Abstract:AbstractColeusin factor is a diterpenoid compound isolated from the root of a tropical plant, Coleus forskohlii. Although Coleusin factor has been reported to suppress proliferation of and induce apoptosis in several types of cancer cells, the effects of Coleusin factor on osteosarcoma and the underlying mechanism are still not fully understood. In this study, we show that Coleusin factor treatment potently inhibits the growth of osteosarcoma cells associated with G1 cell-cycle arrest. Interestingly, apoptosis and cell death are not induced. Instead, Coleusin factor causes osteosarcoma cells to exhibit typical properties of differentiated osteoblasts, including a morphologic alteration resembling osteoblasts, the expression of osteoblast differentiation markers, elevated alkaline phosphatase activity, and increased cellular mineralization. Coleusin factor treatment significantly increases the expression of bone morphogenetic protein-2 (BMP-2), a crucial osteogenic regulator, and runt-related transcription factor 2 (RUNX2), one of the key transcription factors of the BMP pathway. When BMP-2 signaling is blocked, Coleusin factor fails to inhibit cell proliferation and to induce osteoblast differentiation. Thus, upregulation of BMP-2 autocrine is critical for Coleusin factor to induce osteoblast differentiation and exert its anticancer effects on osteosarcoma. Importantly, administration of Coleusin factor inhibits the growth of osteosarcoma xenografted in nude mice without systemic or immunologic toxicity. Osteosarcoma is a highly aggressive cancer marked by the loss of normal differentiation. Coleusin factor represents a new type of BMP-2 inducer that restores differentiation in osteosarcoma cells. Abstract:Background: Embelin is an active compound identified as a novel X-linked inhibitor of apoptosis protein (XIAP) inhibitor from the Embelia ribes that exhibits various medicinal effects including anti-inflammatory and anticancer activities. However, the therapeutic effect of Embelin to human osteosarcoma is not yet determined. Objectives: In this study, we evaluated the sensitizing potential of Embelin on promoting apoptosis to cause osteosarcoma cell death and inhibiting its invasion. Methods: We uesd 3-(4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide to detect the survival rates of osteosarcoma cells, Western blot to detect the expression of proteins in U-2 OS and MG63 cells, and fluorescence microscope to observe the morphology of apoptotic cells. Results: The survival of osteosarcoma cells decreased, When Embelin was used. Obvious condensed and flared fluorescence was observed, when used high-dose Embelin. There was an increase of caspase-3, cleaved caspase-3, caspase-8, and caspase-9 in Embelin group, while PI3K, AKt, p-AKt, X-linked inhibitor of apoptosis protein, and MMP-9 were downregulated. The invasion of Embelin application was significantly lower than that of the control application. Conclusion: Embelin promoted apoptosis via XIAP and PI3K/Akt signaling pathway. XIAP inhibitor Embelin inducing apoptosis could cause osteosarcoma cell death and inhibit its invasion.

Abstract:Gap junctions, formed by connexin (Cx) family proteins, permit direct exchange of regulatory ions and small signal molecules between neighbouring cells. Gap junctional intercellular communication (GJIC) plays an important role in maintaining the homeostasis and preventing cell transformation. Most of the tumour cells feature deficient or aberrant connexin expression and GJIC level, and restoration of connexin expression and GJIC is correlated with cell growth control. Numerous researches has suggested the possibility of connexins as potential anti-tumour targets for chemoprevention and chemotherapy. We investigated the ability of Coleusin Factor (CF, also named FSK88) to regulate the Cx43 expression and GJIC level in rat osteosarcoma UMR106 cells. The results have demonstrated that CF increased the mRNA and protein expression of Cx43 in both in a dose- and timedependent manner, and concomitant with up-regulation of Cx43, CF treatment up-regulated the diminished GJIC level in UMR106 cells as assayed by dye transfer experiments. In addition, Cx43 distribution at the plasma membrane was also enhanced dramatically by CF treatment. Furthermore, we discovered that CF was potent to inhibit the growth and proliferation of UMR106 cells. These results provide the first evidence that CF can regulate connexin and GJIC, indicating that Cx43 may be a target of CF to exert its anti-tumour effects.

