Abstract:Bladder cancer (BC), a urologic disease, commonly occurs globally and is very invasive. Patients with invasive BC have low 5-year survival rate. Hence, the mechanisms underlying BC development and progression should be elucidated. MicroRNAs (miRNAs), as common noncoding RNAs, are receiving increasing attention because of their biological functions. The irregular expression patterns of miRNAs are linked to BC occurrence; therefore, determining the functions of miRNAs in abnormally expressed BC tissues might enable to elucidate the pathogenetic mechanism of BC and offer new markers for the prognosis, diagnosis, and therapy of BC. Here, we consolidate the primary roles of miRNAs with atypical expression in BC development as well as their association with BC pathological grades and chemotherapy resistance development in patients with BC.

Abstract:Abstract Background Metastasis is the leading cause of death in patients with bladder cancer (BC). However, current available treatments exert little effects on metastatic BC. Moreover, traditional grading and staging have only a limited ability to identify metastatic BC. Accumulating evidence indicates that the aberrant expression of microRNA is intimately associated with tumor progression. So far, many miRNAs have been identified as molecular targets for cancer diagnosis and therapy. This study focused on the role of miR‐516a‐5p (miR‐516a) in BC. Methods MiR‐516a expression and its downstream signaling pathway were detected using molecular cell biology and biochemistry approaches and techniques. Fresh clinical BC tissue was used to study the clinicopathological characteristics of patients with different miR‐516a expression. The biological functions of miR‐516a in BC were tested both in vivo and in vitro. Results A more invasive BC phenotype was significantly and positively correlated with miR‐516a overexpression in BC patients. MiR‐516a inhibition significantly decreased BC cell invasion and migration in vitro and in vivo. Furthermore, miR‐516a attenuated the expression of PH domain leucine‐rich repeat‐containing protein phosphatase 2 protein and inhibited SMAD‐specific E3 ubiquitin protein ligase 1 transcription by activating the AKT/Forkhead box O3 signaling pathway, which stabilized MMP9 and slowed down its proteasomal degradation, ultimately promoting BC motility and invasiveness. Conclusions Our findings reveal the crucial function of miR‐516a in promoting BC metastasis, and elucidate the molecular mechanism involved, suggesting that miR‐516a may be a promising novel diagnostic and therapeutic target for BC.
Oncogenic Role of MIR516A in Human Bladder Cancer Was Mediated by Its Attenuating PHLPP2 Expression and BECN1-dependent Autophagy.

Abstract:Although MIR516A has been reported to be downregulated and act as a tumor suppressor in multiple cancers, its expression and potential contribution to human bladder cancer (BC) remain unexplored. Unexpectedly, we showed here that MIR516A was markedly upregulated in human BC tissues and cell lines, while inhibition of MIR516A expression attenuated BC cell monolayer growth in vitro and xenograft tumor growth in vivo, accompanied with increased expression of PHLPP2. Further studies showed that MIR516A was able to directly bind to the 3 '-untranslated region of PHLPP2 mRNA, which was essential for its attenuating PHLPP2 expression. The knockdown of PHLPP2 expression in MIR516A-inhibited cells could reverse BC cell growth, suggesting that PHLPP2 is a MIR516A downstream mediator responsible for MIR516A oncogenic effect. PHLPP2 was able to mediate BECN1/Beclin1 stabilization indirectly, therefore promoting BECN1-dependent macroautophagy/autophagy, and inhibiting BC tumor cell growth. In addition, our results indicated that the increased autophagy by attenuating MIR516A resulted in a dramatic inhibition of xenograft tumor formation in vivo. Collectively, our results reveal that MIR516A has a novel oncogenic function in BC growth by directing binding to PHLPP2 3 '-UTR and inhibiting PHLPP2 expression, in turn at least partly promoting CUL4A-mediated BECN1 protein degradation, thereby attenuating autophagy and promoting BC growth, which is a distinct function of MIR516A identified in other cancers.
Overexpression of Mir-340 Inhibits Cell Proliferation and Induces Apoptosis of Human Bladder Cancer Via Targeting Glut-1

