Abstract:Alzheimer's disease (AD) is the most common neurodegenerative disorder accompanied with memory impairment followed by global cognitive deficits. Accumulation of beta–amyloid peptide 42 (Aβ42) in the brain is widely believed to be central etiology of AD. However, the precise molecular mechanisms underlying neuronal injury and death caused by Aβ42 remain to be elucidated. In vitro, Aβ42 can form a wide array of structures, including multiple monomer conformers, various size of oligomers, Aβ–derived diffusible ligands (ADDLs), protofibrils, fibrils and spheroids. Recent research efforts have been focused on understating the relationship between these different misfolding structures and neurotoxicity and the relevance to AD pathogenesis. In previous work, we have reported that the transgenic flies expressing Aβ42 fused to secretion signal in neurons showed progressive amyloid deposits formation, learning defects and neurodegeneration. Our recent study using immuno–electron microscopy further confirmed that most of Aβ42 accumulation occurred intraneuronally. These results indicate our Aβ42 fly is a useful model system to study intraneuronal toxicity of Aβ42 in vivo. To gain insights into structure–toxicity relationship of Aβ42 in vivo, we established new transgenic fly lines expressing Aβ42 with mutations that either accelerate or inhibit aggregation of Aβ42 in vitro including familial Arctic mutation. We found that Aβ42 with Arctic mutation accumulated in neuronal soma and caused cell body degeneration. In contrast, the mutant Aβ42 that is known to reduce Aβ42 aggregation in vitro accumulated in dendrites and induced neurite degenetarion. Interestingly, wild–type Aβ42 accumulated in both neuronal soma and dendrites, indicating that each mutant Aβ42 may represent one aspect of wild type Aβ42. In addition to different neuronal injury, such as cell death and neurite degeneration, expression of mutant Aβ42 caused different magnitude of locomotor impairment and premature death. We are currently analyzing their effects on learning and memory. These results suggest that each misfolding status of Aβ42 is associated with a specific type of neuronal injury, and raise the possibility that pathologies induced by Aβ42 include multiple mechanisms caused by different misfolding status.