Abstract:Coleusin factor (CF), a kind of diterpenoids, is isolated and purified from the root of a Chinese tropical plant Coleus forskohlii by our laboratory. Our previous studies have demonstrated that CF significantly inhibits growth in some kinds of cancer cell lines. Here, we found that CF remarkably inhibited growth in human gastric cancer BGC-823 cells by decreasing cell proliferation, inducing G0/G1 cell cycle arrest and apoptosis. CF also decreased the mitochondrial membrane potential in BGC-823 cells. Immunoblotting analysis revealed that CF significantly decreased the expressions of cyclinD1, Bcl-2, and Bcl-xL, increased the expressions of cytosol cytochrome c, p53, p21, and Rb. In addition, CF significantly increased the expressions and activities of caspase-3 and -9. More importantly, CF potently inhibited the growth of BGC-823 cells xenografted in athymic nude mice with negligible body weight loss and damage towards the spleen. These results indicate that CF exerts a cytotoxic effect on BGC-823 cells by inducing cell cycle arrest and apoptosis.

Abstract:BACKGROUND:T52 is a steroidal saponin extracted from the traditional Chinese herb Rohdea fargesii (Baill.), and it is reported to possess strong anti-proliferative capabilities in human pharyngeal carcinoma cell lines. However, whether T52 has anti-osteosarcoma properties, and its potential mechanism is remains unknown.PURPOSE:To examine the outcome and underlying mechanism of T52 in osteosarcomas (OS).METHODS/STUDY DESIGNS:The physiological roles of T52 in OS cells were examined using CCK-8, colony formation (CF), EdU staining, cell cycle/apoptosis and cell migration/invasion assays. The relevant T52 targets against OS were assessed via bioinformatics prediction, and the binding sites were analyzed by molecular docking. Western blot analysis was carried out to examine the levels of factors associated with apoptosis, cell cycle, and STAT3 signaling pathway activation.RESULTS:T52 markedly diminished the proliferation, migration, and invasion of OS cells, and promoted G2/M arrest and apoptosis in a dose-dependent fashion (DDF) in vitro. Mechanistically, molecular docking predicted that T52 stably associated with STAT3 Src homology 2 (SH2) domain residues. Western blot revealed that T52 suppressed the STAT3 signaling pathway, as well as the expression of the downstream targets, such as, Bcl-2, Cyclin D1, and c-Myc. In addition, the anti-OS property of T52 were partially reversed by STAT3 reactivation, which confirmed that STAT3 signaling is critical for regulating the anti-OS property of T52.CONCLUSION:We firstly demonstrated that T52 possessed strong anti-osteosarcoma property in vitro, which was brought on by the inhibition of the STAT3 signaling pathway. Our findings provided pharmacological support for treating OS with T52.

Abstract:ALL1 fused gene from chromosome 1q (AF1Q) functions as an oncogene in several types of cancers, but it has not been observed in osteosarcoma. In this study, we revealed that AF1Q was overexpressed in multiple osteosarcoma cell lines, and its expression level increased with the severity of tumor malignancy in osteosarcoma biopsies. AF1Q was coupled with the transcription factor T cell factor 4 (TCF4) to assemble a complex to bind to the promoter of cyclooxygenase 2 (COX2) and activate its expression. The individual knockdown of AF1Q, TCF4, or COX2 in osteosarcoma cell lines significantly decreased cell proliferation and invasion in vitro. The tumor xenograft model also indicated that the individual knockdown of AF1Q, TCF4, or COX2 could inhibit tumor growth and metastasis. On the basis of these promising results, we established an in vitro AlphaScreen method to identify the compounds that disrupted the AF1Q-TCF4 interaction in a naturally derived small molecule pool. We discovered a compound called PSM0537, which showed a strong ability to inhibit the AF1Q-TCF4 interaction at a low dose of half-maximal inhibitory concentration (IC50) (210.3 ± 15.6 nM). The administration of PSM0537 in vitro and in vivo could dramatically inhibit cell proliferation, invasion, and metastasis. Collectively, our findings reveal that the AF1Q-TCF4 transcriptional complex controls the expression of COX2 and that targeting the AF1Q-TCF4 interaction with PSM0537 could inhibit tumor cell growth and metastasis. Our results provide a new path for chemotherapy of osteosarcoma.