Abstract:BACKGROUND:Bladder cancer (BC) has high mortality due to distant metastasis. Previous works suggested that microRNA (miRNA)-340 is a critical regulator for the development and progression of various cancers. The specific biological function of miR-340 in BC is little known.METHODS:In the present study, RT-qPCR was performed to measure the expression of miR-340 in paired BC tissues and adjacent non-tumor tissues. Next, the target gene of miR-340 was identified using dual-luciferase reporter assay and its level was also tested in tissues. Moreover, cell proliferation and apoptosis were analyzed by CCK-8 and flow cytometry. Finally, the expression of PCNA, Bax was detected by RT-qPCR and western blotting, as well as PI3K/AKT signaling measured by western blotting.RESULT:The results demonstrated that miR-340 expression was downregulated and its target Glut-1 level was upregulated in BC tissues. Functionally, overexpression of miR-340 suppressed the proliferation and induced apoptosis in BC cells, while Glut-1 reversed the suppression of proliferation or induction of apoptosis induced by miR-340. Additionally, miR-340 repressed PCNA, p-PI3K and p-AKT levels but enhanced Bax level, while Glut-1 rescued the effects.CONCLUSION:In conclusion, miR-340 functions as a tumor suppressor of BC, which inhibited proliferation and induced apoptosis by targeting Glut-1 partly through regulating PCNA, Bax expression and PI3K/AKT pathway. This study suggested that miR-340 is a potential target for the treatment of BC.

Abstract:The urothelial carcinoma is the most common pathological type of bladder tumor. Creation of lists of miRNAs differentially expressed between this tumor type and normal tissue might help identify new diagnostic and prognostic markers. We therefore performed the present miRNA microarray analysis with 25 cases of bladder urothelial carcinomas and adjacent normal bladder tissue. The results showed a panel of 51 differentially expressed miRNAs with at least 2-fold differences in expression compared with the normal controls, including 20 up-regulated and 31 down-regulated examples. The expression levels of ten of the top dysregulated miRNAs, mir-1, mir-145, mir-143, mir-100, mir-200b, mir-708, mir-133a, mir-133b, mir-125b and mir-99 were experimentally verified using real-time RT-PCR analysis. These findings suggest that these miRNAs may be involved in bladder urothelial carcinoma pathogenesis and have potential as biomarkers.
Genetic Alteration and Prospective Signaling Pathways of Mir-517 A-3 P in Bladder Cancer : a Study Based on Mirna Sequencing Data and Bioinformatics Methods

Abstract:Purpose: To explore the clinicopathological value and prospective function of microRNA-517a-3p (miR517a-3p) in bladder cancer (BC). Methods: The clinical importance of miR-517a-3p expression and genetic alteration in BC was unraveled by using The Cancer Genome Atlas (TCGA), cBioPortal, Gene Expression Omnibus (GEO) and ArrayExpress. The potential target genes of miR-517a-3p were obtained via the combination of predicted genes and down-regulated genes post miR-517a-3p transfection in vitro from microarray (GSE39093). The probable signaling pathways were further evaluated with multiple bioinformatics approaches. Results: MiR-517a-3p expression was significantly higher in the samples of pathologic N stage (N1-N3), pathologic stage (III-IV) and patients with lymphovascular invasion than that of their counterparts (P<0.05) based on data from TCGA. The amplification was the only alteration type which counted for 3% (14/412) in BC as indicated by cBioPortal. A total of 858 genes were gained by prediction from at least two predicting programs. Compared to control cells, there were 4844 genes downexpressed after BC cells (BOY and T24) were transfected with miR-517a-3p. Only “hsa05200: Pathways in cancer” was significantly enriched via Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis (P<0.05). Three significant pathways were discovered in PANTHER pathway analysis (P<0.05). Hub genes, such as CREB1, MAPK1, RPS6KA5, SMAD3 and PPARA, were identified by protein-protein interaction. Conclusion: The results in this study indicate that miR-517a-3p may play a potential role in the occurrence and development of BC, especially its genetic alteration, via various signal pathways. However, the exact mechanism still needs to be ascertained by in vitro and in vivo experiments.

