Increased Growth and Fatty Acid Accumulation Via Overexpression of Hypothetical Protein Unigene 12362 with the SWIB/MDM2 Domain in Schizochytrium

Abstract:Identifying genetic targets that enhance both growth and fatty acid accumulation is crucial to address the low productivity challenge in utilizing Schizochytrium for fatty acid production. In this study, hypothetical protein Unigene12362 containing the SWIB/MDM2 domain, which was upregulated during the late fermentation stage, was discovered and overexpressed in Schizochytrium ZW0, leading to notable increases of growth and fatty acid accumulation, particularly boosting C14 components of fatty acids in the fermentation medium. We found that this protein could activate genes involved in acetyl-CoA and NADPH biosynthesis and promote the expression of most genes in fatty acid synthesis-related pathways. Further functional analysis revealed that it might be glucose-responsive protein and appears to respond to stress, especially under high-glucose or low-phosphorous conditions. In a 5-L fermenter under low-phosphorous conditions, the engineered strain carrying Unigene12362 exhibited a 1.11-fold increase in growth, accompanied by enhancements of 1.27-fold in total fatty acids, 1.14-fold in saturated fatty acid, and 1.43-fold in polyunsaturated fatty acids. These findings demonstrate that this hypothetical protein holds important potential for industrial applications by enhancing both growth and fatty acid synthesis.
