cashmere goats, as an important part of animal husbandry production, make outstanding contributions to animal fiber industry. In recent years, a great deal of research has been done on the molecular regulation mechanism of hair follicle cycle growth. However, there are few reports on the molecular regulation mechanisms of secondary hair follicle growth cycle in cashmere goats. In this study, we used transcriptome sequencing technique to sequence the skin of Inner Mongolia cashmere goats in different periods, Analyze the variation and difference of genes in the whole hair follicle cycle. And then, we verified the regulation mechanism of cashmere goat secondary hair follicle growth cycle by fluorescence quantitative PCR. As the result shows: The results of tissue section showed that the growth cycle of cashmere hair could be divided into three distinct periods: growth period (March-September), regression period (September-December) and resting period (December-March). The results of differential gene analysis showed that March was considered the beginning of the cycle, and the difference of gene expression was the most significant. Cluster analysis of gene expression in the whole growth cycle further supported the key nodes of the three periods of villus growth, and the differential gene expression of keratin corresponding to the villus growth cycle further supported the results of tissue slices. Quantitative fluorescence analysis showed that KAP3.1, KRTAP 8-1 and KRTAP 24-1 genes had close positive correlation with the growth cycle of cashmere, and their regulation was consistent with the growth cycle of cashmere. However, there was a sequence of expression time, indicating that the results of cycle regulation made the growth of cashmere change.

Abstract:Cashmere goat hair follicles are divided into primary hair follicles and secondary hair follicles. The primary hair follicles produce coarse hair, and the secondary hair follicles produce cashmere. The development of hair follicles is affected by a variety of signaling molecules and pathways. Studies have shown that non-coding RNAs are widely involved in the development of hair follicles of the goat, including small RNAs (miRNAs), long non-coding RNAs (lncRNA), and circular RNAs (circRNAs). In recent years, circRNAs, as a new type of circular closed non-coding RNAs, have attracted great attention due to their high stability. However, its regulatory effect on cashmere goat hair follicles mainly focuses on the periodic regulation of secondary hair follicles, and there is no report on the development of cashmere goat hair follicles during the fetal period. Therefore, this study was based on the circRNA, miRNA, and mRNA expression profiles obtained by whole-transcriptional sequencing of the skin tissue of the Inner Mongolia cashmere goats in the fetal period (days 45, 55, 65, and 75) and screening out the morphological changes of hair follicles at different periods. A total of 113 circRNAs related to the development of secondary hair follicles were present. According to the principle of the ceRNA regulatory network, a ceRNA regulatory network composed of 13 circRNAs, 21 miRNAs, and 110 mRNAs related to the development of secondary hair follicles was constructed. Then, qRT-PCR and Sanger sequencing identified circRNA2034, circRNA5712, circRNA888, and circRNA9127 were circRNAs. Next, the dual-luciferase reporter gene verified the targeting relationship of circRNA5712-miR-27b-3p-Dll4. In conclusion, this study constructed a ceRNA regulatory network for the development of cashmere goat secondary hair follicles, laying a foundation for the analysis of circRNAs regulating the morphogenesis and development of cashmere goat secondary hair follicles through the ceRNA mechanism.