Abstract:BACKGROUND:MicroRNAs (miRNAs) are well characterized for their important roles in human cancers by influencing various aspects of malignancy. Till now, the function and mechanism of miR-204, a tumor suppressor in several cancers, remain unclear in bladder cancer (BC). Here, we intend to explore its roles in BC progression.METHODS:qRT-PCR was applied to determine miR-204 and ROBO4 expression in BC tissues and cell lines. miR-204 expression with clinicopathological features was analyzed. The impacts of miR-204 on BC cell growth and metastasis in vitro were evaluated by both loss-of-function and gain-of-function assays (CCK-8, crystal violet staining, wound healing and transwell assays). Furthermore, qRT-PCR, Western blot and luciferase reporter assays were used to validate the targeting of ROBO4 by miR-204. Finally, linear regression was performed to analyze the correlation of miR-204 and ROBO4 in BC tissues.RESULTS:Expression of miR-204 was markedly decreased in BC tissues and cell lines were compared with respective controls. Low miR-204 expression was associated with positive advanced T stage and lymph node metastasis. Cellular function studies revealed that miR-204 inhibited BC cell growth, migration and invasion. Mechanistic exploration found that miR-204 directly targeted ROBO4. Rescue assays indicated that ROBO4 restoration could reverse the antitumor effects of miR-204 in BC. Finally, ROBO4 was significantly correlated with miR-204 levels inversely.CONCLUSION:miR-204 might serve as a tumor suppressor in BC by targeting ROBO4.

Abstract:BACKGROUND:Bladder cancer, the ninth-most-common malignancy worldwide with an estimated 356,000 new cases and 145,000 deaths annually, has a propensity to relapse, requiring lifelong monitoring after diagnosis. 75% patients diagnosed with BC are non-muscle invasive BC and over 50% of them experience recurrences within 6-12 years of initial diagnosis. miRNA are small, noncoding RNA and shown to be oncogenes or anti-oncogenes in bladder cancer, contributing to numerous BC cell processes, including cell proliferation, differentiation, migration and apoptosis.METHODS:RT-qPCR were performed to detect the expression of miR-187-5p in tissues and cell lines, After which, clinicopathological variables and the prognostic value of altered miR-187-5p expression in BC was analyzed with the 48 formalin-fixed paraffin-embedded BC samples. Moreover, Cell functional assays (wound healing assay, CCK-8 assay, transwell assay and flow cytometry assay) were performed to explore the relationship between miR-187-5p expression and cell proliferation, migration, invasion and apoptosis in BC.RESULTS:Up-regulation of miR-187-5p was observed in BC tissues and BC cell lines. Cox proportional hazard regression analysis demonstrated that the patients with low expression of miR-187-5p experience lower risks of recurrence in the univariate and multivariate analysis. The Kaplan-Meier recurrence-free curves suggested that the patients with low expression of miR-187-5p experience lower risks of recurrence. Up-regulation of miR-187-5p promotes cell proliferation and mobility and inhibits the apoptosis of 5637 and UM-UC-3 cell, while down-regulation of miR-187-5p reverses these effects.CONCLUSIONS:The results of our study demonstrated that oncogene miR-187-5p is associated with cellular proliferation, migration, invasion, apoptosis and an increased risk of recurrence in bladder cancer.

Abstract:Objective: To determine miR-1 expression and its potential molecular mechanism in bladder cancer. Methods: We analyzed the correlation between miR-1 expression and different clinicopathological features based on The Cancer Genome Atlas (TCGA). Gene Expression Omnibus (GEO) and ArrayExpress microarray datasets, as well as data from TCGA and the literature, were used to explore miR-1 expression in bladder cancer. A fixed-effects model and a random-effects model were used. We detected the diagnostic value of miR-1 in bladder cancer. Differentially expressed genes in TCGA, genes from predicting tools and upregulated genes in GSE24782 were then cross-referenced. The target genes from the literature were added, and the final genes were considered the promising target genes of miR-1 in bladder cancer. Subsequently, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses were carried out. Results: The combined standardized mean difference was -1.29 (95% CI: -1.57, -2.00) and -1.83 (95% CI: -3.08, -0.58) based on the fixed-and random-effects models, respectively. The area under the curve was 0.9660 (Q* = 0.9135) in the summarized receiver operating characteristic curve. The most significantly related GO annotation items were blood vessel development in biological processes, extracellular region in cellular components, and G-protein-activated inward-rectifier potassium channel activity in molecular functions. KEGG pathway analysis demonstrated that the promising target genes of miR-1 in bladder cancer were related to maturity onset diabetes of the young. Conclusion: The miR-1 was downregulated in bladder cancer. It might play an important role via its target genes in bladder cancer.
Mir-758-3p Suppresses Human Bladder Cancer Cell Proliferation, Migration and Invasion by Targeting NOTCH2.