Abstract:Abstract Coleon C (6,11,12,14,16-pentahydroxyabieta-5,8,11,13-tetraen-7-one), extracted from Coleus forskohlii Briq., was investigated for its anti-tumor activity on eight human tumor cell lines (95-D, A375, HeLa, A431, MKN45, BEL7402, LoVo and HL60) and two normal ones (293, L02) by MTT and colony-forming assay in vitro. The results indicated that A375 was the most sensitive of all the cell lines. Hoechst 33258 staining showed fragmentation and condensation of chromatin. DNA ladder assay indicated the fragments of DNA because of apoptosis. Flow cytometric analysis demonstrated hypodiploid cells existed in A375 after Coleon C treatment. In the acute toxicity studies of C57BL/6 mice, LD50 of Coleon C was 1496±150 mg/kg. In the model of Lewis lung carcinoma, the average tumor weight in groups injected with 80 mg/kg Coleon C decreased by 48.9±14.3% compared with that of the control. These results indicate that Coleon C could effectively inhibit tumor cell proliferation and growth by inducing apoptosis with low toxicity. To our knowledge, this is the first report on the anti-tumor activity of Coleon C both in vitro and in vivo.

Abstract:Genistein, a nontoxic flavonoid compound, has potent antitumor activity in various cancer cell lines. This study was designed to investigate whether combination therapy with gemcitabine and genistein enhances antitumor efficacy in osteosarcoma cell lines (MG-63 and U2OS). Our results show that significant reduction in cell viability and corresponding induction of apoptosis were observed with combination treatment in both cell lines. On the molecular level, we found that gemcitabine alone can activate nuclear factor kappa B (NF-kappa B) in osteosarcoma, suggesting the potential mechanism of acquired chemoresistance. In contrast, genistein reversed the cancer's resistance to gemcitabine through the downregulation of NF-kappa B activity and the suppression of Akt. These findings suggest that the combination of gemcitabine and genistein enhanced the antitumor efficacy by abrogating the Akt/NF-kappa B pathway. The marked ability to induce apoptosis with a combination of gemcitabine and genistein suggests that this could be a rational and novel approach for osteosarcoma preclinical and clinical trials. Abstract:Summary This study was undertaken to investigate the inhibitory effects of triterpenoid compound oleanolic acid and its synthetic derivatives on osteosarcoma cells in order to identify new therapeutic candidates for the treatment of this disease. We used the 3-(4,5-dimethyl-2 thiazolyl)-2,5-diphenyl tetrazolium bromide assay to assess the effect of oleanolic acid compounds on the proliferation of osteosarcoma cells. The effect of dextrose-oleanolic acid (the most potent oleanolic acid derivative) on apoptosis of osteosarcoma cells was evaluated using the Annexin-V method. The cell cycle of dextrose-oleanolic acid-treated cells was examined by flow cytometry, and the in vivo effects of dextrose-oleanolic acid were evaluated in a mouse osteosarcoma model. Oleanolic acid compounds had an overall inhibitory effect on the proliferation of osteosarcoma cells. Our in vitro data showed that the dextrose-oleanolic acid derivative brought about maximal inhibition of proliferation of osteosarcoma cells while inducing apoptosis. It could also inhibit the growth of osteosarcoma and decreased the rate of lung metastasis in vivo . Of the oleanolic acid derivatives, dextrose-oleanolic acid exhibited the most potent anti-osteosarcoma activity; it may represent a new frontier in the treatment of osteosarcoma.

Abstract:Objective To investigate the effect of saikosaponin-d (SS-d) on the proliferation, apoptosis, migration and invasion of the osteosarcoma Saos-2 cells, and the related mechanism.MethodsOsteosarcoma Saos-2 cells were treated with varying concentrations of SS-d.CCK-8 analysis was used to evaluate the proliferation ability of Saos-2 cells;flow cytometry was employed to investigate the apoptosis ability;Transwell Chamber assay was utilized to explore the migration and invasion ability.Next, Western blot was used to assess the effect of SS-d on activation of nuclear factor-κB (NF-κB)/mitogen-activated protein kinase (MAPK) pathways and the downstream apoptosis-related proteins.ResultsSS-d dose-dependently inhibited osteosarcoma Saos-2 cell proliferation, migration and invasion (P<0.05) while promoted apoptosis (P<0.05).Western blot revealed that SS-d could inhibit the expressions of p-IκBα and p-p65, reduce the production of p-Jnk and p-p38, contributing to the inhibition of activated NF-κB/MAPK signaling, the elevated expressions of apoptosis-related proteins Bax and cleaved caspase-3 with decreased Bcl-2/Bax ratio, and the apoptosis of osteosarcoma Saos-2 cells.ConclusionSS-d was capable of downregulating osteosarcoma Saos-2 cells proliferation, migration and invasion, upregulating osteosarcoma Saos-2 cells apoptosis.These effects might be associated with the inhibition of NF-κB/MAPK signaling, decreased Bcl-2/Bax ratio and increased cleaved caspase-3 by SS-d treatment.