Abstract:MicroRNAs (miRs) are widely involved in regulating tumor development and progression. miR-758-3p has been reported to suppress the progression of various cancer types, including hepatocellular carcinoma. However, whether miR-758-3p has a role in bladder cancer (BC) has not been previously reported, and was thus investigated in the present study. It was revealed that miR-758-3p was downregulated in BC tissues and cell lines. Transfection with miR-758-3p mimics suppressed the proliferation, migration and invasion of BC cells, and inhibition of miR-758-3p had the opposite effect. In terms of the underlying mechanisms, a luciferase reporter assay revealed that Notch receptor 2 (NOTCH2) is a direct target gene of miR-758-3p in BC cells. Transfection with miR-758-3p mimics decreased the mRNA and protein levels of NOTCH2. Furthermore, an inverse correlation between miR-758-3p and NOTCH2 levels was identified. Finally, overexpression of NOTCH2 significantly rescued the proliferation, migration and invasion of BC cells transfected with miR-758-3p mimics. Taken together, the present study indicated that miR-758-3p suppresses BC cell proliferation, migration and invasion by targeting NOTCH2.
MicroRNA-31 Functions As a Tumor Suppressor and Increases Sensitivity to Mitomycin-C in Urothelial Bladder Cancer by Targeting Integrin Α5

Abstract:Urothelial bladder cancer (UBC) is a common genitourinary malignancy. MiR-31, a well-identified miRNA, exhibits diverse properties in different cancers. However, the specific functions and mechanisms of miR-31 in UBC have not been investigated. In this study, tumor samples, especially invasive UBC, showed significantly reduced level of miR-31, as compared with normal urothelium. Prognostic analysis using the EORTC model showed that down-regulation of miR-31 correlated with higher risks of recurrence and progression in noninvasive UBC cases. Remarkably, overexpression of miR-31 mimics in UBC cell lines inhibited cell proliferation, migration and invasion. Integrin α5 (ITGA5), an integrin family member, was subsequently identified as a direct target of miR-31 in UBC cells. When treated with mitomycin-C (MMC), miR-31-expressing UBC cells displayed lower survival and higher apoptotic rates, and deactivated Akt and ERK. These effects arising from miR-31 overexpression were abrogated by ITGA5 restoration. Furthermore, miR-31 markedly inhibited tumor growth and increased the effectiveness of MMC in UBC xenografts. In summary, our data suggest that miR-31 is a prognostic predictor and can serve as a potential therapeutic target of UBC.

Abstract:We focused on studying the effects of a key miRNA-mRNA axis in bladder urothelial carcinoma (BUC). Firstly, miRNAs and mRNAs differentially expressed in BUC were analyzed. Clinical information in the TCGA database was used for survival analysis, and the regulator of miRNA-93-5p was predicted. miRNA-93-5p and KLF9 mRNA expression were detected by qRT-PCR. Protein level detection and targeting measurement were, respectively, achieved by western blot and dual-luciferase approaches. The proliferative, invasive, and migratory abilities were tested through CCK-8, Transwell, and wound healing methods. Cell apoptosis in each group was detected through flow cytometry. As discovered, miRNA-93-5p level was markedly high in BUC cells while KLF9 expression was remarkably low. miRNA-93-5p overexpression promoted BUC cell abilities. Besides, miRNA-93-5p inhibited KLF9 expression. Furthermore, KLF9 overexpression dramatically attenuated such promotion on cancer cell abilities. On the whole, miRNA-93-5p/KLF9 axis facilitated BUC progression, offering a new potential target for BUC patients.
Expression of Mir-149-3p Inhibits Proliferation, Migration, and Invasion of Bladder Cancer by Targeting S100A4.

Abstract:MicroRNAs play key roles during various crucial cell processes, such as proliferation, migration, and invasion. In addition, microRNAs have been shown to possess oncogenic and tumor suppressive functions in human cancers. Increasing evidence has clarified that miR-149-3p, a novel cancer-related microRNA, plays an important role in suppression of proliferation, migration, and invasion; however, the effect and mechanisms underlying the miR-149-3p effect in bladder cancer (BCa) remain unclear. In the current study we found that the increased expression of miR-149-3p significantly suppressed cell proliferation, migration, and invasion ability in BCa. The suppressive effect was related to S100A4. A further investigation showed that miR-149-3p negatively regulated S100A4, as verified by the luciferase reporter assay. Furthermore, our study showed that S100A4 mediated the anti-metastatic effects of miR-149-3p on proliferation, migration, and invasion of BCa cells. Analysis of a xenograft mouse model showed that miR-149-3p expression significantly decreased tumor growth by targeting S100A4. Taken together, these data indicate that S100A4 promotes cell growth, migration, and invasion and can by reversed by miR-149-3p in BCa.
Effect of Mir-29B-1* and Mir-29C Knockdown on Cell Growth of the Bladder Cancer Cell Line T24

Abstract:Objective To investigate the role of the microRNAs miR-29b-1-5p (miR-29b-1*) and miR-29c in bladder urothelial cancer (BUC). Methods Levels of miR-29b-1* and miR-29c in normal urothelial cells (HU609) and BUC cells (T24) were determined via quantitative real-time reverse transcription–polymerase chain reaction. T24 cells were transfected with small interfering RNA targeting miR-29b-1* or miR-29c, and cell growth was assessed using 3-(4,5-dimehylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. The predicted targets and oncogenic pathways of these microRNAs were determined using bioinformatics analysis. Results MiR29b-1* and miR-29c levels were higher in T24 cells than normal urothelial cells. Knockdown of miR-29b-1* or miR-29c suppressed T24 cell growth. Bioinformatic analysis showed that miR-29b-1* and miR-29c co-regulated a subset of putative target genes, about 10% of which have been experimentally validated. Conclusion Both miR-29b-1* and miR-29c regulate cell growth in BUC. The targets of miR-29b-1* and miR-29c may be functionally associated with proliferation, cell cycle and apoptosis.
Identification and Bioinformatics Analysis of Mirnas Associated with Human Muscle Invasive Bladder Cancer.

Abstract:Accumulated evidence has indicated that micro (mi)RNAs play vital roles in the occurrence and development of human muscle invasive bladder cancer (MIBC), however, little is known about the miRNAs' regulatory networks. In the present study, the authors aimed to use bioinformatics analysis to identify the key miRNAs and potential target genes, as well as studying the underlying mechanisms for MIBC. They collected several human MIBC tissues to generate a miRNA expression analysis by microarray analysis comparing with normal bladder tissues, identifying 104 differentially expressed miRNAs (102 were downregulated and 2 were upregulated) and predicted 11,884 putative target genes of the dysregulated miRNAs. To understand the function of dysregulated miRNAs in the development of MIBC, networks among miRNAs and genes, gene ontologies and pathways were built. The subsequent bioinformatics analysis indicated that the mitogen-associated protein kinase (MAPK) signaling pathway, apoptosis and pathways in cancer and the cell cycle, were significantly enriched Overall, these results provided comprehensive information on the biological function of dysregulated miRNAs in the development of MIBC. The identification of miRNAs and their putative targets may offer new diagnostic and therapeutic strategies for human muscle invasive bladder cancer.
MicroRNA-30c-5p Arrests Bladder Cancer G2/M Phase and Suppresses Its Progression by Targeting PRC1-mediated Blocking of CDK1/Cyclin B1 Axis.

Abstract:BACKGROUND:MicroRNAs (miRNAs) play a critical role in cancer development and progression, the dis-regulation of miR-30c-5p has been observed in various malignant tumors but no research was done in bladder cancer (BCa). This study aims to investigate the downregulation of miR-30c-5p in BCa, and examine its mechanism and prognostic significance.METHODS:Bioinformatics analyses and clinical specimens were employed to investigate the relationship between miR-30c-5p and clinical information in BCa patients. The expression levels of miR-30c-5p and its target gene were assessed by real-time PCR and western blot. Cell viability was evaluated through clonogenic capacity, CCK-8, and EdU assays. Cell cycle distribution and cell apoptosis were determined by flow cytometry. The anti-tumor effect of miR-30c-5p was also validated in animal models.RESULTS:The expression levels of miR-30c-5p were significantly decreased in both bladder tumor tissue and BCa cell lines. Low miR-30c-5p expression was found to be correlated with unfavorable TNM stages and poor prognosis. Over-expressing miR-30c-5p was observed to hinder BCa cell growth, migration, and invasion abilities and causing cell cycle arrest. Mechanistically, miR-30c-5p directly binds and suppresses PRC1, thereby blocking the CDK1/Cyclin B1 axis in BCa, thus impairing BCa cell viability and inducing cell cycle arrest at G2/M phase.CONCLUSION:Down-regulated miR-30c-5p promotes BCa through its target gene PRC1, miR-30c-5p is a favorable biomarker for predicting clinical outcomes in BCa patients and has the potential to be a therapeutic target.
